Elektronentransfer durch Oligopeptide : Synthese und kinetische Untersuchung von Polyprolin-Modellsystemen
In der vorliegenden Dissertation wurde ein Konzept vorgestellt, das es ermöglicht in einem
Oligopeptid im Sub-Mikrosekunden Zeitraum ein Radikalkation zu erzeugen. Die entwickelte
Methode zeichnet sich dabei durch die hohe Regioselektivität sowie die milden
Reaktionsbedingungen aus.
Bei den untersuchten Modellverbindungen von Typ 37 wurde der C-Terminus von
Oligopeptiden mit einem 2-pivaloyl-modifizierten Tetrahydrofuranderivat verknüpft
(Schema 5.1). Bei der Bestrahlung des Radikalvorläufers 37 kommt es nach einer Norrish-
Typ-I-Eliminierung (ka) und anschließender radikalinduzierter b-Eliminierung der
Diphenylphosphat-Austrittsgruppe (kE) zur Bildung eines Radikalkations der Art 39. Das infolge der Fragmentierung gebildete Radikalkation 39 hat stark oxidierende
Eigenschaften und vermag die Seitenketten geeigneter aromatischer Aminosäuren zu
oxidieren. Bei Flash-Photolyseexperimenten (XeCl-Excimer Laser, l = 308 nm) mit den
Modellverbindungen 67 und 69 (Schema 5.1) konnte gezeigt werden, dass die gebundenen
Aminosäuren Tyrosin und 2,4,6-Trimethoxyphenylalanin, durch das infolge der Bestrahlung
gebildeten Radikalkations der Art 39, oxidiert werden können. Dieser Vorgang wurde anhand der charakteristischen Absorption des gebildeten
Tyrosylradikals (lmax = 410 nm) bzw. des erzeugten Radikalkations von
2,4,6-Trimethoxyphenylalanin (lmax = 550 nm) verfolgt.
Bei anschließenden Experimenten stand die Untersuchung mehrstufiger Elektronentransfer-
Prozesse im Mittelpunkt. Dazu wurden die Modellsysteme 111 - 114 synthetisiert, die drei
verschiedene Redoxzentren aufweisen (Schema 5.2). In den durchgeführten Kaskaden-Experimenten oxidiert das nach der Bestrahlung der
Oligopeptide 111 - 114 entstehende Enolether-Radikalkation 115 in einem ersten
Elektronentransfer-Schritt (kET1) die Seitenkette von 2,4,6-Trimethoxyphenylalanin. Das
dabei gebildete Radikalkation des intermediären Elektronendonors (116) gibt in einem
zweiten Elektronentransfer-Schritt (kET2) die Ladung an Tyrosin, den finalen
Elektronendonor, weiter. Der Abstand des intermediären Elektronendonors zum finalen
Elektronendonor wurde in den Modellsystemen 111 - 114 variiert. Zum Einsatz kamen dabei
n = 0, 1, 3 und 5 verbrückende Prolineinheiten. Der in Schema 5.2 (S. 96) dargestellte Reaktionsablauf konnte durch die Aufnahme von
Transienten-Absorptions-Spektren bestätigt werden (Abbildung 5.2 A). Dabei war es
möglich, die Oxidation und anschließende Reduktion des intermediären Elektronendonors im
Verlauf des mehrstufigen Elektronentransfer-Prozesses zu verfolgen. Aufgrund der spektralen
Eigenschaften der oxidierten Aminosäuren gelang es, den Redoxprozess von
2,4,6-Trimethoxyphenylalanin unbeeinflusst vom Redoxprozess des finalen Elektronendonors
Tyrosin zeitaufgelöst zu untersuchen (Abbildung 5.2 B). Es wurde festgestellt, dass der erste Elektronentransfer-Schritt in den Modellsystemen
113 - 114 konzentrationsunabhängig mit kET1 ≥ 4·107 s-1 abläuft. Durch Auftragung der
beobachteten Geschwindigkeit des zweiten Elektronentransfer-Schritts gegen die
Konzentration gelang es die Geschwindigkeitskonstante für den intramolekularen
Elektronentransfer zwischen dem finalen und intermediären Elektronendonor (kET2,intra) zu
ermitteln. Darüber hinaus konnten durch die Auftragung Informationen zur Geschwindigkeit
des intermolekularen Elektronentransfers (kET2,inter) gewonnen werden (Tabelle 5.1, S. 98).
