    Efectividad de un programa de Mindfulness en familiares de pacientes diagnosticados con enfermedad de Alzheimer

    Trabajo fin de grado en EnfermeríaIntroducción: La enfermedad de Alzheimer acapara un 60-80% de los casos de demencia que se producen en todo el mundo. El cuidado de estas personas provoca un gran impacto en la unidad familiar y ocasiona la sobrecarga del cuidador principal, lo que requiere desarrollar estrategias para mejorar la calidad de vida de estos cuidadores. Las evidencias indican la efectividad de los programas de Mindfulness en la reducción de los síntomas negativos de los cuidadores, no obstante, se precisa realizar más investigaciones que establezcan con claridad la eficacia de estos tratamientos. Objetivo: Determinar la efectividad de una intervención de Mindfulness para la reducción de la percepción de sobrecarga de los familiares de pacientes diagnosticados de Alzheimer que se encuentran en la fase severa de su enfermedad y acuden al Centro de Salud Lucero. Metodología: se presenta un proyecto con diseño cuasi-experimental y mediciones pre-post intervención, a realizar en el mes de julio de 2016 con un grupo de 20 sujetos. Las mediciones se realizarán al inicio, al finalizar la intervención y a los 3 meses. Se medirán las variables: sociodemográficas, atención plena (FFMQ), sobrecarga (Escala de Sobrecarga del Cuidador), estrés (PSS), resiliencia (CD-RISC), y satisfacción. Propuesta de análisis: a dos niveles, descripción de las características de la población y contraste de hipótesis. Implicaciones para el cuidado: la obtención de resultados óptimos en la reducción de la percepción de sobrecarga y mejora de la calidad de vida de los cuidadores, demostrará la efectividad de estas técnicas como estrategia para abordar este problema.Introduction: Alzheimer’s disease is about 60-80% of dementia cases around the world. The care of these people causes a great impact on the family unit and causes the main caregiver’s burden, which requires developing strategies to improve the caregivers’ quality of life. Evidence indicates the effectiveness of Mindfulness programs to reduce the negative symptoms of caregivers. However, is required more researches to establish clearly the efficacy of these treatments. Aim: to determinate the effectiveness of a Mindfulness program for reducing the burden’s perception of caregivers of patients diagnosed with Alzheimer’s disease, whom are in the severe phase of their disease and belong to the Primary Health Center Lucero. Methods: is presented a project with a quasi-experimental design and before-after intervention measures, to be made in July 2016, with a group of 20 subjects. Evaluations will be performed at baseline, at the end of the intervention, and at 3 months. The variables that will be evaluated are: sociodemographic variables, mindfulness (FFMQ), burden (Caregiver Burden Scale), stress (Perceived Stress Scale), resilience (CD-RISC) and satisfaction. Proposal of analysis: on two levels, description of the characteristics of population and hypothesis testing. Care implications: optimal results in reducing the burden’s perception of caregivers and in improving the quality of life of caregivers, will demonstrate the effectiveness of these techniques as a strategy to approach this problem

    Accessibility instruments for planning practice: bridging the gap between academic research and decision-making.

    One of the core objectives of urban planning practice is to provide spatial equity in terms of opportunities and use of public space and facilities. Accessibility is the element that serves this purpose as a concept linking the reciprocal relationship between transport and land use, thus shaping individual potential mobility to reach the desired destinations. Accessibility concepts are increasingly acknowledged as fundamental to understand the functioning of cities and urban regions. Indeed, by introducing them in planning practice, better solutions can be achieved in terms of spatial equity. The COST Action TU1002 "Accessibility instruments for planning practice" was specifically designed to address the gap between scientific research in measuring and modelling accessibility, and the current use of indicators of accessibility in urban planning practice. This paper shows the full process of introducing an easily understandable measure of accessibility to planning practitioners in Madrid, which is one of the case studies of the above-mentioned COST action. Changes in accessibility after the opening of a new metro line using contour measures were analyzed and then presented to a selection of urban planners and practitioners in Madrid as part of a workshop to evaluate the usefulness of this tool for planning practice. Isochrone maps were confirmed as an effective tool, as their utility can be supplemented by other indicators, and being GIS-based, it can be easily computed (when compared with transport models) and integrated with other datasets

    Obesity in Spanish Schoolchildren: Relationship with Lipid Profile and Insulin Resistance.

