    Influences of the technological parameters on the surface twist in grinding

    The requirement towards to the functional surfaces of the machine components became higher over the time. The final steps in the machining are supposed to create a surface which ensures wear resistance, longer lifetime and has special features on demand as well. The twist free surface can be considered as a special surface condition. The following paper presents the effect of the grinding parameters to the surface twist.Требования в отношении функциональных поверхностей деталей машин повысились с течением времени. Предполагается, что на окончательных этапах механической обработки должна создаваться поверхность, которая обеспечивает износостойкость, длительный срок службы, а также обладает специальными требуемыми характеристиками.     Oxidation and Release of Ruthenium from Short Fuel Rods above 1500oC

    The fission product ruthenium is radiotoxic and in oxidized form as ruthenium-tetroxide (RuO4) also chemical toxic. During fuel element change or in case of a leakage in fuel storages, air flows in the containment or fuel storage. If the circulation pump fails, the fuel elements are heated-up, and at high temperatures gaseous ruthenium oxides are formed and rapidly released from the fuel. Due to the significant higher volatility of RuO4 in the Chernobyl Catastrophe the measured concentration of the Ru-isotopes in the fall-outs was comparable to those of iodine and caesium. In order to get more insight in the chemical and physical behaviour of this fission product under severe accident conditions, the Ruthenium release was studied in a series of Separate Effect Tests (RUSET). In the test short fuel rod segments were used and exposed to air and steam atmosphere at high temperatures. The experiments in air atmosphere showed, that the partial pressures of the released Ru-oxides was two orders of magnitudes lower compared to those measured in previous tests with Ru-powder diluted in a ZrO2 matrix. It was found that ruthenium was not released in steam atmosphere in the examined temperature range.     A reduced-function allele reveals that EARLY FLOWERING3 repressive action on the circadian clock is modulated by phytochrome signals in Arabidopsis

    Arabidopsis thaliana EARLY FLOWERING3 (ELF3) is essential for the generation of circadian rhythms. ELF3 has been proposed to restrict light signals to the oscillator through phytochrome photoreceptors, but that has not been explicitly shown. Furthermore, the genetic action of ELF3 within the clock had remained elusive. Here, we report a functional characterization of ELF3 through the analysis of the elf3-12 allele, which encodes an amino acid replacement in a conserved domain. Circadian oscillations persisted, and unlike elf3 null alleles, elf3-12 resulted in a short circadian period only under ambient light. The period shortening effect of elf3-12 was enhanced by the overexpression of phytochromes phyA and phyB. We found that elf3-12 was only modestly perturbed in resetting of the oscillator and in gating light-regulated gene expression. Furthermore, elf3-12 essentially displayed wild-type development. We identified targets of ELF3 transcriptional repression in the oscillator, highlighting the action at the morning gene PSEUDO-RESPONSE REGULATOR9. Taken together, we identified two separable roles for ELF3, one affecting the circadian network and the other affecting light input to the oscillator. This is consistent with a dual function of ELF3 as both an integrator of phytochrome signals and a repressor component of the core oscillator

    Quantifying heterogeneities within cell populations is important for many fields including cancer research and neurobiology; however, techniques to isolate individual cells are limited. Here, we describe a high-throughput, non-disruptive, and cost-effective isolation method that is capable of capturing individually targeted cells using widely available techniques. Using high-resolution microscopy, laser microcapture microscopy, image analysis, and machine learning, our technology enables scalable molecular genetic analysis of single cells, targetable by morphology or location within the sample.Peer reviewe

    T cell immune response predicts survival in severely ill COVID-19 patients requiring venovenous extracorporeal membrane oxygenation support

