    Human resources are one of the resources that most determine the success or failure of an organization. In the era of globalization, human resource issues are in the spotlight and the focus for agencies to survive. Where the success of the organization depends on the performance of the organizational actors concerned. An employee is not only required to have knowledge, abilities, and skills but also must have experience, motivation, self-discipline, work enthusiasm, and good work ethic. This research aims to determine the influence of internal supervision, work discipline, and work motivation on the performance of employees of Seluma Regency Satpol PP and Fire Department. This type of research is quantitative descriptive, namely research that uses mathematical models, theories, and/or hypotheses related to natural phenomena. The sample in this study used a purposive sampling technique, where the population sampled at Satpol PP and Fire Department, Seluma Regency were 37 civil servant employees. Based on the results of research conducted it was found that internal supervision, work discipline and work motivation together had a significant effect on employees’ performance at Satpol PP and Fire Department, Seluma Regency with regression multiple linear Y = 8.106 + 0.352 X1+ 0.393 X2 + 0.50

    PENGARUH CAPITAL ADEQUACY RATIO (CAR), NON PERFORMING LOAN (NPL), BIAYA OPERASIONAL DAN PENDAPATAN OPERASIONAL (BOPO) DAN NET INTEREST MARGIN (NIM) TERHADAP LABA PERUSAHAAN PERBANKAN DENGAN VOLUME PENYALURAN KREDIT SEBAGAI VARIABEL INTERVENING

    Penelitian ini bertujuan untuk mengetahui pengaruh CAR, NPL, BOPO dan NIM terhadap volume penyaluran kredit dan laba perusahaan perbankan, pengaruh volume penyaluran kredit terhadap laba perusahaan perbankan serta mengetahui pengaruh CAR, NPL, BOPO dan NIM terhadap laba perusahaan perbankan melalui volume penyaluran kredit. Teknik pengumpulan data dilakukan dengan metode dokumentasi data sekunder berupa laporan keuangan tahunan Bank Umum Go Public yang dipublikasikan melalui situs resmi Bursa Efek Indonesia (BEI). Teknik pengambilan sampel dengan metode purposive sampling. Jumlah sampel yang digunakan adalah 160. Teknik analisis SEM (Struktural Equation Model) dilakukan untuk menguji pengaruh langsung, pengaruh tidak langsung dan pengaruh total. Hasil penelitian menunjukkan mean CAR dari 160 observasi perusahaan perbankan adalah sebesar 17,4780%, rata-rata NPL perusahaan 1,8221%, rata-rata BOPO yang diungkapkan oleh perusahaan 79,0935%, rata-rata NIM perusahaan sebesar 5,6951%. Hasil penelitian ini juga menunjukkan bahwa terhadap volume penyaluran kredit, CAR dan BOPO berpengaruh negatif, NIM berpengaruh positif, sedangkan NPL tidak berpengaruh. CAR dan BOPO berpengaruh negatif terhadap laba perusahaan sedangkan NPL dan NIM tidak berpengaruh terhadap laba perusahaan perbankan, volume penyaluran kredit berpengaruh positif terhadap laba perusahaan perbankan dan volume penyaluran kredit mampu memediasi CAR, NPL dan NIM terhadap laba perusahaan perbankan tetapi tidak mampu memediasi BOPO terhadap laba perusahaan perbankan.  Kata kunci:      Capital Adequacy Ratio(CAR), Non Performing Loan (NPL), Biaya Operasional dan Pendapatan Operasional (BOPO), Net Interest Margin (NIM), Laba Perusahaan Perbankan, Volume Penyaluran Kredit.

