    Viruses infecting daphne in New Zealand : a thesis presented in partial fulfilment of the requirements for the degree of Master of Horticultural Science

    The genus Daphne belongs to the Thymelaeaceae and contains both evergreen and deciduous shrubs which produce attractive, highly fragrant flowers. Plants are neat and shapely and seldom grow more than 3-4 feet high, fitting well into rock gardens or herbaceous borders. A large number of species of this very popular ornamental are grown in New Zealand, the most common of which is Daphne odora Thunb. This latter species contains several cultivars and one (Daphne odora 'Leucanthe') is almost ubiquitous at least in the North Island. Its importance is illustrated by the fact that several nurseries visited during the study cited production figures for Leucanthe of between 5,000 and 12,000 specimens per annum and one firm has also entered the export trade.     Phylogeny and historical biogeography of Lauraceae

    Phylogenetic relationships among 122 species of Lauraceae representing 44 of the 55 currently recognized genera are inferred from sequence variation in the chloroplast and nuclear genomes. The trnL-trnF, trnT-trnL, psbA-trnH, and rpll6 regions of cpDNA, and the 5' end of 26S rDNA resolved major lineages, while the ITS/5.8S region of rDNA resolved a large terminal lade. The phylogenetic estimate is used to assess morphology-based views of relationships and, with a temporal dimension added, to reconstruct the biogeographic history of the family. Results suggest Lauraceae radiated when trans-Tethyean migration was relatively easy, and basal lineages are established on either Gondwanan or Laurasian terrains by the Late Cretaceous. Most genera with Gondwanan histories place in Cryptocaryeae, but a small group of South American genera, the Chlorocardium-Mezilauruls lade, represent a separate Gondwanan lineage. Caryodaphnopsis and Neocinnamomum may be the only extant representatives of the ancient Lauraceae flora docu- mented in Mid- to Late Cretaceous Laurasian strata. Remaining genera place in a terminal Perseeae-Laureae lade that radiated in Early Eocene Laurasia. Therein, non-cupulate genera associate as the Persea group, and cupuliferous genera sort to Laureae of most classifications or Cinnamomeae sensu Kostermans. Laureae are Laurasian relicts in Asia. The Persea group and Cinnamomum group (of Cinnamomeae) show tropical amphi-Pacific disjunctions here credited to disruption of boreotropical ranges by Eocene-Oligocene climatic cooling. The Ocotea complex accommodates re- maining Cinnamomeae and shows a trans-Atlantic disjunction possibly derived from a Madrean-Tethyan ancestral distribution. These findings support Laurasian ancestry for most extant Lauraceae, with their considerable neotropical representation primarily derived from Early Miocene radiation of the Ocotea complex upon reaching South America

    L’objectif principal de cette étude est d’établir le potentiel salmonicole du lac Cardonnière qui se trouve dans la réserve faunique de Rimouski, afin de bonifier la pêche sportive de l’omble de fontaine (Salvelinus fontinalis). Pour cela, une diagnose a été réalisée les 2, 3 et 4 septembre 2011. La bathymétrie et les données physico-chimiques ont été mesurées. Le potentiel de fraie dans le lac, ses tributaires et son émissaire a été évalué, et une pêche expérimentale a été effectuée. Il en est ressorti que le lac a un bon potentiel salmonicole et que l’omble de fontaine s’en tire plutôt bien, malgré la présence du meunier noir (Catostomus commersoni). Afin d’assurer la viabilité de la pêche sportive à long terme, des mesures sont proposées dans le présent rapport

    A genome-wide analysis of Escherichia coli responses to fosfomycin using TraDIS-Xpress reveals novel roles for phosphonate degradation and phosphate transport systems

    BACKGROUND: Fosfomycin is an antibiotic that has seen a revival in use due to its unique mechanism of action and efficacy against isolates resistant to many other antibiotics. In Escherichia coli, fosfomycin often selects for loss-of-function mutations within the genes encoding the sugar importers, GlpT and UhpT. There has, however, not been a genome-wide analysis of the basis for fosfomycin susceptibility reported to date. METHODS: Here we used TraDIS-Xpress, a high-density transposon mutagenesis approach, to assay the role of all genes in E. coli involved in fosfomycin susceptibility. RESULTS: The data confirmed known fosfomycin susceptibility mechanisms and identified new ones. The assay was able to identify domains within proteins of importance and revealed essential genes with roles in fosfomycin susceptibility based on expression changes. Novel mechanisms of fosfomycin susceptibility that were identified included those involved in glucose metabolism and phosphonate catabolism (phnC-M), and the phosphate importer, PstSACB. The impact of these genes on fosfomycin susceptibility was validated by measuring the susceptibility of defined inactivation mutants. CONCLUSIONS: This work reveals a wider set of genes that contribute to fosfomycin susceptibility, including core sugar metabolism genes and two systems involved in phosphate uptake and metabolism previously unrecognized as having a role in fosfomycin susceptibility

