    Primarna proteoliza bijelog sira u salamuri proizvedenog od sirovog kravljeg mlijeka praćena visokomolarnim SDS-PAGE elektroforetskim sistemom

    The aim of this work was to investigate primary proteolysis of white brined cow cheese prepared from raw milk by SDS-electrophoretic method based on high-molarity Tris buffer system and to correlate with the results of other commonly used parameters. Proteolytic changes of white brined cow cheese were monitored by three parameters: total protein, water soluble proteins and degree of proteolysis. Changes of major casein fractions were followed by SDS-electrophoretic system in reducing conditions. The total protein content and moisture content of white brined cow cheese were significantly affected by ripening. Ripening in brine increased water soluble proteins and degree of proteolysis. Major caseins were differently resistant to proteolysis; αs-CN was more susceptible than β-CN. The αs-CN content was highly and negatively correlated with time of ripening and water soluble proteins whereas no significant correlation (p<0.05) between β-CN content and these parameters was found. Also, a strong significant correlation (p<0.05) between the amount of low molecular weight products and time of ripening, water soluble proteins and αs-CN content was observed. SDS-PAGE method used in this study could be useful for monitoring the white cow cheese proteolysis.Cilj ovog rada bio je istražiti proces primarne proteolize bijelog sira u salamuri pripremljenog od sirovog kravljeg mlijeka metodom SDS-poliakrilamidne gel elektroforeze zasnovane na primjeni visoko molarnog sistema Tris pufera i korelirati s rezultatima uobičajeno korištenih parametara. Proteolitičke promjene bijelog sira u salamuri praćene su pomoću tri parametra: ukupnih proteina, proteina topljivih u vodi i stupnja proteolize. Promjena glavnih kazeinskih frakcija praćena je SDS-elektroforetskom metodom u reducirajućim uvjetima. Zrenje je značajno utjecalo na sadržaj ukupnih proteina i sadržaj vlage u kravljem bijelom siru. Ovaj proces povećao je sadržaj proteina topljivih u vodi i stupanj proteolize. Glavne kazeinske frakcije bile su različito otporne na proteolizu. αs-CN je bio mnogo osjetljiviji na djelovanje proteolitičkih enzima u odnosu na β-CN. Količina αs-CN u jakoj je negativnoj korelaciji sa trajanjem zrenja i proteinima topljivim u vodi, dok nije utvrđena značajna korelacija između sadržaja β-CN i ovih parametara. Također, značajna korelacija utvrđena je između količine produkata male molekulske mase i trajanja zrenja, sadržaja αs-CN i proteina topljivih u vodi. SDS-elektroforetska metoda korištena u ovim istraživanjima može biti korisna za praćenje proteolitičkih promjena tijekom zrenja kravljeg bijelog sira u salamuri

    The trade-off between taxi time and fuel consumption in airport ground movement

    Environmental impact is a very important agenda item in many sectors nowadays, which the air transportation sector is also trying to reduce as much as possible. One area which has remained relatively unexplored in this context is the ground movement problem for aircraft on the airport’s surface. Aircraft have to be routed from a gate to a runway and vice versa and it is still unknown whether fuel burn and environmental impact reductions will best result from purely minimising the taxi times or whether it is also important to avoid multiple acceleration phases. This paper presents a newly developed multi-objective approach for analysing the trade-off between taxi time and fuel consumption during taxiing. The approach consists of a combination of a graph-based routing algorithm and a population adaptive immune algorithm to discover different speed profiles of aircraft. Analysis with data from a European hub airport has highlighted the impressive performance of the new approach. Furthermore, it is shown that the trade-off between taxi time and fuel consumption is very sensitive to the fuel-related objective function which is used

    Extensive sampling sheds light on species-level diversity in Palearctic Placobdella (Annelida: Clitellata: Glossiphoniiformes)

    The bloodfeeding leech genus Placobdella is dominated by North American diversity, with only a single nominal species known from Central America and one from the Palearctic region. This is likely due to considerable underestimation of Palearctic biodiversity, but investigations into potential hidden diversity are lacking. To shed light on this, the present study introduces new data for specimens initially identified as Placobdella costata from Ukraine (close to the type locality), Italy, Germany, Latvia, Montenegro, Bulgaria, Slovenia, Turkey, Azerbaijan, Tunisia, and Algeria, and uses both nuclear (Internal Transcribed Spacer [ITS] region) and mitochondrial (cytochrome c oxidase subunit I [COI]) sequence data in phylogenetic and DNA barcoding frameworks, in order to better understand species-level diversity. Seven independent lineages are present in the trees, five of which show adequate separation at the COI locus to suggest their unique species-level status (COI distances between these clades range from 4.86 to 8.10%). However, the ITS data suggest that speciation is recent or incipient in these clades, and that not enough time has passed for clear separation at this locus. We discuss the evolutionary and taxonomic implications of our findings and speculate on dispersal events that may have contributed to shaping this pattern of geographic distribution

    Variations in concerns reported on the Patient Concerns Inventory (PCI) in head and neck cancer patients from different health settings across the world

    Background: The aim was to collate and contrast patient concerns from a range of different head and neck cancer follow-up clinics around the world. Also, we sought to explore the relationship, if any, between responses to the patient concerns inventory (PCI) and overall quality of life (QOL). Methods: Nineteen units participated with intention of including 100 patients per site as close to a consecutive series as possible in order to minimize selection bias. Results: There were 2136 patients with a median total number of PCI items selected of 5 (2-10). “Fear of the cancer returning” (39%) and “dry mouth” (37%) were most common. Twenty-five percent (524) reported less than good QOL. Conclusion: There was considerable variation between units in the number of items selected and in overall QOL, even after allowing for case-mix variables. There was a strong progressive association between the number of PCI items and QOL

