    Neuronal accumulation of unrepaired DNA in a novel specific chromatin domain: structural, molecular and transcriptional characterization

    There is growing evidence that defective DNA repair in neurons with accumulation of DNA lesions and loss of genome integrity underlies aging and many neurodegenerative disorders. An important challenge is to understand how neurons can tolerate the accumulation of persistent DNA lesions without triggering the apoptotic pathway. Here we study the impact of the accumulation of unrepaired DNA on the chromatin architecture, kinetics of the DNA damage response and transcriptional activity in rat sensory ganglion neurons exposed to 1-to-3 doses of ionizing radiation (IR). In particular, we have characterized the structural, molecular and transcriptional compartmentalization of unrepaired DNA in persistent DNA damaged foci (PDDF). IR induced the formation of numerous transient foci, which repaired DNA within the 24 h post-IR, and a 1-to-3 PDDF. The latter concentrate DNA damage signaling and repair factors, including ?H2AX, pATM, WRAP53 and 53BP1. The number and size of PDDF was dependent on the doses of IR administered. The proportion of neurons carrying PDDF decreased over time of post-IR, indicating that a slow DNA repair occurs in some foci. The fine structure of PDDF consisted of a loose network of unfolded 30 nm chromatin fiber intermediates, which may provide a structural scaffold accessible for DNA repair factors. Furthermore, the transcription assay demonstrated that PDDF are transcriptionally silent, although transcription occurred in flanking euchromatin. Therefore, the expression of ?H2AX can be used as a reliable marker of gene silencing in DNA damaged neurons. Moreover, PDDF were located in repressive nuclear environments, preferentially in the perinucleolar domain where they were frequently associated with Cajal bodies or heterochromatin clumps forming a structural triad. We propose that the sequestration of unrepaired DNA in discrete PDDF and the transcriptional silencing can be essential to preserve genome stability and prevent the synthesis of aberrant mRNA and protein products encoded by damaged genes

    Persistent accumulation of unrepaired DNA damage in rat cortical neurons: nuclear organization and ChIP-seq analysis of damaged DNA

    Neurons are highly vulnerable to DNA damage induced by genotoxic agents such as topoisomerase activity, oxidative stress, ionizing radiation (IR) and chemotherapeutic drugs. To avert the detrimental effects of DNA lesions in genome stability, transcription and apoptosis, neurons activate robust DNA repair mechanisms. However, defective DNA repair with accumulation of unrepaired DNA are at the basis of brain ageing and several neurodegenerative diseases. Understanding the mechanisms by which neurons tolerate DNA damage accumulation as well as defining the genomic regions that are more vulnerable to DNA damage or refractory to DNA repair and therefore constitute potential targets in neurodegenerative diseases are essential issues in the field. In this work we investigated the nuclear topography and organization together with the genome-wide distribution of unrepaired DNA in rat cortical neurons 15 days upon IR. About 5% of non-irradiated and 55% of irradiated cells accumulate unrepaired DNA within persistent DNA damage foci (PDDF) of chromatin. These PDDF are featured by persistent activation of DNA damage/repair signaling, lack of transcription and localization in repressive nuclear microenvironments. Interestingly, the chromatin insulator CTCF is concentrated at the PDDF boundaries, likely contributing to isolate unrepaired DNA from intact transcriptionally active chromatin. By confining damaged DNA, PDDF would help preserving genomic integrity and preventing the production of aberrant proteins encoded by damaged genes.ChIP-seq analysis of genome-wide ?H2AX distribution revealed a number of genomic regions enriched in ?H2AX signal in IR-treated cortical neurons. Some of these regions are in close proximity to genes encoding essential proteins for neuronal functions and human neurodegenerative disorders such as epm2a (Lafora disease), serpini1 (familial encephalopathy with neuroserpin inclusion bodies) and il1rpl1 (mental retardation, X-linked 21).     Ursolic and oleanolic acids as antimicrobial and immunomodulatory compounds for tuberculosis treatment

