    Cross-Talk between Protein Kinase A and Mitogen-Activated Protein Kinases Signalling in the Adaptive Changes Observed during Morphine Withdrawal in the Heart

    ABSTRACT Our previous studies have shown that morphine withdrawal induced an increase in the expression of protein kinase (PK) A and mitogen-activated extracellular kinase (MAPK) pathways in the heart during morphine withdrawal. The purpose of the present study was to evaluate the interaction between PKA and extracellular signal-regulated kinase (ERK) signaling pathways mediating the cardiac adaptive changes observed after naloxone administration to morphine-dependent rats. Dependence on morphine was induced by a 7-day subcutaneous implantation of morphine pellets. Morphine withdrawal was precipitated on day 8 by an injection of naloxone (2 mg/kg). ERK1/2 and tyrosine hydroxylase (TH) phosphorylation was determined by quantitative blot immunolabeling using phosphorylation statespecific antibodies. Naloxone-induced morphine withdrawal activates ERK1/2 and phosphorylates TH at Ser31 in the right and left ventricle, with an increase in the mean arterial blood pressure and heart rate. When N-(2-guanidinoethyl)-5-isoquinolinesulfonamide (HA-1004), a PKA inhibitor, was infused, concomitantly with morphine, it diminished the expression of ERK1/2. In contrast, the infusion of calphostin C (a PKC inhibitor) did not modify the morphine withdrawal-induced activation of ERK1/2. The ability of morphine withdrawal to activate ERK that phosphorylates TH at Ser31 was reduced by HA-1004. The present findings demonstrate that the enhancement of ERK1/2 expression and the phosphorylation state of TH at Ser31 during morphine withdrawal are dependent on PKA and suggest cross-talk between PKA and ERK1/2 transduction pathway mediating morphine withdrawal-induced activation (phosphorylation) of TH

    La presente publicación actualiza las estimaciones del Producto Bruto Geográfico (PBG) del Partido de General Pueyrredon a precios constantes, para la serie 2004-2012, así como el año 2012 a precios corrientes, lo que permite conocer el desempeño de la economía local a nivel agregado y por ramas de actividad, de tipo tradicional y no tradicional, observadas y no observadas. Los estudios precedentes, realizados en convenio entre la Facultad de Ciencias Económicas y Sociales de la Universidad Nacional de Mar del Plata y la Municipalidad de General Pueyrredon, son la estimación del PBG para el período 1993-1999 con año base 1993 y la estimación del año 2004, que sirve como base para este estudio. La importante tarea de recopilación de información estadística constituye un aporte para avanzar en la conformación de un sistema integrado de indicadores útil para la toma de decisiones de distintos actores, tanto públicos como privados. El Producto Bruto es el indicador de desempeño macroeconómico más conocido y utilizado y forma parte del denominado Sistema de Cuentas Nacionales, que constituyen una herramienta esencial para la evaluación, análisis y pronóstico de fenómenos económicos, al poner de relieve el tamaño y la estructura de una economía y todos los sectores que la componen. A nivel territorial, las Cuentas Regionales son un método de desglose de las Cuentas Nacionales que permiten conocer la estructura económica y analizar la evolución temporal de una economía en una jurisdicción geográfica determinada.Fil: Lacaze, María Victoria.     Use of barley straw residues to avoid high erosion and runoff rates on persimmon plantations in Eastern Spain under low frequency-high magnitude simulated rainfall events

    [EN] Soil and water losses due to agricultural mismanagement are high and non-sustainable in many orchards. An experiment was set up with rainfall simulation at 78 mm h(-1) over 1 hour on 20 paired plots of 2 m(2) (bare and straw covered) in new persimmon plantations in Eastern Spain. Effects of straw cover on the control of soil and water losses were assessed. An addition of 60% straw cover (75 gm(-2)) resulted in delayed ponding and runoff generation and consequently reduced water losses from 60% to 13% of total rainfall. The straw cover reduced raindrop impact and thus sediment detachment from 1014 to 47 g plot(-1) h(-1). The erosion rate was reduced from 5.1 to 0.2 Mg ha-(1) h(-1).     Carbonyl reductase 1 catalyzes 20β-reduction of glucocorticoids, modulating receptor activation and metabolic complications of obesity

    Carbonyl Reductase 1 (CBR1) is a ubiquitously expressed cytosolic enzyme important in exogenous drug metabolism but the physiological function of which is unknown. Here, we describe a role for CBR1 in metabolism of glucocorticoids. CBR1 catalyzes the NADPH-dependent production of 20 beta-dihydrocortisol (20 beta-DHF) from cortisol. CBR1 provides the major route of cortisol metabolism in horses and is up-regulated in adipose tissue in obesity in horses, humans and mice. We demonstrate that 20 beta-DHF is a weak endogenous agonist of the human glucocorticoid receptor (GR). Pharmacological inhibition of CBR1 in diet-induced obesity in mice results in more marked glucose intolerance with evidence for enhanced hepatic GR signaling. These findings suggest that CBR1 generating 20 beta-dihydrocortisol is a novel pathway modulating GR activation and providing enzymatic protection against excessive GR activation in obesity
