    Introduction ~Considering that the human intestine is home to almost 100 trillion microorganisms including bacteria (Cryan, 2011), recent research has focused on the role of the microbiome in neurobiological functions such as stress, anxiety and coping responses. ~Focusing on animal models, previous findings indicate that modifications of the gut microbiota via antibiotics and certain probiotics alter the anxiety response via the vagus nerve & immune system mediation (MacQueen et al., 2017). ~Recently, the concept of Psychobiotics has been introduced to refer to the use of microbiota to positively influence mental health outcomes (Foster et al., 2017)

    Enriched Environment Exposure Enhances Social Interactions and Oxytocin Responsiveness in Male Long-Evans Rats

    Both social and physical stimuli contribute to the complexity of an animal’s environment, influencing biobehavioral responses to subsequent challenges. In the current study, male Long-Evans rats were randomly assigned to an isolate (ISO), social control (SC) or social enriched (SE) group (n = 8 per group). The SC and SE conditions were group housed with the SE group exposed to physical enrichment stimuli that were natural as opposed to manufactured (e.g., hollowed out log instead of plastic hiding place). On three occasions during their 40-day enriched environment exposure, night/dark phase videos were obtained for 1 h during the early part of the dark phase. During this time, the SE animals exhibited significantly more social grooming with no differences between the SE and SC in the frequency of play or self-grooming bouts. Subsequently, all animals were assessed in social interaction and problem-solving escape tasks during the last week of the enriched environment exposure. SE rats exhibited increased digging bouts toward the restrained conspecific in the social interaction task whereas the other groups exhibited more escape responses. In the problem-solving task, SE animals exhibited a decreased latency to cross the barrier to escape from the predator odor (i.e., cat urine and fur). Neural analyses indicated increased oxytocin-immunoreactive (OT-ir) tissue in the SE supraoptic and paraventricular nuclei of the hypothalamus compared to the other groups. Interestingly, blood samples indicated lower peripheral corticosterone (CORT) and higher OT levels in the ISO animals when compared to the SC and SE animals, an effect retrospectively attributed to separation anxiety in the SE and SC animals in preparation for histology procedures. When the behavioral, neural and endocrine data were visualized as a multifaceted dataset via a multidimensional scaling analysis, however, an association between social enrichment and higher OT involvement was observed in the SE animals, as well as heightened stress responsivity in the ISO and SC groups. In sum, the SE animals exhibited a facilitation of social responses, problem-solving ability and OT immunoreactive responsiveness. These findings provide new information about the influences of both physical and social stimuli in dynamic and enriched environments

    Analytical Representations for Characterizing the Global Aviation Radiation Environment Based on Model and Measurement Databases

    The Nowcast of Atmospheric Ionizing Radiation for Aviation Safety climatological model and the Automated Radiation Measurements for Aerospace Safety (ARMAS) statistical database are presented as polynomial fit equations. Using equations based on altitude, L shell, and geomagnetic conditions an effective dose rate for any location from a galactic cosmic ray (GCR) environment can be calculated. A subset of the ARMAS database is represented by a second polynomial fit equation for the GCR plus probable relativistic energetic particle (REP; Van Allen belt REP) effective dose rates within a narrow band of L shells with altitudinal and geomagnetic dependency. Solar energetic particle events are not considered in this study since our databases do not contain these events. This work supports a suggestion that there may be a REP contribution having an effect at aviation altitudes. The ARMAS database is rich in Western Hemisphere observations for L shells between 1.5 and 5; there have been many cases of enhanced radiation events possibly related to effects from radiation belt particles. Our work identifies that the combined effects of an enhanced radiation environment in this L shell range are typically 15% higher than the GCR background. We also identify applications for the equations representing the Nowcast of Atmospheric Ionizing Radiation for Aviation Safety and ARMAS databases. They include (i) effective dose rate climatology in comparison with measured weather variability and (ii) climatological and statistical weather nowcasting and forecasting. These databases may especially help predict the radiation environment for regional air traffic management, for airport overflight operations, and for air carrier route operations of individual aircraft

    Cardio-Facio-Cutaneous Syndrome: Clinical Features, Diagnosis, and Management Guidelines

    Cardio-facio-cutaneous syndrome (CFC) is one of the RASopathies that bears many clinical features in common with the other syndromes in this group, most notably Noonan syndrome and Costello syndrome. CFC is genetically heterogeneous and caused by gene mutations in the Ras/mitogen-activated protein kinase pathway. the major features of CFC include characteristic craniofacial dysmorphology, congenital heart disease, dermatologic abnormalities, growth retardation, and intellectual disability. It is essential that this condition be differentiated from other RASopathies, as a correct diagnosis is important for appropriate medical management and determining recurrence risk. Children and adults with CFC require multidisciplinary care from specialists, and the need for comprehensive management has been apparent to families and health care professionals caring for affected individuals. To address this need, CFC International, a nonprofit family support organization that provides a forum for information, support, and facilitation of research in basic medical and social issues affecting individuals with CFC, organized a consensus conference. Experts in multiple medical specialties provided clinical management guidelines for pediatricians and other care providers.     Neuronally enriched microvesicle RNAs are differentially expressed in the serums of Parkinson’s patients