    BACKGROUND: New alternatives for the treatment of Tuberculosis (TB) are urgently needed and medicinal plants represent a potential option. Chamaedora tepejilote and Lantana hispida are medicinal plants from Mexico and their hexanic extracts have shown antimycobacterial activity. Bioguided investigation of these extracts showed that the active compounds were ursolic acid (UA) and oleanolic acid (OA). METHODS: The activity of UA and OA against Mycobacterium tuberculosis H37Rv, four monoresistant strains, and two drug-resistant clinical isolates were determined by MABA test. The intracellular activity of UA and OA against M. tuberculosis H37Rv and a MDR clinical isolate were evaluated in a macrophage cell line. Finally, the antitubercular activity of UA and OA was tested in BALB/c mice infected with M. tuberculosis H37Rv or a MDR strain, by determining pulmonary bacilli loads, tissue damage by automated histomorphometry, and expression of IFN-γ, TNF-α, and iNOS by quantitative RT-PCR. RESULTS: The in vitro assay showed that the UA/OA mixture has synergistic activity. The intracellular activity of these compounds against M. tuberculosis H37Rv and a MDR clinical isolate in a macrophage cell line showed that both compounds, alone and in combination, were active against intracellular mycobacteria even at low doses. Moreover, when both compounds were used to treat BALB/c mice with TB induced by H37Rv or MDR bacilli, a significant reduction of bacterial loads and pneumonia were observed compared to the control. Interestingly, animals treated with UA and OA showed a higher expression of IFN-γ and TNF-α in their lungs, than control animals. CONCLUSION: UA and OA showed antimicrobial activity plus an immune-stimulatory effect that permitted the control of experimental pulmonary TB

    Identification of Epigenetic Interactions between MicroRNA-30c-5p and DNA Methyltransferases in Neuropathic Pain

    Neuropathic pain is a prevalent and severe chronic syndrome, often refractory to treatment, whose development and maintenance may involve epigenetic mechanisms. We previously demonstrated a causal relationship between miR-30c-5p upregulation in nociception-related neural structures and neuropathic pain in rats subjected to sciatic nerve injury. Furthermore, a short course of an miR-30c-5p inhibitor administered into the cisterna magna exerts long-lasting antiallodynic effects via a TGF-?1-mediated mechanism. Herein, we show that miR-30c-5p inhibition leads to global DNA hyper-methylation of neurons in the lumbar dorsal root ganglia and spinal dorsal horn in rats subjected to sciatic nerve injury. Specifically, the inhibition of miR-30-5p significantly increased the expression of the novo DNA methyltransferases DNMT3a and DNMT3b in those structures. Furthermore, we identified the mechanism and found that miR-30c-5p targets the mRNAs of DNMT3a and DNMT3b. Quantitative methylation analysis revealed that the promoter region of the antiallodynic cytokine TGF-?1 was hypomethylated in the spinal dorsal horn of nerve-injured rats treated with the miR-30c-5p inhibitor, while the promoter of Nfyc, the host gene of miR-30c-5p, was hypermethylated. These results are consistent with long-term protection against neuropathic pain development after nerve injury.     Microbiological and Pharmacological Evaluation of the Micropropagated Rubus liebmannii Medicinal Plant

    Rubus liebmannii is an endemic species from Mexico used in traditional medicine primarily to treat dysentery and cough. The in vitro activity against Giardia lamblia and Entamoeba histolytica that produces the ethanolic extract of the aerial parts of the plant led us to expand the pharmacological and phytochemical research of this species. Gastrointestinal disorders including amebiasis remain one of the health problems that need to be addressed and it is of interest to find alternatives that improve their treatment. Also, it is important to emphasize that R. liebmannii grows wild in the country and is not found in abundance; therefore, alternatives that avoid overexploitation of the natural resource are mandatory. Ongoing with the evaluation of the potentialities that R. liebmannii possesses for treating infectious gastrointestinal diseases, the aim of the present study was to evaluate the biological effects and the chemical composition of the micropropagated plant

    Organización y dinámica de los focos nucleares permanentes de reparación del daño del DNA en neuronas

