    Genetic modifiers of ambulation in the cooperative international Neuromuscular Research Group Duchenne natural history study

    OBJECTIVE: We studied the effects of LTBP4 and SPP1 polymorphisms on age at loss of ambulation (LoA) in a multiethnic Duchenne muscular dystrophy (DMD) cohort. METHODS: We genotyped SPP1 rs28357094 and LTBP4 haplotype in 283 of 340 participants in the Cooperative International Neuromuscular Research Group Duchenne Natural History Study (CINRG-DNHS). Median ages at LoA were compared by Kaplan-Meier analysis and log-rank test. We controlled polymorphism analyses for concurrent effects of glucocorticoid corticosteroid (GC) treatment (time-varying Cox regression) and for population stratification (multidimensional scaling of genome-wide markers). RESULTS: Hispanic and South Asian participants (n=18, 41) lost ambulation 2.7 and 2 years earlier than Caucasian subjects (p=0.003, <0.001). The TG/GG genotype at SPP1 rs28357094 was associated to 1.2-year-earlier median LoA (p=0.048). This difference was greater (1.9 years, p=0.038) in GC-treated participants, whereas no difference was observed in untreated subjects. Cox regression confirmed a significant effect of SPP1 genotype in GC-treated participants (hazard ratio = 1.61, p=0.016). LTBP4 genotype showed a direction of association with age at LoA as previously reported, but it was not statistically significant. After controlling for population stratification, we confirmed a strong effect of LTBP4 genotype in Caucasians (2.4 years, p =0.024). Median age at LoA with the protective LTBP4 genotype in this cohort was 15.0 years, 16.0 for those who were treated with GC. INTERPRETATION: SPP1 rs28357094 acts as a pharmacodynamic biomarker of GC response, and LTBP4 haplotype modifies age at LoA in the CINRG-DNHS cohort.     The availability, appropriateness, and integration of services to promote Indigenous Australian youth wellbeing and mental health : Indigenous youth and service provider perspectives

    Concerns about the complexity, fragmentation and inefficiency of Australia’s current youth mental health service systems have led policy makers to seek improvements through a shift to community-based solutions. However, there is little evidence of how communities can make this shift. This paper examines the efforts of one Aboriginal and Torres Strait Islander (hereafter, respectfully, Indigenous) community—Yarrabah in north Queensland—to develop strategies for mental health and wellbeing service system improvements for school-aged youth (5–18 years). The research was co-designed with Yarrabah’s community-controlled health service and explores the perceptions of Yarrabah youth and service providers. Iterative grounded theory methods were used to collect and analyse data from 32 youth aged 11–24 years and 24 service providers. Youth were reluctant to seek help, and did so only if they felt a sense of safety, trust, relationality and consistency with providers. Young people’s four suggestions for improvement were access to (1) information and awareness about mental health; (2) youth facilities, spaces and activities; (3) safe and available points of contact; and (4) support for recovery from mental illness. Service providers highlighted an appetite for youth-guided community change and recommended five improvement strategies: (1) listening to youth, (2) linking with community members, (3) providing wellbeing promotion programs, (4) intervening early, and (5) advocating to address the determinants of youth mental health. Overall, both groups realised a disjunct between youth need and service provision, but a willingness to work together for systems change. This study demonstrates the importance of community-driven efforts that harness both youth and service providers’ perspectives, and suggests a need for ongoing dialogue as the basis for co-designing and implementing improvements to wellbeing supports and mental health services for Indigenous youth

    Implementing the threshold learning outcomes for agriculture at university

    The national Learning and Teaching Academics Standards statement for agriculture (AgLTAS) describes the nature and extent of the discipline and threshold learning outcomes (TLOs) that define what a graduate should know and understand and what core skills they should have at graduation. The AgLTAS statement has been endorsed by the Australian Council of Deans of Agriculture and can be used to communicate to current and future students the minimum standards of their degree, as well as be used to inform curriculum design. While the AgLTAS document provides explanatory notes to assist educators to further understand the intent of the TLOs there are no exemplars on how the AgLTAS standards can be implemented. We will present two case studies of how academics at the University of Tasmania and the University of Adelaide used the AgLTAS to map their respective agriculture curricula. An online Curriculum Mapping Tool (CMT) was used to evaluate the alignment between the curriculum and the TLOs and to identify gaps in the curriculum where improvement may be required. Workshop participants completed pre- and post-workshop questionnaires to evaluate the benefits of mapping the curriculum for the respective degrees against the AgLTAS TLOs. The pre-workshop questionnaire collected demographic data about the participants and their knowledge about the agriculture TLOs, degree and the Australian Qualifications Framework. The post-workshop questionnaire established perceived changes in each participant’s awareness, knowledge, connection with the teaching team and curriculum. It was also used to inform the development of the explanatory notes section in the AgLTAS statement. In addition, four units from each University were chosen randomly for external benchmarking. The output from the CMT demonstrated that the degrees met or in some instances exceeded the graduate level TLOs for agriculture. Determining the proficiency level for a TLO can be an issue, as to the difference between explicit learning outcomes that might be expected at introductory level, but which at advanced levels are regarded as implicit. External reviewers examined the curricula of courses at the two Universities for alignment to the TLOs to identify and enable correction for academic bias in internally mapping units. The external evaluation aligned well with the results from the CMT. A combination of external review and curriculum mapping workshops involving the entire teaching team is however still recommended when mapping degrees to TLOs. The process of curriculum mapping was shown to provide most academics with a more holistic view of how the degree meets the AgLTAS and has the potential to drive innovation in assessment design. Curriculum mapping also assisted academics by reinforcing their understanding of constructive alignment between assessment and learning outcomes at the unit, course and the discipline threshold standards. We will also provide commentary on what we believe is the next steps and implications of the AgLTAS for curriculum development, industry engagement and graduate employability in the agriculture discipline

