The role of neutrophil recruitment in the pathogenesis of salmonella enterica serotype typhimurium-induced enteritis in calves
The role of neutrophils in the pathogenesis of Salmonella
typhimurium-induced ruminant and human enteritis and diarrhea remains
incompletely understood. To address this question, the in vivo bovine ligated ileal
loop model of non-typhoidal salmonellosis was used in calves with the naturallyoccurring
Bovine Leukocyte Adhesion Deficiency (BLAD) mutation whose
neutrophils are unable to extravasate and infiltrate the extravascular matrix. Data
obtained from BLAD calves were compared to those from genetically normal
calves negative for the BLAD mutation. Morphologic studies showed that the
absence of significant tissue influx of neutrophils in intestine infected by S.
typhimurium resulted in less tissue damage, reduced luminal fluid accumulation,
and increased bacterial invasion compared to regular calves. Study of gene
expression profile of cytokines by quantitative Real-Time PCR (qRTPCR)
revealed that the massive tissue influx of neutrophils during acute infection is
mainly driven by the CXC chemokine GRO- α especially in the last stages of acute infection and to a lesser extent, IL-8. In contrast, the pro-inflammatory
cytokines IL-1 β and TNF- α were not significantly correlated with the presence or
absence of tissue neutrophils.
The precise in situ localization of gene expression of these major
cytokines and chemokines was investigated by qRTCPR from specific groups of
intestinal cells captured by Laser Capture Microdissection in S. typhimuriuminfected
ileal loops from BLAD animals. Our data confirmed that gene expression
of IL-8, GRO- α, and IL-1 β was predominantly localized to enterocytes of crypts
with less expression in enterocytes of villi tips and cells that form the domed villi
were not an important source of TNF- α gene expression. Microarray technology
was used to determine the global transcriptional profile of bovine intestinal loops
inoculated with S. typhimurium. The host samples were hybridized on a 13K
bovine-specific oligoarray and microarray data was analyzed using a suite of
gene expression analysis and modeling tools. Analysis of our data revealed that
the tissue influx of neutrophils in ileal loops greatly influenced the host gene
expression. Major differences in gene expression in relevant fields of Salmonella
research including inflammation and immune response, Toll-like receptor
signaling, cytokine profiles, apoptosis, and intracellular defense against infection
are discussed
On the movement analysis of null subjects in Brazilian Portuguese : experimental results from extraction of embedded subjects
There is an on-going debate on the empirical adequacy of the movement approach to definite null subjects in Brazilian Portuguese, BP (cf. Ferreira 2000, Rodrigues 2004, and Nunes 2009). On the one hand, Modesto (2000) observes that the null subject of finite embedded clauses associated with object control verbs like convencer 'to convince' is subject-oriented, rather than object-oriented and this is unexpected from a movement perspective. On the other hand, Rodrigues (2004) claims that these embedded clauses are actually adjuncts in BP and the observed subject orientation can be accounted for in terms of Hornstein's (1999, 2001) movement analysis of adjunct control. This paper aims to contribute to this debate by presenting the results of an experiment on grammaticality judgments by BP speakers on the extraction of embedded subjects out of complement and adjunct clauses, as well as finite embedded clauses associated with convencer. The results show that when a distinctive pattern could be observed, finite clauses associated with convencer behaved like adjunct clauses rather than complement clauses. The experiment thus provides confirming evidence for Rodrigues's (2004) adjunct analysis, invalidating Modesto's (2000) argument against the movement approach to definite null subjects in BP
PolĂticas pĂşblicas e interseccionalidade: debatendo gĂŞnero, raça e classe no sistema socioeducativo
El objetivo del estudio es discutir polĂticas pĂşblicas a partir de la interseccionalidad entre gĂ©nero, raza y clase en el sistema socioeducativo. La metodologĂa comprendiĂł un estudio bibliográfico, que permitiĂł profundizar en los fenĂłmenos a partir de las bases de datos investigadas. Se concluyĂł que la incorporaciĂłn de gĂ©nero, raza y clase en el sistema socioeducativo potencia la discriminaciĂłn, opresiĂłn y castigo de los adolescentes privados de libertad. AsĂ, existe la necesidad de formular polĂticas pĂşblicas interseccionales que señalen las dimensiones estructurantes y atiendan las demandas de los adolescentes como medio de enfrentamiento a la violencia institucional. Se propone discutir el proyecto de privaciĂłn de libertad, que tiene como objetivo mantener los aspectos ideolĂłgicos y conservadores para el mantenimiento del orden.O objetivo do estudo Ă© discutir polĂticas pĂşblicas tendo como base a interseccionalidade entre gĂŞnero, raça e classe no sistema socioeducativo. A metodologia compreendeu estudo bibliográfico, permitindo um aprofundamento dos fenĂ´menos a partir de bases de dados pesquisadas. Concluiu-se que a incorporação do gĂŞnero, raça e classe no sistema socioeducativo potencializa as discriminações, opressões e punições das adolescentes privadas de liberdade. Assim, destaca-se a necessidade de formular polĂticas pĂşblicas interseccionais que apontem as dimensões estruturantes e contemplem as demandas das adolescentes como enfrentamento da violĂŞncia institucional. Propõe-se discutir o projeto de privação de liberdade, a qual objetiva manter os aspectos ideolĂłgicos e conservadores para manutenção da ordem
