Putoaville ratkaisu : Työhönvalmennusta ja päihdekuntoutusta nuorille
PURA – toiminnasta työkyky -kehittämishanke toimi vuosina 2014–2017 Sytyke-Centren/Hengitysliitto ry:n, Kelan, Peruspalvelukuntayhtymä Kallion, Avominne-päihdeklinikan ja Kuntoutussäätiön yhteistyönä. Hankkeen PURA-nimi muodostuu sanoista ”putoaville ratkaisuja”. Hankkeessa kehitettiin ja järjestettiin PURA-valmennusta alle 29-vuotiaille nuorille, jotka tarvitsevat tukea ammattiin opiskelussa tai työelämään pääsyssä. Valmennukseen osallistuneilla nuorilla oli mielenterveyden pulmia, päihteiden käyttö haittasi ammatillisten suunnitelmien etenemistä tai nuoren elämäntilanne oli muuten haastava. Valmennuksen tavoitteena oli edistää syrjäytymisvaarassa olevien nuorten kuntoutumista, työelämävalmiuksien parantumista ja työelämään sijoittumista. Valmennus koostui kolmesta osasta: toiminnallisesta työ- ja toimintakyvyn arviointijaksosta, työkykyvalmennusjaksosta ja tarvittaessa päihdekuntoutusjaksosta. Hankkeen arviointitutkimuksen tehtävänä oli selvittää, minkälaiset elementit edistävät kohderyhmän nuorten edellytyksiä sitoutua kuntoutukseen ja saavuttaa sille asetettuja tavoitteita. Aineiston muodostivat valmennukseen osallistuneilta nuorilta sekä valmennuksesta vastaavilta ammattilaisilta kerätyt kerronnalliset haastattelut. Analyysimetodina käytettiin laadullista aineistolähtöistä sisällönanalyysiä yhteisöllisen vuorovaikutuksen viitekehyksessä. Tuloksena todetaan, että PURA-valmennus tarjoaa mallin nuorten varhaisvaiheen kuntoutukselle. Sen tarkoituksena on saada syrjäytyneet nuoret palvelujen piiriin, jotta nuoren elämään liittyvää ongelmien vyyhtiä voidaan tarkastella yhdessä nuorten, kuntoutuksen moniammatillisen työryhmän ja ammatillisen palveluverkoston toimijoiden kanssa ja etsiä parhaita yksilöllisiä ratkaisuja nuoren ongelmiin. Nuorta tuetaan rakentamaan motiivia irrottautua päihteidenkäytöstä lisäämällä ymmärrystä päihdeongelman haitoista sekä osoittamalla päihteettömyyden olevan mahdollista ja ympäristön tarjoavan hänelle resursseja päihteidenkäytön lopettamiseen. Päihteidenkäytön lopettamisen lisäksi nuori tarvitsee tukea arkisten asioiden hoitamisen opetteluun sekä asumiseen ja toimeentuloon liittyviin asioihin. Kyvykkyyksiin kohdistuva arviointi auttaa nuoria tunnistamaan omia vahvuuksiaan. Arviointi on luonteeltaan monialaista, kannattelevaa työ- ja toimintakyvyn arviointia, joka tukee nuoren työ- ja toimintakykyä ja edistää siten kuntoutuksen tavoitteiden saavuttamista. Lisäksi se muistuttaa nuorta niistä vahvuuksista, joita ammattilaiset ovat todenneet hänellä olevan, mikä on tärkeää varsinkin, jos nuoren motivaatio kuntoutukseen uhkaa notkahtaa. Työkykyvalmennuksella edistetään nuorten työllistymiskykyä kokonaisvaltaisella työotteella, jossa valmennus kohdistetaan niin nuoren vapaa-ajan toimintaan, nuorten keskuudessa vallitsevaan työn tekemisen kulttuuriin kuin tukemaan nuorta onnistumaan työssä ja löytämään motivaation työelämäsiirtymää tukevaan opiskeluun. PURA-valmennuksessa pyritään kontrolloivan arvioinnin sijaan arvioinnilla kannattelemaan nuorta ja tukemaan hänen itsemääräämistään valinnan mahdollisuuksia osoittamalla ja valintoja tukemalla. Näillä keinoin voidaan tukea nuoren sisäistä voimantunnetta, jonka varassa hän rohkenee asettaa tavoitteita ja sitoutua kuntoutukseen. Heikossa asemassa, monien ongelmien vyyhdissä elävät nuoret, jotka ovat kokeneet useita epäonnistumisia ja joiden toimintakyky on alentunut, hyötyvät ammattilaisten nuoria kannattelevasta työskentelyotteesta. Näiden nuorten elämässä ammatillisesta kuntoutuksesta voi muodostua käännekohta ammatillisten suunnitelmien motivoidessa nuorta ratkaisevaan elämänmuutokseen
The Core-Collapse Supernova Rate in Arp299 Revisited
We present a study of the CCSN rate in nuclei A and B1 of the luminous
infrared galaxy Arp299, based on 11 years of Very Large Array monitoring of
their radio emission at 8.4 GHz. Significant variations in the nuclear radio
flux density can be used to identify the CCSN activity in the absence of
high-resolution very long baseline interferometry observations. In the case of
the B1-nucleus, the small variations in its measured diffuse radio emission are
below the fluxes expected from radio supernovae, thus making it well-suited to
detect RSNe through flux density variability. In fact, we find strong evidence
for at least three RSNe this way, which results in a lower limit for the CCSN
rate of 0.28 +/- 0.16 per year. In the A-nucleus, we did not detect any
significant variability and found a SN detection threshold luminosity which
allows only the detection of the most luminous RSNe known. Our method is
basically blind to normal CCSN explosions occurring within the A-nucleus, which
result in too small variations in the nuclear flux density, remaining diluted
by the strong diffuse emission of the nucleus itself. Additionally, we have
attempted to find near-infrared counterparts for the earlier reported RSNe in
the Arp299 nucleus A, by comparing NIR adaptive optics images from the Gemini-N
telescope with contemporaneous observations from the European VLBI Network.
