    Nitric oxide released from luminal s-nitroso-n-acetylcysteine increases gastric mucosal blood flow

    Nitric oxide (NO)-mediated vasodilation plays a key role in gastric mucosal defense, and NO-donor drugs may protect against diseases associated with gastric mucosal blood flow (GMBF) deficiencies. In this study, we used the ex vivo gastric chamber method and Laser Doppler Flowmetry to characterize the effects of luminal aqueous NO-donor drug S-nitroso-N-acetylcysteine (SNAC) solution administration compared to aqueous NaNO2 and NaNO3 solutions (pH 7.4) on GMBF in Sprague-Dawley rats. SNAC solutions (600 μM and 12 mM) led to a rapid threefold increase in GMBF, which was maintained during the incubation of the solutions with the gastric mucosa, while NaNO2 or NaNO3 solutions (12 mM) did not affect GMBF. SNAC solutions (600 μM and 12 mM) spontaneously released NO at 37 °C at a constant rate of 0.3 or 14 nmol·mL-1·min-1, respectively, while NaNO2 (12 mM) released NO at a rate of 0.06 nmol·mL-1·min-1 and NaNO3 (12 mM) did not release NO. These results suggest that the SNAC-induced GMBF increase is due to their higher rates of spontaneous NO release compared to equimolar NaNO2 solutions.     Impact of methodological approaches in the agreement between subjective and objective methods for assessing screen time and sedentary behavior in pediatric population: a systematic review

    Introduction: sedentary behavior is an important target for health promotion. In this systematic review, we aimed to provide evidence to support decisions about measurement approach choices for subjectively assessing sedentary behavior in pediatric population, adopting objective methods as the reference. Methods: in this systematic review with meta-analysis, published studies were retrieved from electronic databases: Medline (PubMed), Web of Science, Embase, SPORTDiscus, BioMed Central and SCOPUS. We considered studies evaluating sedentary behavior agreement through questionnaire and/or diary in comparison with an objective measure. A total of six inclusion criteria v, rere used. We synthesized the data using correlation coefficients (r) as an indicator of agreement estimates. The review protocol is registered in the PROSPERO database (CRD42014015138). Results: a total of 14 studies met the inclusion criteria with ages ranging from 3 to 17.5 years and provided 17 agreement analyses. Thirteen of these agreement analyses (76.5%) reported correlation coefficients. We found two major groups of sedentary activities: screen time (47.1%) and sedentary behaviors (52.9%). The pooled agreement between questionnaires and accelerometers for assessing self-reported screen time was negative (r =-0.15; Cl 95%:-0.17 to-0.13). Conversely, when the sedentary behavior was assessed by questionnaires and accelerometers, the pooled agreement, vas positive for parent-reporting (r = 0.09; Cl 95%; 0.04 to 0.13) and self-reporting (r = 0.43; CI 95%: 0.40 to 0.47) in children and adolescents, respectively. Conclusion: questionnaires have positive agreement with accelerometers for assessing sedentary behavior, whereas the agreement is negative for assessing screen time. Self-reported questionnaires are recommended methods to measure sedentary behavior in adolescents

    Absence of Effective Vesicular-Arbuscular Mycorrhizae on Lecythidaceae in a Primary Forest Area of Central Amazonia.

