    Large Eddy Simulation of acoustic pulse propagation and turbulent flow interaction in expansion mufflers

    A novel hybrid pressure-based compressible solver is developed and validated for low Mach number acoustic flow simulation. The solver is applied to the propagation of an acoustic pulse in a simple expansion muffler, a configuration frequently employed in HVAC and automotive exhaust systems. A set of benchmark results for experimental analysis of the simple expansion muffler both with and without flow are obtained to compare attenuation in forced pulsation for various mean-flow velocities. The experimental results are then used for validation of the proposed pressure-based compressible solver. Compressible, Unsteady Reynolds Averaged Navier-Stokes (URANS) simulation of a muffler with a mean through flow is conducted and results are presented to demonstrate inherent limitations associated with this approach. Consequently, a mixed synthetic inflow boundary condition is developed and validated for compressible Large Eddy Simulation (LES) of channel flow. The mixed synthetic boundary is then employed for LES of a simple expansion muffler to analyse the flow-acoustic and acoustic-pulse interactions inside the expansion muffler. The improvement in the prediction of vortex shedding inside the chamber is highlighted in comparison to the URANS method. Further, the effect of forced pulsation on flow-acoustic is observed in regard to the shift in Strouhal number inside the simple expansion muffler

    Effects of technetium on marine micro-organisms

    Eleven bacterial species have been isolated from the upper layer of intertidal sediments collected along the Belgian coast (Coxyde). Three of them (no. 1, 4 and 11) have been chosen for their halophilous character. One species has been identified as Flavobacterium halmephilum, the other two are being investigated. Effects of technetium (99Tc) have been studied on a mixed bacterial population isolated from sediments, as well as on the three halophilic species.At the concentrations utilized in this work (up to 100 microg ml-1), 99Tc has no evident effects on bacterial growth. Uptake of technetium (99Tc and/or 95mTc) has been investigated in the mixed bacterial population, in the three halophilic bacteria (including Flavobacterium halmephilum) and in the benthic ciliate Uronema marinum. It has been found that technetium is taken up by all these micro-organisms. However, the transfer factor (TF) in bacteria may vary considerably (from 0.5 to 200), but the cause of this variability is not known and deserves further study.The ciliate Uronema marinum, which feeds on living marine bacteria, was found to take up 95mTc added to the culture medium. However, the TF in this ciliate is rather low (from 1.4 to 5.5). Because it feeds on bacteria, Uronema marinum is supposed to take up technetium from water (direct contamination) as well as from contaminated bacteria (indirect contamination). Experiments with 95mTc-labeled bacterial cells might be useful, as they could indicate which form of contamination (direct or indirect) is prevailing

    Using LES to Study Reacting Flows and Instabilities in Annular Combustion Chambers

    Great prominence is put on the design of aeronautical gas turbines due to increasingly stringent regulations and the need to tackle rising fuel prices. This drive towards innovation has resulted sometimes in new concepts being prone to combustion instabilities. In the particular field of annular combustion chambers, these instabilities often take the form of azimuthal modes. To predict these modes, one must compute the full combustion chamber, which remained out of reach until very recently and the development of massively parallel computers. Since one of the most limiting factors in performing Large Eddy Simulation (LES) of real combustors is estimating the adequate grid, the effects of mesh resolution are investigated by computing full annular LES of a realistic helicopter combustion chamber on three grids, respectively made of 38, 93 and 336 million elements. Results are compared in terms of mean and fluctuating fields. LES captures self-established azimuthal modes. The presence and structure of the modes is discussed. This study therefore highlights the potential of LES for studying combustion instabilities in annular gas turbine combustors

    Ecuador Paraiso Escondido virus, a new flavivirus isolated from New World sand flies in Ecuador, is the first representative of a novel clade in the genus flavivirus

