Maximizing and Modeling Malonyl-CoA Production in Escherichia coli

In E. coli, fatty acid synthesis is catalyzed by the enzyme acetyl-CoA carboxylase (ACC), which converts acetyl-CoA into malonyl-CoA. Malonyl-CoA is a major building block for numerous of bioproducts. Multiple parameters regulate the homeostatic cellular concentration of malonyl-CoA, keeping it at a very low level. Understanding how these parameters affect the bacterial production of malonyl-CoA is fundamental to maximizing it and its bioproducts. To this end, competing pathways consuming malonyl-CoA can be eliminated, and optimal nutritional and environmental conditions can be provided to the fermentation broth. Most previous studies utilized genetic modifications, expensive consumables, and high-cost quantification methods, making unfeasible the development of an economically-attractive process with high product yield. In this work, we propose a low-cost, simple, and effective method to maximize and quantify malonyl-CoA production in E. coli. The enzyme, 1,3,6,8-tetrahydroxynapthalenesynthase (THNS) catalyzes the condensation of five molecules of malonyl-CoA forming 1,3,6,8- tetrahydroxynaphthalene (THN), which is auto-oxidized into flaviolin. Flaviolin can be measured spectrophotometrically, meaning that direct quantification of malonyl-CoA is accomplished by measuring the absorbance of flaviolin. Results showed that the main parameters associated with malonyl-CoA maximization are the use of rich medium supplemented with glucose and metals, the incubation temperature of 37C, the inoculum incubation time, and the use of lactose as an inducer. Moreover, a kinetic model of the bacterium metabolism was built in order to guide the maximization of malonyl-CoA. By performing a stability analysis and a metabolic control analysis, the system was determined to be stable and flux controlled by phosphoglucose isomerase. This suggests that this enzyme should be modified to optimize the production of malonyl-CoA

Coffee and cocoa value chains: Gender dynamics in Peru and Nicaragua

The present report assesses women’s and men’s participation in coffee and cocoa value chains in Latin America in order to identify key considerations and next steps for the development of research, policy and practice on value chain upgrading for enhanced smallholder prosperity. In particular, the report analyzes the differential roles that women and men play and the benefits they perceive from their participation in coffee and cocoa value chains. It also considers the role of producer organizations in limiting or enhancing women’s participation and benefits from coffee and cocoa value chains. The report first reviews existing studies of gender and coffee and cocoa value chains in Latin American countries in order to identify principal trends; then, the paper focuses on Peru and Nicaragua as two country cases. In general, the report demonstrates that while factors such as labor burdens, access to productive resources, and socio-cultural norms can limit women’s participation in and returns from coffee and cocoa value chains in comparison to that of men, varied initiatives have sought to address these restricting conditions via producer organizations. The extent and circumstances of their success in advancing gender equality in coffee and cocoa value chains will be an important area for future analysis

Aspectos relativos à promoção do design no Brasil visando a área da gestão: o caso da empresa vencedora do Prêmio CNI José Mindlin 2000

Responde e levanta algumas questões sobre a inserção do Design no processo da gestão. Concentra-se em expor conceitos presentes na literatura que discorrem sobre o que é Design, o que é Gestão, o que é Planejamento Estratégico e Gestão do Design. Articula os primeiros conceitos mencionados com a Gestão de Design, mostrando como se completam e visam os mesmos objetivos no processo administrativo. Descreve um conjunto de ações indutoras da modernização industrial e tecnológica que é encabeçado por programas de promoção do Design, suas missões e objetivos. Apresenta o Prêmio CNI José Mindlin e o estudo de caso de uma empresa, registrando dados sobre sua participação. Aponta considerações acerca da participação da empresa, Interpreta dados obtidos e apresenta os critérios do Prêmio que passaram por uma significativa evolução nos três últimos anos. Enfim, discorre sobre o tema proposto e busca mostrar a importância estratégica do Design nas empresas e as suas reais contribuições para a promoção da competitividade na economia Brasileira. Estimula a inserção de modelos de Gestão do Design, ressaltando sua importância como nova frente de trabalho para os profissionais. This dissertation studies the aspects of promoting Design, aiming Design Management in Brazil. Responds and raises some important issues about the insertion of design in the management process. It focus at showing present concepts in the literature that talk about what Design is, what Management is, what Strategic Planning and Design Management are. Articulates the first concepts with Design Management, showing how they complete each other and aim the same goals in the Administration process. Describes some actions for the industrial and technological modernization that is headed by Design promotion programs, their missions and objectives. Presents the CNI José Mindlin Prize and the case study of a company, registering data about its participation. Points out considerations on the company's participation, interprets data obtained and presents the prize's criterions which have gone through a significant evolution during the past three years. Finally, this work intends to show the strategic importance of Design in the companies and its real contribuition to promote competitiviness in Brazilian economy. Stimulates the insertion of Design Management models, emphasizing its importance as a new career for the professionals