Análise do nĂvel de informação da equipe de enfermagem sobre eventos adversos
Justificativa e Objetivo: Este estudo se insere no campo da avaliação do conhecimento, envolvendo em especĂfico os profissionais de Enfermagem e a ocorrĂŞncia de efeitos adversos, podendo consistir em um agravo evitável. Desta forma, o objetivo foi analisar o nĂvel de informação da equipe de enfermagem sobre os eventos adversos em uma Unidade de Terapia Intensiva. MĂ©todo: Trata-se de um estudo descritivo, exploratĂłrio, de abordagem quantitativa, realizado em uma Unidade de Terapia Intensiva de um hospital estadual do interior de Pernambuco, com 27 integrantes da equipe de Enfermagem. A coleta foi realizada no perĂodo de julho de 2013 por meio de um questionário e os dados foram analisados no software Epi-Info 3.5. Resultados: Houve predominância de 63% com conhecimento pleno em relação Ă definição de eventos adversos; 63% relataram nĂŁo haver formulário prĂłprio para notificação de eventos adversos; 55,6% informam que nĂŁo existe um programa de qualidade do cuidado e 70,4% desconhecem a existĂŞncia de publicação periĂłdica dos indicadores de qualidade do cuidado no serviço. ConclusĂŁo: Possuir conhecimento sobre os eventos adversos nĂŁo elimina os riscos de ocorrĂŞncia; questões estruturais e relacionadas ao cuidado com o paciente favorecem a incidĂŞncia de eventos adversos, acentuada pelo fato da nĂŁo identificar riscos e de nĂŁo promover atividades preventivas e educacionais
Análise do nĂvel de informação da equipe de enfermagem sobre eventos adversos
Background and Objectives: This study is part of the field of knowledge assessment, specifically involving Nursing professionals and the occurrence of adverse effects, and may consist of an avoidable illness. In this way, the objective was to analyze the level of information of the nursing team about the adverse events in an Intensive Care Unit (ICU). Methods: Cross-sectional study, carried out in an ICU of a state hospital in the interior of Pernambuco, Brazil, with 27 members of the Nursing team. The collection was carried out in the period of July 2013 through a questionnaire based on the Predisposition to Occurrence of Adverse Events Scale and the data were analyzed through descriptive statistics in the Epi-Info software 3.5. Results: The nursing staff had knowledge about the definition of adverse events (77.8%), but related to the patient's underlying disease (22.2%); in addition, they did not have a structure to prevent or notify, and the process of care developed in the ICU favored it.
Conclusion: The structural and patient care issues favor the incidence of adverse events, which increase by not identifying the risks and not promoting preventive and educational activities with those involved.JustificaciĂłn y Objetivo: Este estudio se inserta en el campo de la evaluaciĂłn del conocimiento, involucrando en especĂfico a los profesionales de EnfermerĂa y la ocurrencia de efectos adversos, pudiendo consistir en un agravio evitable. La recolecciĂłn fue realizada en el perĂodo de julio de 2013 a travĂ©s de un cuestionario basado en la Escala de PredisposiciĂłn a la Ocurrencia de Eventos Adversos y los datos fueron analizados por medio de estadĂstica descriptiva en el software Epi-Info 3.5. Resultados: El equipo de enfermerĂa tenĂa conocimiento sobre la definiciĂłn de eventos
adversos (77,8%), pero se relacionaban con la enfermedad de base del paciente (22,2%); además, no tenĂan estructura para prevenir o notificar, y era favorecido por el proceso del cuidado desarrollado en la UTI. Avaliação do perfil hematológico (Eritrograma) de peixes-boi-da amazônia (Trichechus inunguis NATTERER, 1883) mantidos em Santarém-PA / Evaluation of the sérum hematological (Erythrogram) of amazon manfish (Trichechus inunguis NATTERER, 1883) maintained in Santarém- PA