    DecSerFlow: Towards a Truly Declarative Service Flow Language

    The need for process support in the context of web services has triggered the development of many languages, systems, and standards. Industry has been developing software solutions and proposing standards such as BPEL, while researchers have been advocating the use of formal methods such as Petri nets and pi-calculus. The languages developed for service flows, i.e., process specification languages for web services, have adopted many concepts from classical workflow management systems. As a result, these languages are rather procedural and this does not fit well with the autonomous nature of services. Therefore, we propose DecSerFlow as a Declarative Service Flow Language. DecSerFlow can be used to specify, enact, and monitor service flows. The language is extendible (i.e., constructs can be added without changing the engine or semantical basis) and can be used to enforce or to check the conformance of service flows. Although the language has an appealing graphical representation, it is grounded in temporal logic

    Structural and functional insights into the candidate genes associated with different developmental stages of flag leaf in bread wheat (Triticum aestivum L.)

    Grain yield is one of the most important aims for combating the needs of the growing world population. The role of development and nutrient transfer in flag leaf for higher yields at the grain level is well known. It is a great challenge to properly exploit this knowledge because all the processes, starting from the emergence of the flag leaf to the grain filling stages of wheat (Triticum aestivum L.), are very complex biochemical and physiological processes to address. This study was conducted with the primary goal of functionally and structurally annotating the candidate genes associated with different developmental stages of flag leaf in a comprehensive manner using a plethora of in silico tools. Flag leaf-associated genes were analyzed for their structural and functional impacts using a set of bioinformatics tools and algorithms. The results revealed the association of 17 candidate genes with different stages of flag leaf development in wheat crop. Of these 17 candidate genes, the expression analysis results revealed the upregulation of genes such as TaSRT1-5D, TaPNH1-7B, and TaNfl1-2B and the downregulation of genes such as TaNAP1-7B, TaNOL-4D, and TaOsl2-2B can be utilized for the generation of high-yielding wheat varieties. Through MD simulation and other in silico analyses, all these proteins were found to be stable. Based on the outcome of bioinformatics and molecular analysis, the identified candidate genes were found to play principal roles in the flag leaf development process and can be utilized for higher-yield wheat production.     Science and Management of Intermittent Rivers and Ephemeral Streams (SMIRES)

    More than half of the global river network is composed of intermittent rivers and ephemeral streams (IRES), which are expanding in response to climate change and increasing water demands. After years of obscurity, the science of IRES has bloomed recently and it is being recognised that IRES support a unique and high biodiversity, provide essential ecosystem services and are functionally part of river networks and groundwater systems. However, they still lack protective and adequate management, thereby jeopardizing water resources at the global scale. This Action brings together hydrologists, biogeochemists, ecologists, modellers, environmental economists, social researchers and stakeholders from 14 different countries to develop a research network for synthesising the fragmented, recent knowledge on IRES, improving our understanding of IRES and translating this into a science-based, sustainable management of river networks. Deliverables will be provided through i) research workshops synthesising and addressing key challenges in IRES science, supporting research exchange and educating young researchers, and ii) researcher-stakeholder workshops translating improved knowledge into tangible tools and guidelines for protecting IRES and raising awareness of their importance and value in societal and decision-maker spheres. This Action is organized within six Working Groups to address: (i) the occurrence, distribution and hydrological trends of IRES; (ii) the effects of flow alterations on IRES functions and services; (iii) the interaction of aquatic and terrestrial biogeochemical processes at catchment scale; (iv) the biomonitoring of the ecological status of IRES; (v) synergies in IRES research at the European scale, data assemblage and sharing; (vi) IRES management and advocacy training

    Declarative Event-Based Workflow as Distributed Dynamic Condition Response Graphs

    We present Dynamic Condition Response Graphs (DCR Graphs) as a declarative, event-based process model inspired by the workflow language employed by our industrial partner and conservatively generalizing prime event structures. A dynamic condition response graph is a directed graph with nodes representing the events that can happen and arrows representing four relations between events: condition, response, include, and exclude. Distributed DCR Graphs is then obtained by assigning roles to events and principals. We give a graphical notation inspired by related work by van der Aalst et al. We exemplify the use of distributed DCR Graphs on a simple workflow taken from a field study at a Danish hospital, pointing out their flexibility compared to imperative workflow models.     Propofol-Induced Changes in Neurotrophic Signaling in the Developing Nervous System In Vivo

    Several studies have revealed a role for neurotrophins in anesthesia-induced neurotoxicity in the developing brain. In this study we monitored the spatial and temporal expression of neurotrophic signaling molecules in the brain of 14-day-old (PND14) Wistar rats after the application of a single propofol dose (25 mg/kg i.p). The structures of interest were the cortex and thalamus as the primary areas of anesthetic actions. Changes of the protein levels of the brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF), their activated receptors tropomyosin-related kinase (TrkA and TrkB) and downstream kinases Akt and the extracellular signal regulated kinase (ERK) were assessed by Western immunoblot analysis at different time points during the first 24 h after the treatment, as well as the expression of cleaved caspase-3 fragment. Fluoro-Jade B staining was used to follow the appearance of degenerating neurons. The obtained results show that the treatment caused marked alterations in levels of the examined neurotrophins, their receptors and downstream effector kinases. However, these changes were not associated with increased neurodegeneration in either the cortex or the thalamus. These results indicate that in the brain of PND14 rats, the interaction between Akt/ERK signaling might be one of important part of endogenous defense mechanisms, which the developing brain utilizes to protect itself from potential anesthesia-induced damage. Elucidation of the underlying molecular mechanisms will improve our understanding of the age-dependent component of anesthesia-induced neurotoxicity
