Validation of a method to elute viruses from different types of face masks
Due to the SARS-CoV-2 pandemic, it is crucial to study the efficiency of face masks in retaining viruses for the upcoming years. The first objective of this study was to validate a method to elute viruses from polyester and cotton face masks. We observed that deionized water followed by 3% beef glycine (pH 9.5 or pH 7.2) was significantly more efficient (p < 0.05) in eluting the bacteriophage phiX174 virus from polyester (4.73% ± 0.25% to 28.67% ± 1.89%), polyester/cotton (3% ± 0.33%), and cotton (1.7% ± 0.21%) face masks than 3% beef glycine only (pH 9.5 or pH 7.2) as a single eluent (3.4% ± 0.16% to 21.33% ± 0.94% for polyester, 1.91% ± 0.08% for polyester/cotton, and 1.47% ± 0.12% for cotton face masks). Also, deionized water was significantly less efficient as a single eluent for eluting bacteriophage phiX174 from all the studied face mask types. The polyethylene glycol (PEG) precipitation method was substantially more efficient (p < 0.05) as a second step concentration method for the viruses in the eluates than the organic flocculation (OF) method. Higher viral loads were eluted from polyester face masks than cotton ones. We also found varying viral loads in the eluate solutions from different commercial polyester face masks, with the highest percentage seen for the N95 face mask. The second objective was to apply the validated method to study the effect of autoclaving on the different face mask materials. Results of the study did not show any significant differences in the viral loads eluted from the studied face masks before and after one and five autoclaving cycles. Moreover, a scanning electron microscope (SEM) analysis revealed no changes in the yarns, elongation, tensile strength, and contact angle measurements of the polyester or cotton materials after one or five autoclaving cycles
Case Study in Refractory Non-Hodgkin's Lymphoma: Successful Treatment with Plerixafor
The present case study describes our experience in treating a young woman diagnosed with a relapsing case of diffuse large cell lymphoma, who was heavily pre-treated with chemotherapy and radiotherapy. Our only chance to improve her survival was by using high-dose chemotherapy, followed by peripheral stem cell rescue. Unfortunately, in this patient, collecting sufficient stem cells for bone marrow transplantation proved to be very difficult since she had already been heavily treated with chemotherapy and radiotherapy. Currently, granulocyte colony-stimulating factor (G-CSF) alone or G-CSF plus chemotherapy are the most commonly used treatments for stem cell mobilization. However, 5–30% of patients do not respond to these agents. Plerixafor is a new hematopoietic stem cell-mobilizing drug that antagonizes the binding of chemokine stromal cell-derived factor-1α to CXC chemokine receptor 4. It is indicated in combination with G-CSF to mobilize hematopoietic stem cells to the peripheral blood for collection and subsequent autologous transplantation in patients with non-Hodgkin's lymphoma and multiple myeloma [Kessans et al.: Pharmacotherapy 2010;30:485–492; Jantunen: Expert Opin Biol Ther 2011;11:1241–1248]. Based on our findings, we consider plerixafor to be a very efficient and practical solution to mobilize and collect stem cells among all patients in such a situation, enabling us to proceed to autologous bone marrow transplantation and peripheral stem cell rescue in order to improve the patients’ overall survival
Improvement of renal oxidative stress markers after ozone administration in diabetic nephropathy in rats
<p>Abstract</p> <p>Background</p> <p>Several complications of diabetes mellitus (DM) e.g. nephropathy (DN) have been linked to oxidative stress. Ozone, by means of oxidative preconditioning, may exert its protective effects on DN.</p> <p>Aim</p> <p>The aim of the present work is to study the possible role of ozone therapy in ameliorating oxidative stress and inducing renal antioxidant defence in streptozotocin (STZ)-induced diabetic rats.</p> <p>Methods</p> <p>Six groups (n = 10) of male Sprague Dawley rats were used as follows: Group C: Control group. Group O: Ozone group, in which animals received ozone intraperitoneally (i.p.) (1.1 mg/kg). Group D: Diabetic group, in which DM was induced by single i.p. injections of streptozotocin (STZ). Group DI: Similar to group D but animals also received subcutaneous (SC) insulin (0.75 IU/100 gm BW.). Group DO: In which diabetic rats received the same dose of ozone, 48 h after induction of diabetes. Group DIO, in which diabetic rats received the same doses of insulin and ozone, respectively. All animals received daily treatment for six weeks. At the end of the study period (6 weeks), blood pressure, blood glycosylated hemoglobin (HbA<sub>1c</sub>), serum creatinine, blood urea nitrogen (BUN), kidney tissue levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxide (GPx), aldose reductase (AR) activities and malondialdehyde (MDA) concentration were measured.</p> <p>Results</p> <p>Induction of DM in rats significantly elevated blood pressure, HbA<sub>1c</sub>, BUN, creatinine and renal tissue levels of MDA and AR while significantly reducing SOD, CAT and GPx activities. Either Insulin or ozone therapy significantly reversed the effects of DM on all parameters; in combination (DIO group), they caused significant improvements in all parameters in comparison to each alone.</p> <p>Conclusions</p> <p>Ozone administration in conjunction with insulin in DM rats reduces oxidative stress markers and improves renal antioxidant enzyme activity which highlights its potential uses in the regimen for treatment of diabetic patients.</p
Surgical site infection after gastrointestinal surgery in high-income, middle-income, and low-income countries: a prospective, international, multicentre cohort study
Background: Surgical site infection (SSI) is one of the most common infections associated with health care, but its importance as a global health priority is not fully understood. We quantified the burden of SSI after gastrointestinal surgery in countries in all parts of the world.
