    Como el control interno es una de las piezas clave para la gestión eficiente, eficaz y transparente de los bienes públicos, tenemos la intención de analizar los procedimientos de control interno adoptados en la sección de almacenamiento y suministro en los municipios de Trás-os-Montes y Alto Douro. En este sentido, se estudió una muestra de 10 ayuntamientos de los distritos de Vila Real y Bragança, preguntándole a los dirigentes de la sección a través de una encuesta, de febrero a marzo de 2011. Se encontraron diferencias significativas en las dimensiones relativas a los documentos/información de requisición y los procedimientos relativos a la devolución de hoja de pedido. Por lo tanto, hemos identificado la necesidad de mejorar la aplicación de procedimientos de control interno en varias dimensiones, excepto en los procedimientos de compromiso, el cambio de las condiciones concertadas en la hoja de pedido y en los procedimientos del almacenamiento, posibilitando de esa forma el cumplimiento de las normas de la contabilidad pública resultadas del POCAL (Decreto-Ley n.º 54-A/99, 22 de febrero)

    Study of the stress intensity factors in the bulk of the material with synchrotron diffraction

    Artículo de Proceedings de Congreso Internacional Fatigue2017In this work we present the results of a hybrid experimental and analytical approach for estimating the stress intensity factor. It uses the elastic strains within the bulk obtained by synchrotron X-ray diffraction data. The stress intensity factor is calculated using a multi-point overdeterministic method where the number of experimental data points is higher than the number of unknowns describing the elastic field surrounding the crack-tip. The tool is tested on X-ray strain measurements collected on a bainitic steel.     Heme metabolism, oxidative and nitrosative markers in a mouse model of Hemochromatosis: Effect of Isoflurane, ethanol and 5-aminolevulinic acid

    Hereditary hemochromatosis (HH) is characterized by iron homeostasis alterations. Association between HH and Porphyria Cutanea Tarda has been reported. The aim was to characterize oxidative and nitrosative stress status and its relationship with heme metabolism in a hemochromatosis mouse model (Hfe-/-), and to evaluate the effects of Isoflurane, ethanol and 5-aminolevulinic acid (ALA). Male and female Hfe−/− and wild-type C57BL/6J mice received Isoflurane (2 ml/kg); ethanol (30%) or ALA (40 mg/kg). In male Hfe-/-, reduced glutathione (GSH) was diminished respect to C57BL/6J mice. Female Hfe-/- showed higher levels of GSH and total antioxidant capacity than male Hfe-/-. Catalase activity was lower in male and female Hfe-/- than in controls. 5-Aminolevulinic acid synthetase activity was higher in male and female Hfe-/- than in controls. In male Hfe-/-, Porphobilinogen deaminase (PBG-D) activity was augmented and Heme oxygenase (HO) activity was diminished probably to avoid iron increase. Isoflurane and ethanol reduced PBG-D and increased HO activities. HO induction would indicate oxidative stress instauration being more striking due to ethanol that also induced Superoxide dismutase activity. Isoflurane reduced Nitric Oxide Synthase expression. ALA altered antioxidant system. In Hfe-/- mice different metabolisms were altered being more affected by the drugs studied. Findings here described would contribute to increase the knowledge about the association between HH and the Porphyrias and about the effects of volatile anaesthetics on different metabolisms in genetic models of Porphyrias and associated diseases.Fil: Buzaleh, Ana Maria.     Adaptation and convergence in circadian‐related genes in Iberian freshwater fish

    The circadian clock is a biological timing system that improves the ability of organisms to deal with environmental fluctuations. At the molecular level it consists of a network of transcription-translation feedback loops, involving genes that activate (bmal and clock - positive loop) and repress expression (cryptochrome (cry) and period (per) - negative loop). This is regulated by daily alternations of light but can also be affected by temperature. Fish, as ectothermic, depend on the environmental temperature and thus are good models to study its integration within the circadian system. Here, we studied the molecular evolution of circadian genes in four Squalius freshwater fish species, distributed across Western Iberian rivers affected by two climatic types with different environmental conditions (e.g., light and temperature). S. carolitertii and S. pyrenaicus inhabit the colder northern region under Atlantic climate type, while S. torgalensis, S. aradensis and some populations of S. pyrenaicus inhabit the warmer southern region affected by summer droughts, under Mediterranean climate type.info:eu-repo/semantics/publishedVersio

