    The transcription factor BATF operates as an essential differentiation checkpoint in early effector CD8+ T cells

    The transcription factor BATF is required for interleukin 17 (IL-17)-producing helper T cell (TH17) and follicular helper T cell (TFH) differentiation. Here, we show that BATF also has a fundamental role in regulating effector CD8+ T cell differentiation. BATF-deficient CD8+ T cells show profound defects in effector expansion and undergo proliferative and metabolic catastrophe early after antigen encounter. BATF, together with IRF4 and Jun proteins, binds to and promotes early expression of genes encoding lineage-specific transcription-factors (T-bet and Blimp-1) and cytokine receptors, while paradoxically repressing genes encoding effector molecules (IFN-γ and granzyme B). Thus, BATF amplifies TCR-dependent transcription factor expression and augments inflammatory signal propagation but restrains effector gene expression. This checkpoint prevents irreversible commitment to an effector fate until a critical threshold of downstream transcriptional activity has been achieved

    Polyfunctional T cell responses in children in early stages of chronic Trypanosoma cruzi infection contrast with monofunctional responses of long-term infected adults

    Background: Adults with chronic Trypanosoma cruzi exhibit a poorly functional T cell compartment, characterized by monofunctional (IFN-γ-only secreting) parasite-specific T cells and increased levels of terminally differentiated T cells. It is possible that persistent infection and/or sustained exposure to parasites antigens may lead to a progressive loss of function of the immune T cells. Methodology/Principal Findings: To test this hypothesis, the quality and magnitude of T. cruzi-specific T cell responses were evaluated in T. cruzi-infected children and compared with long-term T. cruzi-infected adults with no evidence of heart failure. The phenotype of CD4+ T cells was also assessed in T. cruzi-infected children and uninfected controls. Simultaneous secretion of IFN-γ and IL-2 measured by ELISPOT assays in response to T. cruzi antigens was prevalent among T. cruzi-infected children. Flow cytometric analysis of co-expression profiles of CD4+ T cells with the ability to produce IFN-γ, TNF-α, or to express the co-stimulatory molecule CD154 in response to T. cruzi showed polyfunctional T cell responses in most T. cruzi-infected children. Monofunctional T cell responses and an absence of CD4+TNF-α+-secreting T cells were observed in T. cruzi-infected adults. A relatively high degree of activation and differentiation of CD4+ T cells was evident in T. cruzi-infected children. Conclusions/Significance: Our observations are compatible with our initial hypothesis that persistent T. cruzi infection promotes eventual exhaustion of immune system, which might contribute to disease progression in long-term infected subjects.Fil: Albareda, María Cecilia.     Functionalized microelectrodes arrays with integrated microfluidic channels for single-site multiple

    Nowadays, different chemical and physical transfection techniques are used to delivery biomolecules of interest (e.g. DNA, RNA, proteins) into cells. Among the physical methods, electroporation generates transient pores in the plasma membrane by applying electrical pulses to suspended cells. One of its main limitations is the lack of spatio-temporal control over the process: it does not allow to select single cells (desirable requirement especially in highly heterogeneous tissues), and to monitor the transfection results in real-time. To circumvent these disadvantages, alternative microscale approaches are increasingly required. This work presents an integrated platform consisting of a gold microelectrode array (MEA) for singlesite electroporation and fluidic channels for controlled delivery of bio-chemical entities . In order to improve the efficiency of electroporation, the gold electrodes were coated with a thin film of nanostructured of Titanium Dioxide

    O trabalho apresentado na disciplina de Extensão e Pesquisa em Computação no Cursode Bacharelado em Ciências da Computação- trata de uma solução para o setor decompras do Instituto Federal Catarinense cuja necessidade surgiu da atual dinâmica noprocesso de compras de materiais, por exemplo, do IFC. Essas compras são feitas apartir do preenchimento manual penoso e exaustivo de diversos formulários quedemanda tempo e é passível de erros. Com o propósito de estabelecer um procedimentomais rápido, confiável e simplificado, o projeto tem como objetivo desenvolver umsistema capaz de automatizar essas tarefas e informatizar integralmente o processo. Essaabordagem também proporcionará vantagens adicionais, como a maior transparêncianas operações e o aumento na confiabilidade dos dados. Dando importância àcomplexidade e à profundidade dessa metodologia falha do setor de compras doInstituto, tornou-se um propósito apresentar um programa de software comaprofundamento das linguagens de programação, bem como, na construção de bases dedados como meio para armazenamento de informações. Dentre as tecnologias utilizadas, estarão: PHP, HTML, CSS, Javascript e MYSQL. Cabe destacar, ainda, que parafacilitar os afazeres do projeto, foram utilizadas metodologias ágeis de organizaçãocomo a plataforma Notion, cuja eficiência permitiu a distribuição efetiva de tarefas paraque o presente projeto fosse finalizado e, desse modo, a potencialização dos resultadosesperados. Cabe inferir, portanto, que a presente proposta destaca a importância de umaperspectiva sistêmica e atualizada da agenda de compras do IFC. Através dadisponibilização periódica de relatórios abrangentes, o sistema proporcionará melhoriassignificativas nas tomadas de decisões e no gerenciamento de processos. Assim, ficaevidente o quanto o trabalho descrito é necessário dentro do setor de compras além debeneficiar todos os envolvidos no processo e seus receptores, os estudantes e osprofessores. Espera-se auxiliar no crescimento do ambiente estudantil do Instituto paraque esse seja cada vez mais um exemplo de educandário

    Sex Disparity in Cord Blood FoxP3+ CD4 T Regulatory Cells in Infants Exposed to Malaria In Utero.

    Sex differences in the immune response and in infectious disease susceptibility have been well described, although the mechanisms underlying these differences remain incompletely understood. We evaluated the frequency of cord blood CD4 T cell subsets in a highly malaria-exposed birth cohort of mother-infant pairs in Uganda by sex. We found that frequencies of cord blood regulatory T cell ([Treg] CD4+CD25+FoxP3+CD127lo/-) differed by infant sex, with significantly lower frequencies of Tregs in female than in male neonates (P = .006). When stratified by in utero malaria exposure status, this difference was observed in the exposed, but not in the unexposed infants