    Computational prediction of Pho regulons in cyanobacteria

    <p>Abstract</p> <p>Background</p> <p>Phosphorus is an essential element for all life forms. However, it is limiting in most ecological environments where cyanobacteria inhabit. Elucidation of the phosphorus assimilation pathways in cyanobacteria will further our understanding of the physiology and ecology of this important group of microorganisms. However, a systematic study of the Pho regulon, the core of the phosphorus assimilation pathway in a cyanobacterium, is hitherto lacking.</p> <p>Results</p> <p>We have predicted and analyzed the Pho regulons in 19 sequenced cyanobacterial genomes using a highly effective scanning algorithm that we have previously developed. Our results show that different cyanobacterial species/ecotypes may encode diverse sets of genes responsible for the utilization of various sources of phosphorus, ranging from inorganic phosphate, phosphodiester, to phosphonates. Unlike in <it>E. coli</it>, some cyanobacterial genes that are directly involved in phosphorus assimilation seem to not be under the regulation of the regulator SphR (orthologue of PhoB in <it>E coli</it>.) in some species/ecotypes. On the other hand, SphR binding sites are found for genes known to play important roles in other biological processes. These genes might serve as bridging points to coordinate the phosphorus assimilation and other biological processes. More interestingly, in three cyanobacterial genomes where no <it>sphR </it>gene is encoded, our results show that there is virtually no functional SphR binding site, suggesting that transcription regulators probably play an important role in retaining their binding sites.</p> <p>Conclusion</p> <p>The Pho regulons in cyanobacteria are highly diversified to accommodate to their respective living environments. The phosphorus assimilation pathways in cyanobacteria are probably tightly coupled to a number of other important biological processes. The loss of a regulator may lead to the rapid loss of its binding sites in a genome.</p

    This Article considers how corporations are using image advertising in litigation hot spots as a means of influencing litigation outcomes. It describes how Samsung and other companies advertised in the Eastern District of Texas--a patent litigation hot spot --to curry favor with the people who live there, including by sponsoring an ice rink located directly outside the courthouse. To be sure, image advertisements are constitutionally protected speech and might even warrant the highest level of protection under the First Amendment when they are not purely commercial in nature. Still, the Article argues, courts should be able to prohibit such advertisements altogether, or at the very least limit their impact through voir dire and discovery, because they threaten the right to an impartial jury guaranteed by the Seventh Amendment

    The introduction and utilization of Information Systems (IS) in the hospital environment has had a significant and lasting impact on the practice of medicine. The development of this dissertation will attempt to explore a widely overlooked area: The comparison of Chief Information Officers (CIOs) in the United States and the United Kingdom. Aspects of CIO experiences relating to assumed roles, CIO challenges, skills, frustrations, success, failure, leadership, management, involvement and perceptions about the role of Information Technology (IT) in healthcare are discussed with a comparative global model. This study investigates the managerial roles of the Chief Information Officer based on Mintzberg\u27s classical managerial role model. To be successful, CIOs need to have relation-building skills, managerial skills, a broad knowledge of technology, and management, technical and business degrees along with certain personal traits and backgrounds. While high technology has emerged as an important economic issue in all advanced industrial countries, there is much variation between countries in the success of their high technological industries, and in government efforts to encourage high technology in the healthcare sector. This research will attempt to demonstrate that CIOs as a whole believe IT to be indispensable in effective realization of the healthcare mission in a global, information-intensive civilization, and that IT can positively impact the quality, cost and medical issues of healthcare. The overall objective of this study is to examine the following: (1) will an increase in global information technology expand awareness of appropriate differing styles? (2) does global information technology have a positive and significant relationship within the international community? Finally, this study calls for more interdisciplinary research integrating insight from organizational behavior, international business and information technology
