    Quality assessment of truffle-inoculated seedlings in Italy: proposing revised parameters for certification

    Aim of study: the main aims of this study were to evaluate the quality of truffle-inoculated seedlings produced by commercial nurseries in Italy and to identify their minimum requisites in terms of plant age, health, homogeneity, and cut-off percentage of inoculated Tuber and non-Tuber ectomycorrhizae, based on the analysis of an extensive sample of seedlings subjected to quality control and certification.Area of study: truffle-inoculated seedlings produced by Italian commercial nurseries.Material and Methods: analysis of truffle-inoculated seedlings for health and quality standards; recording of presence of inoculated Tuber spp. and other concurrent fungi according to the official Italian method for certification; selective amplification of ectomycorrhizal DNA by PCR species-specific primers.Main results: We showed that mycorrhization levels in truffle-inoculated seedlings increased with time after truffle-spore inoculation. The highest mean percentage of the inoculated Tuber spp., but also the highest presence of contaminants, were recorded after three years. The mycorrhization level of Tuber melanosporum and T. aestivum was higher in Corylus and Ostrya seedlings than in Q. ilex and Q. pubescens, but the latter two host species showed the lowest presence of other ectomycorrhizal fungi. Mycorrhization level distribution in truffle-inoculated seedlings of suitable batches differed very little from the distribution in only all suitable seedlings. Truffle seedlings with other Tuber spp. were very few and even absent after three years.     Transverse dynamics of water across the melting point: A parallel neutron and x-ray inelastic scattering study

    Joint inelastic neutron and x-ray scattering measurements have been performed on heavy water across the melting point. The spectra bear clear evidence of low- and high-frequency inelastic shoulders related to transverse and longitudinal modes, respectively. Upon increasing the momentum transfer, the spectral shape evolves from a viscoelastic regime, where the low-frequency mode is clearly over-damped, toward an elastic one where its propagation becomes instead allowed. The crossover between the two regimes occurs whenever both the characteristic frequency and the linewidth of the low-frequency mode match the inverse of the structural relaxation time. Furthermore, we observe that the frequency of the transverse mode undergoes a discontinuity across the melting, whose extent reduces upon increasing the exchanged momentum

    Stimulated Brillouin scattering at 1 nm-1 wavevector by extreme ultraviolet transient gratings

    We crossed two femtosecond extreme ultraviolet (EUV) pulses in a beta - Ga2O3 (001) single crystal to create transient gratings (TG) of light intensity with sub-100 nm spatial periodicity. The EUV TG excitation launches phonon modes, whose dynamics were revealed via the backward diffraction of a third, time-delayed, EUV probe pulse. In addition to the modes typically observed in this kind of experiment, the phase-matching condition imposed by the TG, combined with the sharp penetration depth of the EUV excitation pulses, permitted to generate and detect phonons with a wavevector tangibly larger (approximately 1 nm-1) than the EUV TG one, via stimulated Brillouin back-scattering (SBBS) of the EUV probe. While SBBS of an optical probe was reported in previous EUV TG experiments, the extension of SBBS to short wavelength radiation can be used as a contact-less experimental tool for filling the gap between the wavevector range accessible through inelastic hard X-ray and thermal neutron scattering techniques, and the one accessible through Brillouin scattering of visible and UV light.Comment: 7 pages, 3 figure

    Signature of a polyamorphic transition in the THz spectrum of vitreous GeO2

    The THz spectrum of density fluctuations, S(Q,ω)S(Q, \omega), of vitreous GeO2_2 at ambient temperature was measured by inelastic x-ray scattering from ambient pressure up to pressures well beyond that of the known α\alpha-quartz to rutile polyamorphic (PA) transition. We observe significant differences in the spectral shape measured below and above the PA transition, in particular, in the 30-80 meV range. Guided by first-principle lattice dynamics calculations, we interpret the changes in the phonon dispersion as the evolution from a quartz-like to a rutile-like coordination. Notably, such a crossover is accompanied by a cusp-like behavior in the pressure dependence of the elastic response of the system. Overall, the presented results highlight the complex fingerprint of PA phenomena on the high-frequency phonon dispersion.Comment: 18 pages, 5 figure