Die Werte für kET2,intra wurden bezüglich ihrer Distanzabhängigkeit untersucht. Dabei wurde
nur eine schwache Abhängigkeit von kET2,intra gegenüber der Anzahl n der verbrückenden
Prolineinheiten zwischen dem intermediären und finalen Elektronendonor festgestellt. Diese
Beobachtung steht im Einklang mit Untersuchungen des Elektronentransfers in anderen
Prolin-verbrückten Modellsystemen.[12, 74]. Die beobachtete Distanzabhängigkeit kann nicht auf der Grundlage eines Elektronentransfers
durch die Bindung erklärt werden. Vielmehr deuten die Versuchsergebnisse auf einen
Elektronentransfer durch den Raum hin. Dazu müssen in den Modellsystemen 111 - 114
Konformationen realisiert werden, bei denen die Redoxzentren deutlich geringere Abstände
aufweisen, als diese Rechnungen vermuten lassen. Bei Untersuchungen der Flexibilität von
Oligoprolin-Modellsystemen wurde kürzlich gezeigt, dass Konformationen, bei denen die
Enden der Oligopeptide in geringem Abstand zueinander vorliegen, im Zeitfenster des
beobachteten Elektronentransfers erreicht werden können.[157-159] Um ein umfassenderes Bild
vom Mechanismus des Elektronentransfers in den Oligopeptiden 111 - 114 zu erhalten, ist es
erforderlich weitere Untersuchungen zur Struktur der Verbindungen in Lösung
durchzuführen.
Damit exakte Messungen der Distanzabhängigkeit des Elektronentransfers in Modellsystemen
der Art 111 - 114 durchgeführt werden können, muss der Polyprolin-Abstandshalter durch ein
rigideres Strukturelement ersetzt werden. Dabei bietet sich der Einsatz von stabilisierten
Peptidstrukturen mit bekannter Röntgenstruktur an.[163]
Darüber hinaus könnte in zukünftigen Experimenten eine Aminosäure mit
2,4-Dimethoxybenzylseitenketten zum Einsatz kommen. Die erwartete Absorption des bei der
Oxidation dieser Aminosäure entstehenden Radikalkations liegt bei etwa 430 nm.[124]
Eventuell kann der Redoxprozess dieser Aminosäure ungestört neben dem Tyrosylradikal und
dem Radikalkation von 2,4,6-Trimethoxyphenylalanin untersucht werden. Damit wäre die
Synthese von Peptiden möglich, in denen drei Elektronentransfer-Schritte nebeneinander
zeitaufgelöst verfolgt werden können

Cold Atmospheric Plasma in the Treatment of Osteosarcoma
Human osteosarcoma (OS) is the most common primary malignant bone tumor occurring most commonly in adolescents and young adults. Major improvements in disease-free survival have been achieved by implementing a combination therapy consisting of radical surgical resection of the tumor and systemic multi-agent chemotherapy. However, long-term survival remains poor, so novel targeted therapies to improve outcomes for patients with osteosarcoma remains an area of active research. This includes immunotherapy, photodynamic therapy, or treatment with nanoparticles. Cold atmospheric plasma (CAP), a highly reactive (partially) ionized physical state, has been shown to inherit a significant anticancer capacity, leading to a new field in medicine called “plasma oncology.” The current article summarizes the potential of CAP in the treatment of human OS and reviews the underlying molecular mode of action
Lösemittelrückgewinnung im Verpackungsdruck - Stoffrecycling versus Verbrennung