Methods: This international, prospective, multicentre cohort study included consecutive patients undergoing elective or emergency gastrointestinal resection within 2-week time periods at any health-care facility in any country. Countries with participating centres were stratified into high-income, middle-income, and low-income groups according to the UN's Human Development Index (HDI). Data variables from the GlobalSurg 1 study and other studies that have been found to affect the likelihood of SSI were entered into risk adjustment models. The primary outcome measure was the 30-day SSI incidence (defined by US Centers for Disease Control and Prevention criteria for superficial and deep incisional SSI). Relationships with explanatory variables were examined using Bayesian multilevel logistic regression models. This trial is registered with ClinicalTrials.gov, number NCT02662231.
Findings: Between Jan 4, 2016, and July 31, 2016, 13 265 records were submitted for analysis. 12 539 patients from 343 hospitals in 66 countries were included. 7339 (58·5%) patient were from high-HDI countries (193 hospitals in 30 countries), 3918 (31·2%) patients were from middle-HDI countries (82 hospitals in 18 countries), and 1282 (10·2%) patients were from low-HDI countries (68 hospitals in 18 countries). In total, 1538 (12·3%) patients had SSI within 30 days of surgery. The incidence of SSI varied between countries with high (691 [9·4%] of 7339 patients), middle (549 [14·0%] of 3918 patients), and low (298 [23·2%] of 1282) HDI (p < 0·001). The highest SSI incidence in each HDI group was after dirty surgery (102 [17·8%] of 574 patients in high-HDI countries; 74 [31·4%] of 236 patients in middle-HDI countries; 72 [39·8%] of 181 patients in low-HDI countries). Following risk factor adjustment, patients in low-HDI countries were at greatest risk of SSI (adjusted odds ratio 1·60, 95% credible interval 1·05–2·37; p=0·030). 132 (21·6%) of 610 patients with an SSI and a microbiology culture result had an infection that was resistant to the prophylactic antibiotic used. Resistant infections were detected in 49 (16·6%) of 295 patients in high-HDI countries, in 37 (19·8%) of 187 patients in middle-HDI countries, and in 46 (35·9%) of 128 patients in low-HDI countries (p < 0·001).
Interpretation: Countries with a low HDI carry a disproportionately greater burden of SSI than countries with a middle or high HDI and might have higher rates of antibiotic resistance. In view of WHO recommendations on SSI prevention that highlight the absence of high-quality interventional research, urgent, pragmatic, randomised trials based in LMICs are needed to assess measures aiming to reduce this preventable complication

Comparison between uniaxial and pull-out tests setup for polyester and HDPE geogrids
Uniaxial tensile and/or pull-out tests are performed to obtain the stress-strain curve of geogrids. The clamp lining in contact with a specimen in the uniaxial tensile test or the grit of a sandpaper clamp liner in a pull-out test affects the results. In this study, one geogrid made of polyester and another one made of high-density polyethylene (HDPE) are tested in the uniaxial tensile test using various clamps and in pull-out test using sandpapers of different grits. Based on the results obtained, it is recommended to test HDPE in uniaxial tests with serrated steel-lined clamps and in pull-out tests with sandpaper (grit 180) lined clamps. A polyester geogrid shall be tested in uniaxial tests with plastic-lined clamps and in pull-out tests with sandpaper (grit 400) lined clamps
Cellulose nanocrystals from agricultural residues (Eichhornia crassipes): Extraction and characterization
International audienceExtraction of cellulose nanocrystals (CNCs) from agro-residues has received much attention, not only for their unique properties supporting a wide range of potential applications, but also their limited risk to global climate change. This research was conducted to assess Nile roses (Eichhornia crassipes) fibers as a natural biomass to extract CNCs through an acid hydrolysis approach. Nile roses fibers (NRFs) were initially subjected to alkaline (pulping) and bleaching pretreatments. Microcrystalline cellulose (MCC) was used as control in comparison to Nile rose based samples. All samples underwent acid hydrolysis process at a mild temperature (45 °C). The impact of extraction durations ranging from 5 to 30 min on the morphology structure and crystallinity index of the prepared CNCs was investigated. The prepared CNCs were subjected to various characterization techniques, namely: X-ray diffraction (XRD), FT-IR analysis, Transmission electron microscopy (TEM), and X-ray Photoelectron spectroscopy (XPS). The outcomes obtained by XRD showed that the crystallinity index increased as the duration of acid hydrolysis was prolonged up to 10 min, and then decreased, indicating optimal conditions for the dissolution of amorphous zones of cellulose before eroding the crystallized domains. These data were confirmed by FT-IR spectroscopy. However, a minor effect of hydrolysis duration on the degree of crystallinity was noticed for MCC based samples. TEM images illustrated that a spherical morphology of CNCs was formed as a result of 30 min acid hydrolysis, highlighting the optimal 20 min acid hydrolysis to obtain a fibrillar structure. Magnetic Graphene Oxide as an Efficient Adsorbent for the Separation and Preconcentration of Cu(II), Pb(II), and Cd(II) from Environmental Samples
The separation and preconcentration of copper(II), lead(II), and cadmium(II) ions on magnetic graphene oxide (MGO) by solid-phase extraction was carried out. Quantitative recovery was obtained by adsorption of analytes on MGO at pH 6 and elution of 3 M HNO3 in 10% acetone. To optimize the presented method, the effects of various parameters-including pH, eluent conditions, and vortex time-were examined. Matrix effects were also investigated. Mean recoveries of the analytes were between 95 and 105%. The proposed method was validated by applying it to certified reference materials. Addition and recovery tests were also performed. The method was applied to verify the analyte content of several water and food samples
