    Etude diachronique (XIIIe - XVe siècles) de la concordance des temps en espagnol médiéval. Approche explicative

    This thesis provides an explicative case study method to explore the "sequence of tenses" in Old Spanish thanks to the theory of moods developed by Gilles Luquet.We first examined the contributions of grammars and linguists on this syntactic phenomenon. Nevertheless, the ongoing controversy on the existence of a rule – the 2007 "new rule" proposed by Jerzy Kowal –, its contradictions and weaknesses ("violations") are all factors that make it essential to rethink the criteria that determine verbal forms. The collected data, drawn from linguistic databases, were classified according to the traditional terminology of embedded clauses : substantive, adjective and adverbial clauses. We attempted to determine the factors underlying the alternation, the emergence or disappearance of certain verbal forms. Variations – diachronic, diaphasic, diastratic and diatopic variations – were also analyzed. Statistics were used to fix the magnitude of the patterns and to guarantee the reliability of the observed evolutions.This study reveals that the medieval speaker wasn't more governed by a ''sequence of tenses'' based on mechanical congruence than the actual speaker. On the contrary, the speaker himself was able to (in)actualize parts of his speech in function of specific communicative purpose.Cette thèse propose une approche explicative de la « concordance des temps » en espagnol médiéval grâce à l'apport de la théorie des modes développée par Gilles Luquet. Nous avons tout d'abord synthétisé les apports des grammaires et des linguistes sur ce phénomène syntaxique. Mais la controverse toujours en vigueur sur l'existence d'une règle – la « new rule » de Jerzy Kowal date de 2007 –, ses contradictions et ses insuffisances (exemples « non concordants ») sont autant de facteurs qui nécessitent de repenser les paramètres qui conditionnent véritablement le choix des formes verbales. Les exemples, issus de corpus informatisés, ont été classés selon la terminologie traditionnelle entre subordonnées substantives, adjectives et adverbiales. Nous avons cherché à déterminer les raisons qui sous-tendaient l'alternance, l'émergence ou la disparition de certaines formes verbales. L'analyse des variations – diachroniques, diaphasiques, diastratiques et diatopiques – est également au cœur de cette étude. L'approche quantitative a pour objectif de déterminer la représentativité des occurrences et la fiabilité des évolutions constatées.Il ressort de cette étude que le locuteur médiéval n'était pas plus contraint à une « concordance des temps » fondée sur un accord mécanique que le locuteur d'aujourd'hui.     Specific patterns of changes in wheat gene expression after treatment with three antifungal compounds

    The two fungicides azoxystrobin and fenpropimorph are used against powdery mildew and rust diseases in wheat (Triticum aestivumL). Azoxystrobin, a strobilurin, inhibits fungal mitochondrial respiration and fenpropimorph, a morpholin, represses biosynthesis of ergosterol, the major sterol of fungal membranes. Although the fungitoxic activity of these compounds is well understood, their effects on plant metabolism remain unclear. In contrast to the fungicides which directly affect pathogen metabolism, benzo(1,2,3) thiadiazole-7-carbothioic acid S-methylester (BTH) induces resistance against wheat pathogens by the activation of systemic acquired resistance in the host plant. In this study, we monitored gene expression in spring wheat after treatment with each of these agrochemicals in a greenhouse trial using a microarray containing 600 barley cDNA clones. Defence-related genes were strongly induced after treatment with BTH, confirming the activation of a similar set of genes as in dicot plants following salicylic acid treatment. A similar gene expression pattern was observed after treatment with fenpropimorph and some defence-related genes were induced by azoxystrobin, demonstrating that these fungicides also activate a defence reaction. However, less intense responses were triggered than with BTH. The same experiments performed under field conditions gave dramatically different results. No gene showed differential expression after treatment and defence genes were already expressed at a high level before application of the agrochemicals. These differences in the expression patterns between the two environments demonstrate the importance of plant growth conditions for testing the impact of agrochemicals on plant metabolis

    Broad spectrum microarray for fingerprint-based bacterial species identification

    BACKGROUND: Microarrays are powerful tools for DNA-based molecular diagnostics and identification of pathogens. Most target a limited range of organisms and are based on only one or a very few genes for specific identification. Such microarrays are limited to organisms for which specific probes are available, and often have difficulty discriminating closely related taxa. We have developed an alternative broad-spectrum microarray that employs hybridisation fingerprints generated by high-density anonymous markers distributed over the entire genome for identification based on comparison to a reference database. RESULTS: A high-density microarray carrying 95,000 unique 13-mer probes was designed. Optimized methods were developed to deliver reproducible hybridisation patterns that enabled confident discrimination of bacteria at the species, subspecies, and strain levels. High correlation coefficients were achieved between replicates. A sub-selection of 12,071 probes, determined by ANOVA and class prediction analysis, enabled the discrimination of all samples in our panel. Mismatch probe hybridisation was observed but was found to have no effect on the discriminatory capacity of our system. CONCLUSIONS: These results indicate the potential of our genome chip for reliable identification of a wide range of bacterial taxa at the subspecies level without laborious prior sequencing and probe design. With its high resolution capacity, our proof-of-principle chip demonstrates great potential as a tool for molecular diagnostics of broad taxonomic groups

