Candidiases are the most recurrent fungal infections, especially among immunosuppressed patients. Although Candida albicans is still the most widespread isolated species, non-Candida albicans Candida species have been increasing. The goal of this work was to determine the susceptibility of C. glabrata biofilms to echinocandins and to evaluate their effect on the biofilm matrix composition, comparing the results with other Candida species. Drug susceptibilities were assessed through the determination of minimum inhibitory concentration (MIC), minimum fungicidal concentration (MFC) and minimum biofilm eradication concentration (MBEC) of caspofungin (Csf) and micafugin (Mcf). The -1,3 glucans content of the matrices was assessed after contact with the drugs. The data suggest that, generally, after contact with echinocandins, the concentration of -1,3 glucans increased. These adjustments in the matrix composition of C. glabrata biofilms and the chemical differences between Csf and Mcf, seem responsible and may determine the effectivity of the drug responses. This study was supported by the Portuguese Foundation for Science and Technology (FCT) under the scope of the strategic funding of UID/BIO/04469/2013 unit and COMPETE 2020 [POCI-01–0145-FEDER-006684] and BioTecNorte operation [NORTE-01–0145-FEDER-000004] funded by the European Regional Development Fund under the scope of Norte2020 - Programa Operacional Regional do Norte, Célia F. IntroductionLittle is known about Quality of Life within the first court of unvaccinated COVID-19 pregnant women exposed to the pandemic stressor. Primary aim of this study was to evaluate 1 year after hospital discharge HRQoL in a cohort of COVID-19 unvaccinated pregnant patients with COVID-19.Methodsin this prospective observational study, all COVID-19 positive pregnant women at any gestational age, admitted to the Obstetric Department at the University Hospital of Udine, Italy, from 1 March 2020 to 1 March 2021, requiring or not oxygen supplementation due to SARS-CoV2 pneumonia were evaluated. Patients with a history of neurological or psychiatric disease, those with a previous abortion, and those who refused to provide written informed consent were excluded from the study. We investigated pregnant positive COVID-19 women Health-related quality of life (HRQoL) with the Short-Form Health Survey-36 (SF-36) and Post-traumatic Stress-Disorder (PTSD) with the Impact of Event Scale-Revised (IES-R).Results62 pregnant women respected the inclusion criteria of the study, and data from 33 patients were analyzed. The mean age was 32 ± 6 years, with a median gestational age of 38 weeks [IQR 34–40]. 15.2% of patients required oxygen therapy through noninvasive respiratory support (with high flow nasal cannula) for a median of 9 days [IQR 6–12]. The median Physical Component Summary (PCS) and Mental Component Summary (MCS) scores were 50.2 [IQR 46.7–53.7] and 56.0 [IQR 46.8–60.6] respectively. Ten patients out of 33 (30%) tested positive for PTSD. Maternal age, gestational age, and history of cardiac-pulmonary-kidney disease significantly affected HRQoL at multivariable analysis.DiscussionIn COVID-19 pregnant unvaccinated women some physical impairments reducing HRQoL are still present 1 year after hospital discharge. Previous medical history such as history of cardiac-pulmonary-kidney disease significantly affected HRQoL. Long and repeated follow-up should be pursued in this category of patients.Clinical trial registration:ClinicalTrials.gov, Identifier NCT04860687

The base of the Priabonian Stage is one of two stage boundaries in the Paleogene that remains to be formalized. The Alano section (NE Italy) was elected by consensus as a suitable candidate for the base of the Priabonian during the Priabonian Working Group meeting held in Alano di Piave in June 2012. Further detailed research on the section is now followed by a formal proposal, which identifies the base of a prominent crystal tuff layer, the Tiziano bed, at meter 63.57 of the Alano section, as a suitable candidate for the Priabonian Stage. The choice of the Tiziano bed is appropriate from the historical point of view and several bio-magnetostratigraphic events are available to approximate this chronostratigraphic boundary and guarantee a high degree of correlatability over wide geographic areas. Events which approximate the base of the Priabonian Stage in the Alano section are the successive extinction of large acarininids and Morozovelloides (planktonic foraminifera), the Base of common and continuous Cribrocentrum erbae and the Top of Chiasmolithus grandis (nannofossils), as well as the Base of Subchron C17n.2n and the Base of Chron C17n (magnetostratigraphy). Cyclostratigraphic analysis of the Bartonian-Priabonian transition of the Alano section as well as radioisotopic data of the Tiziano tuff layer provide an absolute age (37.710 – 37.762 Ma, respectively) of this bed and, consequently, of the base of the Priabonian Stage

Haematological consequences of acute uncomplicated falciparum malaria: a WorldWide Antimalarial Resistance Network pooled analysis of individual patient data

