    The role of bacteria in pine wilt disease: insights from microbiome analysis.

    Pine Wilt Disease (PWD) has a significant impact on Eurasia pine forests. The microbiome of the nematode (the primary cause of the disease), its insect vector, and the host tree may be relevant for the disease mechanism. The aim of this study was to characterize these microbiomes, from three PWD-affected areas in Portugal, using Denaturing Gradient Gel Electrophoresis, 16S rRNA gene pyrosequencing, and a functional inference-based approach (PICRUSt). The bacterial community structure of the nematode was significantly different from the infected trees but closely related to the insect vector, supporting the hypothesis that the nematode microbiome might be in part inherited from the insect. Sampling location influenced mostly the tree microbiome (P < 0.05). Genes related both with plant growth promotion and phytopathogenicity were predicted for the tree microbiome. Xenobiotic degradation functions were predicted in the nematode and insect microbiomes. Phytotoxin biosynthesis was also predicted for the nematode microbiome, supporting the theory of a direct contribution of the microbiome to tree-wilting. This is the first study that simultaneously characterized the nematode, tree and insect-vector microbiomes from the same affected areas, and overall the results support the hypothesis that the PWD microbiome plays an important role in the disease's development

    A method to consider the nonlinear behaviour of reinforced concrete in flexible earth-retaining walls: preliminary results

    This paper presents a numerical methodology to incorporate in finite element analyses of deep excavations the nonlinear behaviour of reinforced concrete walls. This method is based on the interaction between two well validated numerical models: one used for incremental analysis of geotechnical works (but assuming concrete as linear elastic) and the other used for nonlinear analysis of reinforced concrete structures taking into account the characteristics of the concrete and the steel reinforcement at each structural section. The analysis of an excavation in clay supported by a single propped diaphragm wall is presented as an example of application. It is shown how the consideration of the nonlinear behaviour of reinforced concrete influences the global performance of the retaining structure

    A systematic procedure to enhance reproducibility of SWASV cycles in the determination of toxic metals in real samples

    This work presents for the first time a systematic study on the optimization of the electrochemical cleaning time of a mercury film when it is used as a working electrode material in the analysis of toxic metals, such as Pb2+, used as model metal, in real samples by SWASV. The optimization study for the film’s cleaning time aimed at attaining a Pb2+ minimum value in the film after the re-oxidation step of the pre-concentrated metal, given the impossibility of complete removal of traces of the electroactive species from the film. This value was kept constant in each concentration range studied ensuring thus that all assays were performed in initial identical conditions. An assay performed on a synthetic sample was taken as reference. In it, given the absence of matrix effects, and after the electrochemical cleaning step, a direct proportionality was observed between the residual amounts of Pb2+ in the film (which for the cleaning time used was never completely removed) and Pb2+ concentration in the solution. This fact determined a high correlation between Pb2+ peak current and Pb2+ concentration which was not observed when real samples (tree leaves) were analyzed. This behavior may result from the presence of the interfering surfactants always present in real samples of complex matrix. Cleaning time optimization was performed for the following Pb2+ concentration ranges in the real samples of complex matrix: 0.006-0.020, 0.020-0.080, 0.060-0.200 and 0.100-0.600 ppb. As expected, in order to obtain identical levels of film’s cleaning efficiency, the need for longer cleaning times has been observed for higher concentrations.     Lipid composition and dynamics of cell membranes of Bacillus stearothermophilus adapted to amiodarone

    Bacillus stearothermophilus, a useful model to evaluate membrane interactions of lipophilic drugs, adapts to the presence of amiodarone in the growth medium. Drug concentrations in the range of 1-2 [mu]M depress growth and 3 [mu]M completely suppresses growth. Adaptation to the presence of amiodarone is reflected in lipid composition changes either in the phospholipid classes or in the acyl chain moieties. Significant changes are observed at 2 [mu]M and expressed by a decrease of phosphatidylethanolamine (relative decrease of 23.3%) and phosphatidylglycerol (17.9%) and by the increase of phosphoglycolipid (162%). The changes in phospholipid acyl chains are expressed by a decrease of straight-chain saturated fatty acids (relative decrease of 12.2%) and anteiso-acids (22%) with a parallel increase of the iso-acids (9.8%). Consequently, the ratio straight-chain/branched iso-chain fatty acids decreases from 0.38 (control cultures) to 0.30 (cultures adapted to 2 [mu]M amiodarone). The physical consequences of the lipid composition changes induced by the drug were studied by fluorescence polarization of diphenylhexatriene and diphenylhexatriene-propionic acid, and by differential scanning calorimetry. The thermotropic profiles of polar lipid dispersions of amiodarone-adapted cells are more similar to control cultures (without amiodarone) than those resulting from a direct interaction of the drug with lipids, i.e., when amiodarone was added directly to liposome suspensions. It is suggested that lipid composition changes promoted by amiodarone occur as adaptations to drug tolerance, providing the membrane with physico-chemical properties compatible with membrane function, counteracting the effects of the drug.http://www.sciencedirect.com/science/article/B6VNN-419BF60-K/1/8f2d4fae7f9c131d26230cf4123da94