However, we were not able to detect NIR counterparts for the reported radio SNe
within the innermost regions of nucleus A. While our NIR observations were
sensitive to typical CCSNe at 300 mas from the centre of the nucleus A,
suffering from extinction up to A_v~15 mag, they were not sensitive to such
highly obscured SNe within the innermost nuclear regions where most of the EVN

The nuclear starburst in Arp 299-A: From the 5.0 GHz VLBI radio light-curves to its core-collapse supernova rate
The nuclear region of the Luminous Infra-red Galaxy Arp 299-A hosts a recent
( Myr), intense burst of massive star formation which is expected to
lead to numerous core-collapse supernovae (CCSNe). Previous VLBI observations,
carried out with the EVN at 5.0 GHz and with the VLBA at 2.3 and 8.4 GHz,
resulted in the detection of a large number of compact, bright, non-thermal
sources in a region \lsim150 pc in size. We aim at establishing the nature of
all non-thermal, compact components in Arp 299-A, as well as estimating its
core-collapse supernova rate. We use multi-epoch European VLBI Network (EVN)
observations taken at 5.0 GHz to image with milliarcsecond resolution the
compact radio sources in the nuclear region of Arp 299-A. We also use one
single-epoch 5.0 GHz Multi-Element Radio Linked Interferometer Network (MERLIN)
observation to image the extended emission in which the compact radio sources
--traced by our EVN observations-- are embedded. Twenty-six compact sources are
detected, 8 of them are new objects not previously detected. The properties of
all detected objects are consistent with them being a mixed population of CCSNe
and SNRs. We find clear evidence for at least two new CCSNe, implying a lower
limit to the CCSN rate of \nu_{\rm SN}\gsim0.80 SN/yr indicating that the
bulk of the current star formation in Arp 299-A is taking place in the
innermost pc. Our MERLIN observations trace a region of diffuse,
extended emission which is co-spatial to the region where all compact sources
are found. From this diffuse, non-thermal radio emission we obtain an
independent estimate for the core-collapse supernova rate, which is in the
range - 0.65 SN/yr, roughly in agreement with previous
estimates and our direct estimate of the CCSN rate from the compact radio

Modeling of LMNA-Related Dilated Cardiomyopathy Using Human Induced Pluripotent Stem Cells
Dilated cardiomyopathy (DCM) is one of the leading causes of heart failure and heart transplantation. A portion of familial DCM is due to mutations in the LMNA gene encoding the nuclear lamina proteins lamin A and C and without adequate treatment these patients have a poor prognosis. To get better insights into pathobiology behind this disease, we focused on modeling LMNA-related DCM using human induced pluripotent stem cell derived cardiomyocytes (hiPSC-CM). Primary skin fibroblasts from DCM patients carrying the most prevalent Finnish founder mutation (p.S143P) in LMNA were reprogrammed into hiPSCs and further differentiated into cardiomyocytes (CMs). The cellular structure, functionality as well as gene and protein expression were assessed in detail. While mutant hiPSC-CMs presented virtually normal sarcomere structure under normoxia, dramatic sarcomere damage and an increased sensitivity to cellular stress was observed after hypoxia. A detailed electrophysiological evaluation revealed bradyarrhythmia and increased occurrence of arrhythmias in mutant hiPSC-CMs on beta -adrenergic stimulation. Mutant hiPSC-CMs also showed increased sensitivity to hypoxia on microelectrode array and altered Ca2+ dynamics. Taken together, p.S143P hiPSC-CM model mimics hallmarks of LMNA-related DCM and provides a useful tool to study the underlying cellular mechanisms of accelerated cardiac degeneration in this disease
The Submillimeter Array 1.3 mm line survey of Arp 220
We present the first aperture synthesis unbiased spectral line survey toward
an extragalactic object. The survey covered the 40 GHz frequency range between
202 and 242 GHz of the 1.3 mm atmospheric window. We find that 80% of the
observed band shows molecular emission, with 73 features identified from 15
molecular species and 6 isotopologues. The 13C isotopic substitutions of HC3N
and transitions from H2(18)O, 29SiO, and CH2CO are detected for the first time
outside the Galaxy. Within the broad observed band, we estimate that 28% of the
total measured flux is due to the molecular line contribution, with CO only
contributing 9% to the overall flux. We present maps of the CO emission at a
resolution of 2.9"x1.9" which, though not enough to resolve the two nuclei,
recover all the single-dish flux. The 40 GHz spectral scan has been modelled
assuming LTE conditions and abundances are derived for all identified species.
The chemical composition of Arp 220 shows no clear evidence of an AGN impact on
the molecular emission but seems indicative of a purely starburst-heated ISM.
The overabundance of H2S and the low isotopic ratios observed suggest a
chemically enriched environment by consecutive bursts of star formation, with
an ongoing burst at an early evolutionary stage. The large abundance of water
(~10^-5), derived from the isotopologue H2(18)O, as well as the vibrationally
excited emission from HC3N and CH3CN are claimed to be evidence of massive star
forming regions within Arp 220. Moreover, the observations put strong
constraints on the compactness of the starburst event in Arp 220. We estimate
that such emission would require ~2-8x10^6 hot cores, similar to those found in
the Sgr B2 region in the Galactic center, concentrated within the central 700
