    Melanogaster coccolobae sp. nov. (Paxillaceae, Boletales), un hongo hipogeo tropical de las áreas urbanas de Quintana Roo, México

    Background and Aims: The genus Melanogaster is characterized by its hypogeous to semi hypogeous habit, brownish basidiomata, gel-filled gleba locules, and globose to ellipsoid basidiospores. The genus is distributed in temperate zones, but sequences from Coccoloba root tips and a few basidiome collections have revealed its presence in the tropics. The aim of this article is to describe a new species of Melanogaster based on ecological, molecular, and morphological data. Methods: Specimens were collected in urban vegetation of Quintana Roo in the Yucatán Peninsula, Mexico. For morphological description, the classic protocols for sequestrate fungi were followed. The dried material was deposited in the mycological herbarium “José Castillo Tovar” of the Instituto Tecnológico de Ciudad Victoria (ITCV) and the herbarium of the Universidad Autónoma de Yucatán (UADY). Key results: Melanogaster coccolobae is presented as a new species from the urban gardens of Quintana Roo based on ecological, molecular, and morphological evidence. This species is characterized by its hypogeous to semi hypogeous basidioma, greyish orange, brown to reddish brown peridium composed of two layers, sweet smell, subglobose, ellipsoid or piriform basidiospores, and by its mycorrhizal association with Coccoloba spicata. Conclusions: Melanogaster coccolobae is the first species described from the Mexican Caribbean from urban gardens with Coccoloba spicata. More studies about the tropical sequestrate fungi are recommended.Antecedentes y Objetivos: El género Melanogaster se caracteriza por su hábito hipogeo a semi hipogeo, basidiomas parduscos, gleba con lóculos llenos de gel y basidiosporas globosas a elipsoides. El género se distribuye en zonas templadas, pero secuencias de ectomicorrizas de Coccoloba y pocas colecciones de basidiomas han revelado su presencia en los trópicos. El objetivo de este artículo es describir una nueva especie de Melanogaster a partir de datos ecológicos, moleculares y morfológicos. Métodos: Los especímenes fueron recolectados en jardines urbanos de Quintana Roo en la Península de Yucatán, México. Para la descripción morfológica se siguieron los protocolos clásicos para hongos secuestrados. El material se depositó en el herbario micológico “José Castillo Tovar” del Instituto Tecnológico de Ciudad Victoria (ITCV) y en el herbario de la Universidad Autónoma de Yucatán (UADY). Resultados clave: Melanogaster coccolobae se presenta como una nueva especie de los jardines urbanos de Quintana Roo con base en evidencia morfológica, ecológica y molecular. Esta especie se caracteriza por sus basidiomas hipogeos a semi hipogeos, peridio naranja grisáceo, marrón o marrón rojizo, compuesto por dos capas, olor dulce, basidiosporas subglobosas, elipsoides o piriformes y por formar asociación micorrízica con Coccoloba spicata. Conclusiones: Melanogaster coccolobae es la primera especie descrita del Caribe mexicano en jardines urbanos con Coccoloba spicata. Se recomiendan más estudios sobre los hongos secuestrados tropicales

    Substrate-induced dimerization of engineered monomeric variants of triosephosphate isomerase from Trichomonas vaginalis

    "The dimeric nature of triosephosphate isomerases (TIMs) is maintained by an extensive surface area interface of more than 1600 angstrom 2. TIMs from Trichomonas vaginalis (TvTIM) are held in their dimeric state by two mechanisms: a ball and socket interaction of residue 45 of one subunit that fits into the hydrophobic pocket of the complementary subunit and by swapping of loop 3 between subunits. TvTIMs differ from other TIMs in their unfolding energetics. In TvTIMs the energy necessary to unfold a monomer is greater than the energy necessary to dissociate the dimer. Herein we found that the character of residue I45 controls the dimer-monomer equilibrium in TvTIMs. Unfolding experiments employing monomeric and dimeric mutants led us to conclude that dimeric TvTIMs unfold following a four state model denaturation process whereas monomeric TvTIMs follow a three state model. In contrast to other monomeric TIMs, monomeric variants of TvTIM1 are stable and unexpectedly one of them (I45A) is only 29-fold less active than wild-type TvTIM1. The high enzymatic activity of monomeric TvTIMs contrast with the marginal catalytic activity of diverse monomeric TIMs variants. The stability of the monomeric variants of TvTIM1 and the use of cross-linking and analytical ultracentrifugation experiments permit us to understand the differences between the catalytic activities of TvTIMs and other marginally active monomeric TIMs. As TvTIMs do not unfold upon dimer dissociation, herein we found that the high enzymatic activity of monomeric TvTIM variants is explained by the formation of catalytic dimeric competent species assisted by substrate binding.

