    Retroviral Inhibition of cAMP-Dependent Protein Kinase Inhibits Myelination But Not Schwann Cell Mitosis Stimulated by Interaction with Neurons

    Schwann cells are the myelinating glia of the peripheral nervous system. Neuron-Schwann cell contact profoundly affects several aspects of Schwann cell phenotype, including stimulation of mitosis and myelin formation. Many reports suggest that neuronal contact exerts this influence on Schwann cells by elevating Schwann cell cAMP and activating cAMP-dependent protein kinase A (PKA). To elucidate the importance of Schwann cell PKA in neuronal stimulation of Schwann cell mitosis and myelination, the gene encoding the PKA inhibitory protein RIalphaAB or PKIEGFP was delivered to Schwann cells using retroviral vectors. PKA inhibitory retroviral vectors effectively blocked forskolin-stimulated Schwann cell mitosis and morphological change, demonstrating the ability of the vectors to inhibit PKA in infected Schwann cells. Treatment of dorsal root ganglia neuron-Schwann cell cocultures with H-89 (10 microm) or KT5720 (1-10 microm), chemical inhibitors selective for PKA, significantly inhibited neuronal stimulation of Schwann cell mitosis. In contrast, retrovirus-mediated inhibition of Schwann cell PKA had no effect on the ability of neurons to stimulate Schwann cell mitosis. However, markedly fewer myelin segments were formed by Schwann cells expressing PKA inhibitory proteins compared with controls. These results suggest that activation of Schwann cell PKA is required for myelin formation but not for Schwann cell mitosis stimulated by interaction with neurons

    We review the assumptions and the logic underlying the derivation of DLCQ Matrix models. In particular we try to clarify what remains valid at finite NN, the role of the non-renormalization theorems and higher order terms in the supergravity expansion. The relation to Maldacena's conjecture is also discussed. In particular the compactification of the Matrix model on T3T_3 is compared to the AdS5×S5AdS_5\times S_5 N=4{\cal N}=4 super Yang-Mills duality, and the different role of the branes in the two cases is pointed out.Comment: 19 pages, Late

    Lemoine et al. (2006) and Lemoine et al. (2010) showed that applying more detailed models of time-variable gravity (TVG) improved the quality of the altimeter satellite orbits (e.g. TOPEX/Poseidon, Jason-1, Jason-2). This modeling include application of atmospheric gravity derived from 6-hrly pressure fields obtained from the ECMWF and annual gravity variations to degree & order 20x20 in spherical harmonics derived from GRACE data. This approach allowed the development of a consistent geophysical model for application to altimeter satellite orbit determination from 1993 to 2011. In addition, we have also evaluated the impact of TVG modeling on the POD of Jason-1 and Jason-2 by application of a weekly degree & order four gravity coefficient time series developed using data from ten SLR & DORIS-tracked satellites from 1993 to 2011 (Lemoine et al., 2011)

    Expression of lymphotoxin-αÎČ on antigen-specific T cells is required for DC function

    During an immune response, activated antigen (Ag)-specific T cells condition dendritic cells (DCs) to enhance DC function and survival within the inflamed draining lymph node (LN). It has been difficult to ascertain the role of the tumor necrosis factor (TNF) superfamily member lymphotoxin-αÎČ (LTαÎČ) in this process because signaling through the LTÎČ-receptor (LTÎČR) controls multiple aspects of lymphoid tissue organization. To resolve this, we have used an in vivo system where the expression of TNF family ligands is manipulated only on the Ag-specific T cells that interact with and condition Ag-bearing DCs. We report that LTαÎČ is a critical participant required for optimal DC function, independent of its described role in maintaining lymphoid tissue organization. In the absence of LTαÎČ or CD40L on Ag-specific T cells, DC dysfunction could be rescued in vivo via CD40 or LTÎČR stimulation, respectively, suggesting that these two pathways cooperate for optimal DC conditioning

    During the last thirty years, ‘consumption’ has become a major topic in the study of contemporary culture within anthropology, psychology and sociology. For many authors it has become central to understanding the nature of material culture in the modern world but this paper argues that the concept is, in British writing at least, too concerned with its economic origins in the selling and buying of consumer goods or commodities. It is argued that to understand material culture as determined through the monetary exchange for things - the cash nexus - leads to an inadequate sociological understanding of the social relations with objects. The work of Jean Baudrillard is used both to critique the concept of consumption as it leads to a focus on advertising, choice, money and shopping and to point to a more sociologically adequate approach to material culture that explores objects in a system of models and series, ‘atmosphere’, functionality, biography, interaction and mediation

