    Sub-daily rainfall extremes in the Nordic–Baltic region

    Short-duration rainfall extremes are associated with a range of societal hazards, notably pluvial flooding but in addition, e.g., erosion-driven nutrient transport and point-source contamination. Fundamental for all analysis, modelling and risk assessment related to short-duration rainfall extremes is the access to and analysis of high-resolution observations. In this study, sub-daily rainfall observations from 543 meteorological stations in the Nordic–Baltic region were collected, quality-controlled and consistently analyzed in terms of records, return levels, geographical and climatic dependencies, time of occurrence of maxima and trends. The results reflect the highly heterogeneous rainfall climate in the region, with longitudinal and latitudinal gradients as well as local variability, and overall agree with previous national investigations. Trend analyses in Norway and Denmark indicated predominantly positive trends in the period 1980–2018, in line with previous investigations. Gridded data sets with estimated return levels and dates of occurrence (of annual maxima) are provided open access. We encourage further efforts towards international exchange of sub-daily rainfall observations as well as consistent regional analyses in order to attain the best possible knowledge on which rainfall extremes are to be expected in present as well as future climates

    Perennials as Future Grain Crops: Opportunities and Challenges

    Perennial grain crops could make a valuable addition to sustainable agriculture, potentially even as an alternative to their annual counterparts. The ability of perennials to grow year after year significantly reduces the number of agricultural inputs required, in terms of both planting and weed control, while reduced tillage improves soil health and on-farm biodiversity. Presently, perennial grain crops are not grown at large scale, mainly due to their early stages of domestication and current low yields. Narrowing the yield gap between perennial and annual grain crops will depend on characterizing differences in their life cycles, resource allocation, and reproductive strategies and understanding the trade-offs between annualism, perennialism, and yield. The genetic and biochemical pathways controlling plant growth, physiology, and senescence should be analyzed in perennial crop plants. This information could then be used to facilitate tailored genetic improvement of selected perennial grain crops to improve agronomic traits and enhance yield, while maintaining the benefits associated with perennialism

    Framework For a Collective Definition of Regenerative Agriculture in India

    The concept of regenerative agriculture has received increasing attention worldwide as a method to restore and conserve natural resources while maintaining crop productivity. However, there remains a lack of consensus as to what conditions define regenerative agriculture, making it difficult for decision-makers, researchers, the agricultural sector, and the public to adopt regenerative agriculture practices. Here, we present the initial process to create a unified, cross-sectoral definition for regenerative agriculture in India that considers the viewpoints of multiple stakeholders and addresses the current challenges faced by the Indian agricultural sector. To this end, we compiled interactions with individuals from across India to identify the most pressing concerns for India's human and environmental ecosystems.     Study of the "Fast SCR" -like mechanism of H2-assisted SCR of NOx with ammonia over Ag/Al2O3

    It is shown that Ag/Al2O3 is a unique catalytic system for H-2-assisted selective catalytic reduction of NOx by NH3 (NH3-SCR) with both Ag and alumina being necessary components of the catalyst. The ability of Ag/Al2O3 and pure Al2O3 to catalyse SCR of mixtures of NO and NO2 by ammonia is demonstrated, the surface species occurring discussed, and a "Fast SCR"-like mechanism of the process is proposed. The possibility of catalyst surface blocking by adsorbed NOx and the influence of hydrogen on desorption of NOx were evaluated by FTIR and OFT calculations

    Discrimination of conventional and organic white cabbage from a long-term field trial study using untargeted LC-MS-based metabolomics

    The influence of organic and conventional farming practices on the content of single nutrients in plants is disputed in the scientific literature. Here, large-scale untargeted LC-MS-based metabolomics was used to compare the composition of white cabbage from organic and conventional agriculture, measuring 1,600 compounds. Cabbage was sampled in 2 years from one conventional and two organic farming systems in a rigidly controlled long-term field trial in Denmark. Using Orthogonal Projection to Latent Structures-Discriminant Analysis (OPLS-DA), we found that the production system leaves a significant (p = 0.013) imprint in the white cabbage metabolome that is retained between production years. We externally validated this finding by predicting the production system of samples from one year using a classification model built on samples from the other year, with a correct classification in 83% of cases. Thus, it was concluded that the investigated conventional and organic management practices have a systematic impact on the metabolome of white cabbage. This emphasizes the potential of untargeted metabolomics for authenticity testing of organic plant products

