    Cílem diplomové práce je vyhodnotit nástroje personálního marketingu pro omezení fluktuace ve vybrané organizaci. Teoretická část vychází z teoretických východisek personálního marketingu a fluktuace zaměstnanců. Praktická část se zabývá analýzou současného stavu personálního marketingu a analýzou fluktuace zaměstnanců, analyzuje výsledky dotazníkového šetření spokojenosti zaměstnanců. Na závěr jsou navržená opatření, která vedou ke snížení fluktuace a ke spokojenosti zaměstnanců a jsou vyhodnoceny přínosy těchto opatření pro organizaci.The aim of the diploma thesis is to evaluate the tools of personnel marketing to reduce turnover in the selected organization. The theoretical part is based on the theoretical basis of personnel marketing and employee turnover. The practical part deals with the analysis of the current state of personnel marketing and analysis of employee turnover, analyzes the results of a questionnaire survey of employee satisfaction.     Tato práce se zabývá tím, jaký podíl má glotalizace na cizineckém přízvuku. Důraz klade na české mluvčí angličtiny a hodnocení jejich promluv. Mluvčí, jejichž promluvy byly pro účely této práce analyzovány, byli předem ohodnoceni percepčním testem. Podle výsledků tohoto testu byli rozděleni do tří skupin: A, která obsáhla mluvčí hovořící angličtinou blížící se rodilému standardu; C, do které byli naopak zařazeni mluvčí se silným českým přízvukem; a B, do které spadají mluvčí s detekovatelným cizineckým přízvukem, jenž však není hodnocen jako český. Úkolem této práce je zjistit, jakým způsobem používají mluvčí každé skupiny glotalizaci a zda se toto použití shoduje s předpoklady vycházejícími z dosavadního výzkumu.In this paper, we are interested in what the part of glottalization is in a foreign accent. We focus on Czech speakers of English and an evaluation of their utterances. Those speakers analyzed for the purposes of this paper were evaluated in advance by a perceptual test. In accordance with the results of the test, they were divided into three groups: A, containing those speakers with an English close to the native standard; C, containing speakers with a strong Czech accent; and B, containing speakers with a detectable foreign accent which, however, is not evaluated as a Czech one. In this chapter, the individual styles and typical elements of garden architecture from the oldest medieval gardens around the castles and monasteries, through the Renaissance and Baroque ornamental gardens around thy typical Renaissance villas and chateaus to the landscape parks of classicism and romanticism and modern gardening, will be described. For each of the key periods, a few typical representatives of ornamental gardens from the countryside of the South Bohemian region will be described, including their original and contemporary composition, composition of wood and plants and the buildings and furniture in the garden locality

    The Role of Public Spaces in Small Municipality

    Public spaces are a subspace of municipality space: they are its physical type, closely linked to permanent settlements, especially to cities. There are both social communication and movement of people, things and goods. These are classic, usually architecturally designed spaces between buildings: squares, agora, streets and parks. Public spaces in small municipalities have been crucial to the functioning of the community, their social, historically, and also economic life. In various types of rural municipalities, the function and formation of public spaces have changed over the course of history, often in relation to the geographical location of the municipality in terms of location or local conditions and customs. Nowadays, the tourist attraction of the place is also an important driving element in the form of public spaces. This chapter should show how public spaces have changed over time using examples of different types of municipalities and show examples of good and somewhat worse care for them

    Virion structure and genome delivery mechanism of sacbrood honeybee virus

    Infection by sacbrood virus (SBV) from the family Iflaviridae is lethal to honey bee larvae but only rarely causes the collapse of honey bee colonies. Despite the negative effect of SBV on honey bees, the structure of its particles and mechanism of its genome delivery are unknown. Here we present the crystal structure of SBV virion and show that it contains 60 copies of a minor capsid protein (MiCP) attached to the virion surface. No similar MiCPs have been previously reported in any of the related viruses from the order Picornavirales. The location of the MiCP coding sequence within the SBV genome indicates that the MiCP evolved from a C-terminal extension of a major capsid protein by the introduction of a cleavage site for a virus protease. The exposure of SBV to acidic pH, which the virus likely encounters during cell entry, induces the formation of pores at threefold and fivefold axes of the capsid that are 7 angstrom and 12 angstrom in diameter, respectively. This is in contrast to vertebrate picornaviruses, in which the pores along twofold icosahedral symmetry axes are currently considered the most likely sites for genome release. SBV virions lack VP4 subunits that facilitate the genome delivery of many related dicistroviruses and picornaviruses. MiCP subunits induce liposome disruption in vitro, indicating that they are functional analogs of VP4 subunits and enable the virus genome to escape across the endosome membrane into the cell cytoplasm

