    The wild bootstrap for multilevel models

    In this paper we study the performance of the most popular bootstrap schemes for multilevel data. Also, we propose a modified version of the wild bootstrap procedure for hierarchical data structures. The wild bootstrap does not require homoscedasticity or assumptions on the distribution of the error processes. Hence, it is a valuable tool for robust inference in a multilevel framework. We assess the finite size performances of the schemes through a Monte Carlo study. The results show that for big sample sizes it always pays off to adopt an agnostic approach as the wild bootstrap outperforms other techniques

    Multilevel Models with Stochastic Volatility for Repeated Cross-Sections: an Application to tribal Art Prices

    In this paper we introduce a multilevel specification with stochastic volatility for repeated cross-sectional data. Modelling the time dynamics in repeated cross sections requires a suitable adaptation of the multilevel framework where the individuals/items are modelled at the first level whereas the time component appears at the second level. We perform maximum likelihood estimation by means of a nonlinear state space approach combined with Gauss-Legendre quadrature methods to approximate the likelihood function. We apply the model to the first database of tribal art items sold in the most important auction houses worldwide. The model allows to account properly for the heteroscedastic and autocorrelated volatility observed and has superior forecasting performance. Also, it provides valuable information on market trends and on predictability of prices that can be used by art markets stakeholders

    Information behaviour of Poles in area of healthy eating. Results of EATWELL surveyThe rate of obesity in Poland increases rapidly, especially fast among children and adolescents. An important and also fully accepted by the society ways of fighting this phenomenon are social marketing and educational interventions. To make these activities effective they have to keep pace with changes in information behavior of target groups. But the importance of raising information competency of consumers is not fully understood by those who design and implement health programs and health Information behavior and literacy rarely are the subject of research. The goal of this study is to find where and how Polish citizens look for diet related information, what barriers they encounter, and whether they behave differently in comparison with citizens of other European countries. Method: A survey administered through computer-assisted on-line web-interviewing to a probabilistic, stratified by age and gender, sample of respondents in Poland (n = 600). Comparison sample – same number of respondents in Belgium, Denmark, Italy and UK. Results: 49% of surveyed Poles do not know where to look for healthy diet related information and have bigger problems with this task then the respondents in other surveyed countries. In comparison with Danish, a probability that a Pole knows where to look for such information decreases twice. Individual and environmental determinants strongly affect information knowledge and behavior. Men, educated, poorer and sicker persons have bigger problems with finding information and are less likely to attempt to look for it. Majority of respondents uses Internet and Google to look for healthy diet information. Only 23% of Polish respondents would turn to their GPs for healthy eating advice

    The Cooperation between hMena Overexpression and HER2 Signalling in Breast Cancer

    hMena and the epithelial specific isoform hMena11a are actin cytoskeleton regulatory proteins belonging to the Ena/VASP family. EGF treatment of breast cancer cell lines upregulates hMena/hMena11a expression and phosphorylates hMena11a, suggesting cross-talk between the ErbB receptor family and hMena/hMena11a in breast cancer. The aim of this study was to determine whether the hMena/hMena11a overexpression cooperates with HER-2 signalling, thereby affecting the HER2 mitogenic activity in breast cancer. In a cohort of breast cancer tissue samples a significant correlation among hMena, HER2 overexpression, the proliferation index (high Ki67), and phosphorylated MAPK and AKT was found and among the molecular subtypes the highest frequency of hMena overexpressing tumors was found in the HER2 subtype. From a clinical viewpoint, concomitant overexpression of HER2 and hMena identifies a subgroup of breast cancer patients showing the worst prognosis, indicating that hMena overexpression adds prognostic information to HER2 overexpressing tumors. To identify a functional link between HER2 and hMena, we show here that HER2 transfection in MCF7 cells increased hMena/hMena11a expression and hMena11a phosphorylation. On the other hand, hMena/hMena11a knock-down reduced HER3, AKT and p44/42 MAPK phosphorylation and inhibited the EGF and NRG1-dependent HER2 phosphorylation and cell proliferation. Of functional significance, hMena/hMena11a knock-down reduced the mitogenic activity of EGF and NRG1. Collectively these data provide new insights into the relevance of hMena and hMena11a as downstream effectors of the ErbB receptor family which may represent a novel prognostic indicator in breast cancer progression, helping to stratify patients

    How the First Year of the COVID-19 Pandemic Impacted Patients’ Hospital Admission and Care in the Vascular Surgery Divisions of the Southern Regions of the Italian Peninsula

    Background: To investigate the effects of the COVID-19 lockdowns on the vasculopathic population. Methods: The Divisions of Vascular Surgery of the southern Italian peninsula joined this multicenter retrospective study. Each received a 13-point questionnaire investigating the hospitalization rate of vascular patients in the first 11 months of the COVID-19 pandemic and in the preceding 11 months. Results: 27 out of 29 Centers were enrolled. April-December 2020 (7092 patients) vs. 2019 (9161 patients): post-EVAR surveillance, hospitalization for Rutherford category 3 peripheral arterial disease, and asymptomatic carotid stenosis revascularization significantly decreased (1484 (16.2%) vs. 1014 (14.3%), p = 0.0009; 1401 (15.29%) vs. 959 (13.52%), p = 0.0006; and 1558 (17.01%) vs. 934 (13.17%), p < 0.0001, respectively), while admissions for revascularization or major amputations for chronic limb-threatening ischemia and urgent revascularization for symptomatic carotid stenosis significantly increased (1204 (16.98%) vs. 1245 (13.59%), p < 0.0001; 355 (5.01%) vs. 358 (3.91%), p = 0.0007; and 153 (2.16%) vs. 140 (1.53%), p = 0.0009, respectively). Conclusions: The suspension of elective procedures during the COVID-19 pandemic caused a significant reduction in post-EVAR surveillance, and in the hospitalization of asymptomatic carotid stenosis revascularization and Rutherford 3 peripheral arterial disease. Consequentially, we observed a significant increase in admissions for urgent revascularization for symptomatic carotid stenosis, as well as for revascularization or major amputations for chronic limb-threatening ischemia

    Clinical Features, Cardiovascular Risk Profile, and Therapeutic Trajectories of Patients with Type 2 Diabetes Candidate for Oral Semaglutide Therapy in the Italian Specialist Care

    Introduction: This study aimed to address therapeutic inertia in the management of type 2 diabetes (T2D) by investigating the potential of early treatment with oral semaglutide. Methods: A cross-sectional survey was conducted between October 2021 and April 2022 among specialists treating individuals with T2D. A scientific committee designed a data collection form covering demographics, cardiovascular risk, glucose control metrics, ongoing therapies, and physician judgments on treatment appropriateness. Participants completed anonymous patient questionnaires reflecting routine clinical encounters. The preferred therapeutic regimen for each patient was also identified. Results: The analysis was conducted on 4449 patients initiating oral semaglutide. The population had a relatively short disease duration (42%  60% of patients, and more often than sitagliptin or empagliflozin. Conclusion: The study supports the potential of early implementation of oral semaglutide as a strategy to overcome therapeutic inertia and enhance T2D management