    Gastric emptying of low- and high-caloric liquid meals measured using ultrasonography in healthy volunteers

    Purpose Delayed gastric emptying is present in patients with functional dyspepsia (FD), diabetes mellitus, and neurological diseases. Diet may affect gastric emptying symptoms in patients with FD. We sought to determine the extent to which gastric emptying and symptoms of dyspepsia are influenced by caloric content in healthy subjects using ultrasonography. Materials and Methods 32 healthy volunteers were given 2 meals with different caloric content in random order. Gastric emptying was determined using ultrasonography to measure antral area when fasting, and postprandially at intervals of 0, 10, 20, and 30 min. Dyspeptic symptoms including discomfort, nausea, and fullness were graded. Results The antral area following a high-caloric meal compared to a low-caloric meal was significantly increased at 0, 10, 20, and 30 min (P=0.0203,<0.0001<0.0001,<0.0001, respectively), as was the median fullness (P<0.0048, 0.0001, 0.0009, 0.0001, respectively) measured at the same time points. There was a weak correlation (r2=0.1, P<0.0001) between the antral area and subjective fullness. No differences between gastric emptying in males and females were found. Conclusion The caloric content of a meal influences gastric emptying. Using ultrasonography to measure the antral area helps us to assess gastric emptying and therefore to assess patients with functional dyspepsia.publishedVersio

    Usual Dietary Intake, Nutritional Adequacy and Food Sources of Calcium, Phosphorus, Magnesium and Vitamin D of Spanish Children Aged One to <10 Years. Findings from the EsNuPI Study

    Bone problems in the population begin to be establish in childhood. The present study aims to assess the usual calcium, phosphorus, magnesium, and vitamin D intakes, along with the food sources of these nutrients, in Spanish children participating in the EsNuPI (Estudio Nutricional en Población Infantil Española) study. Two 24 h dietary recalls were applied to 1448 children (1 to <10 years) divided into two sub-samples: one reference sample (RS) of the general population [n = 707] and another sample which exclusively included children consuming enriched or fortified milks, here called “adapted milks” (AMS) [n = 741]. Estimation of the usual intake shows that nutrient intake increased with age for all nutrients except vitamin D. Using as reference the Dietary Reference Values from the European Food Safety Authority (EFSA), calcium and magnesium intakes were found to be below the average requirement (AR) and adequate intake (AI), respectively, in a considerable percentage of children. Furthermore, phosphorus exceeded the AI in 100% of individuals and vitamin D was lower than the AI in almost all children studied. The results were very similar when considering only plausible reporters. When analyzing the food sources of the nutrients studied, milk and dairy products contributed the most to calcium, phosphorus, magnesium, and vitamin D. Other sources of calcium were cereals and vegetables; for phosphorus: meat, meat products, and cereals; for magnesium: cereals and fruits; and, for vitamin D: fish and eggs. These results highlight the desirability of improving the intake concerning these nutrients, which are involved in bone and metabolic health in children. The AMS group appeared to contribute better to the adequacy of those nutrients than the RS group, but both still need further improvement.     The green chemistry in the chemical analysis area consists of the design of qualitative or quantitative techniques that make a minimum generation of residues and have a validity in the results comparable to traditional methods. The microscale chemistry has had a relevant role in recent years in order to modify and implement new methodologies for the quantification of chemical analytes. In this project, a technique for quantifying the alcoholic content of tequila samples was developed based on the chemical principles of alcohol oxidation, which is a colorimetric technique that allows identifying the presence of an alcohol. The objective of the project was to apply the color change of the ethanol oxidation and to quantify it´s content based on the intensity of the blue color generated in the oxidation, using a low-cost photocolorimeter. The results were comparable with the obtained with a spectrophotometer, it was validated by a statistical method to know their reliability and finally, a correlation was made so that the same results were obtained with the proposed technique as with the commercial equipment.La química verde abordada en el área de los análisis químicos consiste en el diseño de técnicas cualitativas o cuantitativas que tengan una mínima generación de residuos y que permitan tener una validez en lo resultados comparable con métodos tradicionales. En este sentido, la química a microescala ha tenido un papel relevante en años recientes para poder modificar e implementar nuevas metodologías para la cuantificación de analitos químicos. En el presente proyecto se desarrolló una técnica de cuantificación del grado alcohólico de muestras de tequila basada en los principios químicos de la oxidación de alcoholes, la cual es una técnica colorimétrica que permite identificar la presencia de un alcohol. El objetivo del trabajo fue aprovechar el cambio de color de la oxidación del etanol y cuantificar el contenido alcohólico con base en la intensidad del color azul generado en la oxidación, mismo que se cuantificó con un fotocolorímetro a bajo costo y que mostró resultados comparables con los obtenidos con un espectrofotómetro, además que se realizó una validación estadística de los resultados para conocer la confiabilidad de los mismos y finalmente, se hizo una correlación para que con la técnica propuesta se obtuvieran los mismos resultados que con el equipo comercial

    One-year cardiovascular outcomes after coronavirus disease 2019: The cardiovascular COVID-19 registry