    Expression patterns of microRNAs associated with CML phases and their disease related targets

    <p>Abstract</p> <p>Background</p> <p>MicroRNAs are important regulators of transcription in hematopoiesis. Their expression deregulations were described in association with pathogenesis of some hematological malignancies. This study provides integrated microRNA expression profiling at different phases of chronic myeloid leukemia (CML) with the aim to identify microRNAs associated with CML pathogenesis. The functions of <it>in silico </it>filtered targets are in this report annotated and discussed in relation to CML pathogenesis.</p> <p>Results</p> <p>Using microarrays we identified differential expression profiles of 49 miRNAs in CML patients at diagnosis, in hematological relapse, therapy failure, blast crisis and major molecular response. The expression deregulation of miR-150, miR-20a, miR-17, miR-19a, miR-103, miR-144, miR-155, miR-181a, miR-221 and miR-222 in CML was confirmed by real-time quantitative PCR. <it>In silico </it>analyses identified targeted genes of these miRNAs encoding proteins that are involved in cell cycle and growth regulation as well as several key signaling pathways such as of mitogen activated kinase-like protein (MAPK), epidermal growth factor receptor (EGFR, ERBB), transforming growth factor beta (TGFB1) and tumor protein p53 that are all related to CML. Decreased levels of miR-150 were detected in patients at diagnosis, in blast crisis and 67% of hematological relapses and showed significant negative correlation with miR-150 proved target <it>MYB </it>and with <it>BCR-ABL </it>transcript level.</p> <p>Conclusions</p> <p>This study uncovers microRNAs that are potentially involved in CML and the annotated functions of <it>in silico </it>filtered targets of selected miRNAs outline mechanisms whereby microRNAs may be involved in CML pathogenesis.</p

    The current views on the Roma expressed by Czechprimary and secondary school teachers In this article, we deal with the topic ofattitudes towards the Roma minority expressed byprimary and secondary school teachers. We presentthe findings of our research carried out in 2014–2015.First, we introduce the basic theoretical concepts behind the term “multiculturalism” as well as the broadterm “Roma identity”. We then describe our methodology and research techniques. In the central part ofour paper, we focus our interpretation on the specificways teachers perceive the Roma minority, such as theascription of characteristics or traits. Their perceptionis usually influenced by personal experience with Romastudents. We presume that the teachers’ attitudes caninfluence the attitudes of their students. We will alsoshow that their negative approach towards the Romacan be transferred onto the students in the form ofmisrepresentations and stereotyping

    Characterization of insulin crystalline form in isolated β-cell secretory granules

    Insulin is stored in vivo inside the pancreatic β-cell insulin secretory granules. In vitro studies have led to an assumption that high insulin and Zn2+ concentrations inside the pancreatic β-cell insulin secretory granules should promote insulin crystalline state in the form of Zn2+-stabilized hexamers. Electron microscopic images of thin sections of the pancreatic β-cells often show a dense, regular pattern core, suggesting the presence of insulin crystals. However, the structural features of the storage forms of insulin in native preparations of secretory granules are unknown, because of their small size, fragile character and difficult handling. We isolated and investigated the secretory granules from MIN6 cells under near-native conditions, using cryo-electron microscopic (Cryo-EM) techniques. The analysis of these data from multiple intra-granular crystals revealed two different rhomboidal crystal lattices. The minor lattice has unit cell parameters (a ≃ b ≃ 84.0 Å, c ≃ 35.2 Å), similar to in vitro crystallized human 4Zn2+-insulin hexamer, whereas the largely prevalent unit cell has more than double c-axis (a ≃ b ≃ c ≃ 96.5 Å) that probably corresponds to two or three insulin hexamers in the asymmetric unit. Our experimental data show that insulin can be present in pancreatic MIN6 cell granules in a microcrystalline form, probably consisting of 4Zn2+-hexamers of this hormone
