    High prey-predator size ratios and unselective feeding in copepods: A seasonal comparison of five species with contrasting feeding modes

    There has been an upsurge of interest in trait-based approaches to zooplankton, modelling the seasonal changes in the feeding modes of zooplankton in relation to phytoplankton traits such as size or motility. We examined this link at two English Channel plankton monitoring sites south of Plymouth (L4 and E1). At L4 there was a general transition from diatoms in spring to motile microplankton in summer and autumn, but this was not mirrored in the succession of copepod feeding traits; for example the ambushing Oithona similis dominated during the spring diatom bloom. At nearby E1 we measured seasonality of food and grazers, finding strong variation between 2014 and 2015 but overall low mesozooplankton biomass (median 4.5 mg C m−3). We also made a seasonal grazing study of five copepods with contrasting feeding modes (Calanus helgolandicus, Centropages typicus, Acartia clausi, Pseudocalanus elongatus and Oithona similis), counting the larger prey items from the natural seston. All species of copepod fed on all food types and differences between their diets were only subtle; the overriding driver of diet was the composition of the prey field. Even the smaller copepods fed on copepod nauplii at significant rates, supporting previous suggestions of the importance of intra-guild predation. All copepods, including O. similis, were capable of tackling extremely long (> 500 μm) diatom chains at clearance rates comparable to those on ciliates. Maximum observed prey:predator length ratios ranged from 0.12 (C. helgolandicus) up to 0.52 (O. similis). Unselective feeding behaviour and the ability to remove highly elongated cells have implications for how copepod feeding is represented in ecological and biogeochemical models

    Diversity and occurrence of siphonophores in Irish coastal waters

    Siphonophores are at times amongst the most abundant invertebrate zooplankton predators in the oceans. Historically, siphonophores have been under-sampled and of the studies conducted there has been a bias towards oceanic oligotrophic waters where they are considered to be more important. In temperate coastal regions, comparatively less is known about the diversity and abundance of siphonophores, where periodic blooms can restructure the plankton communities and have been correlated with high mortalities in the salmon aquaculture industry. To address this lack of knowledge, plankton samples were collected during two periods (March 2009-March 2011 and April 2014-November 2015) from a coastal embayment in the southwest of Ireland. In total, three siphonophore species were found, the calycophoran Muggiaea atlantica, and the physonects, Nanomia bijuga and Agalma elegans. Muggiaea atlantica was the most abundant species (250 colonies m−3), with densities an order of magnitude higher than either physonect. Muggiaea atlantica displayed a distinct seasonality, whereas the physonect species were sporadic in occurrence. Comparing siphonophores in Bantry Bay and the Western English Channel (Plymouth Marine Laboratory's L4 station) indicates both regions share a similar pattern of inter-annual occurrence and provides novel information on the seasonality and occurrence of siphonophores in Irish coastal waters

    Metabolically active, non-nitrogen fixing,Trichodesmiumin UK coastal waters during winter

    Trichodesmium, a colonial cyanobacterium typically associated with tropical waters, was observed between January and April 2014 in the western English Channel. Sequencing of the heterocyst differentiation (hetR) and 16S rRNA genes placed this community within the Clade IV Trichodesmium, an understudied clade previously found only in low numbers in warmer waters. Nitrogen fixation was not detected although measurable rates of nitrate uptake and carbon fixation were observed. Trichodesmium RuBisCO transcript abundance relative to gene abundance suggests the potential for viable and potentially active Trichodesmium carbon fixation. Observations of Trichodesmium when coupled with a numerical advection model indicate that Trichodesmium communities can remain viable for >3.5 months at temperatures lower than previously expected. The results suggest that Clade IV Trichodesmium occupies a different niche to other Trichodesmium species, and is a cold- or low-light-adapted variant

    Serum methylarginines and spirometry-measured lung function in older adults

    Rationale: Methylarginines are endogenous nitric oxide synthase inhibitors that have been implicated in animal models of lung disease but have not previously been examined for their association with spirometric measures of lung function in humans. Objectives: This study measured serum concentrations of asymmetric and symmetric dimethylarginine in a representative sample of older community-dwelling adults and determined their association with spirometric lung function measures. Methods: Data on clinical, lifestyle, and demographic characteristics, methylated arginines, and L-arginine (measured using LC-MS/MS) were collected from a population-based sample of older Australian adults from the Hunter Community Study. The five key lung function measures included as outcomes were Forced Expiratory Volume in 1 second, Forced Vital Capacity, Forced Expiratory Volume in 1 second to Forced Vital Capacity ratio, Percent Predicted Forced Expiratory Volume in 1 second, and Percent Predicted Forced Vital Capacity. Measurements and Main Results: In adjusted analyses there were statistically significant independent associations between a) higher asymmetric dimethylarginine, lower Forced Expiratory Volume in 1 second and lower Forced Vital Capacity; and b) lower L-arginine/asymmetric dimethylarginine ratio, lower Forced Expiratory Volume in 1 second, lower Percent Predicted Forced Expiratory Volume in 1 second and lower Percent Predicted Forced Vital Capacity. By contrast, no significant associations were observed between symmetric dimethylarginine and lung function. Conclusions: After adjusting for clinical, demographic, biochemical, and pharmacological confounders, higher serum asymmetric dimethylarginine was independently associated with a reduction in key measures of lung function. Further research is needed to determine if methylarginines predict the decline in lung function

