Microbial shelf life of chub-packaged ground beef from four large U.S. processing plants
Ten pound chubs of coarsely ground beef of two different lean:fat specifications (73:27 and 81:19) were stored at three temperatures (34, 38 or 45 ÌŠF) to monitor the effects of storage temperature on microbial condition of the product. Ground beef from four U.S. plants was tested (2 trials each), and microbial analyses were conducted on storage days 0, 6, 10, 14, and 18 using seven different media to estimate counts of total aerobic and anaerobic, lactic acid bacteria (LAB), and Gram-negative bacteria. Bacterial counts for a given culture medium were similar among plants and meat types. At day 10, total mi crobial counts from chubs stored at 38 or 45 ÌŠF were approximately 8 log10 CFU/g, whereas total counts from chubs stored at 34 ÌŠF were approximately 4.5 log 10 CFU/g (4 log=10,000, CFU is colony forming units). Regardless of storage temperature and meat type, LAB predominated. Growth of gram-negative enteric bacteria was delayed in chubs stored at 34 ÌŠF throughout the 18 day study, whereas counts increased in chubs stored at 38 or 45 ÌŠ F
Haemophilia patients aged 0 - 18 years in the Western Cape
Objectives. To record the number of haemophiliacs aged 0- 18 years in the Western Cape (WC), what event led to the diagnosis, the level of clotting factor, treatment, functional status of their joints and impact of the disease on the family.Design. A prospective study of patients registered with the South African National Haemophilia Registry and new patients, utilising the patients' paediatricians, hospital records, patient and guardian interviews, physicalexamination and provincial nurse haemophilia co-ordinators.Setting. Haemophilia care centres at the three WC academic hospitals, regional hospitals and homes of patients. Two elective medical students, MHH and JJH, collected the information.Subjects. All boys with confirmed haemophilia A or B in the WC.Outcome measures. Events that led to diagnosis, degree of haemophilia, use of clotting factor, functional status, and effect on family. Results. Of 78 patients (59 haemophilia A, 19 haemophilia B) identified, 49 could be studied. Forty-three per cent had severe, 29% moderate and 22% mild disease (6% unknown). Family history was present in 49%, but led to diagnosis in only 12%. The most common first symptoms were subcutaneous and mucosal bleeding. Delay in diagnosis varied from 0 to 9 months. Twenty-nine per cent of guardians were suspected of child abuse. RSA produced clotting factor was used 'on demand' in 73% of patients, for periodic prophylaxis in 20% and as continuous prophylaxis in 7%. Joints were functionally restricted in 43% of patients. The majority of guardians (59%) said the disease had a major impact on the family.Conclusions. The diagnosis of haemophilia in children with a positive family history was often delayed. Haemophilia causes significant morbidity in our patients and their families

Objective - Emerging evidence suggests that human blood contains bone marrow (BM)-derived endothelial progenitor cells that contribute to postnatal neovascularization. Clinical trials demonstrated that administration of BM-cells can enhance neovascularization. Most studies, however, used crude cell populations. Identifying the role of different cell populations is important for developing improved cellular therapies. Methods and Results - Effects of the hematopoietic stem cell-containing CD34+ cell population on migration, proliferation, differentiation, stimulation of, and participation in capillary-like tubule formation were assessed in an in vitro 3-dimensional matrix model using human microvascular endothelial cells. During movement over the endothelial monolayer, CD34+ cells remained stuck at sites of capillary tube formation and time- and dose-dependently formed cell clusters. Immunohistochemistry confirmed homing and proliferation of CD34+ cells in and around capillary sprouts. CD34+ cells were transduced with the LNGFR marker gene to allow tracing. LNGFR gene-transduced CD34 + cells integrated in the tubular structures and stained positive for CD31 and UEA-1. CD34+ cells alone stimulated neovascularization by 17%. Coculture with CD34- cells led to 68% enhancement of neovascularization, whereas CD34- cells displayed a variable response by themselves. Cell-cell contact between CD34+ and CD34- cells facilitated endothelial differentiation of CD34+ cells. Conclusions - Our data suggest that administration of CD34+-enriched cell populations may significantly improve neovascularization and point at an important supportive role for (endogenous or exogenous) CD34- cells. © 2005 American Heart Association, Inc. Single particle flame-combustion studies on solid biomass fuels
Combustion of solid biomass in large scale power generation has been recognized as a key technology for the transition to a decarbonized electricity sector in the UK by 2050. Much of the near-term forecast capacity is likely to be by the conversion of existing coal-fired pulverized fuel plant (DECC, 2012). In such applications, it will be necessary to ensure that the combustion behaviour of the solid biomass fuels is engineered to match, as far as practical, that of the original plant design. While biomass feedstock characteristics vary considerably, one controllable variable for pulverized fuel is the size of the particles.Useful modelling for adaptation and design of boiler plant can be improved with more detailed measurement of the real behaviour of individual particles of the varying fuels. Typical power plant biomass fuels including pine, eucalyptus and willow with particle sizes ranging from up to 3. mm (Van Loo and Koppejan, 2008) and with differing moisture content and aspect ratios were selected for study. Single particles were supported in a water-cooled cover and then exposed above a flame, simulating biomass combustion in a furnace. Measurements of ignition delay, volatile burning time and char burn-out time were undertaken using high speed image capture. Temperatures of the surrounding environment and near to the particle surface were measured with thermocouples and thermometric imaging. Thermo-gravimetric measurements on separate samples complement the single particle measurements as a means of verifying the demarcation between the different stages of combustion and providing kinetic data.Analysis of the data identified correlations between the biomass fundamental characteristics, particle size, and the observed combustion profiles. Empirical expressions for the duration of each combustion stage have been derived. These have been validated with basic modelling including the predicted devolatilisation stage calculated by the FG-Biomass model (Chen et al.,1998)