Política de indexação nas bibliotecas das universidades federais do Brasil

TCC (graduação em Biblioteconomia) - Universidade Federal de Santa Catarina, FlorianópolisPesquisa desenvolvida sobre o tema política de indexação nas bibliotecas das universidades federais do Brasil. Objetiva identificar os principais aspectos que constituem a política de indexação; verificar se as bibliotecas das universidades federais do Brasil adotam uma política de indexação; identificar o conteúdo das políticas de indexação utilizadas pelas bibliotecas de universidades federais brasileiras e sintetizar os aspectos referentes às políticas de indexação adotadas por essas bibliotecas. Realiza uma pesquisa exploratória e descritiva, estudando as bibliotecas das universidades federais do Brasil. Com relação aos procedimentos técnicos, realiza uma pesquisa bibliográfica e de levantamento, aborda os aspectos publicados acerca do tema e coleta dados com o uso de questionário como ferramenta. As respostas analisadas e interpretadas apontam que 55% das bibliotecas das universidades federais do Brasil utilizam política de indexação. Conclui que os elementos que podem constar na política de indexação não são totalmente contemplados pelas bibliotecas, no entanto, é presente a preocupação com a política de indexação por parte dos respondentes da pesquis

Tidal evolution of a close-in planet with a more massive outer companion

We investigate the motion of a two-planet coplanar system under the combined effects of mutual interaction and tidal dissipation. The secular behavior of the system is analyzed using two different approaches, restricting to the case of a more massive outer planet. First, we solve the exact equations of motion through the numerical simulation of the system evolution. We also compute the stationary solutions of the mean equations of motion based on a Hamiltonian formalism. The influence of oral health on glycemic control, under interdisciplinar guidance

O controle glicêmico é essencial não só para o paciente diabético, mas também para qualquer outro paciente que zele pelo seu bem-estar geral. Saber que uma boa saúde bucal interfere positivamente nesse controle favorece o paciente, desde que ele adquira esse conhecimento durante seus atendimentos médicos e odontológicos. Esse estudo é do tipo quantitativo transversal e por meio de um questionário avaliou-se o conhecimento de 196 pacientes do XVI Projeto Antonio Prudente (Feira de Saúde 2016) sobre a influência da saúde bucal no controle glicêmico, além de verificar a orientação médica e odontológica aos pacientes sobre essa influência e relacionar o tipo de orientação profissional com a glicemia deles. Poucos entrevistados consideraram a saúde bucal importante para controlar a glicemia, pelo fato de que a minoria dos pacientes é orientada em conjunto pelos médicos e dentistas sobre a necessidade do cuidado da saúde como um todo. Concluiu-se que é extremamente importante ter serviços de saúde públicos e privados multidisciplinares aos diabéticos para educá-los no autocuidado, gerar melhor comunicação entre os profissionais e garantir melhor qualidade de vida aos pacientes após controle eficaz da glicemia capilar.Glycemic control is essential not only for diabetic patients, but also for any other patient who watches over his general well-being. Therefore, knowing that good oral health interferes positively in the former mentioned control is something that favors the patient, as long as he acquires such knowledge during his medical and dental appointments. This is a cross-sectional quantitative study, which aimed, through a questionnaire, to evaluate the knowledge of 196 patients of the project entitled XVI Antonio Prudente - Health Fair 2016 on the influence of oral health on glycemic control, as well as to verify the medical and dental orientation given to these patients about this influence and how to relate the professional orientation with the patient’s blood glucose. Few interviewees considered oral health as something important for glycemic control because the minority of patients is jointly advised by doctors and dentists about the need for multidisciplinary diabetic care in order to better control the disease. It was concluded that it is extremely important to have multidisciplinary public and private health services for diabetics to educate them in self-care, to generate better communication among professionals and to guarantee better quality of life for patients after effective control of capillary glycemia

