    Investigation on the Effect of Shapes on the Drying Kinetics and Sensory Evaluation Study of Dried Jackfruit

    Jackfruits are seasonal and highly nutritional fruits indigenous to the Southwestern rainforests of India. However much of the produce are spoilt annually due to poor preservation techniques. Minimal studies have been conducted on the drying kinetics of jackfruit and the effect of shapes on the drying kinetics. In this research, drying curves of three different shaped jackfruit slices were obtained using a convective oven at 40oC, 50oC, 60oC and 70oC. Modified Midilli-Kucuk Model was found to be the best kinetic model for drying of jackfruits. At all temperatures, effective moisture diffusivity values and activation energy varied from 2.66 x 10-10 - 4.85 x 10-10 m2/s and 16.08 - 20.07 kJ/mol respectively. Drying was found to be most efficient at 50oC using the square shaped slices with a R2, RMSE and SSE value of 0.9984, 0.01127 and 0.002668 respectively. Sensory evaluation of untreated and additive-added dried jackfruit slices was conducted by 40 untrained sensory panelists. Jackfruit with ascorbic acid and sugar coating had highest aesthetics value due to better retention of colour by ascorbic acid. However sugar coated jackfruit had the most favorable taste and smell. Further optimization must be done to satisfy consumers collectively to enable a highly marketable product

    A current review of the diagnostic and treatment strategies of hepatic encephalopathy

    Hepatic encephalopathy (HE) is a serious and potentially fatal complication in patients with cirrhotic liver disease. It is a spectrum ranging from minimal hepatic encephalopathy (MHE) without recognizable clinical symptoms or signs, to overt HE with risk of cerebral edema and death. HE results in diminished quality of life and survival. The broad range of neuropsychiatric manifestations reflects the range of pathophysiological mechanisms and impairment in neurotransmission that are purported to cause HE including hyperammonemia, astrocyte swelling, intra-astrocytic glutamine, upregulation of 18-kDa translocator protein (TSPO) (formerly known as peripheral benzodiazepine receptor or PBTR), and manganese. There is a myriad of diagnostic tools including simple bedside clinical assessment, and more complex neuropsychological batteries and neurophysiological tests available today. Current treatment strategies are directed at reducing ammonia, with newer agents showing some early promise. This paper describes the pathophysiology of the disease and summarises current diagnostic and treatment therapies available

    Alcohol promotes breast cancer cell invasion by regulating the Nm23-ITGA5 pathway

    <p>Abstract</p> <p>Background</p> <p>Alcohol consumption is an established risk factor for breast cancer metastasis. Yet, the mechanism by which alcohol promotes breast cancer metastases is unknown. The ability of cancer cells to invade through tissue barriers (such as basement membrane and interstitial stroma) is an essential step towards establishing cancer metastasis. In the present study, we identify and examine the roles of two genes, <it>Nm23 </it>and <it>ITGA5</it>, in alcohol-induced breast cancer cell invasion.</p> <p>Methods</p> <p>Human breast cancer T47D cells were treated with ethanol at various concentrations. Boyden chamber invasion assays were used to measure cellular invasive ability. The mRNA expression level of metastasis suppressor genes including <it>Nm23 </it>was determined by qRT-PCR. <it>ITGA5 </it>was identified using a qRT-PCR array of 84 genes important for cell-cell and cell-extracellular matrix interactions. <it>Nm23 </it>overexpression in addition to <it>Nm23</it>- and <it>ITGA5 </it>knock-down were used to determine the role of the Nm23-ITGA5 pathway on cellular invasive ability of T47D cells. Protein expression levels were verified by Western blot.</p> <p>Results</p> <p>Alcohol increased the invasive ability of human breast cancer T47D cells in a dose-dependent manner through the suppression of the <it>Nm23 </it>metastatic suppressor gene. In turn, <it>Nm23 </it>down-regulation increased expression of fibronectin receptor subunit <it>ITGA5</it>, which subsequently led to increased cellular invasion. Moreover, <it>Nm23 </it>overexpression was effective in suppressing the effects of alcohol on cell invasion. In addition, we show that the effects of alcohol on invasion were also inhibited by knock-down of <it>ITGA5</it>.</p> <p>Conclusions</p> <p>Our results suggest that the Nm23-ITGA5 pathway plays a critical role in alcohol-induced breast cancer cell invasion. Thus, regulation of this pathway may potentially be used to prevent the establishment of alcohol-promoted metastases in human breast cancers.</p

    Air Pollution and Lymphocyte Phenotype Proportions in Cord Blood

    Effects of air pollution on morbidity and mortality may be mediated by alterations in immune competence. In this study we examined short-term associations of air pollution exposures with lymphocyte immunophenotypes in cord blood among 1,397 deliveries in two districts of the Czech Republic. We measured fine particulate matter < 2.5 ÎŒm in diameter (PM(2.5)) and 12 polycyclic aromatic hydrocarbons (PAHs) in 24-hr samples collected by versatile air pollution samplers. Cord blood samples were analyzed using a FACSort flow cytometer to determine phenotypes of CD3(+) T-lymphocytes and their subsets CD4(+) and CD8(+), CD19(+) B-lymphocytes, and natural killer cells. The mothers were interviewed regarding sociodemographic and lifestyle factors, and medical records were abstracted for obstetric, labor and delivery characteristics. During the period 1994 to 1998, the mean daily ambient concentration of PM(2.5) was 24.8 ÎŒg/m(3) and that of PAHs was 63.5 ng/m(3). In multiple linear regression models adjusted for temperature, season, and other covariates, average PAH or PM(2.5) levels during the 14 days before birth were associated with decreases in T-lymphocyte phenotype fractions (i.e., CD3(+) CD4(+), and CD8(+)), and a clear increase in the B-lymphocyte (CD19(+)) fraction. For a 100-ng/m(3) increase in PAHs, which represented approximately two standard deviations, the percentage decrease was −3.3% [95% confidence interval (CI), −5.6 to −1.0%] for CD3(+), −3.1% (95% CI, −4.9 to −1.3%) for CD4(+), and −1.0% (95% CI, −1.8 to −0.2%) for CD8(+) cells. The corresponding increase in the CD19(+) cell proportion was 1.7% (95% CI, 0.4 to 3.0%). Associations were similar but slightly weaker for PM(2.5).     Preservation of mesophilic mixed culture for anaerobic palm oil mill effluent treatment by convective drying methods