O Trichechus inunguis (Natterer, 1883), mais conhecido como peixe-boi-da-amazĂ´nia, Ă© o menor mamĂfero aquático da ordem Sirenia, endĂŞmico em praticamente todo o territĂłrio da bacia amazĂ´nica. Sua população foi drasticamente reduzida no perĂodo prĂ© e pĂłs-colonial pela exploração de gordura e couro, e atualmente está na lista dos animais ameaçados de extinção. No municĂpio de SantarĂ©m-Pará, e cidades vizinhas, os animais oriundos de resgates realizados por ĂłrgĂŁos ambientais e Corpo de Bombeiros, sĂŁo recebidos no ZoolĂłgico da Universidade da AmazĂ´nia (ZOOUNAMA), e permanecem em um projeto de 2 fases, composto por reabilitação e soltura. Uma das ferramentas de suporte na clinica de animais em fase de reabilitação Ă© o hemograma, que, quando necessário, permite instituir uma conduta terapĂŞutica imediata, monitora a sanidade dos indivĂduos e contribui para otimizar o manejo desses sirĂŞnios de água doce cativos. Dessa forma, a presente pesquisa objetivou avaliar as variaveis hematologicas dos peixes-boi em fase de reabilitação mantidos no ZOUNAMA. Foram avaliados os hemogramas de 37 animais sendo separados por grupos de acordo com a faixa etária e o sexo dos animais. Foi definido como filhote os animais entre 1 a 24 meses (grupo 1), jovens, os animais entre 24 a 72 meses (grupo 2) e adultos, com idade a partir de 73 meses (grupo 3). Os resultados nĂŁo revelaram diferenças significativas (p>0,05) nos parametros hematologicos dos grupos avaliados no presnte estudo. TambĂ©m foi constatado que as variaveis sanguineas se mantiveram dentro dos valores de referencia para a especie
Role of SPI-1 Secreted Effectors in Acute Bovine Response to Salmonella enterica Serovar Typhimurium: A Systems Biology Analysis Approach
Salmonella enterica Serovar Typhimurium (S. Typhimurium) causes enterocolitis with diarrhea and polymorphonuclear cell (PMN) influx into the intestinal mucosa in humans and calves. The Salmonella Type III Secretion System (T3SS) encoded at Pathogenicity Island I translocates Salmonella effector proteins SipA, SopA, SopB, SopD, and SopE2 into epithelial cells and is required for induction of diarrhea. These effector proteins act together to induce intestinal fluid secretion and transcription of C-X-C chemokines, recruiting PMNs to the infection site. While individual molecular interactions of the effectors with cultured host cells have been characterized, their combined role in intestinal fluid secretion and inflammation is less understood. We hypothesized that comparison of the bovine intestinal mucosal response to wild type Salmonella and a SipA, SopABDE2 effector mutant relative to uninfected bovine ileum would reveal heretofore unidentified diarrhea-associated host cellular pathways. To determine the coordinated effects of these virulence factors, a bovine ligated ileal loop model was used to measure responses to wild type S. Typhimurium (WT) and a ΔsipA, sopABDE2 mutant (MUT) across 12 hours of infection using a bovine microarray. Data were analyzed using standard microarray analysis and a dynamic Bayesian network modeling approach (DBN). Both analytical methods confirmed increased expression of immune response genes to Salmonella infection and novel gene expression. Gene expression changes mapped to 219 molecular interaction pathways and 1620 gene ontology groups. Bayesian network modeling identified effects of infection on several interrelated signaling pathways including MAPK, Phosphatidylinositol, mTOR, Calcium, Toll-like Receptor, CCR3, Wnt, TGF-β, and Regulation of Actin Cytoskeleton and Apoptosis that were used to model of host-pathogen interactions. Comparison of WT and MUT demonstrated significantly different patterns of host response at early time points of infection (15 minutes, 30 minutes and one hour) within phosphatidylinositol, CCR3, Wnt, and TGF-β signaling pathways and the regulation of actin cytoskeleton pathway
Global Island Monitoring Scheme (GIMS) : a proposal for the long-term coordinated survey and monitoring of native island forest biota
Islands harbour evolutionary and ecologically unique biota, which are currently disproportionately threatened by a multitude of anthropogenic factors, including habitat loss, invasive species and climate change. Native forests on oceanic islands are important refugia for endemic species, many of which are rare and highly threatened. Long-term monitoring schemes for those biota and ecosystems are urgently needed: (i) to provide quantitative baselines for detecting changes within island ecosystems, (ii) to evaluate the effectiveness of conservation and management actions, and (iii) to identify general ecological patterns and processes using multiple island systems as repeated 'natural experiments'. In this contribution, we call for a Global Island Monitoring Scheme (GIMS) for monitoring the remaining native island forests, using bryophytes, vascular plants, selected groups of arthropods and vertebrates as model taxa. As a basis for the GIMS, we also present new, optimized monitoring protocols for bryophytes and arthropods that were developed based on former standardized inventory protocols. Effective inventorying and monitoring of native island forests will require: (i) permanent plots covering diverse ecological gradients (e.g. elevation, age of terrain, anthropogenic disturbance); (ii) a multiple-taxa approach that is based on standardized and replicable protocols; (iii) a common set of indicator taxa and community properties that are indicative of native island forests' welfare, building on, and harmonized with existing sampling and monitoring efforts; (iv) capacity building and training of local researchers, collaboration and continuous dialogue with local stakeholders; and (v) long-term commitment by funding agencies to maintain a global network of native island forest monitoring plots.Peer reviewe
