    Adaptive Dispersion Compensation for Remote Fiber Delivery of NIR Femtosecond Pulses

    We report on remote delivery of 25 pJ broadband near-infrared femtosecond light pulses from a Ti:sapphire laser through 150 meters of single-mode optical fiber. Pulse distortion due to dispersion is overcome with pre-compensation using adaptive pulse shaping techniques, while nonlinearities are mitigated using an SF10 rod for the final stage of pulse compression. Near transform limited pulse duration of 130 fs is measured after the final compression.Comment: 3 pages, 4 figure

    Aged lipid-laden microglia display impaired responses to stroke

    Microglial cells of the aged brain manifest signs of dysfunction that could contribute to the worse neurological outcome of stroke in the elderly. Treatment with colony-stimulating factor 1 receptor antagonists enables transient microglia depletion that is followed by microglia repopulation after treatment interruption, causing no known harm to mice. We tested whether this strategy restored microglia function and ameliorated stroke outcome in old mice. Cerebral ischemia/reperfusion induced innate immune responses in microglia highlighted by type I interferon and metabolic changes involving lipid droplet biogenesis. Old microglia accumulated lipids under steady state and displayed exacerbated innate immune responses to stroke. Microglia repopulation in old mice reduced lipid-laden microglia, and the cells exhibited reduced inflammatory responses to ischemia. Moreover, old mice with renewed microglia showed improved motor function 2 weeks after stroke. We conclude that lipid deposits in aged microglia impair the cellular responses to ischemia and worsen functional recovery in old mice.© 2022 The Authors.     Parameter estimation with the current generation of phenomenological waveform models applied to the black hole mergers of GWTC-1

    We consider the ten confidently detected gravitational-wave signals in theGWTC-1 catalog which are consistent with mergers of binary black hole systems,and perform a thorough parameter estimation re-analysis. This is made possibleby using computationally efficient waveform models of the current (fourth)generation of the IMRPhenom family of phenomenological waveform models, whichconsists of the IMRPhenomX frequency-domain modelsand the IMRPhenomTtime-domain models. The analysis is performed with both precessing andnon-precessing waveform models with and without subdominant spherical harmonicmodes. Results for all events are validated with convergence tests, discussingin particular the events GW170729 and GW151226. For the latter and the othertwo lowest-mass events, we also compare results between two independentsampling codes, Bilby and LALInference. We find overall consistent results withthe original GWTC-1 results, with all Jensen-Shannon divergences between theprevious results using IMRPhenomPv2 and our default IMRPhenomXPHM posteriorsbelow 0.045 bits, but we also discuss cases where including subdominantharmonics and/or precession influences the posteriors.<br

    New twists in compact binary waveform modeling: A fast time-domain model for precession

    We present IMRPhenomTPHM, a phenomenological model for the gravitational wave signals emitted by the coalescence of quasi-circular precessing binary black holes systems. The model is based on the "twisting up" approximation, which maps non-precessing signals to precessing ones in terms of a time dependent rotation described by three Euler angles, and which has been utilized in several frequency domain waveform models that have become standard tools in gravitational wave data analysis. Our model is however constructed in the time domain, which allows several improvements over the frequency domain models: we do not use the stationary phase approximation, we employ a simple approximation for the precessing Euler angles for the ringdown signal, and we implement a new method for computing the Euler angles through the evolution of the spin dynamics of the system, which is more accurate and also computationally efficient

    Ecological niche and phylogeography elucidate complex biogeographic patterns in Loxosceles rufescens (Araneae, Sicariidae) in the Mediterranean Basin

    Background: Understanding the evolutionary history of morphologically cryptic species complexes is difficult, and made even more challenging when geographic distributions have been modified by human-mediated dispersal. This situation is common in the Mediterranean Basin where, aside from the environmental heterogeneity of the region, protracted human presence has obscured the biogeographic processes that shaped current diversity. Loxosceles rufescens (Araneae, Sicariidae) is an ideal example: native to the Mediterranean, the species has dispersed worldwide via cohabitation with humans. A previous study revealed considerable molecular diversity, suggesting cryptic species, but relationships among lineages did not correspond to geographic location. Results: Delimitation analyses on cytochrome c oxidase subunit I identified 11 different evolutionary lineages, presenting two contrasting phylogeographic patterns: (1) lineages with well-structured populations in Morocco and Iberia, and (2) lineages lacking geographic structure across the Mediterranean Basin. Dating analyses placed main diversification events in the Pleistocene, and multiple Pleistocene refugia, identified using ecological niche modeling (ENM), are compatible with allopatric differentiation of lineages. Human-mediated transportation appears to have complicated the current biogeography of this medically important and synanthropic spider. Conclusions: We integrated ecological niche models with phylogeographic analyses to elucidate the evolutionary history of L. rufescens in the Mediterranean Basin, with emphasis on the origins of mtDNA diversity. We found support for the hypothesis that northern Africa was the center of origin for L. rufescens, and that current genetic diversity originated in allopatry, likely promoted by successive glaciations during the Pleistocene. We corroborated the scenario of multiple refugia within the Mediterranean, principally in northern Africa, combining results from eight atmosphereocean general circulation models (AOGCMs) with two different refugium-delimitation methodologies. ENM results were useful for providing general views of putative refugia, with fine-scale details depending on the level of stringency applied for agreement among models

