    Growth and lipid class composition of the Arctic pelagic amphipod Themisto libellula

    Carnivorous zooplankton is a key element to the energy transfer through the arctic food web, linking lipid rich herbivores to the top predators. We investigated the growth and lipid dynamic of the Arctic pelagic amphipod Themisto libellula in Kongsfjorden (Svalbard, 79°N) from May to October 2007. Additional samplings were performed in spring and summer 2006 and further north in Rijpfjorden (80°N), in September 2006 and 2007. In Kongsfjorden, the first free-swimming stages (3 mm) appeared early May and reached their adult length (25 mm), in October. During their first year, they grew according to a Von Bertalanffy model and most probably constituted a single cohort. Juveniles had the highest growth rate (0.19 mm day−1) and revealed relatively low total lipid (TL) content (about 2.5% wet weight (WW)) with phospholipids as the major lipid class. Sub-adults showed a distinct decrease of growth rates which coincided with the increase of neutral lipid storage, reflecting a switch in energy allocation, from somatic growth to lipid storage. Indeed wax esters (WE) increased up to 48.5% TL on average in adults in 2006 while triacylglycerols (TAG) remained almost constant below 25.2% TL. The absence of lipid accumulation (in disproportion of the weight) in 2007 could be explained by a higher metabolism of T. libellula or preys of lower quality. In Rijpfjorden, adults in their second year continued accumulating lipid (up to 10% WW) with high and similar proportions of both lipid classes, WE and TAG. We highlighted that T. libellula exhibited a variable lipid metabolism along its life cycle depending on its physiological needs and environmental conditions

    International audienceOver the last decade, there has been a significant deployment of systems dedicated to surveillance. These systems make use of real-time sensors that generate continuous streams of data. Despite their success in many cases, the increased number of sensors leads to a cognitive overload for the operator in charge of their analysis. However, the context and the application requires an ability to react in real-time. The research presented in this paper introduces a spatio-temporal-based approach the objective of which is to provide a qualitative interpretation of the behavior of an entity (e.g., a human or vehicle). The process is formally supported by a fuzzy logic-based approach, and designed in order to be as generic as possible

    Non-target screening with high-resolution mass spectrometry: critical review using a collaborative trial on water analysis

    In this article, a dataset from a collaborative nontarget screening trial organised by the NORMAN Association is used to review the state-of-the-art and discuss future perspectives of non-target screening using high-resolution mass spectrometry in water analysis. A total of 18 institutes from 12 European countries analysed an extract of the same water sample collected from the River Danube with either one or both of liquid and gas chromatography coupled with mass spectrometry detection. This article focuses mainly on the use of high resolution screening techniques with target, suspect, and non-target workflows to identify substances in environmental samples. Specific examples are given to emphasise major challenges including isobaric and co-eluting substances, dependence on target and suspect lists, formula assignment, the use of retention information, and the confidence of identification. Approaches and methods applicable to unit resolution data are also discussed. Although most substances were identified using high resolution data with target and suspect-screening approaches, some participants proposed tentative non-target identifications. This comprehensive dataset revealed that nontarget analytical techniques are already substantially harmonised between the participants, but the data processing remains time-consuming. Although the objective of a Bfullyautomated identification workflow^ remains elusive in the short term, important steps in this direction have been taken, exemplified by the growing popularity of suspect screening approaches. Major recommendations to improve non-target screening include better integration and connection of desired features into software packages, the exchange of target and suspect lists, and the contribution of more spectra from standard substances into (openly accessible) databases.This work was supported in part by the SOLUTIONS project, which received funding from the European Union’s Seventh Framework Programme for research, technological development and demonstration under Grant Agreement No. 603437

    Trophic level stability-inducing effects of predaceous early juvenile fish in an estuarine mesocosm study.