The role of data visualization in Railway Big Data Risk Analysis
Big Data Risk Analysis (BDRA) is one of the possible alleys for the further development of risk models in the railway transport. Big Data techniques allow a great quantity of information to be handled from different types of sources (e.g. unstructured text, signaling and train data). The benefits of this approach may lie in improving the understanding of the risk factors involved in railways, detecting possible new threats or assessing the risk levels for rolling stock, rail infrastructure or railway operations. For the efficient use of BDRA, the conversion of huge amounts of data into a simple and effective display is particularly challenging. Especially because it is presented to various specific target audiences. This work reports a literature review of risk communication and visualization in order to find out its applicability to BDRA, and beyond the visual techniques, what human factors have to be considered in the understanding and risk perception of the infor-mation when safety analysts and decision-makers start basing their decisions on BDRA analyses. It was found that BDRA requires different visualization strategies than those that have normally been carried out in risk analysis up to now
Pulsar timing arrays and the challenge of massive black hole binary astrophysics
Pulsar timing arrays (PTAs) are designed to detect gravitational waves (GWs)
at nHz frequencies. The expected dominant signal is given by the superposition
of all waves emitted by the cosmological population of supermassive black hole
(SMBH) binaries. Such superposition creates an incoherent stochastic
background, on top of which particularly bright or nearby sources might be
individually resolved. In this contribution I describe the properties of the
expected GW signal, highlighting its dependence on the overall binary
population, the relation between SMBHs and their hosts, and their coupling with
the stellar and gaseous environment. I describe the status of current PTA

A protocol for developing reporting guidelines for laboratory studies in endodontology
Laboratory‐based research studies are the most common form of research endeavour and make up the majority of manuscripts that are submitted for publication in the field of Endodontology. The scientific information derived from laboratory studies can be used to design a wide range of subsequent studies and clinical trials and may have translational potential to benefit clinical practice. Unfortunately, the majority of laboratory‐based articles submitted for publication fail the peer‐review step, because unacceptable flaws or substantial limitations are identified. Even when apparently well‐conducted laboratory‐based articles are peer‐reviewed, they can often require substantial corrections prior to the publication. It is apparent that some authors and reviewers may lack the training and experience to have developed a systematic approach to evaluate the quality of laboratory studies. Occasionally, even accepted manuscripts contain limitations that may compromise interpretation of data. To help authors avoid manuscript rejection and correction pitfalls, and to aid editors/reviewers to evaluate manuscripts systematically, the purpose of this project is to establish and publish quality guidelines for authors to report laboratory studies in the field of Endodontology so that the highest standards are achieved. The new guidelines will be named–‘Preferred Reporting Items for Laboratory studies in Endodontology’ (PRILE). A steering committee was assembled by the project leads to develop the guidelines through a five‐phase consensus process. The committee will identify new items as well as review and adapt items from existing guidelines. The items forming the draft guidelines will be reviewed and refined by a PRILE Delphi Group (PDG). The items will be evaluated by the PDG on a nine‐point Likert scale for relevance and inclusion. The agreed items will then be discussed by a PRILE face‐to‐face consensus meeting group (PFCMG) formed by 20 individuals to further refine the guidelines. This will be subject to final approval by the steering committee. The approved PRILE guidelines will be disseminated through publication in relevant journals, presented at congresses/meetings, and be freely available on a dedicated website. Feedback and comments will be solicited from researchers, editors and peer reviewers, who are invited to contact the steering committee with comments to help them update the guidelines periodically