    Lecythidaceae comprise one of the four most common tree families in the Amazon forest, but there have been few studies of vesicular-arbuscular mycorrhizae in this taxon. In this study roots of 31 species in six genera of Lecythidaceae were collected in a primary forest on an acid oxisol. No vesicular-arbuscular mycorrhizae, as indicated by the presence of vesicles or arbuscules were detected. Only some hyfas were found on some species roots, but not enough for increasing the nutrient absorption from the soil. This indicated that under the edaphic condiction and the time which the samples were collected, these species worked as no mycor-rhizal plants. These informations confirm datas found for other researches, and may be important for exploring more efficiently and by a not predatory way, the species of Lecythidaceae on agroforestry systems in the Amazon.As Lecythidaceae compreendem uma das quatro famílias de árvores mais abundantes na floresta amazônica, mas foram pouco estudadas no que sc refere à simbiose com fungos micorrízicos dos solos. Foram coletadas raízes de 31 espécies de seis gêneros desta família numa floresta de terra firme da Amazônia Central, próxima a Manaus, sob um latossolo amarelo. Em algumas espécies observou-se uma ausência total de fungos nas raízes, enquanto que em outras, apenas algumas hifas de fungos, mas sem vesículas ou arbúsculos, estruturas que serviriam para confirmar a presença de micorrizas vesículo-arbusculares efetivas nas mesmas. As poucas bifas encontradas não seriam suficientes para aumentar a absorção de nutrientes do solo, indicando que nas condições edáficas e épocas de coletas do presente estudo, as plantas se comportaram como não micorrizicas. Estas informações reforçam dados obtidos por outros autores e podem servir de embasamento para uma exploração mais eficiente e não predatória de espécies desta família em sistemas agroflorestais na Amazônia

    Metabolism of alcaligenes denitrificans in biofilm vs planktonic cells

    Aims: To compare the effect of phosphorous concentration (200 mg P 1-1 and 20 mg P 1-1) on the denitrifying efficiency of Alcaligenes denitrificans when in the form of planktonic cells or in the form of a biofilm, and to select the most adequate C/N ratio. Methods and Results: Two types of assays were carried out: with planktonic cells and with cells in biofilm form. Anoxic bottles with the appropriate C/N and phosphorous concentration were incubated at 30°C and submitted to orbital shaking at 150 rev min-1. The specific activity of cells in biofilm form, in terms of substrate consumption, was significantly higher than cells in planktonic form. With regard to the effect of increasing phosphorous concentration, an increase in specific activity was also only evident when the cells were in biofilm form. Conclusions: The two forms showed different performances and phosphorous concentration only markedly affected the biofilm form. Significance and Impact of the Study: The importance of the C/N/P ratio in the denitrification process is demonstrated. As there was no report in the literature about the stoichiometric relationship of heterotrophic denitrification with citrate, its stoichiometry, including the requirement for cell synthesis, was determined.Instituto de Biotecnologia e Química Fina (IBQF).     Histopathological features of the brain extracellular matrix from dogs with canine distemper

    ABSTRACT Canine distemper causes demyelinating leucoencephalitis, like human multiple sclerosis. The encephalic microenvironment, including the extracellular matrix, is an important key factor of this lesion, already described in multiple sclerosis but not proved in canine distemper. Thereby, the aim of this work is to characterize the extracellular matrix in the encephalon of dogs with canine distemper. Samples of cortex and cerebellum of 14 naturally infected dogs with canine distemper virus were collected after being sent for necropsy in the Animal Pathology Laboratory of the Veterinary Hospital of Uberlândia Federal University. The samples were processed as routine, stained with Hematoxylin and Eosin (H.E), Masson Trichrome (MT), Periodic Acid-Schiff (PAS) and Reticulin, and then described. Areas of demyelination and necrosis were quantified in percentage of stain. The TM samples showed blue stain around vessels and meninge, which indicates a higher deposition of collagen in lesioned areas. At necrotic areas, reticulin stain pointed to a disorganization in the vascular wall and PAS-stained pink granules in macrophages. We conclude that the extracellular matrix seems to participate in the pathogeny of canine distemper. More research should be done to better detail the involvement of these molecules in the course of this disease

    Morphological and electrophysiological properties of pyramidal-like neurons in the stratum oriens of Cornu ammonis 1 and Cornu ammonis 2 area of Proechimys