    Analysis of CTNS gene transcripts in nephropathic cystinosis

    Nephropathic cystinosis (NC) is an autosomal recessive disorder caused by mutations of the CTNS gene that encodes for a cystine transmembrane transporter. Several mutations have been described in the coding and promoter regions of the CTNS gene in affected individuals. We selected three patients with NC from two unrelated families, in whom sequence analysis of the CTNS gene detected only one or no mutations. Total RNA was isolated from peripheral blood mononuclear cells or fibroblasts and CTNS transcripts were analyzed. We observed a skipping of exon 5 (85 bp) in two siblings and an intron 9 retention of 75 bp associated with partial replication of exon 9 in the third patient. Genomic DNA analysis of intron regions surrounding exon 5 showed a point mutation in the hypothetical lariat branch site of intron 4 at position –24 (c.141–24 T > C) in the first two patients and a duplication of 266 bp including a part of exon and intron 9 in the third patient. Analysis of CTNS gene transcripts allowed identification of mutations in patients in whom CTNS mutations could not be detected by traditional DNA sequencing. These results support the hypothesis that cystinosis is a monogenic disorder

    The newtonian approach in the meteorological tide waves forecasting: preliminary observations in the East Ligurian harbours

    Sea level oscillations are the superposition of many contributions, among which the main are astronomic and meteorological low-frequency tides. In Ligurian Sea meteo-tide components, being most ample than astronomic fluctuations, drive water exchange in harbours. The present note shows first results about port of Genoa concerning a coherency study between atmospheric variation and corresponding sea level adjustment (meteorological tide). The newtonian forecasting method of meteorological tides is based on measurements of time elapsing between barometric sea level unbalance (Δg) and its meteorological tide compensation (inverse barometer component). Meteorological tide component is independent on the Earth-Moon-Sun gravitational relationships, moreover parameters related to the shifted water mass are too many to describe the phenomenon analytically (basin topography, barometric strength position and time, chemical water quality, off-shore sea circulation, etc.); then, meteorological tide can’t be accurately foreseen by atmospheric pressure measurements only. A gravimeter can detect the geodetic unbalance starting time and a tide-gauge can detect the newtonian compensation (tide wave) coming time. The difference between these two times is the meteorological tide delay. An opportune statistic of this delay provides an experimental law typical for each harbour to forecast the meteo-tide compensation wave delay.     Biomarkers of aging in frailty and age-associated disorders: State of the art and future perspective

    According to the Geroscience concept that organismal aging and age-associated diseases share the same basic molecular mechanisms, the identification of biomarkers of age that can efficiently classify people as biologically older (or younger) than their chronological (i.e. calendar) age is becoming of paramount importance. These people will be in fact at higher (or lower) risk for many different age-associated diseases, including cardiovascular diseases, neurodegeneration, cancer, etc. In turn, patients suffering from these diseases are biologically older than healthy age-matched individuals. Many biomarkers that correlate with age have been described so far. The aim of the present review is to discuss the usefulness of some of these biomarkers (especially soluble, circulating ones) in order to identify frail patients, possibly before the appearance of clinical symptoms, as well as patients at risk for age-associated diseases. An overview of selected biomarkers will be discussed in this regard, in particular we will focus on biomarkers related to metabolic stress response, inflammation, and cell death (in particular in neurodegeneration), all phenomena connected to inflammaging (chronic, low-grade, age-associated inflammation). In the second part of the review, next-generation markers such as extracellular vesicles and their cargos, epigenetic markers and gut microbiota composition, will be discussed. Since recent progresses in omics techniques have allowed an exponential increase in the production of laboratory data also in the field of biomarkers of age, making it difficult to extract biological meaning from the huge mass of available data, Artificial Intelligence (AI) approaches will be discussed as an increasingly important strategy for extracting knowledge from raw data and providing practitioners with actionable information to treat patients

    Diabetes mellitus (DM) and Alzheimer’s disease (AD) are two highly prevalent conditions in the elderly population and major public health burden. In the past decades, a pathophysiological link between DM and AD has emerged and central nervous system insulin resistance might play a significant role as a common mechanism; however, other factors such as inflammation and oxidative stress seem to contribute to the shared pathophysiological link. Both preclinical and clinical studies have evaluated the possible neuroprotective mechanisms of different classes of antidiabetic medications in AD, with some promising results. Here, we review the evidence on the mechanisms of action of antidiabetic drugs and their potential use in AD

    Nanoscale transient polarization gratings

    We present the generation of transient polarization gratings at the nanoscale, achieved using a tailored accelerator configuration of the FERMI free electron laser. We demonstrate the capabilities of such a transient polarization grating by comparing its induced dynamics with the ones triggered by a more conventional intensity grating on a thin film ferrimagnetic alloy. While the signal of the intensity grating is dominated by the thermoelastic response of the system, such a contribution is suppressed in the case of the polarization grating. This exposes helicity-dependent magnetization dynamics that have so-far remained hidden under the large thermally driven response. We anticipate nanoscale transient polarization gratings to become useful for the study of any physical, chemical and biological systems possessing chiral symmetry