    Background: The long-term cardiovascular (CV) outcomes of COVID-19 have not been fully explored. Methods: This was an international, multicenter, retrospective cohort study conducted between February and December 2020. Consecutive patients ?18 years who underwent a real-time reverse transcriptase-polymerase chain reaction (RT-PCR) for SARS-CoV2 were included. Patients were classified into two cohorts depending on the nasopharyngeal swab result and clinical status: confirmed COVID-19 (positive RT-PCR) and control (without suggestive symptoms and negative RT-PCR). Data were obtained from electronic records, and clinical follow-up was performed at 1-year. The primary outcome was CV death at 1-year. Secondary outcomes included arterial thrombotic events (ATE), venous thromboembolism (VTE), and serious cardiac arrhythmias. An independent clinical event committee adjudicated events. A Cox proportional hazards model adjusted for all baseline characteristics was used for comparing outcomes between groups. A prespecified landmark analysis was performed to assess events during the post-acute phase (31-365 days). Results: A total of 4,427 patients were included: 3,578 (80.8%) in the COVID-19 and 849 (19.2%) control cohorts. At one year, there were no significant differences in the primary endpoint of CV death between the COVID-19 and control cohorts (1.4% vs. 0.8%; HRadj 1.28 [0.56-2.91]; p = 0.555), but there was a higher risk of all-cause death (17.8% vs. 4.0%; HRadj 2.82 [1.99-4.0]; p = 0.001). COVID-19 cohort had higher rates of ATE (2.5% vs. 0.8%, HRadj 2.26 [1.02-4.99]; p = 0.044), VTE (3.7% vs. 0.4%, HRadj 9.33 [2.93-29.70]; p = 0.001), and serious cardiac arrhythmias (2.5% vs. 0.6%, HRadj 3.37 [1.35-8.46]; p = 0.010). During the post-acute phase, there were no significant differences in CV death (0.6% vs. 0.7%; HRadj 0.67 [0.25-1.80]; p = 0.425), but there was a higher risk of deep vein thrombosis (0.6% vs. 0.0%; p = 0.028). Re-hospitalization rate was lower in the COVID-19 cohort compared to the control cohort (13.9% vs. 20.6%; p = 0.001). Conclusions: At 1-year, patients with COVID-19 experienced an increased risk of all-cause death and adverse CV events, including ATE, VTE, and serious cardiac arrhythmias, but not CV death

    Long-term effects of coronavirus disease 2019 on the cardiovascular system, CV COVID registry: A structured summary of a study protocol

    Background: Patients presenting with the coronavirus-2019 disease (COVID-19) may have a high risk of cardiovascular adverse events, including death from cardiovascular causes. The long-term cardiovascular outcomes of these patients are entirely unknown. We aim to perform a registry of patients who have undergone a diagnostic nasopharyngeal swab for SARS-CoV-2 and to determine their long-term cardiovascular outcomes. Study and design: This is a multicenter, observational, retrospective registry to be conducted at 17 centers in Spain and Italy (ClinicalTrials.gov number: NCT04359927). Consecutive patients older than 18 years, who underwent a real-time reverse transcriptase-polymerase chain reaction (RT-PCR) for SARS-CoV2 in the participating institutions, will be included since March 2020, to August 2020. Patients will be classified into two groups, according to the results of the RT-PCR: COVID-19 positive or negative. The primary outcome will be cardiovascular mortality at 1 year. The secondary outcomes will be acute myocardial infarction, stroke, heart failure hospitalization, pulmonary embolism, and serious cardiac arrhythmias, at 1 year. Outcomes will be compared between the two groups. Events will be adjudicated by an independent clinical event committee. Conclusion: The results of this registry will contribute to a better understanding of the long-term cardiovascular implications of the COVID19

    Impact of Diabetes on 10‐Year Outcomes Following ST‐Segment–Elevation Myocardial Infarction: Insights From the EXAMINATION‐EXTEND Trial

    BACKGROUND: Long-term outcomes of ST-segment-elevation myocardial infarction in patients with diabetes have been barely investigated. The objective of this analysis from the EXAMINATION-EXTEND (10-Years Follow-Up of the EXAMINATION trial) trial was to compare 10-year outcomes of patients with ST-segment-elevation myocardial infarction with and without diabetes. METHODS AND RESULTS: Of the study population, 258 patients had diabetes and 1240 did not. The primary end point was patient-oriented composite end point of all-cause death, any myocardial infarction, or any revascularization. Secondary end points were the individual components of the primary combined end point, cardiac death, target vessel myocardial infarction, target lesion revascularization, and stent thrombosis. All end points were adjusted for potential confounders. At 10 years, patients with diabetes showed a higher incidence of patient-oriented composite end point compared with those without (46.5% versus 33.0%; adjusted hazard ratio [HR], 1.31 [95% CI, 1.05-1.61]; P=0.016) mainly driven by a higher incidence of any revascularization (24.4% versus 16.6%; adjusted HR, 1.61 [95% CI, 1.19-2.17]; P=0.002). Specifically, patients with diabetes had a higher incidence of any revascularization during the first 5 years of follow-up (20.2% versus 12.8%; adjusted HR, 1.57 [95% CI, 1.13-2.19]; P=0.007) compared with those without diabetes. No statistically significant differences were found with respect to the other end points. CONCLUSIONS: Patients with ST-segment-elevation myocardial infarction who had diabetes had worse clinical outcome at 10 years compared with those without diabetes, mainly driven by a higher incidence of any revascularizations in the first 5 years