    Deficit of homozygosity among 1.52 million individuals and genetic causes of recessive lethality

    Genotypes causing pregnancy loss and perinatal mortality are depleted among living individuals and are therefore difficult to find. To explore genetic causes of recessive lethality, we searched for sequence variants with deficit of homozygosity among 1.52 million individuals from six European populations. In this study, we identified 25 genes harboring protein-altering sequence variants with a strong deficit of homozygosity (10% or less of predicted homozygotes). Sequence variants in 12 of the genes cause Mendelian disease under a recessive mode of inheritance, two under a dominant mode, but variants in the remaining 11 have not been reported to cause disease. Sequence variants with a strong deficit of homozygosity are over-represented among genes essential for growth of human cell lines and genes orthologous to mouse genes known to affect viability. The function of these genes gives insight into the genetics of intrauterine lethality.     Albiglutide and cardiovascular outcomes in patients with type 2 diabetes and cardiovascular disease (Harmony Outcomes): a double-blind, randomised placebo-controlled trial

    Background: Glucagon-like peptide 1 receptor agonists differ in chemical structure, duration of action, and in their effects on clinical outcomes. The cardiovascular effects of once-weekly albiglutide in type 2 diabetes are unknown. We aimed to determine the safety and efficacy of albiglutide in preventing cardiovascular death, myocardial infarction, or stroke. Methods: We did a double-blind, randomised, placebo-controlled trial in 610 sites across 28 countries. We randomly assigned patients aged 40 years and older with type 2 diabetes and cardiovascular disease (at a 1:1 ratio) to groups that either received a subcutaneous injection of albiglutide (30–50 mg, based on glycaemic response and tolerability) or of a matched volume of placebo once a week, in addition to their standard care. Investigators used an interactive voice or web response system to obtain treatment assignment, and patients and all study investigators were masked to their treatment allocation. We hypothesised that albiglutide would be non-inferior to placebo for the primary outcome of the first occurrence of cardiovascular death, myocardial infarction, or stroke, which was assessed in the intention-to-treat population. If non-inferiority was confirmed by an upper limit of the 95% CI for a hazard ratio of less than 1·30, closed testing for superiority was prespecified. This study is registered with ClinicalTrials.gov, number NCT02465515. Findings: Patients were screened between July 1, 2015, and Nov 24, 2016. 10 793 patients were screened and 9463 participants were enrolled and randomly assigned to groups: 4731 patients were assigned to receive albiglutide and 4732 patients to receive placebo. On Nov 8, 2017, it was determined that 611 primary endpoints and a median follow-up of at least 1·5 years had accrued, and participants returned for a final visit and discontinuation from study treatment; the last patient visit was on March 12, 2018. These 9463 patients, the intention-to-treat population, were evaluated for a median duration of 1·6 years and were assessed for the primary outcome. The primary composite outcome occurred in 338 (7%) of 4731 patients at an incidence rate of 4·6 events per 100 person-years in the albiglutide group and in 428 (9%) of 4732 patients at an incidence rate of 5·9 events per 100 person-years in the placebo group (hazard ratio 0·78, 95% CI 0·68–0·90), which indicated that albiglutide was superior to placebo (p<0·0001 for non-inferiority; p=0·0006 for superiority). The incidence of acute pancreatitis (ten patients in the albiglutide group and seven patients in the placebo group), pancreatic cancer (six patients in the albiglutide group and five patients in the placebo group), medullary thyroid carcinoma (zero patients in both groups), and other serious adverse events did not differ between the two groups. There were three (<1%) deaths in the placebo group that were assessed by investigators, who were masked to study drug assignment, to be treatment-related and two (<1%) deaths in the albiglutide group. Interpretation: In patients with type 2 diabetes and cardiovascular disease, albiglutide was superior to placebo with respect to major adverse cardiovascular events. Evidence-based glucagon-like peptide 1 receptor agonists should therefore be considered as part of a comprehensive strategy to reduce the risk of cardiovascular events in patients with type 2 diabetes.