    CO emission and associated HI absorption from a massive gas reservoir surrounding the z=3 radio galaxy B3 J2330+3927

    We present results of a comprehensive multi-frequency study of the radio galaxy B3 J2330+3927. The 1.9" wide radio source, consisting of 3 components, is bracketed by 2 objects in our Keck K-band image. Optical and near-IR Keck spectroscopy of these two objects yield z=3.087+-0.004. The brightest (K=18.8) object has a standard type II AGN spectrum, and is the most likely location of the AGN, which implies a one-sided jet radio morphology. Deep 113 GHz observations with the IRAM Plateau de Bure Interferometer reveal CO J=4-3 emission, which peaks at the position of the AGN. The CO line is offset by 500 km/s from the systemic redshift of the AGN, but corresponds very closely to the velocity shift of an associated HI absorber seen in Lya. This strongly suggests that both originate from the same gas reservoir surrounding the AGN host galaxy. Simultaneous 230 GHz interferometer observations find a ~3x lower integrated flux density when compared to single dish 250 GHz observations with MAMBO at the IRAM 30m telescope. This can be interpreted as spatially resolved thermal dust emission at scales of 0.5" to 6". Finally, we present a tau <1.3% limit to the HI 21 cm absorption against the radio source, which represents the seventh non-detection out of 8 z>2 radio galaxies observed to date with the WSRT. We present mass estimates for the atomic, neutral, and ionized hydrogen, and for the dust, ranging from M(HI)=2x10^7 M_Sun derived from the associated HI absorber in Lya up to M(H_2)=7x10^{10} M_Sun derived from the CO emission. This indicates that the host galaxy is surrounded by a massive reservoir of gas and dust. The K-band companion objects may be concentrations within this reservoir, which will eventually merge with the central galaxy hosting the AGN.Comment: 16 Pages, including 11 PostScript figures.     Bloodlines: mammals, leeches, and conservation in southern Asia

    Southern Asia is a biodiversity hotspot both for terrestrial mammals and for leeches. Many small-mammal groups are under-studied in this region, while other mammals are of known conservation concern. In addition to standard methods for surveying mammals, it has recently been demonstrated that residual bloodmeals within leeches can be sequenced to find mammals in a given area. While these invertebrate-parasite-derived DNA (iDNA) methods are promising, most of the leech species utilized for this type of survey remain unevaluated, notwithstanding that their diversity varies substantially. Here we examine approximately 750 individual leech specimens in the genus Haemadipsa across a large range in southern Asia (Bangladesh, Cambodia, and China), specifically reviewing the diversity of mammals they feed on and their own genetic structuring. Leeches were found to feed on a considerable variety of mammals, corroborating prior studies. Additionally, leeches were found to have fed both on bats and on birds, neither of which has previously been recorded with this method. The genetic structuring of the leeches themselves revealed 15 distinct clades of which only two precisely corresponded to previously characterized species, indicating that much work is needed to finalize classifications in this genus. Most importantly, with regards to mammal conservation, leeches in these clades appear to feed on a broad range of mammals

    M(atrix) Theory: Matrix Quantum Mechanics as a Fundamental Theory

    A self-contained review is given of the matrix model of M-theory. The introductory part of the review is intended to be accessible to the general reader. M-theory is an eleven-dimensional quantum theory of gravity which is believed to underlie all superstring theories. This is the only candidate at present for a theory of fundamental physics which reconciles gravity and quantum field theory in a potentially realistic fashion. Evidence for the existence of M-theory is still only circumstantial---no complete background-independent formulation of the theory yet exists. Matrix theory was first developed as a regularized theory of a supersymmetric quantum membrane. More recently, the theory appeared in a different guise as the discrete light-cone quantization of M-theory in flat space. These two approaches to matrix theory are described in detail and compared. It is shown that matrix theory is a well-defined quantum theory which reduces to a supersymmetric theory of gravity at low energies. Although the fundamental degrees of freedom of matrix theory are essentially pointlike, it is shown that higher-dimensional fluctuating objects (branes) arise through the nonabelian structure of the matrix degrees of freedom.
