    First detection of Gongylonema species in Geotrupes mutator in Europe

    The detection of three Gongylonema sp. infective larvae in two specimens of the dung beetle Geotrupes mutator (Marsham, 1802) from western Spain is reported here for the first time in Europe. Scanning electron microscopy confirmed that the analyzed specimens belong to the genus Gongylonema, but it was not possible to determine the species identity by the lack of morphological information in the literature and because many of the phenotypic characteristics had not yet fully developed at this juvenile stage. Nevertheless, a phylogenetic analysis using amplified cox1 nucleotide sequences has revealed that the studied larvae could be clearly discriminated ( 89% identity) from all the other Gongylonema cox1 sequences available in public genetic databases. While our results are limited by the scarcity of genetic information available for this genus, the possibility that the analyzed specimens might correspond to a new species should not be ruled out, and more studies are needed.     Interaction between charged lipid vesicles and point- or rod-like trivalent ions

    This work examines the influence of the charge distribution of trivalent cations on their interaction with soft anionic particles, using a combination of experimental measurements and theoretical modelling. In particular, we perform electrophoresis measurements to determine the zeta-potential of anionic liposomes in the presence of spermidine and lanthanum cations. We work in a range of electrolyte concentration where a reversal in the electrophoretic mobility of the liposomes is expected; however, unlike the case of lanthanum cations, spermidine does not induce mobility reversal of liposomes. As a result, the charge distribution within the counterion appears to be a key factor. This conclusion is supported by a theory that accounts for intra-ionic correlations, which has previously been successfully used to describe the colloidal electric double layer. It allows us to model spermidine as rod-like ions and lanthanum cations as point-like ions in order to test the importance of the ionic geometry in the interactions with soft particles such as lipid vesicles

    The Central role of KNG1 gene as a genetic determinant of coagulation pathway-related traits: Exploring metaphenotypes

    Traditional genetic studies of single traits may be unable to detect the pleiotropic effects involved in complex diseases. To detect the correlation that exists between several phenotypes involved in the same biological process, we introduce an original methodology to analyze sets of correlated phenotypes involved in the coagulation cascade in genome-wide association studies. The methodology consists of a two-stage process. First, we define new phenotypic meta-variables (linear combinations of the original phenotypes), named metaphenotypes, by applying Independent Component Analysis for the multivariate analysis of correlated phenotypes (i.e. the levels of coagulation pathway–related proteins). The resulting metaphenotypes integrate the information regarding the underlying biological process (i.e. thrombus/clot formation). Secondly, we take advantage of a family based Genome Wide Association Study to identify genetic elements influencing these metaphenotypes and consequently thrombosis risk. Our study utilized data from the GAIT Project (Genetic Analysis of Idiopathic Thrombophilia). We obtained 15 metaphenotypes, which showed significant heritabilities, ranging from 0.2 to 0.7. These results indicate the importance of genetic factors in the variability of these traits. We found 4 metaphenotypes that showed significant associations with SNPs. The most relevant were those mapped in a region near the HRG, FETUB and KNG1 genes. Our results are provocative since they show that the KNG1 locus plays a central role as a genetic determinant of the entire coagulation pathway and thrombus/clot formation. Integrating data from multiple correlated measurements through metaphenotypes is a promising approach to elucidate the hidden genetic mechanisms underlying complex diseases.Postprint (published version

    Extracellular Vesicles of Hypoxic Adipocytes and Obese Subjects Reduce Insulin-stimulated Glucose Uptake

    Scope We investigate the effects of extracellular vesicles (EVs) obtained from in vitro adipocyte cell models and from obese subjects on glucose transport and insulin responsiveness. Methods and results EVs are isolated from the culture supernatant of adipocytes cultured under normoxia, hypoxia (1% oxygen), or exposed to macrophage conditioned media (15% v/v). EVs are isolated from the plasma of lean individuals and subjects with obesity. Cultured adipocytes are incubated with EVs and activation of insulin signalling cascades and insulin‐stimulated glucose transport are measured. EVs released from hypoxic adipocytes impair insulin‐stimulated 2‐deoxyglucose uptake and reduce insulin mediated phosphorylation of AKT. Insulin‐mediated phosphorylation of extracellular regulated kinases (ERK1/2) is not affected. EVs from individuals with obesity decrease insulin stimulated 2‐deoxyglucose uptake in adipocytes (p = 0.0159). Conclusion EVs released by stressed adipocytes impair insulin action in neighboring adipocytes

