    Visual histological grading system for the evaluation of in vitro-generated neocartilage

    Here we present the development of a visual evaluation system for routine assessment of in vitro-engineered cartilaginous tissue. Neocartilage was produced by culturing human articular chondrocytes in pellet culture systems or in a scaffold-free bioreactor system. All engineered tissues were embedded in paraffin and were sectioned and stained with Safranin O-fast green. The evaluation of each sample was broken into 3 categories (uniformity and intensity of Safranin O stain, distance between cells/amount of matrix produced, and cell morphology), and each category had 4 components with a score ranging from 0 to 3. Three observers evaluated each sample, and the new system was independently tested against an objective computer-based histomorphometry system. Pellets were also assessed biochemically for glycosaminoglycan (GAG) content. Pellet histology scores correlated significantly with GAG contents and were in agreement with the computer-based histomorphometry system. This system allows a valid and rapid assessment of in vitro-generated cartilaginous tissue that has a relevant association with objective parameters indicative of cartilage quality

    Canine distemper virus neutralization activity is low in human serum and it is sensitive to an amino acid substitution in the hemagglutinin protein

    © 2015 Elsevier Inc.Serum was analyzed from 146 healthy adult volunteers in eastern Africa to evaluate measles virus (MV) and canine distemper virus (CDV) neutralizing antibody (nAb) prevalence and potency. MV plaque reduction neutralization test (PRNT) results indicated that all sera were positive for MV nAbs. Furthermore, the 50% neutralizing dose (ND50) for the majority of sera corresponded to antibody titers induced by MV vaccination. CDV nAbs titers were low and generally were detected in sera with high MV nAb titers. A mutant CDV was generated that was less sensitive to neutralization by human serum. The mutant virus genome had 10 nucleotide substitutions, which coded for single amino acid substitutions in the fusion (F) and hemagglutinin (H) glycoproteins and two substitutions in the large polymerase (L) protein. The H substitution occurred in a conserved region involved in receptor interactions among morbilliviruses, implying that this region is a target for cross-reactive neutralizing antibodies

    Angular dependent vortex pinning mechanisms in YBCO coated conductors and thin films

    We present a comparative study of the angular dependent critical current density in YBa2Cu3O7 films deposited on IBAD MgO and on single crystal MgO and SrTiO3 substrates. We identify three angular regimes where pinning is dominated by different types of correlated and uncorrelated defects. We show that those regimes are present in all cases, indicating that the pinning mechanisms are the same, but their extension and characteristics are sample dependent, reflecting the quantitative differences in texture and defect density. In particular, the more defective nature of the films on IBAD turns into an advantage as it results in stronger vortex pinning, demonstrating that the critical current density of the films on single crystals is not an upper limit for the performance of the IBAD coated conductors.Comment: 14 pages, 3 figures.     Nonstomatal limitations are responsible for drought-induced photosynthetic inhibition in four C4 grasses

    Here, the contribution of stomatal and nonstomatal factors to photosynthetic inhibition under water stress in four tropical C(4) grasses was investigated (Panicum coloratum, Bothriochloa bladhii, Cenchrus ciliaris and Astrebla lappacea ). Plants were grown in well watered soil, and then the effects of soil drying were measured on leaf gas exchange, chlorophyll a fluorescence and water relations. During the drying cycle, leaf water potential (Psi(leaf)) and relative water content (RWC) decreased from c. -0.4 to -2.8 MPa and 100-40%, respectively. The CO(2) assimilation rates (A) and quantum yield of PSII (Phi(PSII)) of all four grasses decreased rapidly with declining RWC. High CO(2) concentration (2500 mul l(-1)) had no effect on A or Phi(PSII) at any stage of the drying cycle. Electron transport capacity and dark respiration rates were unaltered by drought. The CO(2) compensation concentrations of P. coloratum and C. ciliaris rose sharply when leaf RWC fell below 70%. In P. coloratum, 5% CO(2) did not prevent the decline of O(2) evolution rates under water stress. We conclude that inhibition of photosynthesis in the four C(4) grasses under water stress is dependent mainly on biochemical limitations

    Barley crops are at risk of being attacked by several leaf diseases. Net blotch, brown rust, Rhynchosporium and Ramularia leaf spot are among the most widespread and can cause severe attack and yield losses. Two trial protocols targeting Ramularia and net blotch, respectively, have been tested in several countries in 2021 and 2022. Ramularia trials were situated in Germany, Ireland, Scotland, and Denmark. The net blotch trials were placed in Denmark, Belgium, the UK, Germany, Finland, and France. In the two protocols, 12–13 different fungicide solutions including co-formulations of DMIs, SDHIs, QoIs, and multi-site inhibitors have been tested to compare efficacy and yield responses. Against Ramularia leaf spot, the fungicides were applied at GS 47–51 and against net blotch at GS 37–45. In six trials, the efficacy against Ramularia leaf spot was scored. The results showed a superior control from the co-formulation fluxapyroxad + metyltetraprole (78–100% control), but also solo mefentrifluconazole and the mixtures fluxapyroxad + mefentrifluconazole performed well (average 74–76% control). The mixture fluxapyroxad + metyltetraprole provided the best yield increase followed by Ascra Xpro. Folpet as a solo solution was inferior. Following the net blotch protocol, only three trials developed enough disease to rank the different fungicides; however, in five trials ranking against brown rust was also possible. Most treatments gave very good control of net blotch, and brown rust (&gt; 80% control). The mixture fluxapyroxad + metyltetraprole delivered the best control against all diseases overall. Average yield responses from eight trials showed very similar increases from the tested fungicides.</p

    PubChem: a public information system for analyzing bioactivities of small molecules

    PubChem (http://pubchem.ncbi.nlm.nih.gov) is a public repository for biological properties of small molecules hosted by the US National Institutes of Health (NIH). PubChem BioAssay database currently contains biological test results for more than 700 000 compounds. The goal of PubChem is to make this information easily accessible to biomedical researchers. In this work, we present a set of web servers to facilitate and optimize the utility of biological activity information within PubChem. These web-based services provide tools for rapid data retrieval, integration and comparison of biological screening results, exploratory structure–activity analysis, and target selectivity examination. This article reviews these bioactivity analysis tools and discusses their uses. Most of the tools described in this work can be directly accessed at http://pubchem.ncbi.nlm.nih.gov/assay/.