    IntroductionThere is a critical gap in understanding which SARS-CoV-2 patients would benefit most from venovenous extracorporeal membrane oxygenation (VV-ECMO) support. The potential role of a dysregulated immune response is still unclear in this patient population.ObjectivesTo assess the potential predictive value of SARS-CoV-2 specific cellular and humoral immune responses for survival in critically ill COVID-19 patients requiring VV-ECMO.MethodsWe conducted a prospective single-center observational study of unvaccinated patients requiring VV-ECMO support treated at the intensive care unit of Semmelweis University Heart and Vascular Center between March and December 2021. Peripheral blood samples were collected to measure the humoral and cellular immune statuses of the patients at the VV-ECMO cannulation. Patients were followed until hospital discharge.ResultsOverall, 35 COVID-19 patients (63% men, median age 37 years) on VV-ECMO support were included in our study. The time from COVID-19 verification to ECMO support was a median (IQR) of 10 (7-14) days. Of the patients, 9 (26%) were discharged alive and 26 (74%) died during their hospital stay. Immune tests confirmed ongoing SARS-CoV-2 infection in all the patients, showing an increased humoral immune response. SARS-CoV-2-specific cellular immune response was significantly higher among survivors compared to the deceased patients. A higher probability of survival was observed in patients with markers indicating a higher T cell response detected by both QuantiFeron (QF) and flow cytometry (Flow) assays. (Flow S1 CD8+ ≥ 0.15%, Flow S1 CD4+ ≥ 0.02%, QF CD4 ≥ 0.07, QF whole genome ≥ 0.59). In univariate Cox proportional hazard regression analysis BMI, right ventricular (RV) failure, QF whole genome T cell level, and Flow S1 CD8+ T cell level were associated with mortality, and we found that an increased T cell response showed a significant negative association with mortality, independent of BMI and RV failure.ConclusionEvaluation of SARS-CoV-2 specific T cell response before the cannulation can aid the risk stratification and evaluation of seriously ill COVID-19 patients undergoing VV-ECMO support by predicting survival, potentially changing our clinical practice in the future

    Der vorliegende Tagungsband eröffnet Einblicke in ausgewählte Beiträge, Workshops, Gruppenimprovisationen und Performances der Arbeitstagung „DaZwischen 2023“: einer Initiative für eine interdisziplinäre musikpädagogische Begegnung von Lehrenden und Studierenden, Mitwirkenden und Teilnehmenden verschiedener Hochschulen und Universitäten. Das Dazwischen entfaltete sich zwischen den Disziplinen und Arbeitsfeldern und eröffnete sich in pädagogischen und spezifisch musikpädagogischen Aspekten. Im Kontext der Vermittlung des Ästhetischen, des ästhetischen Erlebens und der Intention, ästhetisches Erleben zu inszenieren, wurden unbestimmbare, widersprüchliche und ambivalente Zustände beschrieben und diskutiert. Geschieht Improvisation aktiv oder passiv, vollzieht sich Bildung in Freiheit oder intentional, was bewegt – oder lässt sich bewegen? Im DaZwischen erfüllt sich Unerfüllbares, verwirklicht sich Unplanbares. Der vorliegende Band fasst die ersten Impulse des in der Entwicklung begriffenen Symposiums zusammen und bietet Anknüpfungspunkte für eine interdisziplinäre Betrachtung der Didaktiken ästhetischer und musikalischer Bildung.The conference report offers an insight into lectures, group improvisations and performances from the 'DaZwischen 2023' labour conference. This event was created as an initiative for an interdisciplinary music education encounter between teachers and students from various institutions. In the context of conveying the aesthetic - the aesthetic experience and the intention of staging aesthetic experience - indeterminable, contradictory and ambivalent states of aesthetic consciousness and experiencing were described and discussed. This volume summarizes the initial impulses of this specific symposium and its stage in development and offers points of departure for an interdisciplinary consideration of the didactics of aesthetic and musical education

    Real-life experience with switching TNF-α inhibitors in ankylosing spondylitis

    Objective: The aim of this study was to evaluate the efficacy, reasons for switching and drug survival of TNF-α inhibitors (TNFis) used as first- and second-line drugs in ankylosing spondylitis (AS). Methods: Data on patients suffering from AS and treated with at least one TNFi between November 2005 and 2013 were extracted retrospectively from the database of a single clinical centre. Beside demographic data, the disease activity measured by the Bath Ankylosing Spondylitis Disease Activity Index (BASDAI), the response rates (BASDAI50), reasons for switching and survival curves of TNFis were analysed in general and in subgroups of patients treated with each of the available TNFis. The reasons for switching were defined as inefficacy, side effects of the given drug, patient's request and occurrence of extra-articular manifestations. Results: Altogether, 175 patients were on TNFis and 77 of them received at least two TNFis. The patients' age at the initiation of the first TNFi was higher among switchers compared to non-switchers (42.5 ± 12.6 vs. 38.8 ± 11.2 years, p = 0.049); otherwise, gender, disease duration and initial disease activity had no influence on the risk of switching. The decrease of the BASDAI was similar among non-switchers and switchers using either the first or second TNFi, but the response rates to the first and second TNFi were worse in switchers than in non-switchers. Following the failure of the first TNFi, the retention on therapy was unfavourable, especially in patients on infliximab after 1 year of treatment. The main reason for switching from the first drug was inefficacy. The frequency of side effects that led to switching was higher in the infliximab group than in patients treated with other agents. Conclusion: Although the retention rate to a second-line TNFi was somewhat worse than that to the first-line TNFi, the switching of TNFis is a good therapeutic option in AS patients who failed to respond to the first TNFi. © Springer-Verlag 2014
