    A projection method for general form linear least-squares problems

    One of the possible approaches for the solution of underdetermined linear least-squares problems in general form, for a chosen regularization operator L, projects the problem in the null space of L and in its orthogonal complement. In this paper, we show that the projected problem cannot be solved by the generalized singular value decomposition, and propose some approaches to overcome this issue. Numerical experiments ascertain the stability of the new procedures

    Using Artificial Intelligence for COVID-19 Detection in Blood Exams: A Comparative Analysis

    COVID-19 is an infectious disease that was declared a pandemic by the World Health Organization (WHO) in early March 2020. Since its early development, it has challenged health systems around the world. Although more than 12 billion vaccines have been administered, at the time of writing, it has more than 623 million confirmed cases and more than 6 million deaths reported to the WHO. These numbers continue to grow, soliciting further research efforts to reduce the impacts of such a pandemic. In particular, artificial intelligence techniques have shown great potential in supporting the early diagnosis, detection, and monitoring of COVID-19 infections from disparate data sources. In this work, we aim to make a contribution to this field by analyzing a high-dimensional dataset containing blood sample data from over forty thousand individuals recognized as infected or not with COVID-19. Encompassing a wide range of methods, including traditional machine learning algorithms, dimensionality reduction techniques, and deep learning strategies, our analysis investigates the performance of different classification models, showing that accurate detection of blood infections can be obtained. In particular, an F-score of 84% was achieved by the artificial neural network model we designed for this task, with a rate of 87% correct predictions on the positive class. Furthermore, our study shows that the dimensionality of the original data, i.e. the number of features involved, can be significantly reduced to gain efficiency without compromising the final prediction performance. These results pave the way for further research in this field, confirming that artificial intelligence techniques may play an important role in supporting medical decision-making

    Ascertaining the Ideality of Photometric Stereo Datasets under Unknown Lighting

    The standard photometric stereo model makes several assumptions that are rarely verified in experimental datasets. In particular, the observed object should behave as a Lambertian reflector, and the light sources should be positioned at an infinite distance from it, along a known direction. Even when Lambert’s law is approximately fulfilled, an accurate assessment of the relative position between the light source and the target is often unavailable in real situations. The Hayakawa procedure is a computational method for estimating such information directly from data images. It occasionally breaks down when some of the available images excessively deviate from ideality. This is generally due to observing a non-Lambertian surface, or illuminating it from a close distance, or both. Indeed, in narrow shooting scenarios, typical, e.g., of archaeological excavation sites, it is impossible to position a flashlight at a sufficient distance from the observed surface. It is then necessary to understand if a given dataset is reliable and which images should be selected to better reconstruct the target. In this paper, we propose some algorithms to perform this task and explore their effectiveness

    Circulating fatty acids and endocannabinoidome-related mediator profiles associated to human longevity

    To evaluate whether a peculiar plasma profile of fatty acids and endocannabinoidome (eCBome)-related mediators may be associated to longevity, we assessed them in octogenarians (Old; n=42) living in the east-central mountain area of Sardinia, a High-Longevity Zone (HLZ), compared to sexagenarian (Young; n=21) subjects from the same area, and to Olds (n=22) from the Northern Sardinia indicated as Lower-Longevity Zone (LLZ). We found significant increases in conjugated linoleic acid (CLA) and heptadecanoic acid (17:0) levels in Old-HLZ with respect to younger subjects and Old-LLZ subjects. Young-HLZ subjects exhibited higher circulating levels of pentadecanoic acid (15:0) and retinol. Palmitoleic acid (POA) was elevated in both Young and Old subjects from the HLZ. eCBome profile showed a significantly increased plasma level of the two endocannabinoids, N-arachidonoyl-ethanolamine (AEA) and 2-arachidonoyl-glycerol (2-AG) in Old-HLZ subjects compared to Young-HLZ and Old-LLZ respectively. In addition, we found increased N-oleoyl-ethanolamine (OEA), 2-linoleoyl-glycerol (2-LG) and 2-oleoyl-glycerol (2-OG) levels in Old-HLZ group with respect to Young-HLZ (as for OEA an d 2-LG) and both the Old-LLZ and Young-HLZ for 2-OG. The endogenous metabolite of docosahexaenoic acid (DHA), N-docosahexaenoyl-ethanolamine (DHEA) was significantly increased in Old-HLZ subjects. In conclusion, our results suggest that in the HLZ area, Young and Old subjects exhibited a favourable, albeit distinctive, fatty acids and eCBome profile that may be indicative of a metabolic pattern potentially protective from adverse chronic conditions. These factors could point to a suitable physiological metabolic pattern that may counteract the adverse stimuli leading to age-related disorders such as neurodegenerative and metabolic diseases

