Non-extractable proanthocyanidins from grapes are a source of bioavailable (epi)catechin and derived metabolites in rats

The non-extractable fraction of many fruit and vegetables contains putatively bioactive polyphenolic compounds that, in most cases, have not been well characterised structurally. Non-extractable proanthocyanidins (NEPA) of a polymeric nature are part of the dietary fibre fraction of food. Using liquid chromatography coupled to a mass spectrometer equipped with an electrospray ionisation chamber and a triple quadrupole mass analyser for tandem analysis (HPLC–ESI–QqQ–MS/MS) techniques, we examine the phenolic metabolites present in urine and faeces from rats 24 h after ingestion of an NEPA-rich fraction. We show that NEPA are partially depolymerised during their transit along the intestinal tract, as evidenced by the presence of (epi)catechin (EC) monomers and dimers in faeces and phase II conjugates of EC in urine. Moreover, NEPA are further metabolised by the intestinal microbiota into smaller metabolites including phenolic acids that are present in urine as both free phenolics and conjugates with glucuronate or sulphate moieties. For the first time, we report evidence that NEPA behave in vivo as a source of phenolics that are released progressively and deliver phenolic species that come into contact with the intestinal walls and are bioavailable for at least 24 h after ingestion. The present work was supported by the Spanish Ministry of Education and Science (AGL2009-12374-C03-03/ALI).Peer reviewe

Advanced Genetic Studies on Powdery Mildew Resistance in TGR-1551

[EN] Cucurbits powdery mildew (CPM) is one of the main limiting factors of melon cultivation worldwide. Resistance to races 1, 2, and 5 has been reported in the African accession TGR-1551, whose resistance is controlled by a dominant-recessive epistasis. The dominant and recessive quantitative trail loci (QTL) have previously been located in chromosomes 5 and 12, respectively. We used several densely genotyped BC3 families derived from the cross between TGR-1551 and the susceptible cultivar 'Bola de Oro' to finely map these resistance regions. The further phenotyping and genotyping of the selected BC5, BC5S1, BC5S2, BC4S1, BC(4)xPS, and (BC(4)xPS) S-1 offspring allowed for the narrowing of the candidate intervals to a 250 and 381 kb region in chromosomes 5 and 12, respectively. Moreover, the temperature effect over the resistance provided by the dominant gene has been confirmed. High resolution melting markers (HRM) were tightly linked to both resistance regions and will be useful in marker-assisted selection programs. Candidate R genes with variants between parents that caused a potential modifier impact on the protein function were identified within both intervals. These candidate genes provide targets for future functional analyses to better understand the resistance to powdery mildew in melons. This research was funded by the Spanish Ministerio de Ciencia e Innovacion (MCIN/AEI/10.13039/501100011033), grant number PID2020-116055RB (C21 and C22), and by the Conselleria d'Educacio, Investigacio, Cultura i Esports de la Generalitat Valenciana, grant number PROMETEO/2021/072 (to promote excellence groups, cofinanced with FEDER funds). The use of GIS to evaluate and map extreme maximum and minimum temperatures in Spain

    Get PDF
Spanish building legislation has recently changed and now requires an updated and restructured Technical Building Code which is in accordance with European directives. The norm contained in this Code is based on studies of extreme values for climatic elements such as temperature, precipitation and wind. Revised maps of extreme values for climatic elements with a 50-year recurrence interval are required. Here, extreme maximum and minimum temperature maps for Spain are evaluated and mapped by means of geographical information technology. The data are extracted from the historical database held by the Spanish Meteorological Institute. Daily extreme temperatures from 1,181 stations with records going back more than 30 years have been used. The maximum and minimum temperatures are determined as 50-year mean recurrence interval values. To obtain these values, a Gumbel distribution is fitted to the extreme annual values extracted from the database. Spatial interpolation in a regular 5 km×5 km grid of the annual maximum temperature is made by ordinary kriging. Meanwhile, for the annual minimum temperature a residual kriging has been applied due to its strong dependence on altitude

Preliminary evaluation of a short-range ensemble prediction system over western Mediterranean

Comunicación presentada en: 9th WRF Users’ Workshop celebrado del 23 al 27 de junio de 2008 en Colorado.A generation of a short-range ensemble prediction system, based on a set of mesoscale models with different subgrid-scale physic schemes and two different initial conditions, is developed, providing flow-dependent probabilistic forecasts by means of predictive probability distributions over the Western Mediterranean. A ten members short-range ensemble forecast system has been constructed over western Mediterranean area as a result of combining two different initial conditions from global models and five different physics configurations of the non-hydrostatic Mesoscale Model (MM5, version 3). The simulations obtained from this ensemble have been investigated during October 2006 period. An overview of the mean model performance and forecast variability, together with an evaluation of the ensemble accuracy, by means of comparison between the ensemble system and observations is provided. Calculations of the ensemble probability distribution functions for precipitation are displayed, providing explicit information on ensemble forecast uncertainty and constituting one of the major advantages of the ensemble methods over deterministic forecasting. The quality and value of precipitation forecasts have been evaluated against Spanish Climatic Network. The verification scores exhibit hopeful results encouraging the extension of this preliminary research to other verification periods and studying cases

Recent Approaches for the Manufacturing of Polymeric Cranial Prostheses by Incremental Sheet Forming

    Get PDF
This paper presents recent research experiences developed with the aim of manufacturing cranial prostheses in polymeric sheet using Incremental Sheet Forming (ISF) technologies. With this purpose, different approaches have been carried out in Single-Point Incremental Forming (SPIF) and Two-Point Incremental Forming (TPIF) in order to produce customized cranial implants using different polymeric materials. In this context, this research work provides a methodology to design and manufacture polymer customized cranial prostheses using the ISF technologies starting from a patient’s computerized tomography (CT). The results demonstrate the potential of manufacturing polymeric cranial prostheses by ISF in terms of the high formability achievable and show the appropriate geometrical accuracy at affordable manufacturing costs provided by these processes.Ministerio de Economía y Competitividad DPI2015-64047-