    Energy spread of ultracold electron bunches extracted from a laser cooled gas

    Ultrashort and ultracold electron bunches created by near-threshold femtosecond photoionization of a laser-cooled gas hold great promise for single-shot ultrafast diffraction experiments. In previous publications the transverse beam quality and the bunch length have been determined. Here the longitudinal energy spread of the generated bunches is measured for the first time, using a specially developed Wien filter. The Wien filter has been calibrated by determining the average deflection of the electron bunch as a function of magnetic field. The measured relative energy spread σUU=0.64±0.09%\frac{\sigma_{U}}{U} = 0.64 \pm 0.09\% agrees well with the theoretical model which states that it is governed by the width of the ionization laser and the acceleration length

    Children\u27s experiences of companion animal maltreatment in households characterized by intimate partner violence

    Cruelty toward companion animals is a well-documented, coercive tactic used by abusive partners to intimidate and control their intimate partners. Experiences of co-occurring violence are common for children living in families with intimate partner violence (IPV) and surveys show that more than half are also exposed to abuse of their pets. Given children\u27s relationships with their pets, witnessing such abuse may be traumatic for them. Yet little is known about the prevalence and significance of this issue for children. The present study examines the experiences of children in families with co-occurring pet abuse and IPV. Using qualitative methods, 58 children ages 7–12 who were exposed to IPV were asked to describe their experiences of threats to and harm of their companion animals. Following the interviews, template analysis was employed to systematically develop codes and themes. Coding reliability was assessed using Randolph\u27s free-marginal multirater kappa (kfree = .90). Five themes emerged from the qualitative data, the most common being children\u27s exposure to pet abuse as a power and control tactic against their mother in the context of IPV. Other themes were animal maltreatment to discipline or punish the pet, animal cruelty by a sibling, children intervening to prevent pet abuse, and children intervening to protect the pet during a violent episode. Results indicate that children\u27s experiences of pet abuse are multifaceted, potentially traumatic, and may involve multiple family members with diverse motives

    Dissemination and Effectiveness of the Peer Marketing and Messaging of a Web-Assisted Tobacco Intervention: Protocol for a Hybrid Effectiveness Trial

    BACKGROUND: Smoking continues to be the leading preventable cause of death. Digital Interventions for Smoking Cessation (DISCs) are health communication programs accessible via the internet and smartphones and allow for greater reach and effectiveness of tobacco cessation programs. DISCs have led to increased 6-month cessation rates while also reaching vulnerable populations. Despite this, the impact of DISCs has been limited and new ways to increase access and effectiveness are needed. OBJECTIVE: We are conducting a hybrid effectiveness-dissemination study. We aim to evaluate the effectiveness of a machine learning-based approach (recommender system) for computer-tailored health communication (CTHC) over a standard CTHC system based on quit rates and risk reduction. In addition, this study will assess the dissemination of providing access to a peer recruitment toolset on recruitment rate and variability of the sample. METHODS: The Smoker-to-Smoker (S2S) study is a 6-month hybrid effectiveness dissemination trial conducted nationally among English-speaking, current smokers aged \u3e /=18 years. All eligible participants will register for the DISC (Decide2quit) and be randomized to the recommender system CTHC or the standard CTHC, followed by allocation to a peer recruitment toolset group or control group. Primary outcomes will be 7-day point prevalence and risk reduction at the 6-month follow-up. Secondary outcomes include recruitment rate, website engagement, and patient-reported outcomes collected via the 6-month follow-up questionnaire. All primary analyses will be conducted on an intent-to-treat basis. RESULTS: The project is funded from 2017 to 2020 by the Patient Centered Outcomes Research Institute. Enrollment was completed in early 2019, and 6-month follow-ups will be completed by late 2019. Preliminary data analysis is currently underway. CONCLUSIONS: Conducting a hybrid study with both effectiveness and dissemination hypotheses raises some unique challenges in the study design and analysis.     Academic, industry and student perspectives on the inclusion of “vocational knowledge” in a ‘learning and teaching academic standards statement’ for agriculture

    We report on the perspective of academic, student and industry stakeholders in a national project that has developed a Learning and Teaching Academic Standards (LTAS) Statement for the Agriculture discipline. Agricultural research and teaching relies on strong links with industry due to the applied nature of the discipline. Without these links, sustainable and profitable practice change in agricultural systems cannot be achieved. A pilot project, in 2011-2012, with academic staff from three Australian universities identified vocational knowledge as a potential focus for a TLO. The AgLTAS project provided the opportunity to validate or refute this TLO by seeking input from a wider group of stakeholders, including industry. National consensus was sought by a process of iterative consultation with academics, students and industry stakeholders and tested across four Australian universities. We have collected qualitative and quantitative data from industry participants who attended a series of workshops across most Australian States and Territories. We also conducted an online survey. Surprisingly, and contrary to the findings of the pilot project, industry representatives considered vocational knowledge of lesser importance to the need for students to attain highly developed problem solving and communication skills that can generate new opportunities and innovation in agriculture. Industry-specific (vocational) knowledge was generally regarded as attainable during on-the-job training after graduation. This finding prompts the question whether the AgLTAS Statement should be linked to professional accreditation that may be attained either before or after graduation. The next phase of the project aims to trial the Standards Statement for Agriculture by benchmarking the academic standards achieved in four universities that teach Agriculture and related disciplines, using an online Curriculum Mapping Tool