    A detailed analysis of GW190521 with phenomenological waveform models

    In this paper we present an extensive analysis of the GW190521 gravitational wave event with the current (fourth) generation of phenomenological waveform models for binary black hole coalescences. GW190521 stands out from other events since only a few wave cycles are observable. This leads to a number of challenges, one being that such short signals are prone to not resolve approximate waveform degeneracies, which may result in multi-modal posterior distributions. The family of waveform models we use includes a new fast time-domain model IMRPhenomTPHM, which allows us extensive tests of different priors and robustness with respect to variations in the waveform model, including the content of spherical harmonic modes. We clarify some issues raised in a recent paper [Nitz&Capano], associated with possible support for a high-mass ratio source, but confirm their finding of a multi-modal posterior distribution, albeit with important differences in the statistical significance of the peaks. In particular, we find that the support for both masses being outside the PISN mass-gap, and the support for an intermediate mass ratio binary are drastically reduced with respect to what Nitz&Capano found. We also provide updated probabilities for associating GW190521 to the potential electromagnetic counterpart from ZTF

    Plasmodium falciparum Apicomplexan-Specific Glucosamine-6-Phosphate N-Acetyltransferase Is Key for Amino Sugar Metabolism and Asexual Blood Stage Development

    UDP-N-acetylglucosamine (UDP-GlcNAc), the main product of the hexosamine biosynthetic pathway, is an important metabolite in protozoan parasites since its sugar moiety is incorporated into glycosylphosphatidylinositol (GPI) glycolipids and N- and O-linked glycans. Apicomplexan parasites have a hexosamine pathway comparable to other eukaryotic organisms, with the exception of the glucosamine-phosphate N-acetyltransferase (GNA1) enzymatic step that has an independent evolutionary origin and significant differences from nonapicomplexan GNA1s. By using conditional genetic engineering, we demonstrate the requirement of GNA1 for the generation of a pool of UDP-GlcNAc and for the development of intraerythrocytic asexual Plasmodium falciparum parasites. Furthermore, we present the 1.95 A resolution structure of the GNA1 ortholog from Cryptosporidium parvum, an apicomplexan parasite which is a leading cause of diarrhea in developing countries, as a surrogate for P. falciparum GNA1. The indepth analysis of the crystal shows the presence of specific residues relevant for GNA1 enzymatic activity that are further investigated by the creation of site-specific mutants. The experiments reveal distinct features in apicomplexan GNA1 enzymes that could be exploitable for the generation of selective inhibitors against these parasites, by targeting the hexosamine pathway. This work underscores the potential of apicomplexan GNA1 as a drug target against malaria. IMPORTANCE Apicomplexan parasites cause a major burden on global health and economy. The absence of treatments, the emergence of resistances against available therapies, and the parasite''s ability to manipulate host cells and evade immune systems highlight the urgent need to characterize new drug targets to treat infections caused by these parasites. We demonstrate that glucosamine-6-phosphate N-acetyltransferase (GNA1), required for the biosynthesis of UDP-N-acetylglucosamine (UDP-GlcNAc), is essential for P. falciparum asexual blood stage development and that the disruption of the gene encoding this enzyme quickly causes the death of the parasite within a life cycle. The high-resolution crystal structure of the GNA1 ortholog from the apicomplexan parasite C. parvum, used here as a surrogate, highlights significant differences from human GNA1. These divergences can be exploited for the design of specific inhibitors against the malaria parasite

    Reactivity of homoleptic and heteroleptic core paddle wheel Cu(II) compounds

    The compound [Cu(μ-Pip)(μ-OAc)(MeOH)]2 (1) (Pip=Piperonylate, OAc=acetate, MeOH=methanol) has been obtained in high percentage yield. Its reactivity with pyridine/pyrazole derivative ligands (pyridine (py), 3-phenylpyridine (3-Phpy) and 4-acetylpyridine (4-Acpy)) and 3,5-dimethylpyrazole (3,5-dmpz) leads to four monomeric compounds: [Cu(Pip)2(dPy)2(H2O)] (dPy=py (2), 3-Phpy (3) and 4-Acpy (4a)) and [Cu(Pip)2(3,5-dmpz)2] (5). Furthermore, the reaction of 1 with HPip in MeOH:DMF solvent under reflux conditions yields the homoleptic core paddle-wheel compound [Cu(μ-Pip)2(DMF)]2·2DMF (6). The reaction between 6 and 2-benzylpyridine (2-Bzpy) yields the paddle-wheel core compound [Cu(Pip)2(2-Bzpy)]2 (7). All compounds have been fully characterized by analytical and spectroscopic techniques and their X-ray crystal structures have been determined. In this set of compounds, the carboxylate ligand (Pip) displays different coordination modes (monodentate (2-4), bidentate chelate (5) and bridged (1, 6 and 7)). Moreover, their extended structures are discussed: the crystal packing indicates hydrogen bond propagation, which defines 1D (2-5) or 2D (6 and 7) supramolecular networks