    Daily Regulation of Key Metabolic Pathways in Two Seagrasses Under Natural Light Conditions

    The circadian clock is an endogenous time-keeping mechanism that enables organisms to adapt to external environmental cycles. It produces rhythms of plant metabolism and physiology, and interacts with signaling pathways controlling daily and seasonal environmental responses through gene expression regulation. Downstream metabolic outputs, such as photosynthesis and sugar metabolism, besides being affected by the clock, can also contribute to the circadian timing itself. In marine plants, studies of circadian rhythms are still way behind in respect to terrestrial species, which strongly limits the understanding of how they coordinate their physiology and energetic metabolism with environmental signals at sea. Here, we provided a first description of daily timing of key core clock components and clock output pathways in two seagrass species, Cymodocea nodosa and Zostera marina (order Alismatales), co-occurring at the same geographic location, thus exposed to identical natural variations in photoperiod. Large differences were observed between species in the daily timing of accumulation of transcripts related to key metabolic pathways, such as photosynthesis and sucrose synthesis/transport, highlighting the importance of intrinsic biological, and likely ecological attributes of the species in determining the periodicity of functions. The two species exhibited a differential sensitivity to light-to-dark and dark-to-light transition times and could adopt different growth timing based on a differential strategy of resource allocation and mobilization throughout the day, possibly coordinated by the circadian clock. This behavior could potentially derive from divergent evolutionary adaptations of the species to their bio-geographical range of distributions

    Direct-acting antivirals used in HCV-related liver disease do not affect thyroid function and autoimmunity

    Purpose It is well known that interferon-alpha (IFN-alpha), used for long time as the main therapy for HCV-related disease, induces thyroid alterations, but the impact of the new direct-acting antivirals (DAAs) on thyroid is not established. Aim of this prospective study was to evaluate if DAAs therapy may induce thyroid alterations.Methods A total of 113 HCV patients, subdivided at the time of the enrollment in naive group (n = 64) and in IFN-alpha group (n = 49) previously treated with pegylated interferon-alpha and ribavirin, were evaluated for thyroid function and autoimmunity before and after 20-32 weeks of DAAs.Results Before starting DAAs, a total of 8/113 (7.1%) patients showed Hashimoto's thyroiditis (HT) all belonging to IFN-alpha group (8/49, 16.3%), while no HT cases were found in the naive group. Overall, 7/113 (6.2%) patients were hypothyroid: 3/64 (4.7%) belonging to naive group and 4/49 (8.2%) to IFN-alpha group. Furthermore, a total of 8/113 patients (7.1%) showed subclinical hyperthyroidism: 2/64 (3.1%) were from naive group and 6/49 (12.2%) from IFN-alpha group. Interestingly, after DAAs therapy, no new cases of HT, hypothyroidism and hyperthyroidism was found in all series, while 6/11 (54.5%) patients with non-autoimmune subclinical thyroid dysfunction became euthyroid. Finally, the only association between viral genotypes and thyroid alterations was genotype 1 and hypothyroidism.Conclusions This study supports evidence that DAAs have a limited or missing influence on thyroid in patients with HCV-related diseases. Moreover, it provides preliminary evidence that subclinical non-autoimmune thyroid dysfunction may improve after HCV infection resolution obtained by DAAs

    Emerging therapies in pheochromocytoma and paraganglioma: Immune checkpoint inhibitors in the starting blocks

    Pheochromocytoma and paraganglioma are neuroendocrine neoplasms, originating in the adrenal medulla and in parasympathetic and sympathetic autonomic nervous system ganglia, respec-tively. They usually present as localized tumours curable with surgery. However, these tumours may exhibit heterogeneous clinical course, ranging from no/minimal progression to aggressive (progres-sive/metastatic) behavior. For this setting of patients, current therapies are unsatisfactory. Immune checkpoint inhibitors have shown outstanding results for several types of solid cancers. We therefore aimed to summarize and discuss available data on efficacy and safety of current FDA-approved immune checkpoint inhibitors in patients with pheochromocytoma and paraganglioma. After an extensive search, we found 15 useful data sources (four full-published articles, four supplements of scientific journals, seven ongoing registered clinical trials). The data we detected, even with the limit of the small number of patients treated, make a great expectation on the therapeutic use of immune checkpoint inhibitors. Besides, the newly detected predictors of response will (hopefully) be of great helps in selecting the subset of patients that might benefit the most from this class of drugs. Finally, new trials are in the starting blocks, and they are expected to shed in the next future new light on a therapy, which is considered a milestone in oncology
