    Maize Nitrogen Use Efficiency: QTL Mapping in a U.S. Dent x Argentine-Caribbean Flint RILs population

    This study was aimed to identify quantitative trait loci (QTL) for nitrogen use efficiency(NUE) and related traits in a maize population derived from a cross between two lines with different genetic background (B100 and LP2). Recombinant inbred lines (181) from this population were evaluated under fieldconditions during two growing seasons, and significant(P < 0.01) phenotypic and genotypic variability was detected for most evaluated traits. Two different mapping methods were applied for detecting QTLs. Firstly, a trait by trait approach was performed on across environments, and 19 QTLs were identified.Secondly, a multi-trait multi-environment analysis detected seven joint QTLs. Almost all joint QTLs had inconsistent additive effects from one environment to another, which would reflectpresence of QTL × Environment interaction. Most joint QTLs co-localized with QTLs detected by indi-vidual mapping. We detected consistent additive effects for grain yield per plant and NUE, as well as for biomass and nitrogen harvest index in some joint QTLs, especially QTL-1 and QTL-6. These QTLs had positive and stable effects across environments, and presence of some genes within these QTL intervals could be relevant for selecting for both NUE and grain yield simultaneously. Up today, this is a firstreport on the co-localization of QTLs for enhan-ced allocation of biomass allocation to grains with NUE, and NUE candidate gene identificaion. Fine mapping of these regions could allow to detect additional markers more closely linked to these QTLs that could be used for marker assisted selection for NUE

    Efecto del control de alquileres en el área metropolitana de Cataluña

    Los elevados precios de las viviendas en alquiler es un debate muy habitual los últimos años puesto que conlleva a diversas consecuencias negativas como es la difícil emancipación de los jóvenes que existe en la actualidad. En 2020, en Cataluña se aprobó una regulación al respecto, limitando los precios del alquiler en aquellos municipios que habían sufrido mayores incrementos de los mismos. Aun con la interposición del control de los precios de los alquileres el debate sobre su efecto sigue siendo habitual tanto a nivel político como social, especialmente respecto a su efectividad para contener los precios, y sobre su efecto en la oferta, pudiendo generar una reducción significativa sobre el número de contratos de alquiler formalizados en el mercado. El presente estudio pretende precisamente analizar que impacto ha tenido la reciente regulación del precio del alquiler en Catalunya, tanto en los precios pagados por los consumidores, como en el número de contratos firmados. Para ello, hemos hecho uso del estimador en diferencias, donde los municipios bajo la regulación de precios son nuestro grupo afectado, y el resto de municipios de Catalunya son nuestro grupo de control. Los resultados obtenidos indican que la regulación de precios habría sido efectiva para contener los precios del alquiler, ya que ha causado una pequeña disminución en los precios, y que no ha causado efectos negativos en el número de contratos firmados, que no se habrían visto afectados por la regulación. Además, podemos observar como la disminución de precios se produce desde el primer trimestre de aplicación de la regulación, y es especialmente relevante en los municipios más pequeños, disminuyendo su efecto a medida que aumenta la población del municipio. Asimismo, hemos analizado si ha causado efectos de desbordamiento en los municipios limítrofes. Nuestros resultados muestran que los efectos de la regulación se circunscriben a los municipios afectados, y que no generarían un efecto significativo sobre los municipios limítrofes, descartando así la existencia de efectos desbordamiento

    Kernel weight in maize: genetic control of its physiological and compositional determinants in a dent × flint-caribbean RIL population

    The genetic control of maize kernel weight (KW) determination could be studied through its physiological and/ or compositional determinants. Our objective was to dissect the genetic control of maize KW by analyzing its physiological (KGR: kernel growth rate; KFD: kernel filling duration) and compositional (protein, oil, starch) determinants in a dent×flint Caribbean RIL population, which combines a broad genetic background with grains of high added value for industry. An additional objective was to determine the stability of the genetic control under contrasting growing conditions, for which soil nitrogen offer was modified across experiments. Heritability (H2) values were high for KW (H2 = 0.74) and intermediate for the other traits (from 0.62 to 0.42). Kernel weight had a strong correlation with KFD (r = 0.69), KGR (r = 0.60) and protein concentration (r = 0.56). Ten joint QTL with inconsistent effects across years and seven epistatic interactions were detected. Despite changes in effect size, most QTL were significant under both environments. Nine QTL were associated with variations in potential KW (KW ), mean KW, KGR and oil concentration, eight with variations in protein and starch concentration and seven with KFD. Epistatic interactions were related to regions with significant main effects. The most important finding was the existence of a common QTL for KW , KGR and KFD on chromosome 5, for which there was no previous report. Results increased our knowledge on the genetic control of KW through its phenotypic and genetic correlation with KFD, confirming the need to explore different physiological strategies in different genetic backgrounds

