    OGC SWE-based Data Acquisition System Development for EGIM on EMSODEV EU Project

    The EMSODEV[1] (European Multidisciplinary Seafloor and water column Observatory DEVelopment) is an EU project whose general objective is to set up the full implementation and operation of the EMSO distributed Research Infrastructure (RI), through the development, testing and deployment of an EMSO Generic Instrument Module (EGIM). This research infrastructure will provide accurate records on marine environmental changes from distributed local nodes around Europe. These observations are critical to respond accurately to the social and scientific challenges such as climate change, changes in marine ecosystems, and marine hazards. In this paper we present the design and development of the EGIM data acquisition system. EGIM is able to operate on any EMSO node, mooring line, sea bed station, cabled or non-cabled and surface buoy. In fact a central function of EGIM within the EMSO infrastructure is to have a number of ocean locations where the same set of core variables are measured homogeneously: using the same hardware, same sensor references, same qualification methods, same calibration methods, same data format and access, and same maintenance procedures.Peer ReviewedPostprint (published version

    Species-Specific Discrimination of Insect Meals for Aquafeeds by Direct Comparison of Tandem Mass Spectra

    Insect protein has the potential to become a sustainable feed ingredient for the rapidly growing aquaculture industry. In the European Union, insect derived protein is placed under the same legislation as processed animal proteins (PAP). It is therefore of interest to develop methods for regulatory use, which unambiguously identify the species origin of insect-based ingredients. We performed (i) total protein quantification of insect samples using the traditional nitrogen-to-protein conversion factor of 6.25 and the sum of anhydrous amino acids, (ii) quantitative amino acid profiling and (iii) high-throughput tandem mass spectrometry to describe and differentiate 18 different commercial-grade insect meal samples derived from Hermetia illucens (8), Tenebrio molitor (5), Alphitobius diaperinus (3) and Acheta domesticus (2). In addition, we investigated and compared different protein extraction and digestion protocols for proteomic analysis. We found that irrespective of sample preparation, shotgun proteomics in combination with direct spectral comparison were able to differentiate insect meal according to their taxonomic classification. The insect specific spectral libraries created in the present work can in future be used to develop more sensitive targeted methods of insect PAP identification and quantification in commercial feed mixtures

    Future feed control – Tracing banned bovine material in insect meal

    In the present study, we assessed if different legacy and novel molecular analyses approaches can detect and trace prohibited bovine material in insects reared to produce processed animal protein (PAP). Newly hatched black soldier fly (BSF) larvae were fed one of the four diets for seven days; a control feeding medium (Ctl), control feed spiked with bovine hemoglobin powder (BvHb) at 1% (wet weight, w/w) (BvHb 1%, w/w), 5% (BvHb 5%, w/w) and 10% (BvHb 10%, w/w). Another dietary group of BSF larvae, namely *BvHb 10%, was first grown on BvHb 10% (w/w), and after seven days separated from the residual material and placed in another container with control diet for seven additional days. Presence of ruminant material in insect feed and in BSF larvae was assessed in five different laboratories using (i) real time-PCR analysis, (ii) multi-target ultra-high performance liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS/MS), (iii) protein-centric immunoaffinity-LC-MS/MS, (iv) peptide-centric immunoaffinity-LC-MS/MS, (v) tandem mass spectral library matching (SLM), and (vi) compound specific amino acid analysis (CSIA). All methods investigated detected ruminant DNA or BvHb in specific insect feed media and in BSF larvae, respectively. However, each method assessed, displayed distinct shortcomings, which precluded detection of prohibited material versus non-prohibited ruminant material in some instances. Taken together, these findings indicate that detection of prohibited material in the insect-PAP feed chain requires a tiered combined use of complementary molecular analysis approaches. We therefore advocate the use of a combined multi-tier molecular analysis suite for the detection, differentiation and tracing of prohibited material in insect-PAP based feed chains and endorse ongoing efforts to extend the currently available battery of PAP detection approaches with MS based techniques and possibly δ13CAA fingerprinting.publishedVersio

    The Efficacy of Short-Term Supplemental Nutrition Education on Nutrition Knowledge and Dietary Behaviors of Urban Middle School Children