    Understanding the biophysical interaction of LTX-315 with tumoral model membranes

    Host defense peptides are found primarily as natural antimicrobial agents among all lifeforms. These peptides and their synthetic derivatives have been extensively studied for their potential use as therapeutic agents. The most accepted mechanism of action of these peptides is related to a nonspecific mechanism associated with their interaction with the negatively charged groups present in membranes, inducing bilayer destabilization and cell death through several routes. Among the most recently reported peptides, LTX-315 has emerged as an important oncolytic peptide that is currently in several clinical trials against different cancer types. However, there is a lack of biophysical studies regarding LTX-315 and its interaction with membranes. This research focuses primarily on the understanding of the molecular bases of LTX-315′s interaction with eukaryotic lipids, based on two artificial systems representative of non-tumoral and tumoral membranes. Additionally, the interaction with individual lipids was studied by differential scanning calorimetry and Fourier-transformed infrared spectroscopy. The results showed a strong interaction of LTX-315 with the negatively charged phosphatidylserine. The results are important for understanding and facilitating the design and development of improved peptides with anticancer activity

However, PQ repair is difficult when using conventional approach, which is necessary to release PQ for volar plate fixation of distal radius fracture. In addition, anatomical reduction is difficult in patients with distal radius metaphyseal comminution fracture. Therefore, the purpose of this study is to compare the results of volar locking plate fixation using PQ sparing technique and conventional volar approach technique for patients with distal radius metaphyseal comminution fracture

    Calcium-dependent mitochondrial permeability transition is augmented in the kidney of Goto-Kakizaki diabetic rat

    Renal disease associated with diabetes mellitus is a major problem among diabetic patients. The role of mitochondria in the pathogenesis of diabetes has received a large amount of attention in the last years, but many aspects of this subject are still poorly understood. In the present study, we studied the susceptibility of the mitochondrial permeability transition (MPT) on kidney mitochondria from the Goto-Kakizaki (GK) rat, an animal model featuring physiological and pathological alterations characteristic of type 2 diabetes.Kidney mitochondria were isolated by differential centrifugations; mitochondrial electric transmembrane potential and calcium loading capacity were evaluated with a TPP+-selective electrode and with a calcium-sensitive fluorescent probe. Coenzyme Q9, Q10 and vitamin E were evaluated by high-performance liquid chromatography (HPLC).Kidney mitochondria from the diabetic animals had an increased susceptibility to the induction of the MPT by calcium. We observed a loss of calcium-loading capacity and a higher calcium-induced mitochondrial depolarization. Vitamin E and coenzyme Q9 were also increased in kidney mitochondria from GK rats.The results show an enhanced MPT activation in kidney mitochondria from GK rats, which lead us to suggest that this condition may be one major alteration triggered by chronic diabetes in kidney cells, ultimately leading to cell dysfunction.     BSA-PEG Hydrogel: A Novel Protein-Ligand Binding 3D Matrix

    Hydrogel materials have good biomimetic properties and high potential for biomedical and bioanalytical applications. In this work, a hydrogel of serum albumin crosslinked with poly-(ethylene glycol) was prepared and characterized for its water content, protein structure and stability. The ability of the hydrogel to bind small molecule ligands with different hydrophobicity was evaluated using a homologous series of amphiphiles (NBD-Cn, n = 4, 6 and 8) and the calculated binding affinities were similar to that of free protein in solution.     Changes in soil biological properties in different management and tyllage systems in petrocalcic argiudoll

    We analyzed the effect of different tillage systems on soils under different land-use histories, on biological properties of soil during one year. The experiment was carried out at a Petrocalcic Argiudoll of Tres Arroyos (Buenos Aires, Argentina). The specific aim was to describe and compare the soil organic carbon (SOC), the soil basal respiration (BR) and the activities of the enzyme dehydrogenase, urease and acid-phosphomonoesterase under zero and conventional tillage on soils under pasture and intensive agriculture. The SOC concentration was highest in summer (postharvest) independently of tillage system or land-use history. However, in autumn the plots under conventional tillage showed higher values of SOC than those with zero tillage, independently of land-use history. The BR had a significant benefit in favour of summer pasture soils. The effect of land-use history or the tillage system on the enzymes activity was dependent of sampling season. Therefore, the soil enzymes were more sensible than SOC and BR.
