    Determinants of antibody persistence across doses and continents after single-dose rVSV-ZEBOV vaccination for Ebola virus disease: an observational cohort study.

    BACKGROUND: The recombinant vesicular stomatitis virus (rVSV) vaccine expressing the Zaire Ebola virus (ZEBOV) glycoprotein is efficacious in the weeks following single-dose injection, but duration of immunity is unknown. We aimed to assess antibody persistence at 1 and 2 years in volunteers who received single-dose rVSV-ZEBOV in three previous trials. METHODS: In this observational cohort study, we prospectively followed-up participants from the African and European phase 1 rVSV-ZEBOV trials, who were vaccinated once in 2014-15 with 300 000 (low dose) or 10-50 million (high dose) plaque-forming units (pfu) of rVSV-ZEBOV vaccine to assess ZEBOV glycoprotein (IgG) antibody persistence. The primary outcome was ZEBOV glycoprotein-specific IgG geometric mean concentrations (GMCs) measured yearly by ELISA compared with 1 month (ie, 28 days) after immunisation. We report GMCs up to 2 years (Geneva, Switzerland, including neutralising antibodies up to 6 months) and 1 year (Lambaréné, Gabon; Kilifi, Kenya) after vaccination and factors associated with higher antibody persistence beyond 6 months, according to multivariable analyses. Trials and the observational study were registered at ClinicalTrials.gov (Geneva: NCT02287480 and NCT02933931; Kilifi: NCT02296983) and the Pan-African Clinical Trials Registry (Lambaréné PACTR201411000919191). FINDINGS: Of 217 vaccinees from the original studies (102 from the Geneva study, 75 from the Lambaréné study, and 40 from the Kilifi study), 197 returned and provided samples at 1 year (95 from the Geneva study, 63 from the Lambaréné, and 39 from the Kilifi study) and 90 at 2 years (all from the Geneva study). In the Geneva group, 44 (100%) of 44 participants who had been given a high dose (ie, 10-50 million pfu) of vaccine and who were seropositive at day 28 remained seropositive at 2 years, whereas 33 (89%) of 37 who had been given the low dose (ie, 300 000 pfu) remained seropositive for 2 years (p=0·042). In participants who had received a high dose, ZEBOV glycoprotein IgG GMCs decreased significantly between their peak (at 1-3 months) and month 6 after vaccination in Geneva (p0·05). Neutralising antibodies seem to be less durable, with seropositivity dropping from 64-71% at 28 days to 27-31% at 6 months in participants from the Geneva study. INTERPRETATION: Antibody responses to single-dose rVSV-ZEBOV vaccination are sustained across dose ranges and settings, a key criterion in countries where booster vaccinations would be impractical.     Evolution of Linear Absorption and Nonlinear Optical Properties in V-Shaped Ruthenium(II)-Based Chromophores

    In this article, we describe a series of complexes with electron-rich cis-{Ru^(II)(NH_3)_4}^(2+) centers coordinated to two pyridyl ligands bearing N-methyl/arylpyridinium electron-acceptor groups. These V-shaped dipolar species are new, extended members of a class of chromophores first reported by us (Coe, B. J. et al. J. Am. Chem. Soc. 2005, 127, 4845−4859). They have been isolated as their PF_6− salts and characterized by using various techniques including ^1H NMR and electronic absorption spectroscopies and cyclic voltammetry. Reversible Ru^(III/II) waves show that the new complexes are potentially redox-switchable chromophores. Single crystal X-ray structures have been obtained for four complex salts; three of these crystallize noncentrosymmetrically, but with the individual molecular dipoles aligned largely antiparallel. Very large molecular first hyperpolarizabilities β have been determined by using hyper-Rayleigh scattering (HRS) with an 800 nm laser and also via Stark (electroabsorption) spectroscopic studies on the intense, visible d → π^* metal-to-ligand charge-transfer (MLCT) and π → π^* intraligand charge-transfer (ILCT) bands. The latter measurements afford total nonresonant β_0 responses as high as ca. 600 × 10^(−30) esu. These pseudo-C_(2v) chromophores show two substantial components of the β tensor, β_(zzz) and β_(zyy), although the relative significance of these varies with the physical method applied. According to HRS, β_(zzz) dominates in all cases, whereas the Stark analyses indicate that β_(zyy) is dominant in the shorter chromophores, but β_(zzz) and β_(zyy) are similar for the extended species. In contrast, finite field calculations predict that β_(zyy) is always the major component. Time-dependent density functional theory calculations predict increasing ILCT character for the nominally MLCT transitions and accompanying blue-shifts of the visible absorptions, as the ligand π-systems are extended. Such unusual behavior has also been observed with related 1D complexes (Coe, B. J. et al. J. Am. Chem. Soc. 2004, 126, 3880−3891)

The model agreement is substantially improved if newly measured rate coefficients for the OH + NO2 and OH + HNO3 reactions are used. When included in 2-D models, the new rate coefficients significantly increase the calculated ozone loss due to NOx and modestly change the calculated ozone abundances in the lower stratosphere.     Analysing causal structures with entropy

    A central question for causal inference is to decide whether a set of correlations fit a given causal structure. In general, this decision problem is computationally infeasible and hence several approaches have emerged that look for certificates of compatibility. Here we review several such approaches based on entropy. We bring together the key aspects of these entropic techniques with unified terminology, filling several gaps and establishing new connections regarding their relation, all illustrated with examples. We consider cases where unobserved causes are classical, quantum and post-quantum and discuss what entropic analyses tell us about the difference. This has applications to quantum cryptography, where it can be crucial to eliminate the possibility of classical causes. We discuss the achievements and limitations of the entropic approach in comparison to other techniques and point out the main open problems.Comment: 19 (+3) pages, 5 (+1) figures.     Human impact parameterizations in global hydrological models improves estimates of monthly discharges and hydrological extremes: a multi-model validation study

    Human activities have a profound influence on river discharge, hydrological extremes, and water-related hazards. In this study, we compare the results of five state-of-the-art global hydrological models (GHMs) with observations to examine the role of human impact parameterizations (HIP) in the simulation of the mean, high, and low flows. The analysis is performed for 471 gauging stations across the globe and for the period 1971-2010. We find that the inclusion of HIP improves the performance of GHMs, both in managed and near-natural catchments. For near-natural catchments, the improvement in performance results from improvements in incoming discharges from upstream managed catchments. This finding is robust across GHMs, although the level of improvement and reasons for improvement vary greatly by GHM. The inclusion of HIP leads to a significant decrease in the bias of long-term mean monthly discharge in 36-73% of the studied catchments, and an improvement in modelled hydrological variability in 31-74% of the studied catchments. Including HIP in the GHMs also leads to an improvement in the simulation of hydrological extremes, compared to when HIP is excluded. Whilst the inclusion of HIP leads to decreases in simulated high-flows, it can lead to either increases or decreases in low-flows. This is due to the relative importance of the timing of return flows and reservoir operations and their associated uncertainties. Even with the inclusion of HIP, we find that model performance still not optimal. This highlights the need for further research linking the human management and hydrological domains, especially in those areas with a dominant human impact. The large variation in performance between GHMs, regions, and performance indicators, calls for a careful selection of GHMs, model components, and evaluation metrics in future model applications