    Background: Children and adolescents are not meeting the dietary guidelines and overweight and obesity related diseases are on the rise among youth in the United States. However, research is inconsistent with regard to the most effective method of educating urban youth. Purpose: To determine the effect of a supplemental nutrition education on the knowledge and behaviors of urban school students. Methods: Six nutrition lessons were provided to students attending the Boys and Girls Club of Metro Atlanta (BGCMA) in Atlanta, GA. A pre- and posttest was administered to assess differences in students’ nutrition knowledge and behaviors. Chi-square analysis compared the number of correct answers for the entire intervention population and after stratification by gender and number of classes missed. The Wilcoxon signed rank test compared the median number of correct answers in pre- vs. posttest responses for all participants and by the number of classes missed. Results: 15 students enrolled in the study. 2 students did not complete the study. A significant but negative change in knowledge was found in 1 of 15 nutrition questions. A significant negative change in the median number of total answers correct on the pre- vs. posttest was also observed (p=0.049). Approximately 60% of the participants reported that they were more likely to make positive, nutrition-related behavior changes after the intervention. Conclusion: We found a negative association between the nutrition education intervention and knowledge gain. This study suggests that nutrition education alone does not have a significant effect on nutrition knowledge of urban youth

    Etude Ecologique Et Entomologique Des Culicides Larvaires Des Gites De La Province De Kenitra (Maroc)

    Mosquitoes are considered the source of nuisances and used as vectors of major pathogens for humans and animals. Despite the existence of many ways of control, supervising their populations remains a challenge forced by a delicate knowledge of their dynamics in space and time. Hence the need to conduct an entomological and ecological study of breeding sites in the province of Kenitra in order to acheive an inventory of culicidiennes species collected at eight stations of surface water with the measurement of physical-chemical parameters of these waters during several month from April to June 2012. The results revealed the presence of two species of mosquitoes, the Anopheles labranchiae and the Culex pipiens. The first type has a very low density at three stations, while the second one has a very high density in five stations. Furthermore, concentrations and measured values of the physico-chemical parameters allowed to highlight a correlation between the quality of waterin the stations and dynamics of mosquitoes. Thus, the Anopheles attend frequently the well-oxygenated and unpolluted stations (O2 between 2,53 and 3,65 mg/l; BOD5 between 3 and 15 mg/l) against the Culex (O2 between 3,85 and 8,12mg/l, BOD5 between 10 and 100 mg/ l)

    In neutrality theory the recipients of vouchers, grants, and purchase-of-service contracts are eligible to participate as providers in government social service programs without regard to their religious character. Indeed, religious beliefs and practices are prohibited bases for screening out those who want to be welfare program providers. Notable examples of congressional social service legislation conforming to the rule of religious neutrality are the ‘charitable choice‘ feature imbedded in the Welfare Reform Act of 1996 and the Community Services Block Grant Act of 1998, as well as the provision allowing issuance of child care vouchers to indigent parents in the Child Care and Development Block Grant Program of 1990. Likewise, federal grants-in-aid programs, for example the Church Arson Prevention Act of 1996, the Telecommunication Act of 1996, and the President\u27s Values-Based Violence Prevention Initiative to reduce youth violence and gang activity, adhere to the principle of religious neutrality

    Obsea: a decadal balance for a cabled observatory deployment

    The study of the effects of climate change on the marine environment requires the existence of sufficiently long time series of key parameters. The study of these series allows both to characterize the range of variability in each particular region and to detect trends or changes that could be attributed to anthropogenic causes. For this reason, networks of permanent cabled observation systems are being deployed in the ocean. This paper presents a balance of a decade of activity at the OBSEA cabled observatory, as an example of ocean monitoring success and drawbacks. It is not the objective of this article to analyze the scientific and technical aspects already presented by the authors in different publications (Table 4). We will evaluate the overall experience by retracing the different steps of infrastructure deployment and maintenance, focusing on routines for in situ control, damages experienced, breakdowns and administrative constraints by local administrations. We will conclude by providing a set of guidelines to improve cabled observatories scientific outreach, societal projection, and economic efficiency.