    RESUMEN: Cada vez hay más pruebas de que una reparación defectuosa del DNA en neuronas conlleva una acumulación de lesiones en el DNA y pérdida de la integridad del genoma, lo que tiene se asocia a envejecimiento y muchos trastornos neurodegenerativos. Un desafío importante es entender cómo las neuronas pueden tolerar la acumulación de lesiones persistentes de DNA sin desencadenar la vía apoptótica. Aquí se estudia el impacto de la acumulación de DNA no reparado en la arquitectura de la cromatina, la cinética de la respuesta al daño del DNA y la actividad transcripcional en neuronas ganglionares sensoriales de ratas expuestas a 1 a 3 dosis de radiación ionizante. En particular, hemos caracterizado la compartimentación estructural, molecular y transcripcional del DNA no reparado en los focos dañados con DNA persistente (PDDF). Proponemos que el secuestro de DNA no reparado en PDDF discretos y el silenciamiento transcripcional pueden ser procesos esenciales para preservar la estabilidad del genoma y evitar la síntesis de mRNAs aberrantes y productos proteicos codificados por genes dañados.ABSTRACT: There is growing evidence that defective DNA repair in neurons with accumulation of DNA lesions and loss of genome integrity underlies aging and many neurodegenerative disorders.     Introducción: la histoplasmosis es una infección sistémica causada por un hongo dimorfo, histoplasma capsulatum. Crece en suelos que contienen grandes cantidades de excretas tanto de aves como de murciélagos, en las proximidades del suelo de los gallineros y alrededor de árboles que albergan aves y murciélagos, en el interior o alrededor de las cuevas. La aerosolización de las microconidias residentes en estos suelos es la causante de las manifestaciones de la enfermedad Objetivo: dar a conocer un brote de esta enfermedad relacionado con una actividad de remoción de suelos en espacio abierto. Método: estudio observacional descriptivo de reporte de serie de casos que se inició con la identificación del caso índice. Se realizaron tomas de muestras tanto a los niños del colegio como al personal docente, así como de las fuentes de contagio común. Resultados: se identificaron 7 niños, edad promedio de 11 años, predominantemente del sexo femenino y sin enfermedad predisponente condicionante. El diagnóstico de la enfermedad se realizó por estudio de material clínico (esputo, médula ósea, tejido pulmonar) con coloraciones especiales. Se aisló histoplasma capsulatum del suelo donde estos niños residen y donde se desarrolló la actividad de remoción y rastrillo. Se estimó una tasa de ataque del 58 % con una letalidad del 14 %. Conclusión: se ha realizado el primer reporte para el país de un brote de histoplasmosis en espacio abierto relacionado con remoción y rastrillo de suelo, reconociendo un área de endemicidad previamente no determinada. It grows in soils with the consequent contamination of large amounts of birds and bats excreta, near barn soils and around trees that harbor birds and bats, and also inside or around caves. Aerosolization of microconidia residents in these soils is the cause of the transmision of the disease Objective: to report an outbreak of this disease related to an activity of soil removal in an open space. Methods: this is a descriptive observational study of a case series that began with the identification of the index case. Samples were obtained from school children and teachers, as well as from common sources of infection. Results: 7 children, predominantly females with an average age of 11 years, with no predisposing disease were included. Diagnosis was performed by special colorations of sputum, bone marrow and lung tissue. Histoplasma capsulatum was isolated from the soil where these children reside and where the activity of removal and rake was performed. An attack rate of 58% was estimated with a fatality rate of 14%.     Ecological specificity of polar lichen microbiomes

    Comunicación oral en este encuentro virtual que tuvo lugar del 1 al 6 de agosto de 2021Polar regions are likely the areas of the world where lichen communities show their highest degree of development leading to the formation of dense cryptogamic covers on soils. In these areas, lichens and bryophytes growing on the soil surface can be associated with different microorganisms, such as cyanobacteria and heterotrophic bacteria. Under that surface, edaphic prokaryotic (bacteria and archaea) and eukaryotic (algae and fungi) microorganisms coexist forming specific microbial communities. However, the diversity and roles of the microbiome in lichens from Polar Regions remains underexplored. Hence, we proposed to characterize the bacterial community structure of different lichens and mosses species colonizing glacier forefields from both Polar Regions, and the soils under them, in order to assess its specificity. To do so, we combined high-throughput amplicon sequencing with electron microscopy. Our results showed that bacterial communities differed less among microbiomes of cryptogamic covers dominated by different cryptogam species, than between these microbiomes and the bacterial communities from soils beneath. The edaphic bacterial community presented higher diversity than the cryptogamic cover microbiome, as shown by the higher abundance of specific edaphic OTUs. However, certain species specificity was also found in the microbiomes of different lichens and moss species. On the other hand, the microbial community structure of soils with cryptogamic cover was markedly different to that of the bare soils, suggesting a strong influence of the cryptogamic cover development on the edaphic communities. Finally, the electron microscopy revealed that bacteria appear mainly in non-healthy parts of the cryptogams, but also associated to the cell walls of some healthy cells and to soil particles integrated in the cover. All these results suggest that bacteria associated to cryptogamic covers might play an important role in their establishment and functioning, and consequently in the colonization of deglaciated areas.Funding: (MINECO, CTM2015-64728-C2-2-R)Peer reviewe