    BackgroundCirculating small RNAs (smRNAs) originate from diverse tissues and organs. Previous studies investigating smRNAs as potential biomarkers for Parkinson’s disease (PD) have yielded inconsistent results. We investigated whether smRNA profiles from neuronally-enriched serum exosomes and microvesicles are altered in PD patients and discriminate PD subjects from controls.MethodsDemographic, clinical, and serum samples were obtained from 60 PD subjects and 40 age- and sex-matched controls. Exosomes and microvesicles were extracted and isolated using a validated neuronal membrane marker (CD171). Sequencing and bioinformatics analyses were used to identify differentially expressed smRNAs in PD and control samples. SmRNAs also were tested for association with clinical metrics. Logistic regression and random forest classification models evaluated the discriminative value of the smRNAs.ResultsIn serum CD171 enriched exosomes and microvesicles, a panel of 29 smRNAs was expressed differentially between PD and controls (false discovery rate (FDR) < 0.05). Among the smRNAs, 23 were upregulated and 6 were downregulated in PD patients. Pathway analysis revealed links to cellular proliferation regulation and signaling. Least absolute shrinkage and selection operator adjusted for the multicollinearity of these smRNAs and association tests to clinical parameters via linear regression did not yield significant results. Univariate logistic regression models showed that four smRNAs achieved an AUC ≥ 0.74 to discriminate PD subjects from controls. The random forest model had an AUC of 0.942 for the 29 smRNA panel.ConclusionCD171-enriched exosomes and microvesicles contain the differential expression of smRNAs between PD and controls. Future studies are warranted to follow up on the findings and understand the scientific and clinical relevance

    Forces Shaping the Fastest Evolving Regions in the Human Genome

    Comparative genomics allow us to search the human genome for segments that were extensively changed in the last ~5 million years since divergence from our common ancestor with chimpanzee, but are highly conserved in other species and thus are likely to be functional. We found 202 genomic elements that are highly conserved in vertebrates but show evidence of significantly accelerated substitution rates in human. These are mostly in non-coding DNA, often near genes associated with transcription and DNA binding. Resequencing confirmed that the five most accelerated elements are dramatically changed in human but not in other primates, with seven times more substitutions in human than in chimp. The accelerated elements, and in particular the top five, show a strong bias for adenine and thymine to guanine and cytosine nucleotide changes and are disproportionately located in high recombination and high guanine and cytosine content environments near telomeres, suggesting either biased gene conversion or isochore selection. In addition, there is some evidence of directional selection in the regions containing the two most accelerated regions. A combination of evolutionary forces has contributed to accelerated evolution of the fastest evolving elements in the human genome

    NASA's Soil Moisture Active and Passive (SMAP) Mission

    The Soil Moisture Active-Passive (SMAP) Mission is one of the first Earth observation satellites being formulated by NASA in response to the 2007 National Research Council s Decadal Survey. SMAP will make global measurements of soil moisture at the Earth's land surface and its freeze-thaw state. These measurements will allow significantly improved estimates of water, energy and carbon transfers between the land and atmosphere. Soil moisture measurements are also of great importance in assessing flooding and monitoring drought. Knowledge gained from SMAP observations can help mitigate these natural hazards, resulting in potentially great economic and social benefits. SMAP observations of soil moisture and freeze/thaw timing over the boreal latitudes will also reduce a major uncertainty in quantifying the global carbon balance and help to resolve an apparent missing carbon sink over land. The SMAP mission concept will utilize an L-band radar and radiometer sharing a rotating 6-meter mesh reflector antenna flying in a 680 km polar orbit with an 8-day exact ground track repeat aboard a 3-axis stabilized spacecraft to provide high-resolution and high-accuracy global maps of soil moisture and freeze/thaw state every two to three days. In addition, the SMAP project will use these surface observations with advanced modeling and data assimilation to provide estimates of deeper root-zone soil moisture and net ecosystem exchange of carbon. SMAP recently completed its Phase A Mission Concept Study Phase for NASA and transitioned into Phase B (Formulation and Detailed Design). A number of significant accomplishments occurred during this initial phase of mission development. The SMAP project held several open meetings to solicit community feedback on possible science algorithms, prepared preliminary draft Algorithm Theoretical Basis Documents (ATBDs) for each mission science product, and established a prototype algorithm testbed to enable testing and evaluation of the performance of candidate algorithms. SMAP conducted an Applications Workshop in September 2009 to coordinate with potential application users interested in the mission data. A draft Applications Plan describing the Project s planned outreach to potential applications users has been prepared and will be updated during Phase B. SMAP made a significant evaluation of the potential terrestrial radio frequency interference (RFI) source environment and established radiometer and radar flight hardware and ground processing mitigation approaches. SMAP finalized its science orbit and orbit injection approach to optimize launch mass and prepared launch and commissioning scenarios and timeline. A science data communications approach was developed to maximize available science data volume to improve science margins while maintaining moderately short data product latencies to support many potential applications using existing ground assets and with minimum impact to the flight system. SMAP developed rigid multi-body and flexible body dynamics and control models and system designs for the 6-meter rotating instrument reflector-boom assembly (RBA) and flight system to confirm pointing and control performance, and devised strategies to efficiently implement on-orbit balancing if needed. Industry partners were selected for the spin mechanism assembly (SMA) and RBA. Preliminary designs for the radar and radiometer were initiated, including constructing breadboards of key assemblies