Im Verpackungsdruck werden aus Qualitätsanforderungen an das Druckerzeugnis or-ganische Lösemittel (vorwiegend Ethylacetat und Ethanol) eingesetzt. Diese verdampfen beim Trocknungsprozess, gelangen in die Abluft und anschließend in eine Abgas-reinigungsanlage. Bei Lösemittelmassenströmen von 1 t/h und mehr ist hierbei heute eine Lösemittelrückgewinnungsanlage mit adsorptiver Aufkonzentration (Aktivkohle, Zeolithe), Wasserdampfdesorption und Kondensation sowie aufwändiger Kondensataufarbeitung wirtschaftlich tragfähig darstellbar. Für Lösemittelmassenströme unterhalb der benannten Schwelle ist der Einsatz von regenerativen Nachverbrennungsanlagen Stand der Technik, die Lösemittel oxidieren und wandeln sich in die nichttoxischen Substanzen Kohlendioxid und Wasser um. Zur Erhöhung der Ressourceneffizienz und Senkung des Kohlendioxidausstoßes ist der neuartige Ansatz gewählt worden, kein ad- sondern ein absorptives Aufkonzentrationsverfahren auf der Basis von Hochsiedern (Genosorb) und Inertgasdesorption (Stickstoff) mit dem Ziel zu erproben, die wirtschaftliche Tragfähigkeit einer Lösemittelrückgewinnung auch schon bei niedrigeren Lösemittelmassenströmen nachzuweisen. Der besondere Vorteil dieses Verfahrensansatzes liegt in der niedrigen Desorptionstemperatur mit Inertgas (100...130) °C. Im Fall von Adsorbentien würde hierfür eine deutlich erhöhte Desorptionstemperatur (> 200 °C) vonnöten sein, so dass in Verbindung mit dem Wasser in der Umgebungsluft und der großen, ggf. katalytisch unterstützenden Oberfläche des Adsorbens eine Zersetzung vorwiegend von Ethylacetat zu Essigsäure auftreten würde. Die bisher durchgeführten Forschungsanstrengungen lassen den Schluss zu, dass der neue Technologieansatz sich offenbar zielführend wirtschaftlich gestaltet und die Kohlenstoffdioxidemissionen gegenüber dem Verbrennungsverfahren deutlich (40 bis 50 %) gesenkt werden können
Patient preferences for development in MRI scanner design: a survey of claustrophobic patients in a randomized study
Objective: To investigate which magnetic resonance imaging (MRI) scanner designs claustrophobic patients prefer.
Material/methods: We analyzed questionnaires completed by 160 patients at high risk for claustrophobia directly after a scan in either a short-bore or open panoramic scanner as part of a prospective randomized trial Enders et al (BMC Med Imaging 11:4, 2011). Scanner preferences were judged based on schematic drawings of four scanners. Information on the diagnostic performance of the depicted scanners was provided, too.
Results: A majority of patients suggested upright open (59/160, 36.9%) and open panoramic (53/160, 33.1%) before short-bore designs (26/160, 16.3%, for all p < 0.001) for future development. When asked about patients' preferred scanner choice for an upcoming examination, information about a better diagnostic performance of a short-bore scanner significantly improved its preference rates (from 6/160 to 49/160 or 3.8 to 30.5%, p < 0.001). Patients with a claustrophobic event preferred open designs significantly more often than patients without a claustrophobic event (p = 0.047). Patients scanned in a short-bore scanner in our trial preferred this design significantly more often (p = 0.003). Noise reduction (51/160, 31.9%), more space over the head (44/160, 27.5%), and overall more space (33/160, 20.6%) were the commonest suggested areas of improvement.
Conclusion: Patients at high risk for claustrophobia visually prefer open- over short-bore MRI designs for further development. Education about a better diagnostic performance of a visually less-attractive scanner can increase its acceptance. Noise and space were of most concern for claustrophobic patients. This information can guide individual referral of claustrophobic patients to scanners and future scanner development.

• Patients at high risk for claustrophobia visually favor the further development of open scanners as opposed to short- and closed-bore scanner designs.