    This article reports cross-sectional data from a total of 1048 children, 6 to 8 years of age, categorized by presence or absence of obesity, who participated in a voluntary survey of cardiovascular risk factors in Spain over the period of 1998 to 2000, to establish the relationship between obesity and its metabolic consequences at this age. The prevalence of obesity and overweight were 9.4% and 15.7%, respectively, in boys and 10.5% and 18.0%, respectively, in girls. We observed that, in both sexes, obese children had higher triglycerides and lower high-density lipoprotein-cholesterol levels than non-obese children. No differences were found in plasma glucose or low-density lipoprotein-cholesterol levels between normal and obese children. However, we observed that insulin levels and the homeostasis model assessment for insulin resistance were significantly (p < 0.001) higher in obese children of both sexes but that free fatty acid levels were lower in obese children than in nonobese children, with a statistical significance in girls (0.72 ± 0.30 vs. 0.61 ± 0.16 mEq/liter). In summary, our survey found some metabolic consequences of obesity similar to those found in adults (elevated triglycerides, insulin, and the homeostasis model assessment for insulin resistance, and lower high-density lipoprotein-cholesterol). However, other features (glucose, total cholesterol, low-density lipoprotein-cholesterol, and free fatty acid levels) were found to behave differently, indicating that the association of obesity with risk factors seems to change as the children age and may depend on the chronology of sexual maturation.post-print51 K

    New Trends in Aging Drug Discovery

    Aging is considered the main risk factor for many chronic diseases that frequently appear at advanced ages. However, the inevitability of this process is being questioned by recent research that suggests that senescent cells have specific features that differentiate them from younger cells and that removal of these cells ameliorates senescent phenotype and associated diseases. This opens the door to the design of tailored therapeutic interventions aimed at reducing and delaying the impact of senescence in life, that is, extending healthspan and treating aging as another chronic disease. Although these ideas are still far from reaching the bedside, it is conceivable that they will revolutionize the way we understand aging in the next decades. In this review, we analyze the main and well-validated cellular pathways and targets related to senescence as well as their implication in aging-associated diseases. In addition, the most relevant small molecules with senotherapeutic potential, with a special emphasis on their mechanism of action, ongoing clinical trials, and potential limitations, are discussed.     Development of an Inverted Epifluorescence Microscope for Long-Term Monitoring of Bacteria in Multiplexed Microfluidic Devices

    Developing more efficient methods for antibiotic susceptibility testing is a pressing issue in novel drug development as bacterial resistance to antibiotics becomes increasingly common. Microfluidic devices have been demonstrated to be powerful platforms that allow researchers to perform multiplexed antibiotic testing. However, the level of multiplexing within microdevices is limited, evidencing the need of creating simple, low-cost and high-resolution imaging systems that can be integrated in antibiotic development pipelines. This paper describes the design and development of an epifluorescence inverted microscope that enables long-term monitoring of bacteria inside multiplexed microfluidic devices. The goal of this work is to provide a simple microscope powerful enough to allow single-cell analysis of bacteria at a reduced cost. This facilitates increasing the number of microscopes that are simultaneously used for antibiotic testing. We prove that the designed system is able to accurately detect fluorescent beads of 100 nm, demonstrating comparable features to high-end commercial microscopes and effectively achieving the resolution required for single-cell analysis of bacteria. The proposed microscope could thus increase the efficiency in antibiotic testing while reducing cost, size, weight, and power requirements, contributing to the successful development of new antibiotic drugs.This work was partially funded by project TEC2016-78052-R from the Spanish Ministry of Economy and PID2019-109820RB-I00 from the Spanish Ministry of Science and Innovation.     Effectiveness of a universal personalized intervention for the prevention of anxiety disorders: Protocol of a randomized controlled trial (the prevANS project)