    Electrospun composite cellulose acetate/iron oxide nanoparticles non-woven membranes for magnetic hyperthermia applications

    In the present work composite membranes were produced by combining magnetic nanoparticles (NPs) with cellulose acetate (CA) membranes for magnetic hyperthermia applications. The non-woven CA membranes were produced by electrospinning technique, and magnetic NPs were incorporated by adsorption at fibers surface or by addition to the electrospinning solution. Therefore, different designs of composite membranes were obtained. Superparamagnetic NPs synthesized by chemical precipitation were stabilized either with oleic acid (OA) or dimercaptosuccinic acid (DMSA) to obtain stable suspensions at physiological pH. The incorporation of magnetic NP into CA matrix was confirmed by scanning and transmission electron microscopy. The results showed that adsorption of magnetic NPs at fibers' surface originates composite membranes with higher heating ability than those produced by incorporation of magnetic NPs inside the fibers. However, adsorption of magnetic NPs at fibers' surface can cause cytotoxicity depending on the NPs concentration.     Analysis of Grip Amplitude on Velocity in Paralympic Powerlifting

    (1) Background: Paralympic Powerlifting (PP) is a Paralympic modality that is predominantly about developing maximal force, as there are athletes who lift three times their body weight. Our objective was to evaluate the averages of the velocity for 30% and 50% of 1 Maximum Repetition (1 RM) on different amplitudes of the footprint in PP athletes; (2) Methods: The intervention happened over two weeks, with the first being devoted to the familiarization and testing of 1 RM, while in the second week, through the use of a linear Encoder, tests of velocity average (VA), velocity average propulsive (VAP), and velocity peak (VP) were carried out with loads of 30% and 50% of a maximum repetition 1 RM for 1× of the biacromial distance (BAD) 1.3 × BAD, 1.5 × BAD; (3) Results: There was a significant difference in the average velocity of 1 × BAD (1.16 ± 0.14 m/s, 1.07–1.26 IC; η2p 0.20) when compared to 1.3 × BAD (1.00 ± 0.17 m/s, 0.90–1.09 IC; η2p 0.20) over 30% of 1 RM. For the other velocity variables for 30% and 50% of 1 RM with different grip amplitudes, there were no significant differences; (4) Conclusions: In PP, the 1 × BAD footprint contributes significantly to VA at 30% of 1 RM when compared to the 1.3 × BAD and 1.5 × BAD footprints. For loading at 50% of 1 RM the VA, VAP and VP decreased when compared to 30% of 1 RM, to the extent that the VAP and VP generated with the 1.3 × BAD and 1.5 × BAD footprints were higher than those with 1 × BAD, other than for VA 50% of 1 RM, where the 1 × BAD footprint was superior to the others

    Boat noise impacts Lusitanian toadfish breeding males and reproductive outcome

    Anthropogenic noise is a growing threat to marine organisms, including fish. Yet very few studies have addressed the impact of anthropogenic noise on fish reproduction, especially in situ. In this study, we investigated the impacts of boat noise exposure in the reproductive success of wild Lusitanian toadfish (Halobatrachus didactylus), a species that relies on advertisement calls for mate attraction, using behavioural, physiological and reproductive endpoints. Two sets of artificial nests were deployed in the Tagus estuary and exposed to either ambient sound or boat noise during their breeding season. Toadfish males spontaneously used these nests to breed. We inspected nests for occupation and the presence of eggs in six spring low tides (in two years) and assessed male vocal activity and stress responses. Boat noise did not affect nest occupation by males but impacted reproductive success by decreasing the likelihood of receiving eggs, decreasing the number of live eggs and increasing the number of dead eggs, compared to control males. Treatment males also showed depressed vocal activity and slightly higher cortisol levels. The assessment of oxidative stress and energy metabolism-related biomarkers revealed no oxidative damage in noise exposed males despite having lower antioxidant responses and pointed towards a decrease in the activity levels of energy metabolism-related biomarkers. These results suggest that males exposed to boat noise depressed their metabolism and their activity (such as parental care and mate attraction) to cope with an acoustic stressor, consistent with a freezing defensive response/behaviour. Together, our study demonstrates that boat noise has severe impacts on reproductive fitness in Lusitanian toadfish.