    A new flavivirus, Ecuador Paraiso Escondido virus (EPEV), named after the village where it was discovered, was isolated from sand flies (Psathyromyia abonnenci, formerly Lutzomyia abonnenci) that are unique to the New World. This represents the first sand fly-borne flavivirus identified in the New World. EPEV exhibited a typical flavivirus genome organization. Nevertheless, the maximum pairwise amino acid sequence identity with currently recognized flaviviruses was 52.8%. Phylogenetic analysis of the complete coding sequence showed that EPEV represents a distinct clade which diverged from a lineage that was ancestral to the nonvectored flaviviruses Entebbe bat virus, Yokose virus, and Sokoluk virus and also the Aedes-associated mosquito-borne flaviviruses, which include yellow fever virus, Sepik virus, Saboya virus, and others. EPEV replicated in C6/36 mosquito cells, yielding high infectious titers, but failed to reproduce either in vertebrate cell lines (Vero, BHK, SW13, and XTC cells) or in suckling mouse brains. This surprising result, which appears to eliminate an association with vertebrate hosts in the life cycle of EPEV, is discussed in the context of the evolutionary origins of EPEV in the New World.The flaviviruses are rarely (if ever) vectored by sand fly species, at least in the Old World. We have identified the first representative of a sand fly-associated flavivirus, Ecuador Paraiso Escondido virus (EPEV), in the New World. EPEV constitutes a novel clade according to current knowledge of the flaviviruses. Phylogenetic analysis of the virus genome showed that EPEV roots the Aedes-associated mosquito-borne flaviviruses, including yellow fever virus. In light of this new discovery, the New World origin of EPEV is discussed together with that of the other flaviviruses

    The predictive and prognostic potential of plasma telomerase reverse transcriptase (TERT) RNA in rectal cancer patients

    Background: Preoperative chemoradiotherapy (CRT) followed by surgery is the standard care for locally advanced rectal cancer, but tumour response to CRT and disease outcome are variable. The current study aimed to investigate the effectiveness of plasma telomerase reverse transcriptase (TERT) levels in predicting tumour response and clinical outcome. Methods: 176 rectal cancer patients were included. Plasma samples were collected at baseline (before CRT\ubcT0), 2 weeks after CRT was initiated (T1), post-CRT and before surgery (T2), and 4\u20138 months after surgery (T3) time points. Plasma TERT mRNA levels and total cell-free RNA were determined using real-time PCR. Results: Plasma levels of TERT were significantly lower at T2 (Po0.0001) in responders than in non-responders. Post-CRT TERT levels and the differences between pre- and post-CRT TERT levels independently predicted tumour response, and the prediction model had an area under curve of 0.80 (95% confidence interval (CI) 0.73\u20130.87). Multiple analysis demonstrated that patients with detectable TERT levels at T2 and T3 time points had a risk of disease progression 2.13 (95% CI 1.10\u20134.11)-fold and 4.55 (95% CI 1.48\u201313.95)-fold higher, respectively, than those with undetectable plasma TERT levels. Conclusions: Plasma TERT levels are independent markers of tumour response and are prognostic of disease progression in rectal cancer patients who undergo neoadjuvant therapy

    Novel virus discovery and genome reconstruction from field RNA samples reveals highly divergent viruses in dipteran hosts.

    We investigated whether small RNA (sRNA) sequenced from field-collected mosquitoes and chironomids (Diptera) can be used as a proxy signature of viral prevalence within a range of species and viral groups, using sRNAs sequenced from wild-caught specimens, to inform total RNA deep sequencing of samples of particular interest. Using this strategy, we sequenced from adult Anopheles maculipennis s.l. mosquitoes the apparently nearly complete genome of one previously undescribed virus related to chronic bee paralysis virus, and, from a pool of Ochlerotatus caspius and Oc. detritus mosquitoes, a nearly complete entomobirnavirus genome. We also reconstructed long sequences (1503-6557 nt) related to at least nine other viruses. Crucially, several of the sequences detected were reconstructed from host organisms highly divergent from those in which related viruses have been previously isolated or discovered. It is clear that viral transmission and maintenance cycles in nature are likely to be significantly more complex and taxonomically diverse than previously expected

    Molecular evolution of the insect-specific flaviviruses

    There has been an explosion in the discovery of ‘insect-specific’ flaviviruses and/or their related sequences in natural mosquito populations. Herein we review all ‘insect-specific’ flavivirus sequences currently available and conduct phylogenetic analyses of both the ‘insect-specific’ flaviviruses and available sequences of the entire genus Flavivirus. We show that there is no statistical support for virus–mosquito co-divergence, suggesting that the ‘insect-specific’ flaviviruses may have undergone multiple introductions with frequent host switching. We discuss potential implications for the evolution of vectoring within the family Flaviviridae. We also provide preliminary evidence for potential recombination events in the history of cell fusing agent virus. Finally, we consider priorities and guidelines for future research on ‘insect-specific’ flaviviruses, including the vast potential that exists for the study of biodiversity within a range of potential hosts and vectors, and its effect on the emergence and maintenance of the flaviviruses
