    Bone marrow chimeric mice reveal a dual role for CD36 in Plasmodium berghei ANKA infection

    BACKGROUND: Adhesion of Plasmodium-infected red blood cells (iRBC) to different host cells, ranging from endothelial to red blood cells, is associated to malaria pathology. In vitro studies have shown the relevance of CD36 for adhesion phenotypes of Plasmodium falciparum iRBC such as sequestration, platelet mediated clumping and non-opsonic uptake of iRBC. Different adhesion phenotypes involve different host cells and are associated with different pathological outcomes of disease. Studies with different human populations with CD36 polymorphisms failed to attribute a clear role to CD36 expression in human malaria. Up to the present, no in vivo model has been available to study the relevance of different CD36 adhesion phenotypes to the pathological course of Plasmodium infection. METHODS: Using CD36-deficient mice and their control littermates, CD36 bone marrow chimeric mice, expressing CD36 exclusively in haematopoietic cells or in non-haematopoietic cells, were generated. Irradiated CD36(-/- )and wild type mice were also reconstituted with syngeneic cells to control for the effects of irradiation. The reconstituted mice were infected with Plasmodium berghei ANKA and analysed for the development of blood parasitaemia and neurological symptoms. RESULTS: All mice reconstituted with syngeneic bone marrow cells as well as chimeric mice expressing CD36 exclusively in non-haematopoietic cells died from experimental cerebral malaria between day 6 and 12 after infection. A significant proportion of chimeric mice expressing CD36 only in haematopoietic cells did not die from cerebral malaria. CONCLUSION: The analysis of bone marrow chimeric mice reveals a dual role of CD36 in P. berghei ANKA infection. Expression of CD36 in haematopoietic cells, most likely macrophages and dendritic cells, has a beneficial effect that is masked in normal mice by adverse effects of CD36 expression in non-haematopoietic cells, most likely endothelial cells

    BACKGROUND: Scavenger receptors (SRs) recognize endogenous molecules modified by pathological processes as well as components of diverse microorganisms. Mice deficient for both SR-AI and II are more susceptible to infections by a variety of bacterial and viral pathogens. RESULTS: Here we show that SR-A deficient mice and wild type mice are equally susceptible to malaria infection both during liver and blood stages. Moreover, like wild type mice, SR-A deficient mice are able to mount a protective immune response against radiation attenuated sporozoites. CONCLUSION: Our results do not reveal a function of SR-A I and II receptors in the Plasmodium berghei ANKA infection, both in the development of CM and parasitemia control. Moreover, these receptors appear not to be required for the establishment of a protective immune response against the malaria liver stages

    Os indicadores da saúde oral permitem mensurar o estado de saúde e higiene da boca. O presente, estudo de natureza descritiva e cariz transversal, foi realizado numa amostra constituída por 189 adolescentes com idades compreendidas entre os 11 e 17 anos, com uma média de idades de 13 anos (Dp=1.17), do concelho de Sátão, distrito de Viseu. A pesquisa teve como finalidade avaliar o Índice de Dentes Cariados, Perdidos e Obturados (CPOD), o Índice de Placa Bacteriana Simplificado e caracterizar as práticas de higiene oral dos adolescentes. Para o efeito, a avaliação dos indicadores de saúde oral foi efetuada por observação clínica. Em face dos resultados inferiu-se que os adolescentes apresentam saúde oral precária, com 34,9% de dentes cariados, a maioria apresenta placa bacteriana reveladora de deficientes práticas de higiene oral.Os resultados reforçam que o planeamento das intervenções de educação para a saúde em adolescentes devem considerar os valores dos indicadores de saúde oral.The oral health indicators allow measuring health status and oral hygiene. This study is of descriptive and cross-sectional nature. It was conducted on a sample composed of 189 adolescents aged between 11 and 17 years old, with an average age of 13 years old (SD = 1.17), from Sátão, district of Viseu. The research aimed to evaluate the index decayed, missing and filled teeth (DMFT), the Plaque Index Simplified and characterize Index of plaque and the oral hygiene practices of adolescents. The assessment of oral health indicators was made by clinical observation. From the results it was concluded that adolescents have poor oral health, with 34.9% of decayed teeth and most of the adolescents present plaque, revealing deficient oral hygiene practices. The results emphasize that the planning of the interventions of health education in adolescents should consider the values of the indicators of oral health