    Seabird species vary in behavioural response to drone census

Available from the publisher via the DOI in this record.Unmanned aerial vehicles (UAVs) provide an opportunity to rapidly census wildlife in remote areas while removing some of the hazards. However, wildlife may respond negatively to the UAVs, thereby skewing counts. We surveyed four species of Arctic cliff-nesting seabirds (glaucous gull Larus hyperboreus, Iceland gull Larus glaucoides, common murre Uria aalge and thick-billed murre Uria lomvia) using a UAV and compared censusing techniques to ground photography. An average of 8.5% of murres flew off in response to the UAV, but >99% of those birds were non-breeders. We were unable to detect any impact of the UAV on breeding success of murres, except at a site where aerial predators were abundant and several birds lost their eggs to predators following UAV flights. Furthermore, we found little evidence for habituation by murres to the UAV. Most gulls flew off in response to the UAV, but returned to the nest within five minutes. Counts of gull nests and adults were similar between UAV and ground photography, however the UAV detected up to 52.4% more chicks because chicks were camouflaged and invisible to ground observers. UAVs provide a less hazardous and potentially more accurate method for surveying wildlife. We provide some simple recommendations for their use.We thank T.     The effect of distractions in the operating room during endourological procedures

    Contains fulltext : 98421.pdf (publisher's version ) (Closed access)BACKGROUND: Professionals working in the operating room (OR) are subject to various distractions that can be detrimental to their task performance and the quality of their work. This study aimed to quantify the frequency, nature, and effect on performance of (potentially) distracting events occurring during endourological procedures and additionally explored urologists' and residents' perspectives on experienced ill effects due to distracting factors. METHODS: First, observational data were collected prospectively during endourological procedures in one OR of a teaching hospital. A seven-point ordinal scale was used to measure the level of observed interference with the main task of the surgical team. Second, semistructured interviews were conducted with eight urologists and seven urology residents in two hospitals to obtain their perspectives on the impact of distracting factors. RESULTS: Seventy-eight procedures were observed. A median of 20 distracting events occurred per procedure, which corresponds to an overall rate of one distracting event every 1.8 min. Equipment problems and procedure-related and medically irrelevant communication were the most frequently observed causes of interruptions and identified as the most distracting factors in the interviews. Occurrence of distracting factors in difficult situations requiring high levels of concentration was perceived by all interviewees as disturbing and negatively impacting performance. The majority of interviewees (13/15) thought distracting factors impacted more strongly on residents' compared to urologists' performance due to their different levels of experience. CONCLUSION: Distracting events occur frequently in the OR. Equipment problems and communication, the latter both procedure-related and medically irrelevant, have the largest impact on the sterile team and regularly interrupt procedures. Distracting stimuli can influence performance negatively and should therefore be minimized. Further research is required to determine the direct effect of distraction on patient safety

    Comment. What drives plankton seasonality in a stratifying shelf sea? Some competing and complementary theories

    The Plymouth L4 time plankton series in the Western English Channel is a textbook example of a shallow, stratifying shelf sea system. Over its 30 yr of weekly sampling, this site has provided a diverse and contrasting suite of numerical and conceptual models of plankton bloom formation, phenology, and seasonal succession. The most recent of these papers, Kenitz et al. (2017) has initiated this comment, partly because we feel that it has presented a slightly misleading picture of the plankton composition at this site, and of a robust, recurring seasonal succession. We address this by illustrating the extent of inter‐annual variability in phenology that occurs at the site, and which needs to be captured better within models. However our main aim is to foster a much better integration of the variety of top‐down and bottom‐up processes that have all been suggested to be key in driving seasonal succession. Some of these, particularly the multiple grazing and growth controls contributing to the so‐called “loophole hypothesis” may be complementary, but others, such as the role of copepod feeding traits in driving species succession (Kenitz et al. 2017) offer testable competing hypotheses. The basic assumptions and outputs of all these models need to be validated more critically, both against time series data and process studies that include the finding of unselective feeding. We suggest that the variability in plankton phenology (and not just mean timing and amplitude) could be used to diagnose the performance of alternative models of plankton succession

    Seasonal measurements of the nitrogenous osmolyte glycine betaine in marine temperate coastal waters