    Effects of minor compounds from virgin olive oil on inflammatory response in human leukocytes

    Mediterranean diet protects against cardiovascular diseases such as atherosclerosis, now considered an inflammatory disorder. Virgin olive oil is one of the most important foods in this diet, leading to the suggestion that health benefits of Mediterranean diet are achieved at least partially due to the consumption of virgin olive oil. Extra virgin olive oil (EVOO) is the highest quality oil based on its chemical and sensorial properties. Our aim was to evaluate the anti-inflammatory effects of minor compounds found in the unsaponifibale fraction of EVOO on activated human monocytes and neutrophils. For this purpose, we isolated monocytes and neutrophils from blood samples of healthy volunteers. We also isolated the unsaponifiable fraction (UF) from EVOO. Cells were incubated with UF at different concentrations and the pro-inflammatory stimulus LPS. Afterwards, RNA was obtained from cells and then converted into cDNA. The relative gene expression of pro-inflammatory and anti-inflammatory markers was assessed by qRT-PCR. We found that UF from EVOO reduced relative gene expression of pro-inflammatory markers in a dose-dependent manner both in human monocytes and neutrophils. In contrast, UF from EVOO increased relative gene expression of anti-inflammatory markers in these cells. These findings unveil a role of UF from EVOO in the benefits from consumption of EVOO in disorders related to inflammation

    Simple and rapid one-step electrochemical synthesis of nanogranular Cu2O films

    In the present work, we report a simple experimental strategy for the one-step electrochemical synthesis of nanogranular Cu2O films by copper anodization in fluoride-containing ethylene glycol media. Microscopic exploration using field emission scanning electron microscopy (FESEM), atomic force microscopy (AFM) and transmission electron microscopy (TEM), shows the formation of spherical shape grains with sizes ranging from 20 to 40 nm. Raman and X-Ray Photoemission Spectroscopy (XPS) results indicate that only CuI oxide is obtained. A band gap energy Eg=2.01 eV is estimated from UV–vis reflectance spectroscopy indicating that an indirect transition mechanism between semiconductor bands takes place. These evidences indicate that the present synthesis of nanogranular Cu2O films is a promising method for obtaining improved properties of materials for the design of photoelectronic devices.Fil: OyarzĂșn Jerez, Diego Patricio.     Legume plants are able to fix nitrogen in symbiotic association with rhizobia and, like many crops, are sensitive to high salt conditions. However, very few molecular markers can be associated to stress tolerance in legume crops. A Kruppel-like transcription factor, Mtzpt2-1, required for the formation of the nitrogen-fixing region, confers salt tolerance to yeast cells. Here, legume responses to salt stresses were studied using alfalfa and its close relative Medicago truncatula, a model legume species. Salt stress induces the Mszpt2-1 gene both in roots and root harbouring nodules. In addition, Sinorhizobium meliloti strains tolerating up to 700 mM NaCl, were used in nodulation assays to assess salt tolerance of the symbiotic response of M. truncatula. Few nodules, mainly in the upper part of the root, could be detected in plants treated with 200 mM NaCl, suggesting that nodule initiation was particularly sensitive to salt stress. We have also defined for M. truncatula the threshold of NaCl tolerance after which recovery of stressed plants is irreversible under laboratory conditions. After analysing several times of salt treatment (150 mM NaCl), M. truncatula 108R plants stressed for 7 days could not recover (less than 5%), whereas a 4-day treatment allowed at least 75% recovery. Transgenic M. truncatula plants expressing Mtzpt2-1 in antisense configuration are more sensitive to `recover' from salt stress than the wild type. These results identify Mtzpt2-1 as a molecular marker potentially linked to stress tolerance in M. truncatula and suggest its participation in a transcriptional program induced in these plants to cope with salt stress.Facultad de Ciencias ExactasInstituto de BiotecnologĂ­a y BiologĂ­a Molecula