    Evaluasi Validitas Human Cortisol Enzyme-Linked Immunosorbent Assay (ELISA) Kit dan Waktu Sentrifugasi Sampel Darah untuk Pengukuran Konsentrasi Hormon Kortisol pada Kambing Kacang

    ABSTRAK. Penggunaan human cortisol enzyme-linked immunosorbent assay (ELISA) kit untuk pengukuran hormon kortisol pada hewan dan keterlambatan waktu sentrifugasi sampel darah untuk analisis hormon perlu dievaluasi. Penelitian ini bertujuan untuk mengevaluasi validitas human cortisol ELISA kit (EIA-1887, DRG Instruments GmbH, Germany) untuk pengukuran konsentrasi kortisol dan menguji pengaruh keterlambatan waktu sentrifugasi sampel darah terhadap stabilitas konsentrasi kortisol pada kambing kacang. Sampel darah dikoleksi dari delapan ekor kambing kacang. Uji validitas kit EIA-1887 dilakukan secara: a) analitik (uji parallelism, akurasi, dan presisi), dan b) biologis (pengukuran kortisol sebelum dan setelah transportasi). Uji keterlambatan waktu sentrifugasi terhadap stabilitas konsentrasi kortisol dilakukan dengan 5 perlakuan yaitu disentrifugasi kurang dari 1 jam (P1/kontrol), 6 jam (P6), 12 jam (P12), 18 jam (P18),dan 24 jam (P24) setelah darah dikoleksi. Data uji parallelism dianalisis dengan uji persamaan kemiringan, uji presisi dihitung % CV (coefficient variation) intra-assay dan inter-assay, uji akurasi dihitung % recovery, uji T untuk validasi biologis, dan uji ragam (One Way Anova) untuk pengaruh waktu sentrifugasi. Hasil uji parallelism menunjukkan kurva sampel kambing kacang sejajar/parallel dengan kurva standar kortisol. Akurasi kit EIA-1887 adalah 103,43±7,85%, dan % CV intra-assay dan inter-assay adalah 10%. Konsentrasi kortisol setelah transportasi secara signifikan lebih tinggi daripada sebelum transportasi (p0,05). Adanya penurunan secara nyata konsentrasi kortisol pada darah yang disentrifugasi 24 jam (P24) setelah koleksi (p0,05). Kesimpulan, human cortisol ELISA kit (EIA-1887) memiliki validitas yang baik secara analitik dan biologis untuk digunakan dalam pengukuran konsentrasi kortisol pada kambing kacang. Keterlambatan waktu sentrifugasi selama 24 jam berpengaruh terhadap konsentrasi kortisol.  (Evaluation the Validity of Human Cortisol Enzyme-Linked Immunosorbent Assay (ELISA) Kit and Centrifugation Time of Blood Sample for Measuring the Concentration of Cortisol in Kacang Goats)  ABSTRACT. The use of human cortisol ELISA kit for measuring cortisol in animals and delayed to blood centrifugation time for hormone measurement need to be evaluated. This study aimed to evaluate the validity of human cortisol ELISA kit (EIA-1887, DRG Instruments GmbH, Germany) for cortisol measurement and effect of delayed to blood centrifugation time on cortisol concentrations in kacang goats. Blood was collected from eight kacang goats. Validation test of EIA-1887 kit was performed through: a) analytical (parallelism, accuracy, and precision tests), and b) biological validations (measuring cortisol concentrations before and after transportation). Five treatments were performed to test delayed to centrifugation time: blood centrifuged at 1 h (control, P1), 6 h (P6), 12 h (P12), 18 h (P18), and 24 h (P24) after collection. Parallelism data were analyzed by slope equality test, precision and accuracy calculated by % CV of intra-and inter-assay, and % recovery, respectively. Data of biological validation and centrifugation time effects were analyzed by Student t-test, and one way ANOVA, respectively. Results of parallelism showed that serial dilution curve of kacang goat plasma was parallel with cortisol standard curves. Accuracy of EIA-1887 kit was 103.43±7.85%, and % CV of intra-and inter-assay were 10%. Concentration of cortisol after transportation was significantly higher than before transportation (p0.05). Concentration of cortisol was significantly decreased when blood was centrifuged at 24 h after collection (P0.05). In conclusion, human cortisol ELISA kit (EIA-1887) is a reliable assay for measuring cortisol in plasma of kacang goat. Delayed to blood centrifugation time affect cortisol concentrations