    Cell wall components and pectin esterification levels as markers of proliferation and differentiation events during pollen development and pollen embryogenesis in Capsicum annuum L.

    Plant cell walls and their polymers are regulated during plant development, but the specific roles of their molecular components are still unclear, as well as the functional meaning of wall changes in different cell types and processes. In this work the in situ analysis of the distribution of different cell wall components was performed during two developmental programmes, gametophytic pollen development, which is a differentiation process, and stress-induced pollen embryogenesis, which involves proliferation followed by differentiation processes. The changes in cell wall polymers were compared with a system of plant cell proliferation and differentiation, the root apical meristem. The analysis was also carried out during the first stages of zygotic embryogenesis. Specific antibodies recognizing the major cell wall polymers, xyloglucan (XG) and the rhamnogalacturonan II (RGII) pectin domain, and antibodies against high- and low-methyl-esterified pectins were used for both dot-blot and immunolocalization with light and electron microscopy. The results showed differences in the distribution pattern of these molecular complexes, as well as in the proportion of esterified and non-esterified pectins in the two pollen developmental pathways. Highly esterified pectins were characteristics of proliferation, whereas high levels of the non-esterified pectins, XG and RGII were abundant in walls of differentiating cells. Distribution patterns similar to those of pollen embryos were found in zygotic embryos. The wall changes reported are characteristic of proliferation and differentiation events as markers of these processes that take place during pollen development and embryogenesis

    The Radially Swollen 4 Separase Mutation of Arabidopsis thaliana Blocks Chromosome Disjunction and Disrupts the Radial Microtubule System in Meiocytes

    The caspase-family protease, separase, is required at the onset of anaphase to cleave the cohesin complex that joins replicated sister chromatids. However, in various eukaryotes, separase has acquired additional and distinct functions. A single amino-acid substitution in separase is responsible for phenotypes of the Arabidopsis thaliana mutant, radially swollen 4 (rsw4). This is a conditional mutant, resembling the wild type at the permissive temperature (∼20°C) and expressing mutant phenotypes at the restrictive temperature (∼30°C). Root cells in rsw4 at the restrictive temperature undergo non-disjunction and other indications of the loss of separase function. To determine to what extent separase activity remains at 30°C, we examined the effect of the mutation on meiosis, where the effects of loss of separase activity through RNA interference are known; and in addition, we examined female gametophyte development. Here, we report that, at the restrictive temperature, replicated chromosomes in rsw4 meiocytes typically fail to disjoin and the cohesin complex remains at centromeres after metaphase. Meiotic spindles appear normal in rsw4 male meiocytes; however the mutation disrupts the radial microtubule system, which is replaced by asymmetric arrays. Surprisingly, female gametophyte development was relatively insensitive to loss of separase activity, through either rsw4 or RNAi. These effects confirm that phenotypes in rsw4 result from loss of separase activity and establish a role for separase in regulating cell polarization following male meiosis

    Effectiveness of an mHealth intervention combining a smartphone app and smart band on body composition in an overweight and obese population: Randomized controlled trial (EVIDENT 3 study)