• Educating claustrophobic patients about a higher diagnostic performance of a short-bore scanner can significantly increase their acceptance of this otherwise visually less-attractive design.
• A medical history of earlier claustrophobic events in a given MRI scanner type and focusing on the features "more space" and "noise reduction" can help to guide referral of patients who are at high risk for claustrophobia

A Neutrophil Proteomic Signature in Surgical Trauma Wounds
Non-healing wounds continue to be a clinical challenge for patients and medical staff. These wounds have a heterogeneous etiology, including diabetes and surgical trauma wounds. It is therefore important to decipher molecular signatures that reflect the macroscopic process of wound healing. To this end, we collected wound sponge dressings routinely used in vacuum assisted therapy after surgical trauma to generate wound-derived protein profiles via global mass spectrometry. We confidently identified 311 proteins in exudates. Among them were expected targets belonging to the immunoglobulin superfamily, complement, and skin-derived proteins, such as keratins. Next to several S100 proteins, chaperones, heat shock proteins, and immune modulators, the exudates presented a number of redox proteins as well as a discrete neutrophil proteomic signature, including for example cathepsin G, elastase, myeloperoxidase, CD66c, and lipocalin 2. We mapped over 200 post-translational modifications (PTMs; cysteine/methionine oxidation, tyrosine nitration, cysteine trioxidation) to the proteomic profile, for example, in peroxiredoxin 1. Investigating manually collected exudates, we confirmed presence of neutrophils and their products, such as microparticles and fragments containing myeloperoxidase and DNA. These data confirmed known and identified less known wound proteins and their PTMs, which may serve as resource for future studies on human wound healing
Design and Analysis of a LWIR Detector Electronics for Space Applications
µBolometers based on a-Si technology provide a marketable performance while at the same time requiring only moderate resources in terms of cooling requirements. The PICO1024 is a high resolution (1024x768 pixels) infrared image sensor for thermography applications. It makes use of LYNRED latest, state of the art, 17 µm pixel pitch technology. The PICO1024 sensor is a 2-dimensional detector array sensitive to infrared radiation in the long wave spectral range (LWIR) from 8 to 14 m. It uses microbolometer technology to convert infrared radiation into electronic signal for use in thermal imaging cameras. Starting from an analysis of the Capacitance Trans-Impedance Amplifier (CTIA) the front-end electronics architecture is described and analyzed. This includes transient and AC Spice simulations as well as an analysis of the readout signal chain noise performance. The paper also presents a possible replacement of key components (ADC, opamp) by low power, high performance space qualified IEEE parts. Clinical pre-test probability for obstructive coronary artery disease: insights from the European DISCHARGE pilot study.
To test the accuracy of clinical pre-test probability (PTP) for prediction of obstructive coronary artery disease (CAD) in a pan-European setting. Patients with suspected CAD and stable chest pain who were clinically referred for invasive coronary angiography (ICA) or computed tomography (CT) were included by clinical sites participating in the pilot study of the European multi-centre DISCHARGE trial. PTP of CAD was determined using the Diamond-Forrester (D+F) prediction model initially introduced in 1979 and the updated D+F model from 2011. Obstructive coronary artery disease (CAD) was defined by one at least 50% diameter coronary stenosis by both CT and ICA. In total, 1440 patients (654 female, 786 male) were included at 25 clinical sites from May 2014 until July 2017. Of these patients, 725 underwent CT, while 715 underwent ICA. Both prediction models overestimated the prevalence of obstructive CAD (31.7%, 456 of 1440 patients, PTP: initial D+F 58.9% (28.1-90.6%), updated D+F 47.3% (34.2-59.9%), both p < 0.001), but overestimation of disease prevalence was higher for the initial D+F (p < 0.001). The discriminative ability was higher for the updated D+F 2011 (AUC of 0.73 95% confidence interval [CI] 0.70-0.76 versus AUC of 0.70 CI 0.67-0.73 for the initial D+F; p < 0.001; odds ratio (or) 1.55 CI 1.29-1.86, net reclassification index 0.11 CI 0.05-0.16, p < 0.001).