    Glycine betaine (GBT) is a nitrogenous osmolyte ubiquitous throughout the marine environ�ment. Despite its widespread occurrence and sig�nifcance in microbial cycling, knowledge of the seasonality of this compound is lacking. Here, we present a seasonal dataset of GBT concentrations in marine suspended particulate material. Analysing coastal waters in the Western English Channel, GBT peaked in summer and autumn but did not follow the observed maxima in total phytoplankton biomass orchlorophyll a. Instead, we found evidence that GBT concentrations were associated with specifc phyto�plankton groups or species, particularly in the sum�mer when GBT correlated with dinofagellate bio�mass. In contrast, autumn maxima corresponded with a period of rapidly changing salinity and nutrient availability, with potential contributions from some phytoplankton species and Harpacticoid copepods. This suggests distinct environmental drivers for dif�ferent periods of the GBT seasonality. Building on evidence that GBT and dinofagellate biomass peak in summer, concomitantly with low nutrients, we propose that GBT positively afects dinofagellate ftness, allowing them to outcompete other plank�ton when inorganic nutrients are depleted. By using this assumption, we improved the performance of a marine ecosystem model to reproduce the observed increase in dinofagellates biomass in the transition from spring to summer. This work sheds light on the interplay between phytoplankton succession, compet�itive advantage and changing environmental factors relevant to climate change. It paves the way for future multidisciplinary research aiming to understand the importance of dinofagellates in key coastal ecosys�tems and their potential signifcance for methylamine production, compounds relevant for particle growth in atmospheric chemistry

    The fibroblast mitogen platelet-derived growth factor -BB (PDGF-BB) induces a transient expression of the orphan nuclear receptor NR4A1 (also named Nur77, TR3 or NGFIB). The aim of the present study was to investigate the pathways through which NR4A1 is induced by PDGF-BB and its functional role. We demonstrate that in PDGF-BB stimulated NIH3T3 cells, the MEK1/2 inhibitor CI-1040 strongly represses NR4A1 expression, whereas Erk5 downregulation delays the expression, but does not block it. Moreover, we report that treatment with the NF-κB inhibitor BAY11-7082 suppresses NR4A1 mRNA and protein expression. The majority of NR4A1 in NIH3T3 was found to be localized in the cytoplasm and only a fraction was translocated to the nucleus after continued PDGF-BB treatment. Silencing NR4A1 slightly increased the proliferation rate of NIH3T3 cells; however, it did not affect the chemotactic or survival abilities conferred by PDGF-BB. Moreover, overexpression of NR4A1 promoted anchorage-independent growth of NIH3T3 cells and the glioblastoma cell lines U-105MG and U-251MG. Thus, whereas NR4A1, induced by PDGF-BB, suppresses cell growth on a solid surface, it increases anchorage-independent growth

    Harmful Algal Blooms and their impacts on shellfish mariculture follow regionally distinct patterns of water circulation in the western English Channel during the 2018 heatwave

    Harmful algal blooms (HABs) can have severe ecological, societal and economic impacts upon marine ecosystems, human health and the seafood industry. We evaluated changes in marine plankton communities with prevailing physico-chemical conditions throughout an exceptionally warm summer (2018), to elucidate key factors governing HABs and their impacts on shellfish mariculture in the western English Channel. Despite warm, stable weather conditions and widespread seasonal stratification throughout the summer, divergent plankton community compositions were observed at two rope-grown mussel (Mytilus edulis) farms (St Austell Bay and Lyme Bay) and a long-term ecological research LTER site (Plymouth L4). There were significant differences between sites in the abundances of HAB species, including Dinophysis spp. and Karenia mikimotoi, whose cell counts bloomed in excess of UK Food Standards Agency (FSA) advisory ‘trigger’ levels at Plymouth L4 and St Austell Bay, but not at the Lyme Bay site. The K. mikimotoi bloom occurred over two weeks in August and comprised up to 88% of the standing phytoplankton biomass in St Austell Bay. Dinophysis spp. also bloomed here from May to September, constituting up to 28% of phytoplankton biomass. This protracted bloom resulted in concentrations of Dinophysis toxins 1 & 2 and pectenotoxins and okadaic acid in shellfish, which closed shellfish harvesting operations on farms located in St Austell Bay, and other shellfish sites in the west of the western English Channel (but not in the east of the region). Inter-site differences in the abundances of these and other HAB species were associated with variations in water circulation and co-occurring phytoplankton and zooplankton communities. Furthermore, plankton monitoring data obtained from the L4 site over the past 3 decades showed HAB species (including Dinophysis spp.) with abundances commonly occurring above advisory trigger levels during warmer periods, such as that coinciding with our study. Under projected climate warming these blooms are likely to continue to be governed by regionally distinct patterns of water circulation, which need to be taken into account in marine spatial planning, when assessing the suitability of new shellfish mariculture site