    Classically, estuarine planktonic research has focussed largely on the physico-chemical drivers of community assemblages leaving a paucity of information on important biological interactions.Within the context of trophic cascades, various treatments using in situ mesocosms were established in a closed estuary to highlight the importance of predation in stabilizing estuarine plankton abundances. Through either the removal (filtration) or addition of certain planktonic groups, five different trophic systems were established. These treatments contained varied numbers of trophic levels and thus different "predators" at the top of the food chain. The abundances of zooplankton (copepod and polychaete), ciliate, micro-flagellate, nano-flagellate and bacteria were investigated in each treatment, over time. The reference treatment containing apex zooplanktivores (early juvenile mullet) and plankton at natural densities mimicked a natural, stable state of an estuary. Proportional variability (PV) and coefficient of variation (CV) of temporal abundances were calculated for each taxon and showed that apex predators in this experimental ecosystem, when compared to the other systems, induced stability. The presence of these predators therefore had consequences for multiple trophic levels, consistent with trophic cascade theory.PV and CV proved useful indices for comparing stability. Apex predators exerted a stabilizing pressure through feeding on copepods and polychaetes which cascaded through the ciliates, micro-flagellates, nano-flagellates and bacteria. When compared with treatments without apex predators, the role of predation in structuring planktonic communities in closed estuaries was highlighted

    The total lipids and fatty acid composition of natural particulate matter and nutritional quality for zooplankton grazers was studied on a seasonal basis in the Arctic fjord Kongsfjorden (Svalbard) during the spring, summer of 2007 and during the early summer of 2006. Both years were abnormally warm, and the study attempted to evaluate the potential impact of the intrusion of North Atlantic waters. Samples were collected in surface layers and at deep chlorophyll maximum (DCM when present). Both years, chlorophyll concentrations were low (< 2 mu g L-1) even during bloom periods. Species determination indicated Phaeocystis spp. as main constituent of the May bloom while ciliates and flagellates dominated the rest of the survey period. Total lipids showed similar changes at both depths with maximum values in mid-summer of 2007, while it showed reverse patterns between surface and DCM in 2006. Total fatty acid composition was dominated by saturates and monoenoic acids at both depths with significant percentages of pentaenoic acids and 22:6n-3 (DHA) recorded at all times. The 2007 fatty acid dynamics identified four main successions in term of particulate assemblage related mainly to the succession of living cells versus detrital material and to a lesser extent to phytoplankton community changes (diatoms versus non-diatoms). Redundancy analysis confirmed that live phytoplankton is one of the main drivers in the fatty acid changes. Temperature and density of the surface water are also influential in relation to water mass dynamics. Concentrations of fatty acids available to consumers showed n-3 PUFA ranging from 2 to 15 mu g L-1 and n-6 PUFA ranging from 0.3 to 2 mu g L-1. Concentration of EPA (20:5) and DHA are potentially limiting, suggesting a negative impact of Phaeocystis pouchetti-type phytoplankton linked to advection of Atlantic waters in relation to global warming of Arctic waters

    Lipid composition of the three co-existing Calanus species in the Arctic: impact of season, location and environment

    Arctic species of Calanus are critical to energy transfer between higher and lower trophic levels and their relative abundance, and lipid content is influenced by the alternation of cold and warm years. All three species of Calanus were collected during different periods in Kongsfjorden (Svalbard, 79A degrees N) and adjacent shelf during the abnormally warm year of 2006. Lipid composition and fatty acid structure of individual lipid classes were examined in relation with population structure. Wax esters dominated the neutral lipid fraction. Phosphatidylcholine (PC) dominated the structural lipids followed by phosphatidylethanolamine (PE). PC/PE ratios of 3-6 suggested an increase in PC proportions compared to earlier studies. Depending on the time scale, fatty acids of wax esters illustrated either trophic differences between fjord and offshore conditions for C. hyperboreus and C. finmarchicus or trophic differences related to seasonality for C. glacialis. Similarly, seasonality and trophic conditions controlled the changes in fatty acids of triglycerides, but de novo synthesis of long-chain monoenes suggested energy optimization to cope with immediate metabolic needs. Polar lipids fatty acid composition was species specific and on the long-term (comparison with data from the past decade) composition appears related to changes in trophic environment. Fatty acid composition of PC and PE indicated relative dominance of 20:5n-3 in PC and 22:6n-3 in PE for all three species. The combination of PE and PC acyl chain and phospholipid head group restructuring indicates an inter-annual variability and suggests that membrane lipids are the most likely candidate to evaluate adaptive changes in Arctic copepods to hydrothermal regime