RELAÇÃO ENTRE O DESENVOLVIMENTO PSICOMOTOR E AS DIFICULDADES DE APRENDIZAGEM EM CRIANÇAS DE 8 ANOS DE IDADE

A relevância social e científica deste estudo consiste em compreender os fatores relacionados ao desenvolvimento psicomotor e às dificuldades de aprendizagem, possibilitando futuras intervenções que possam melhorar o desenvolvimento das crianças, buscando alternativas que possibilitem as mudanças nesse cenário e ampliando os conhecimentos científicos na área. O objetivo nesta pesquisa foi investigar as relações estabelecidas entre as dificuldades de aprendizagem e o desenvolvimento psicomotor de escolares de 8 anos do Ensino Fundamental da Escola de Educação Básica São Francisco do Município de Chapecó. A população deste estudo compreende 211 alunos, e a amostra foi composta por 29 alunos com faixa etária de oito anos, de ambos os sexos. Este é um estudo de campo não probabilístico, de natureza quantitativa e qualitativa. Para a coleta de dados, foram utilizados como instrumentos os testes do Manual de Desempenho Escolar Análise da Leitura e da Escrita, cuja autoria pertence a Rosa Neto, Santos e Toro (2010). Para identificar e avaliar as crianças com disgrafia, foi utilizada a Escala de Ajuriaguerra adaptada por Lorenzini (1993) e para identificar o perfil motor dos escolares, foi aplicada a Escala de Desenvolvimento Motor – EDM (ROSA NETO, 2002). Para a análise dos dados, utilizou-se o programa de dados estatísticos Excel e o SPSS (versão 20.0 Windows). Os resultados demonstram que 41,3%, dos escolares avaliados apresentaram dificuldades de aprendizagem e de desenvolvimento motor, ficando classificados como normal médio 33,3%, normal baixo 58,3% e inferior 8,3%. Assim, confirma-se que todos os alunos apresentaram nível de atraso motor de 12 meses, ficando clara a importância de o professor realizar avaliações e testes, identificando essas problemáticas e saná-las no decorrer do ensino fundamental, ainda nas séries iniciais.Palavras-chave: Dificuldades de aprendizagem. AÇÕES INTERDISCIPLINARES MEDIADAS PELA TECNOLOGIA PARA CRIANÇAS COM TEA

This article presents a bibliographic study, which aims at proposing a retrospect about the historic and main concepts related to the Autism Spectrum Disorder (ASD), interdisciplinary, digital literacy as well as creative learning, pointing to the issues that embrace the development of literacy of children with ASD. Concerning the interdisciplinary, the article was directed to the contributions of practical actions, in order to value the knowledge of all people involved in the process for the benefit of citizenship formation and students’ autonomy as well as for the teaching qualification in an inclusive proposition. About digital literacy thematic, contributions of dynamic practical activities are presented directed to ludicity. Finally, the concepts of creative learning are approached through propositions elaborated through the spiral learning. After the research accomplishment, the concepts have been deepened and the benefits of proposing interdisciplinary actions mediated by technology have been established, which favor and impulse the literacy process of children with ASD, since these actions have helped to build a more inclusive school and society, as well as improving the quality of life and autonomy of these students, who have shown an atypical development, contributing for the reflection of the intertwining of all the themes involved in the conducted studies.Este artigo apresenta um estudo bibliográfico, tendo como objetivo propor um retrospecto sobre o histórico e os principais conceitos relativos ao Transtorno do Espectro Autista (TEA), à Interdisciplinaridade, à Literacia Digital e à Aprendizagem Criativa, apontando as questões que abarcam o desenvolvimento da alfabetização de crianças com TEA. Sobre a Interdisciplinaridade direcionou-se para as contribuições de práticas e ações, valorizando os saberes de todas as pessoas envolvidas no processo em prol da formação da cidadania e autonomia dos estudantes bem como, para a qualificação do ensino em uma proposta inclusiva. Na temática da Literacia Digital, são apresentadas as contribuições de práticas dinâmicas com direcionamento para a ludicidade. Por fim, são abordados os conceitos da Aprendizagem Criativa através de propostas elaboradas a partir da espiral da aprendizagem. Diante da pesquisa realizada, aprofundaram-se os conceitos e constataram-se os benefícios de propor ações interdisciplinares mediadas pela tecnologia, favorecendo e impulsionando os processos de alfabetização das crianças com TEA, pois essas ações agregam na construção de uma escola e sociedade mais inclusiva, assim como na qualidade de vida e autonomia dos estudantes que apresentam desenvolvimento atípico, contribuindo para a reflexão dos entrelaçamentos dos temas envolvidos no estudo
