    Perspectives of High School Photography Teachers Regarding Visual Literacy

    This qualitative study investigated the perspectives of high school photography teachers regarding visual literacy. A qualitative methodology that used a phenomenographic research design was employed to gain understanding about the perspectives of high school photography teachers in their conceptualization, perceptions, and experiences surrounding visual literacy. A survey/ questionnaire was used to explore participants’ paths towards becoming a high school photography teacher, the amount of years they have been teaching, and their geographic location. Participants perception of school demographics such as school size, community contexts, racial, ethnic, and economic diversity were also collected. Additional prompts were designed to investigate curricular influences, pressing concerns, and pedagogical considerations regarding the various aspects of visual literacy. 100 survey/ questionnaire responses were collected along with five semi-structured interviews. Data were analyzed through constant comparison and in vivo coding was used to construct categories of description. The categories of description were further organized into three main themes: Curricular Influences, Pedagogical Considerations and Creative Applications. Data were then sorted into structural and referential aspects of experience to provide an overview of the outcome space. The structural aspects are described as teachers’ influential experiences and the referential described as the teaching of visual literacy. The results of this study were organized into five major discussion points: 1.) Participants’ lived experiences directly impact their teaching of visual literacy, 2.) Curriculum standards have limited impact on the teaching of visual literacy, 3.) Participants employ dialogue-based strategies when teaching visual literacy, 4.) Both analyzing and creating images are important aspects of visual literacy, 5.) Visual literacy addresses the needs of high school photography students

    The 2015 qualitative election study of Britain

    Related data set “Qualitative Election Study of Britain, 2015” with DOI http://dx.doi.org/10.5255/UKDA-SN-8117-1 in repository “UK Data Service”.The Qualitative Election Study of Britain (QESB) is the first (and only) qualitative longitudinal dataset to investigate political attitudes and voting behaviour over multiple elections and referendums in the United Kingdom. During the 2015 UK general election over 90 voters participated in 23 focus groups across England, Scotland, and Wales before and after polling day. These participants represented a range of political party supporters and independent voters, age groups, and economic back-grounds. They discussed a range of political issues including their vote choice in the election, their impressions of the major party leaders, why they would consider voting (or never voting) for a political party, and their expectations for the country moving forward. Special focus groups were also held around the three leaders’ debates. The 2015 QESB also brought back participants who had participated in the 2010 QESB focus groups and the 2014 Scottish referendum focus groups. The 2015 QESB has created a unique panel of participants whose political opinions can be tracked across multiple elections. The project also includes questions that were asked in prior election focus groups and has replicated, with some modifications, the research design of the previous wave of the study

    Online Transgender Health Information Seeking: Facilitators, Barriers, and Future Directions

    Transgender people face difficulties accessing healthcare from providers, and thus often turn to online sources to seek health information. However, online platforms may not properly support trans health information seeking, and health information found online may be limited in accuracy. To examine how online platforms can best support trans health information seeking, we conducted online focus groups with trans people (n = 26) about their experiences with online health information and their needs and desires for online health information seeking platforms. We found that trans people face both facilitators and barriers to finding accurate, actionable information online. Facilitators include online community discovery, group privacy features, and the dual synchronous and asynchronous nature of online content. Barriers include platform censorship, misinformation, hate speech, and lack of tools to flag inaccurate content.     Production of high levels of poly-3-hydroxybutyrate in plastids of Camelina sativa seeds

    Poly-3-hydroxybutyrate (PHB) production in plastids of Camelina sativa seeds was investigated by comparing levels of polymer produced upon transformation of plants with five different binary vectors containing combinations of five seed-specific promoters for expression of transgenes. Genes encoding PHB biosynthetic enzymes were modified at the N-terminus to encode a plastid targeting signal. PHB levels of up to 15% of the mature seed weight were measured in single sacrificed T1 seeds with a genetic construct containing the oleosin and glycinin promoters. A more detailed analysis of the PHB production potential of two of the best performing binary vectors in a Camelina line bred for larger seed size yielded lines containing up to 15% polymer in mature T2 seeds. Transmission electron microscopy showed the presence of distinct granules of PHB in the seeds. PHB production had varying effects on germination, emergence and survival of seedlings. Once true leaves formed, plants grew normally and were able to set seeds. PHB synthesis lowered the total oil but not the protein content of engineered seeds. A change in the oil fatty acid profile was also observed. High molecular weight polymer was produced with weight-averaged molecular weights varying between 600 000 and 1 500 000, depending on the line. Select lines were advanced to later generations yielding a line with 13.7% PHB in T4 seeds. The levels of polymer produced in this study are the highest reported to date in a seed and are an important step forward for commercializing an oilseed-based platform for PHB production

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    Dissecting the Shared Genetic Architecture of Suicide Attempt, Psychiatric Disorders, and Known Risk Factors

    Background Suicide is a leading cause of death worldwide, and nonfatal suicide attempts, which occur far more frequently, are a major source of disability and social and economic burden. Both have substantial genetic etiology, which is partially shared and partially distinct from that of related psychiatric disorders. Methods We conducted a genome-wide association study (GWAS) of 29,782 suicide attempt (SA) cases and 519,961 controls in the International Suicide Genetics Consortium (ISGC). The GWAS of SA was conditioned on psychiatric disorders using GWAS summary statistics via multitrait-based conditional and joint analysis, to remove genetic effects on SA mediated by psychiatric disorders. We investigated the shared and divergent genetic architectures of SA, psychiatric disorders, and other known risk factors. Results Two loci reached genome-wide significance for SA: the major histocompatibility complex and an intergenic locus on chromosome 7, the latter of which remained associated with SA after conditioning on psychiatric disorders and replicated in an independent cohort from the Million Veteran Program. This locus has been implicated in risk-taking behavior, smoking, and insomnia. SA showed strong genetic correlation with psychiatric disorders, particularly major depression, and also with smoking, pain, risk-taking behavior, sleep disturbances, lower educational attainment, reproductive traits, lower socioeconomic status, and poorer general health. After conditioning on psychiatric disorders, the genetic correlations between SA and psychiatric disorders decreased, whereas those with nonpsychiatric traits remained largely unchanged. Conclusions Our results identify a risk locus that contributes more strongly to SA than other phenotypes and suggest a shared underlying biology between SA and known risk factors that is not mediated by psychiatric disorders.Peer reviewe

    The Trichoptera barcode initiative: a strategy for generating a species-level Tree of Life

    DNA barcoding was intended as a means to provide species-level identifications through associating DNA sequences from unknown specimens to those from curated reference specimens. Although barcodes were not designed for phylogenetics, they can be beneficial to the completion of the Tree of Life. The barcode database for Trichoptera is relatively comprehensive, with data from every family, approximately two-thirds of the genera, and one-third of the described species. Most Trichoptera, as with most of life’s species, have never been subjected to any formal phylogenetic analysis. Here, we present a phylogeny with over 16 000 unique haplotypes as a working hypothesis that can be updated as our estimates improve. We suggest a strategy of implementing constrained tree searches, which allow larger datasets to dictate the backbone phylogeny, while the barcode data fill out the tips of the tree. We also discuss how this phylogeny could be used to focus taxonomic attention on ambiguous species boundaries and hidden biodiversity. We suggest that systematists continue to differentiate between ‘Barcode Index Numbers’ (BINs) and ‘species’ that have been formally described. Each has utility, but they are not synonyms. We highlight examples of integrative taxonomy, using both barcodes and morphology for species description. This article is part of the themed issue ‘From DNA barcodes to biomes’