    AbstractProechimys (Rodentia: Echimyidae) is a neotropical rodent of the Amazon region that has been successfully colonized in the laboratory and used for experimental medicine. Preliminary studies indicated that Proechimys (casiragua) rodents express an atypical resistance to developing a chronic epileptic condition in common models of temporal lobe epilepsy. Moreover, previous investigation of our laboratory described a remarkably different Proechimy's cytoarchitecture organization of the hippocampal CA2 subfield. In the present study, we investigated the intrinsic neuronal properties and morphological characteristics of the Proechimys's hippocampal pyramidal neurons of the CA1 and CA2 areas. A comparative approach was performed using neurons recorded in Wistar rats. A striking finding in Proechimys rodents was the presence of large pyramidal-like neurons throughout the stratum oriens from CA2 to CA1 area. In order to confirm such distinctive feature of the Proechimys's hippocampus, we performed Nissl staining and immunohistochemistry for neurofilament protein SM311. CA2 pyramidal neurons in the stratum pyramidale of Proechimys exhibited a significantly higher input resistance and lower time constant when compared to corresponding cell groups in the same area of the Wistar rat's. This newly identified population of pyramidal-shaped neurons in stratum oriens of Proechimys exhibited distinct electrophysiological and morphological properties. This included larger capacitance, lower input resistance, larger rheobase, long latency to first action potential and slower firing frequency. In addition, the apical dendrites of these neurons were oriented in parallel to apical dendrites of regular pyramidal neurons in stratum pyramidale. Moreover, these neurons were immunoreactive to SM311 as the majority of the neurons of the pyramidal layer. The functional role of these hippocampal neurons of the rodent Proechimys deserves further investigation

    Surface and capillary transitions in an associating binary mixture model

    We investigate the phase diagram of a two-component associating fluid mixture in the presence of selectively adsorbing substrates. The mixture is characterized by a bulk phase diagram which displays peculiar features such as closed loops of immiscibility. The presence of the substrates may interfere the physical mechanism involved in the appearance of these phase diagrams, leading to an enhanced tendency to phase separate below the lower critical solution point. Three different cases are considered: a planar solid surface in contact with a bulk fluid, while the other two represent two models of porous systems, namely a slit and an array on infinitely long parallel cylinders. We confirm that surface transitions, as well as capillary transitions for a large area/volume ratio, are stabilized in the one-phase region.     Cytogenetic characterization and evaluation of c-MYC gene amplification in PG100, a new Brazilian gastric cancer cell line

    Gastric cancer is the fourth most frequent type of cancer and the second cause of cancer mortality worldwide. The genetic alterations described so far for gastric carcinomas include amplifications and mutations of the c-ERBB2, KRAS, MET, TP53, and c-MYC genes. Chromosomal instability described for gastric cancer includes gains and losses of whole chromosomes or parts of them and these events might lead to oncogene overexpression, showing the need for a better understanding of the cytogenetic aspects of this neoplasia. Very few gastric carcinoma cell lines have been isolated. The establishment and characterization of the biological properties of gastric cancer cell lines is a powerful tool to gather information about the evolution of this malignancy, and also to test new therapeutic approaches. The present study characterized cytogenetically PG-100, the first commercially available gastric cancer cell line derived from a Brazilian patient who had a gastric adenocarcinoma, using GTG banding and fluorescent in situ hybridization to determine MYC amplification. Twenty metaphases were karyotyped; 19 (95%) of them presented chromosome 8 trisomy, where the MYC gene is located, and 17 (85%) presented a deletion in the 17p region, where the TP53 is located. These are common findings for gastric carcinomas, validating PG100 as an experimental model for this neoplasia. Eighty-six percent of 200 cells analyzed by fluorescent in situ hybridization presented MYC overexpression.     A large community outbreak of Legionnaires’ disease in Vila Franca de Xira, Portugal, October to November 2014

    An outbreak of Legionnaires’ disease with 334 confirmed cases was identified on 7 November 2014 in Vila Franca de Xira, Portugal and declared controlled by 21 November. Epidemiological, environmental and microbiological analysis identified industrial wet cooling systems to be the probable source of infection. Preliminary results from sequence-based typing of clinical specimens and environmental isolates confirmed this link. A series of meteorological phenomena are likely to have contributed to the scale of this outbreak