    SeniorTec: senior tutorial program to teach technologies to students

    The SeniorTec program was carried out in the framework of the European project Cordon Gris, its objective was to promote intergenerational relationships. The activity was hosted by the old persons who participate in the European project Cordon Gris and was attended by university students from Higher School of Health of Alcoitão and ISCTE-University Institute of Lisbon. Learning sessions were delivered in this action: a Nutrition Workshop and a Financial Education Workshop. After this, an intergenerational contact session took place by using the Cordon Gris app. The SeniorTec program could promote a more positive image of aging and deconstruct negative stereotypes.     Tratamento pré-hospitalar da dor traumática aguda: um estudo observacional

    Objective: To describe and analyze the effectiveness of nurses’ interventions in pain reduction among patients with traumatic injury. Methods: Prospective cohort study conducted in the Immediate Life Support Ambulances in Portugal from March 1, 2019 to April 30, 2020. We have collected data on the kind of interventions implemented and the time elapsed during rescue procedures. To investigate the course of acute trauma pain, a 11-point Numeric Rating Scale was used. Changes in the level of pain registered throughout the three assessment moments were studied using linear mixed-effects models with random intercepts to account for the repeated measurements conducted on the same patient. These changes were assessed before and after the administration of the pain relief interventions. Results: 596 patients were included in this study. Most of them were male (65.9%) and had a mean age of 53.05±19.72 years. There was a reduction in the average pain intensity of 2.44 points (pinfo:eu-repo/semantics/publishedVersio

    The prognostic value of worsening renal function and its timing

All rights reserved.INTRODUCTION: Acute decompensated heart failure (ADHF) admissions are frequently complicated by different patterns of serum creatinine (SCr) elevation. We aimed to assess the prognostic impact of worsening renal function (WRF) based on the timing of its occurrence. METHODS: This was a retrospective cohort of patients admitted for ADHF. Standard WRF was defined as an increase in SCr of ≥0.3 mg/dl during hospitalization. WRF timing was classified as early (within 48 hours of admission) or late (>48 hours). Acute kidney injury (AKI) at admission was defined as a rise in SCr of ≥0.3 mg/dl from outpatient baseline measurement to first measurement at admission. The primary endpoint was a composite of all-cause mortality or hospitalization for cardiovascular events at one-year follow-up. RESULTS: Overall, 249 patients were included (mean age 77±11 years, 62% with preserved left ventricular ejection fraction). Early WRF occurred in 49 patients (19.7%) and was associated with a higher risk of the primary outcome (HR 2.49; 95% CI 1.66-3.73), whereas late WRF was not (p=0.411). After stratification for the presence of early WRF and/or AKI at admission, only patients with early WRF but no AKI at admission and patients with both AKI at admission and early WRF showed a higher risk of the primary outcome after multivariate Cox regression. CONCLUSION: Early WRF was associated with a higher risk of the primary outcome. The timing of WRF seems to be an important factor to take into account when considering the prognostic impact of creatinine variations during hospitalization for ADHF.proofepub_ahead_of_prin

    IL-10 Overexpression After BCG Vaccination Does Not Impair Control of Mycobacterium tuberculosis Infection

    Control of tuberculosis depends on the rapid expression of protective CD4+ T-cell responses in the Mycobacterium tuberculosis (Mtb)-infected lungs. We have recently shown that the immunomodulatory cytokine IL-10 acts intrinsically in CD4+ T cells and impairs their parenchymal migratory capacity, thereby preventing control of Mtb infection. Herein, we show that IL-10 overexpression does not impact the protection conferred by the established memory CD4+ T-cell response, as BCG-vaccinated mice overexpressing IL-10 only during Mtb infection display an accelerated, BCG-induced, Ag85b-specific CD4+ T-cell response and control Mtb infection. However, IL-10 inhibits the migration of recently activated ESAT-6-specific CD4+ T cells into the lung parenchyma and impairs the development of ectopic lymphoid structures associated with reduced expression of the chemokine receptors CXCR5 and CCR7. Together, our data support a role for BCG vaccination in preventing the immunosuppressive effects of IL-10 in the fast progression of Mtb infection and may provide valuable insights on the mechanisms contributing to the variable efficacy of BCG vaccination