    The purpose of this research is to produce a physics learning module on fluid material and to know the student's perception of the module. This research includes research and development. The development Model used is ADDIE (Analyze, Design, Development, Implementation, and Evaluation). The Physics learning module on the fluid material of basic Physics 1 courses has been declared worthy by the validator. Test Result module obtained 85 (very good) for the design aspects of learning, the material aspect is 85.07 (very good), the aspect of readability module is 85.22 (very good), the visualization aspect of the module is 84.11 (very good). The basic Physics Module 1 development fluid material is worth using as one of the learning teaching materials in the classroom.Tujuan penelitian ini adalah menghasilkan modul pembelajaran fisika pada materi fluida dan mengetahui persepsi mahasiswa terhadap modul tersebut. Penelitian ini termasuk penelitian dan pengembangan (Research and Development). Model pengembangan yang digunakan adalah ADDIE (Analyze, Design, Development, Implementation, and Evaluation). Modul pembelajaran fisika pada materi fluida mata kuliah fisika dasar 1 telah dinyatakan layak oleh validator. Hasil ujicoba modul diperoleh 85 (sangat baik) untuk aspek desain pembelajaran, aspek materi yaitu 85,07 (sangat baik), aspek keterbacaan modul yaitu 85,22 (sangat baik), aspek visualisasi modul yaitu 84,11 (sangat baik).     KAJIAN KEBUTUHAN ANGKUTAN LAYANAN PERGERAKAN INTERNAL KAMPUS INSTITUT TEKNOLOGI SUMATERA

    Besides of the lecture buildings and laboratories facilities, transportation modes are also needed to facilitate academicians’ movement needs. Institut Teknologi Sumatera (ITERA) is the first public technology institution on Sumatra, which is located on the border of South Lampung regencies and Bandar Lampung City. From the survey results, it is known that 27% of the academic community, consisting of lecturers, education staff, and students, do not own private vehicles. It caused an accessibility problem, which is the absence of public transportation that serves internal movements for the captive group. In campus internal service transportation’s planning, there are several analyzes will be carried out to find out how important the internal movement facilities in the ITERA campus area, and the types of vehicles that can serve the movement requests that occur. Based on the survey results, it is known that the internal campus movements are carried out by all ITERA academicians, with the peak transfer hour at 15.00-16.00 WIB. From the analysis of the existing geometric conditions, it can be seen that the lane width on the ITERA internal ring road is more than 2.5 m, which means that it is sufficient to serve the movement Light Vehicles (LV) and medium or large buses according to the type classification. land public transport vehicles. In addition, from the choice group, more than 50% of respondents stated that they are willing to use public transportation that serves the internal movements of the ITERA campus if these facilities are already available.Selain fasilitas fisik berupa gedung perkuliahan dan laboratorium, fasilitas berupa moda transportasi juga dibutuhkan untuk memudahkan pergerakan sivitas akademika dalam memenuhi kebutuhan akademik maupun non akademik. Institut Teknologi Sumatera (ITERA) merupakan institusi teknologi negeri pertama di pulau Sumatera, yang berada di perbatasan kabupaten Lampung Selatan dan Kota Bandar Lampung. Dari hasil survei diketahui bahwa sebesar 27% dari sivitas akademika yaitu yang terdiri dari dosen, tenaga kependidikan/staf, dan mahasiswa tidak memiliki kendaraan pribadi. Hal ini menimbulkan suatu permasalahan aksesibilitas yaitu tidak adanya angkutan umum yang melayani pergerakan internal bagi golongan captive tersebut. Dalam merencanakan angkutan layanan internal kampus, akan dilakukan beberapa analisis untuk mengetahui seberapa pentingnya sarana pergerakan internal di wilayah kampus ITERA, dan jenis kendaraan yang dapat melayani permintaan pergerakan yang terjadi. Berdasarkan hasil survei, diketahui bahwa pergerakan internal kampus dilakukan oleh seluruh sivitas akademika ITERA, dengan waktu perpindahan puncak yaitu pada pukul 15.00-16.00 WIB. Dari analisis kondisi geometri eksisting, dapat diketahui bahwa lebar lajur pada ruas jalan lingkar internal ITERA yaitu melebihi 2,5 m yang artinya memadai untuk melayani pergerakan dengan jenis kendaraan mobil penumpang ringan atau Light Vehicle (LV) dan bus sedang atau besar sesuai dengan klasifikasi jenis kendaraan angkutan umum darat. Selain itu, dari golongan kelompok choice didapatkan lebih dari 50% responden yang menyatakan untuk bersedia menggunakan angkutan umum yang melayani pergerakan internal kampus ITERA apabila fasilitas tersebut telah tersedia