    A rare retrorectal presentation of a bronchogenic cyst: A case report

    AbstractIntroductionBronchogenic cysts are rare abnormalities and a retrorectal presentation is exceptional. Its natural history is not known, but malignant transformation is quite rare. Retrorectal bronchogenic cysts are usually asymptomatic.Presentation of the caseWe present the case of a 36-year-old young man with a past medical history of HIV seropositivity who underwent a procedure to excise a sacral coccyx cyst at another surgical center in February 2009. A histological examination confirmed it was a sacral cyst that was resected in sano. The patient presented with a recurrence of the cyst, and this report describes the combined surgical procedure using a double sacrococcygeal and abdominal approach.DiscussionA complete excision without cyst rupture is recommended to reduce the risk of local recurrence and malignant transformation, as previously reported. Resection can ben performed using multiple approaches depending on the cyst’s locationConclusionHerein, we report the case of a retrorectal bronchogenic cyst in a 36 years old man who was initially treated for a pilonidal cyst. A double surgical approach (abdominal and Kraske) resulted in complete resectioning with no reccurrence

    Intraoperative carcinoid syndrome during small-bowel neuroendocrine tumour surgery

    Only few descriptions of intraoperative carcinoid syndrome (ioCS) have been reported. The primary objective of this study was to describe ioCS. A second aim was to identify risk factors of ioCS. We retrospectively analysed patients operated for small-bowel neuroendocrine tumour in our institution between 2007 and 2015, and receiving our preventive local regimen of octreotide continuous administration. ioCS was defined as highly probable in case of rapid (<5 min) arterial blood pressure changes ≥40%, not explained by surgical/anaesthetic management and regressive ≥20% after octreotide bolus injection. Probable cases were ioCS which did not meet all criteria of highly-probable ioCS. Suspected ioCS were detected on the anaesthesia record by an injection of octreotide due to a manifestation which did not meet the criteria for highly-probable or probable ioCS. A total of 81 patients (liver metastases: 59, prior carcinoid syndrome: 49, carcinoid heart disease: 7) were included; 139 ioCS occurred in 45 patients: 45 highly probable, 67 probable and 27 suspected. ioCs was hypertensive (91%) and/or hypotensive (29%). There was no factor, including the use of vasopressors, significantly associated with the occurrence of an ioCS. All surgeries were completed and one patient died from cardiac failure 4 days after surgery. After preoperative octreotide continuous infusion, ioCS were mainly hypertensive. No ioCS risk factors, including vasopressor use, were identified. No intraoperative carcinoid crisis occurred, suggesting the clinical relevance of a standardized octreotide prophylaxis protocol

    A composite transcriptional signature differentiates responses towards closely related herbicides in Arabidopsis thaliana and Brassica napus

    In this study, genome-wide expression profiling based on Affymetrix ATH1 arrays was used to identify discriminating responses of Arabidopsis thaliana to five herbicides, which contain active ingredients targeting two different branches of amino acid biosynthesis. One herbicide contained glyphosate, which targets 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS), while the other four herbicides contain different acetolactate synthase (ALS) inhibiting compounds. In contrast to the herbicide containing glyphosate, which affected only a few transcripts, many effects of the ALS inhibiting herbicides were revealed based on transcriptional changes related to ribosome biogenesis and translation, secondary metabolism, cell wall modification and growth. The expression pattern of a set of 101 genes provided a specific, composite signature that was distinct from other major stress responses and differentiated among herbicides targeting the same enzyme (ALS) or containing the same chemical class of active ingredient (sulfonylurea). A set of homologous genes could be identified in Brassica napus that exhibited a similar expression pattern and correctly distinguished exposure to the five herbicides. Our results show the ability of a limited number of genes to classify and differentiate responses to closely related herbicides in A. thaliana and B. napus and the transferability of a complex transcriptional signature across species

Continental-scale acoustic telemetry networks are increasingly being established worldwide, producing large volumes of information-rich geospatial data. During the last decade, the Integrated Marine Observing System’s Animal Tracking Facility (IMOS ATF) established a permanent array of acoustic receivers around Australia. Simultaneously, IMOS developed a centralised national database to foster collaborative research across the user community and quantify individual behaviour across a broad range of taxa. Here we present the database and quality control procedures developed to collate 49.6 million valid detections from 1891 receiving stations. This dataset consists of detections for 3,777 tags deployed on 117 marine species, with distances travelled ranging from a few to thousands of kilometres. Connectivity between regions was only made possible by the joint contribution of IMOS infrastructure and researcher-funded receivers. This dataset constitutes a valuable resource facilitating meta-analysis of animal movement, distributions, and habitat use, and is important for relating species distribution shifts with environmental covariates

    Observations and models to support the first Marine Ecosystem Assessment for the Southern Ocean (MEASO)

    Assessments of the status and trends of habitats, species and ecosystems are needed for effective ecosystem-based management in marine ecosystems. Knowledge on imminent ecosystem changes (climate change impacts) set in train by existing climate forcings are needed for adapting management practices to achieve conservation and sustainabililty targets into the future. Here, we describe a process for enabling a marine ecosystem assessment (MEA) by the broader scientific community to support managers in this way, using a MEA for the Southern Ocean (MEASO) as an example. We develop a framework and undertake an audit to support a MEASO, involving three parts. First, we review available syntheses and assessments of the Southern Ocean ecosystem and its parts, paying special attention to building on the SCAR Antarctic Climate Change and Environment report and the SCAR Biogeographic Atlas of the Southern Ocean. Second, we audit available field observations of habitats and densities and/or abundances of taxa, using the literature as well as a survey of scientists as to their current and recent activities. Third, we audit available system models that can form a nested ensemble for making, with available data, circumpolar assessments of habitats, species and food webs. We conclude that there is sufficient data and models to undertake, at least, a circumpolar assessment of the krill-based system. The auditing framework provides the basis for the first MEASO but also provides a repository (www.SOKI.aq/display/MEASO) for easily amending the audit for future MEASOs. We note that an important outcome of the first MEASO will not only be the assessment but also to advise on priorities in observations and models for improving subsequent MEASOs