    Background: Mobile health (mHealth) is currently among the supporting elements that may contribute to an improvement in health markers by helping people adopt healthier lifestyles. mHealth interventions have been widely reported to achieve greater weight loss than other approaches, but their effect on body composition remains unclear. Objective: This study aimed to assess the short-term (3 months) effectiveness of a mobile app and a smart band for losing weight and changing body composition in sedentary Spanish adults who are overweight or obese. Methods: A randomized controlled, multicenter clinical trial was conducted involving the participation of 440 subjects from primary care centers, with 231 subjects in the intervention group (IG; counselling with smartphone app and smart band) and 209 in the control group (CG; counselling only). Both groups were counselled about healthy diet and physical activity. For the 3-month intervention period, the IG was trained to use a smartphone app that involved self-monitoring and tailored feedback, as well as a smart band that recorded daily physical activity (Mi Band 2, Xiaomi). Body composition was measured using the InBody 230 bioimpedance device (InBody Co., Ltd), and physical activity was measured using the International Physical Activity Questionnaire. Results: The mHealth intervention produced a greater loss of body weight (–1.97 kg, 95% CI –2.39 to –1.54) relative to standard counselling at 3 months (–1.13 kg, 95% CI –1.56 to –0.69). Comparing groups, the IG achieved a weight loss of 0.84 kg more than the CG at 3 months. The IG showed a decrease in body fat mass (BFM; –1.84 kg, 95% CI –2.48 to –1.20), percentage of body fat (PBF; –1.22%, 95% CI –1.82% to 0.62%), and BMI (–0.77 kg/m2, 95% CI –0.96 to 0.57). No significant changes were observed in any of these parameters in men; among women, there was a significant decrease in BMI in the IG compared with the CG. When subjects were grouped according to baseline BMI, the overweight group experienced a change in BFM of –1.18 kg (95% CI –2.30 to –0.06) and BMI of –0.47 kg/m2 (95% CI –0.80 to –0.13), whereas the obese group only experienced a change in BMI of –0.53 kg/m2 (95% CI –0.86 to –0.19). When the data were analyzed according to physical activity, the moderate-vigorous physical activity group showed significant changes in BFM of –1.03 kg (95% CI –1.74 to –0.33), PBF of –0.76% (95% CI –1.32% to –0.20%), and BMI of –0.5 kg/m2 (95% CI –0.83 to –0.19). Conclusions: The results from this multicenter, randomized controlled clinical trial study show that compared with standard counselling alone, adding a self-reported app and a smart band obtained beneficial results in terms of weight loss and a reduction in BFM and PBF in female subjects with a BMI less than 30 kg/m2 and a moderate-vigorous physical activity level. Nevertheless, further studies are needed to ensure that this profile benefits more than others from this intervention and to investigate modifications of this intervention to achieve a global effect

    Circulating microRNAs in sera correlate with soluble biomarkers of immune activation but do not predict mortality in ART treated individuals with HIV-1 infection: A case control study

    Introduction: The use of anti-retroviral therapy (ART) has dramatically reduced HIV-1 associated morbidity and mortality. However, HIV-1 infected individuals have increased rates of morbidity and mortality compared to the non-HIV-1 infected population and this appears to be related to end-organ diseases collectively referred to as Serious Non-AIDS Events (SNAEs). Circulating miRNAs are reported as promising biomarkers for a number of human disease conditions including those that constitute SNAEs. Our study sought to investigate the potential of selected miRNAs in predicting mortality in HIV-1 infected ART treated individuals. Materials and Methods: A set of miRNAs was chosen based on published associations with human disease conditions that constitute SNAEs. This case: control study compared 126 cases (individuals who died whilst on therapy), and 247 matched controls (individuals who remained alive). Cases and controls were ART treated participants of two pivotal HIV-1 trials. The relative abundance of each miRNA in serum was measured, by RTqPCR. Associations with mortality (all-cause, cardiovascular and malignancy) were assessed by logistic regression analysis. Correlations between miRNAs and CD4+ T cell count, hs-CRP, IL-6 and D-dimer were also assessed. Results: None of the selected miRNAs was associated with all-cause, cardiovascular or malignancy mortality. The levels of three miRNAs (miRs -21, -122 and -200a) correlated with IL-6 while miR-21 also correlated with D-dimer. Additionally, the abundance of miRs -31, -150 and -223, correlated with baseline CD4+ T cell count while the same three miRNAs plus miR- 145 correlated with nadir CD4+ T cell count. Discussion: No associations with mortality were found with any circulating miRNA studied. These results cast doubt onto the effectiveness of circulating miRNA as early predictors of mortality or the major underlying diseases that contribute to mortality in participants treated for HIV-1 infection