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Implementasi Kurikulum 2013 pada Masa Pandemi

    Virus COVID-19 sangat berdampak di semua bidang, salah satunya bidang pendidikan. Dengan adanya pandemi COVID-19 ini, pemerintah menerapkan kebijakan semua pembelajaran dilakukan jarak jauh/daring. Sebelum pandemi COVID-19 ini beberapa pihak sekolah menggunakan Kurikulum 2013. Kurikulum 2013 ini membuktikan bahwa kurikulum tersebut kurang sesuai dengan keadaan saat ini.  Hal itu menjadikan pemerintah memiliki peran penting untuk mengatasi masalah tersebut yaitu dengan mengeluarkannya kurikulum darurat. Tujuan dari penelitian ini adalah untuk mengetahui implementasi Kurikulum 2013 pada saat pandemi COVID-19 yang lebih rincinya meliputi proses pembelajaran, kelebihan dan kekurangan penerapan Kurikulum 2013, efektifkah pembelajaran Kurikulum 2013 dilakukan saat pandemi, dan persiapan guru untuk pembelajaran selanjutnya di masa pandemi. Penelitian ini dilakukan melalui angket google form. Hasil penelitian menunjukkan proses pembelajaran Kurikulum 2013 yang dilakukan saat pandemi ternyata tidak efektif karena sangat memberatkan guru, siswa, dan orang tua. Untuk kelebihannya yaitu siswa dituntut untuk aktif, adanya penilaian dari semua aspek, standar penilaian berbasis kompetensi, dan munculnya pendidikan karakter, sedangkan kekurangannya adalah banyak guru yang belum siap, penguasaan teknologi dan informasi masih terbatas, dan kesusahan sinyal. Persiapan guru untuk pembelajaran selanjutnya di masa pandemi adalah guru dituntut untuk kreatif dan inovatif agar pembelajaran dapat berjalan secara efektif meskipun pembelajaran dilakukan secara daring. Kata Kunci: kurikulum, pandemi, pembelajaran. Abstract The COVID-19 virus has had an impact on all fields, one of which is education. With the COVID-19 pandemic, the government has implemented a policy of all learning done remotely / online. Before the COVID-19 pandemic, several schools used the 2013 Curriculum. The 2013 Curriculum proved that the curriculum was not in accordance with the current situation. This makes the government have an important role to overcome this problem, namely by issuing an emergency curriculum. The purpose of this study was to determine the implementation of the 2013 Curriculum during the COVID-19 pandemic, which in more detail included the learning process, the advantages and disadvantages of implementing the 2013 Curriculum, whether the 2013 Curriculum learning was effective during a pandemic, and the preparation of teachers for further learning during the pandemic. This research was conducted through a google form questionnaire. The results showed that the 2013 Curriculum learning process which was carried out during the pandemic was not effective because it was very burdensome for teachers, students, and parents. The advantages are that students are required to be active, there are assessments from all aspects, competency-based assessment standards, and the emergence of character education, while the drawbacks are that many teachers are not ready, the mastery of technology and information is still limited, and signal difficulties. Teacher preparation for further learning during the pandemic is that teachers are required to be creative and innovative so that learning can run effectively even though learning is done online.     Individual based simulation for online marketplace diffusion among trading small medium enterprises: A conceptual framework

    The number of Indonesian SMEs and Indonesian Internet users are increasing. This situation is of concern to the Indonesian government. The government then launched a SME go online program with a target of 8 million SMEs using the online platform in 2019. One of the online platforms that SMEs may use in Indonesia is the online marketplace such as Tokopedia, Bukalapak and others. Previous research studied the factors that support or hinder the adoption of e-commerce in SMEs environment but had not explored how the interaction of those factors in the adoption of online marketplace. In a social system, the diffusion and adoption of online marketplaces in SMEs is influenced by socio-economic characteristics, personality variables and communication behaviours that exist and are acquired by individuals. This paper describes a conceptual framework for the development of an individual-based simulation commonly referred to as an agent-based simulation to understand the adoption by SMEs and the diffusion of online marketplace in SME environments