    Background: To date, all preventive anxiety disorders interventions are one-fit-all and none of them are based on individual level and risk profile. The aim of this project is to design, develop and evaluate an online personalized intervention based on a risk algorithm for the universal prevention of anxiety disorders in the general population. Methods: A randomized controlled trial (RCT) with two parallel arms (prevANS vs usual care) and 1-year follow- up including 2000 participants without anxiety disorders from Spain and Portugal will be conducted. The prevANS intervention will be self-guided and can be implemented from the prevANS web or from the participants' Smartphone (through an App). The prevANS intervention will have different intensities depending on the risk level of the population, evaluated from the risk algorithm for anxiety: predictA. Both low and moderate-high risk participants will receive information on their level and profile (risk factors) of anxiety disorders, will have access to stress management tools and psychoeducational information periodically. In addition, participants with a moderate-high risk of anxiety disorders will also have access to cognitive-behavioral training (problem-solving, decision-making, communication skills, and working with thoughts). The control group will not receive any intervention, but they will fill out the same questionnaires as the intervention group. Assessments will be completed at baseline, 6 and 12-month follow-up. The primary outcome is the cumulative incidence of anxiety disorders. Secondary outcomes include depressive and anxiety symptoms, risk probability of anxiety disorders (predictA algorithm) and depression (predictD algorithm), improvement in physical and mental quality of life, and acceptability and satisfaction with the intervention. In addition, cost-effectiveness and cost-utility analyses will also be carried out from two perspectives, societal and health system, and analyses of mediators and moderators will also be performedSpanish Ministry of Health, the Institute of Health Carlos III, co-funded by the European Social Fund “Investing in your future” (grant references: CP19/00056), and the Chronicity, Primary Care and Health Promotion Research Network ‘RICAPPS’ (RD21/0016/0012); and Spanish Ministry of Science and Innovation, the State Investigation Agency (PID2020-119652RA-l00).     Cannabinoid receptor CB2 drives HER2 pro-oncogenic signaling in breast cancer

    Pharmacological activation of cannabinoid receptors elicits antitumoral responses in different models of cancer. However, the biological role of these receptors in tumor physio-pathology is still unknown. We analyzed CB2 cannabinoid receptor protein expression in two series of 166 and 483 breast tumor samples operated in the University Hospitals of Kiel, Tübingen and Freiburg between 1997 and 2010. CB2 mRNA expression was also analyzed in previously published DNA microarray datasets. The role of CB2 in oncogenesis was studied by generating a mouse line that expresses the HER2 rat ortholog (neu) and lacks CB2, and by a variety of biochemical and cell biology approaches in human breast cancer cells in culture and in vivo, upon modulation of CB2 expression by si/shRNAs and overexpression plasmids. CB2-HER2 molecular interaction was studied by co-localization, coimmunoprecipitation and proximity ligation assays. We show an association between elevated CB2 expression in HER2+ breast tumors and poor patient prognosis. We also demonstrate that genetic inactivation of CB2 impairs tumor generation and progression in MMTV-neu mice. Moreover, we show that HER2 upregulates CB2 expression by activating the transcription factor ELK1 via the ERK cascade, and that an increased CB2 expression activates the HER2 prooncogenic signaling machinery at the level of the tyrosine kinase c-SRC. Finally, HER2 and CB2 form heteromers in cancer cells. Our findings reveal an unprecedented role of CB2 as a pivotal regulator of HER2 pro-oncogenic signaling in breast cancer, and suggest that CB2 may be a biomarker with prognostic value in these tumors

    A long non-coding RNA that harbors a SNP associated with type 2 diabetes regulates the expression of TGM2 gene in pancreatic beta cells

    IntroductionMost of the disease-associated single nucleotide polymorphisms (SNPs) lie in non- coding regions of the human genome. Many of these variants have been predicted to impact the expression and function of long non-coding RNAs (lncRNA), but the contribution of these molecules to the development of complex diseases remains to be clarified. MethodsHere, we performed a genetic association study between a SNP located in a lncRNA known as LncTGM2 and the risk of developing type 2 diabetes (T2D), and analyzed its implication in disease pathogenesis at pancreatic beta cell level. Genetic association study was performed on human samples linking the rs2076380 polymorphism with T2D and glycemic traits. The pancreatic beta cell line EndoC-bH1 was employed for functional studies based on LncTGM2 silencing and overexpression experiments. Human pancreatic islets were used for eQTL analysis. ResultsWe have identified a genetic association between LncTGM2 and T2D risk. Functional characterization of the LncTGM2 revealed its implication in the transcriptional regulation of TGM2, coding for a transglutaminase. The T2Dassociated risk allele in LncTGM2 disrupts the secondary structure of this lncRNA, affecting its stability and the expression of TGM2 in pancreatic beta cells. Diminished LncTGM2 in human beta cells impairs glucose-stimulated insulin release.