Background: Plasmodium falciparum malaria is associated with anaemia-related morbidity, attributable to host, parasite and drug factors. We quantified the haematological response following treatment of uncomplicated P. falciparum malaria to identify the factors associated with malarial anaemia. Methods: Individual patient data from eligible antimalarial efficacy studies of uncomplicated P. falciparum malaria, available through the WorldWide Antimalarial Resistance Network data repository prior to August 2015, were pooled using standardised methodology. The haematological response over time was quantified using a multivariable linear mixed effects model with nonlinear terms for time, and the model was then used to estimate the mean haemoglobin at day of nadir and day 7. Multivariable logistic regression quantified risk factors for moderately severe anaemia (haemoglobin < 7 g/dL) at day 0, day 3 and day 7 as well as a fractional fall ≥ 25% at day 3 and day 7. Results: A total of 70,226 patients, recruited into 200 studies between 1991 and 2013, were included in the analysis: 50,859 (72.4%) enrolled in Africa, 18,451 (26.3%) in Asia and 916 (1.3%) in South America. The median haemoglobin concentration at presentation was 9.9 g/dL (range 5.0–19.7 g/dL) in Africa, 11.6 g/dL (range 5.0–20.0 g/dL) in Asia and 12.3 g/dL (range 6.9–17.9 g/dL) in South America. Moderately severe anaemia (Hb < 7g/dl) was present in 8.4% (4284/50,859) of patients from Africa, 3.3% (606/18,451) from Asia and 0.1% (1/916) from South America. The nadir haemoglobin occurred on day 2 post treatment with a mean fall from baseline of 0.57 g/dL in Africa and 1.13 g/dL in Asia. Independent risk factors for moderately severe anaemia on day 7, in both Africa and Asia, included moderately severe anaemia at baseline (adjusted odds ratio (AOR) = 16.10 and AOR = 23.00, respectively), young age (age < 1 compared to ≥ 12 years AOR = 12.81 and AOR = 6.79, respectively), high parasitaemia (AOR = 1.78 and AOR = 1.58, respectively) and delayed parasite clearance (AOR = 2.44 and AOR = 2.59, respectively). In Asia, patients treated with an artemisinin-based regimen were at significantly greater risk of moderately severe anaemia on day 7 compared to those treated with a non-artemisinin-based regimen (AOR = 2.06 [95%CI 1.39–3.05], p < 0.001). Conclusions: In patients with uncomplicated P. falciparum malaria, the nadir haemoglobin occurs 2 days after starting treatment. Although artemisinin-based treatments increase the rate of parasite clearance, in Asia they are associated with a greater risk of anaemia during recovery

Looking for Candida nivariensis and C. bracarensis among a large Italian collection of C. glabrata isolates: results of the FIMUA working group

    No full text
Two recently described pathogenic Candida species, C. nivariensis and C. bracarensis, share many phenotypic characteristics with C. glabrata and are easily misidentified as such. The aim of this study was to determine the occurrence of these cryptic species in Italy. One thousand yeast isolates collected in 14 Italian regions and identified as C. glabrata by phenotypic and biochemical methods were included in this study: 928 were screened on CHROMagar and 72 were analysed by a multiplex PCR. None of these cryptic species was identified despite the nationwide distribution and the variety of biological origin of the isolates

The Alano Section: The Candidate GSSP for the Priabonian Stage

    No full text
The deep-water hemipelagic Alano section is located in the Southern Alps of northern–eastern Italy close to the village of Alano di Piave in the type area of the Priabonian. Since 2005, the section has been presented as the potential Global Boundary Stratotype Section and Point (GSSP) for the late Eocene Priabonian Stage at several meetings of the International Subcommission on Palaeogene Stratigraphy (ISPS). Because of the favourable qualities of the section, we have been asked to produce the data necessary for the formal definition of the GSSP. In 2011, Agnini et al. published most of the documentation needed (lithostratigraphy, calcareous plankton biostratigraphy, magnetostratigraphy, and chemostratigraphy, including oxygen and carbon isotopes and carbonate content). The Agnini et al. (2011) paper also contained a motivated proposal for the definition of the Priabonian at the base of a prominent crystal tuff layer, the Tiziano bed, located at the 63.57 m level in the Alano section. Following the publication of this proposal, a unanimous consensus on the Alano section as the GSSP of the Priabonian (i.e., Gradstein et al. 2012; Wade et al. 2012) has been reached during an ad hoc workshop held in Alano in June 2012. However, not unexpectedly, alternative criteria for the definition of the GSSP have been subsequently advanced (Gradstein et al. 2012; Wade et al. 2012; Less and Özcan 2012). During 2012, we further refined the stratigraphy of the Alano section by obtaining cyclostratigraphic data. Moreover, U–Pb dating of zircons from seven levels has been undertaken. Here, we present the new cyclostratigraphic data and discuss various proposals for the definition of the Priabonian at Alano. We reiterate that our original proposal is the most balanced and in better keeping with Hedberg’s (1976) principles of chronostratigraphy

This study, which is guided by a social constructionist and qualitative approach, aimed to get to know the meanings Family Health Program (FHP) professionals attribute to family members living with mentally-ill patients. Data were collected by means of the focus group technique, involving members from two FHP teams in Ilhéus/Bahia, Brazil. As a result, we describe the family members and their conceptual registers, as well as the following meanings: caring, motivating and suffering prejudice; powerless and lacking resources; producing abuse and unbalanced. It was observed that some of these meanings reinforce the community's exclusion of mental patients and their family members, while others oppose this discourse and are thus able to favor a closer approximation between these actors and the professionals, in view of a new practice that improves their living conditions