    [EN] While anaerobic digestion is a reliable method that treats the waste and produces renewable biomethane fuel, the necessary sludge in liquid form is difficult to handle due to the constant biogas generation. Therefore, this study investigates the possibility of convective air drying, namely heat pump and hot air circulation oven as preservation methods for anaerobic microbial sludge. Drying was conducted at various temperatures, ranging from 22℃ to 70℃.     The implausibility of ‘usual care’ in an open system: sedation and weaning practices in Paediatric Intensive Care Units (PICUs) in the United Kingdom (UK)

    Background: The power of the randomised controlled trial depends upon its capacity to operate in a closed system whereby the intervention is the only causal force acting upon the experimental group and absent in the control group, permitting a valid assessment of intervention efficacy. Conversely, clinical arenas are open systems where factors relating to context, resources, interpretation and actions of individuals will affect implementation and effectiveness of interventions. Consequently, the comparator (usual care) can be difficult to define and variable in multi-centre trials. Hence outcomes cannot be understood without considering usual care and factors that may affect implementation and impact on the intervention. Methods: Using a fieldwork approach, we describe PICU context, ‘usual’ practice in sedation and weaning from mechanical ventilation, and factors affecting implementation prior to designing a trial involving a sedation and ventilation weaning intervention. We collected data from 23 UK PICUs between June and November 2014 using observation, individual and multi-disciplinary group interviews with staff. Results: Pain and sedation practices were broadly similar in terms of drug usage and assessment tools. Sedation protocols linking assessment to appropriate titration of sedatives and sedation holds were rarely used (9 % and 4 % of PICUs respectively). Ventilator weaning was primarily a medical-led process with 39 % of PICUs engaging senior nurses in the process: weaning protocols were rarely used (9 % of PICUs). Weaning methods were variably based on clinician preference. No formal criteria or use of spontaneous breathing trials were used to test weaning readiness. Seventeen PICUs (74 %) had prior engagement in multi-centre trials, but limited research nurse availability. Barriers to previous trial implementation were intervention complexity, lack of belief in the evidence and inadequate training. Facilitating factors were senior staff buy-in and dedicated research nurse provision. Conclusions: We examined and identified contextual and organisational factors that may impact on the implementation of our intervention. We found usual practice relating to sedation, analgesia and ventilator weaning broadly similar, yet distinctively different from our proposed intervention, providing assurance in our ability to evaluate intervention effects. The data will enable us to develop an implementation plan; considering these factors we can more fully understand their impact on study outcomes

    Discovery of very-high-energy emission from RGB J2243+203 and derivation of its redshift upper limit

    Very-high-energy (VHE; >> 100 GeV) gamma-ray emission from the blazar RGB J2243+203 was discovered with the VERITAS Cherenkov telescope array, during the period between 21 and 24 December 2014. The VERITAS energy spectrum from this source can be fit by a power law with a photon index of 4.6±0.54.6 \pm 0.5, and a flux normalization at 0.15 TeV of (6.3±1.1)×10−10 cm−2s−1TeV−1(6.3 \pm 1.1) \times 10^{-10} ~ \textrm{cm}^{-2} \textrm{s}^{-1} \textrm{TeV}^{-1}. The integrated \textit{Fermi}-LAT flux from 1 GeV to 100 GeV during the VERITAS detection is (4.1±0.8)×10-8 cm-2s-1(4.1 \pm 0.8) \times 10^{\textrm{-8}} ~\textrm{cm}^{\textrm{-2}}\textrm{s}^{\textrm{-1}}, which is an order of magnitude larger than the four-year-averaged flux in the same energy range reported in the 3FGL catalog, (4.0±0.1×10-9 cm-2s-14.0 \pm 0.1 \times 10^{\textrm{-9}} ~ \textrm{cm}^{\textrm{-2}}\textrm{s}^{\textrm{-1}}). The detection with VERITAS triggered observations in the X-ray band with the \textit{Swift}-XRT. However, due to scheduling constraints \textit{Swift}-XRT observations were performed 67 hours after the VERITAS detection, not simultaneous with the VERITAS observations. The observed X-ray energy spectrum between 2 keV and 10 keV can be fitted with a power-law with a spectral index of 2.7±0.22.7 \pm 0.2, and the integrated photon flux in the same energy band is (3.6±0.6)×10−13 cm−2s−1(3.6 \pm 0.6) \times 10^{-13} ~\textrm{cm}^{-2} \textrm{s}^{-1}. EBL model-dependent upper limits of the blazar redshift have been derived. Depending on the EBL model used, the upper limit varies in the range from z < 0.9<~0.9 to z < 1.1<~1.1
