    La idea central del proyecto DRAMA es la informatización de las diferentes etapas del proceso de creación teatral. Su objetivo final es desarrollar una herramienta multimedia que permita trabajar en distintas aplicaciones, desde al análisis del texto hasta la memorización del proceso de creación, pasando por todas las etapas de la puesta en escena. Este instrumento vendría a reemplazar al tradicional cuaderno de dirección, convirtiéndose en la herramienta de trabajo del director de escena en el siglo XXI

    En un país -Francia- donde el campo del teatro está estructurado culturalmente durante décadas, el Teatro Aplicado es una noción que a menudo aparece como ancillar, frente a un arte institucionali-zado, incluso mirificado. Por un lado, estaría el Teatro, puro, noble, auténtico y por otro, estarían sus avatares: el teatro de empresa, el teatro para el desarrollo personal, el teatro para patologías, etc. Si tienen la misma fuente, su consanguinidad no deja de asustar. ¿cómo pueden unos artistas que crean alejados de cualquier coacción exterior pertenecer a la misma familia del teatro que unos actores o directores que "obedecen" a un encargo, en un contexto específico, con un público muchas veces participantes de talleres ... y que son por tanto prisioneros, en cierto modo, de un arte instru-mental izado? A este problema ético, este libro intenta responder, a través de ejemplos concretos, para una mayor comprensión inrerculrural Francia/ Colombia.In a country -France- where the field of theater has been culturally structured for decades, Applied Theater is a notion that often appears as an ancillary, in the face of an institutionalized, even mirified, art. On the one hand, there would be the Theater, pure, noble, authentic and on the other, there would be its ups and downs: company theater, theater for personal development, theater for pathologies, etc. If they have the same source, their consanguinity does not stop frightening. How can some artists who create far from any external coercion belong to the same theater family as some actors or directors who "obey" a commission, in a specific context, with an audience that is often workshop participants... are they therefore prisoners, in a certain way, of an instrumented art?     Co-variations of cerebral blood volume and single neurons discharge during resting state and visual cognitive tasks in non-human primates

    Summary: To better understand how the brain allows primates to perform various sets of tasks, the ability to simultaneously record neural activity at multiple spatiotemporal scales is challenging but necessary. However, the contribution of single-unit activities (SUAs) to neurovascular activity remains to be fully understood. Here, we combine functional ultrasound imaging of cerebral blood volume (CBV) and SUA recordings in visual and fronto-medial cortices of behaving macaques. We show that SUA provides a significant estimate of the neurovascular response below the typical fMRI spatial resolution of 2mm3. Furthermore, our results also show that SUAs and CBV activities are statistically uncorrelated during the resting state but correlate during tasks. These results have important implications for interpreting functional imaging findings while one constructs inferences of SUA during resting state or tasks

    In vivo optogenetic stimulation of the primate retina activates the visual cortex after long-term transduction

    International audienceOver the last 15 years, optogenetics has changed fundamental research in neuroscience and is now reaching toward therapeutic applications. Vision restoration strategies using optogenetics are now at the forefront of these new clinical opportunities. But applications to human patients suffering from retinal diseases leading to blindness raise important concerns on the long-term functional expression of optogenes and the efficient signal transmission to higher visual centers. Here, we demonstrate in non-human primates continued expression and functionality at the retina level $20 months after delivery of our construct. We also performed in vivo recordings of visually evoked potentials in the primary visual cortex of anesthetized animals. Using synaptic blockers, we isolated the in vivo cortical activation resulting from the direct optogenetic stimulation of primate retina. In conclusion, our work indicates long-term transgene expression and transmission of the signal generated in the macaque retina to the visual cortex, two important features for future clinical applications

    In vivo characterization of physiological and metabolic changes related to isocitrate dehydrogenase 1 mutation expcression by multiparametric MRI and MRS in a rat model with orthotopically grafted human-derived glioblastoma cell lines

    International audienceThe physiological mechanism induced by the isocitrate dehydrogenase 1 (IDH1) mutation, associated with better treatment response in gliomas, remains unknown. The aim of this preclinical study was to characterize the IDH1 mutation through in vivo multiparametric MRI and MRS. Multiparametric MRI, including the measurement of blood flow, vascularity, oxygenation, permeability, and in vivo MRS, was performed on a 4.7 T animal MRI system in rat brains grafted with human‐derived glioblastoma U87 cell lines expressing or not the IDH1 mutation by the CRISPR/Cas9 method, and secondarily characterized with additional ex vivo HR‐MAS and histological analyses. In univariate analyses, compared with IDH1−, IDH1+ tumors exhibited higher vascular density (p < 0.01) and better perfusion (p = 0.02 for cerebral blood flow), but lower vessel permeability (p < 0.01 for time to peak (TTP), p = 0.04 for contrast enhancement) and decreased T1 map values (p = 0.02). Using linear discriminant analysis, vascular density and TTP values were found to be independent MRI parameters for characterizing the IDH1 mutation (p < 0.01). In vivo MRS and ex vivo HR‐MAS analysis showed lower metabolites of tumor aggressiveness for IDH1+ tumors (p < 0.01). Overall, the IDH1 mutation exhibited a higher vascularity on MRI, a lower permeability, and a less aggressive metabolic profile. These MRI features may prove helpful to better pinpoint the physiological mechanisms induced by this mutation

    Therapeutic antibodies – natural and pathological barriers and strategies to overcome them

    International audienceAntibody-based therapeutics have become a major class of therapeutics with over 120 recombinant antibodies approved or under review in the EU or US. This therapeutic class has experienced a remarkable expansion with an expected acceleration in 2021-2022 due to the extraordinary global response to SARS-CoV2 pandemic and the public disclosure of over a hundred anti-SARS-CoV2 antibodies. Mainly delivered intravenously, alternative delivery routes have emerged to improve antibody therapeutic index and patient comfort. A major hurdle for antibody delivery and efficacy as well as the development of alternative administration routes, is to understand the different natural and pathological barriers that antibodies face as soon as they enter the body up to the moment they bind to their target antigen. In this review, we discuss the well-known and more under-investigated extracellular and cellular barriers faced by antibodies. We also discuss some of the strategies developed in the recent years to overcome these barriers and increase antibody delivery to its site of action. A better understanding of the biological barriers that antibodies have to face will allow the optimization of antibody delivery near its target. This opens the way to the development of improved therapy with less systemic side effects and increased patients' adherence to the treatment
