    Production and Evaluation of Activated Carbon from Sludge Generated in a Sewage Treatment Plant

    Indexación: ScieloEn este trabajo se evaluó el carbón activado producido a partir de lodo generado en una planta de tratamiento de aguas servidas. El lodo utilizado proviene de un lodo activado tipo aireación extendida y la transformación fue realizada mediante un proceso de activación química con cloruro de zinc y pirólisis. La pirólisis se realizó a diferentes temperaturas (550, 600, 650°C) y tiempos (30, 45 y 60 minutos). El rendimiento en carbón obtenido fluctuó entre el 42 y 66%, registrándose el mayor rendimiento a la temperatura de 550°C. La capacidad de adsorción, para el carbón activado obtenido, evaluada mediante la constante de adsorción lineal, para el colorante cristal violeta, fluctuó entre los 0,05 y 0,132 L/g. De acuerdo a esto, el mejor carbón se obtuvo cuando la temperatura de pirólisis fue de 650 °C. No resulta estadísticamente significativo el efecto del tiempo de pirólisis, en la constante de adsorción lineal. Abstract This study evaluated the activated carbon produced from the sludge generated in a sewage treatment plant. The sewage sludge used came from an extended aeration plant, and the transformation was carried out by means of a process of chemical activation with zinc chloride and pyrolysis. Pyrolysis was done at different temperatures (550, 600, 650°C) and times (30, 45 and 60 minutes). The yield of the carbon obtained fluctuated between 42% and 66%, with the highest yield recorded at 550°C. The adsorption capacity for the activated charcoal obtained, evaluated by means of the constant of linear adsorption for the dye crystal violet, fluctuated between 0,05 and 0,132 L/g. According to this, the best charcoal was obtained when pyrolysis temperature was 650°C. The effect of the pyrolysis time on the constant of linear adsorption was not statistically significant

    A systematic review of application of multi-criteria decision analysis for aging-dam management

    [EN] Decisions for aging-dam management requires a transparent process to prevent the dam failure, thus to avoid severe consequences in socio-economic and environmental terms. Multiple criteria analysis arose to model complex problems like this. This paper reviews specific problems, applications and Multi-Criteria Decision Making techniques for dam management. Multi-Attribute Decision Making techniques had a major presence under the single approach, specially the Analytic Hierarchy Process, and its combination with Technique for Order of Preference by Similarity to Ideal Solution was prominent under the hybrid approach; while a high variety of complementary techniques was identified. A growing hybridization and fuzzification are the two most relevant trends observed. The integration of stakeholders within the decision making process and the inclusion of trade-offs and interactions between components within the evaluation model must receive a deeper exploration. Despite the progressive consolidation of Multi-Criteria Decision Making in dam management, further research is required to differentiate between rational and intuitive decision processes.     Presencia de Fusarium graminearum en muestras de trigo destinado al consumo humano

    ResumenLa fusariosis es una de las enfermedades más importantes de los cereales, Fusarium graminearum es su principal agente etiológico. Este hongo posee la capacidad de producir distintos tipos y niveles de toxinas, en especial deoxinivalenol (DON). En la campaña 2012–2013 se dieron condiciones ambientales predisponentes para el desarrollo de esta enfermedad. El objetivo de este trabajo fue evaluar la presencia del hongo y el contenido de DON en 50 muestras de trigo. Los resultados demostraron la presencia de Fusarium graminearum en el 80 % de las muestras analizadas. El 24 % de las muestras presentó valores de DON ≥ 1μg/g, el 26 % varió entre 0,5 y 0,99μg/g, mientras que el 50 % restante mostró valores inferiores a 0,5μg/g. Se observó correlación entre la presencia de Fusarium graminearum y de DON. Es necesario establecer valores límites de DON en granos de trigo destinados al consumo humano.AbstractOne of the most important diseases in cereal crops is Fusarium head blight, being Fusarium graminearum the main etiological agent. This fungus has the ability to produce a wide spectrum and quantity of toxins, especially deoxynivalenol (DON). During the last crop season (2012–2013) the climatic conditions favored Fusarium colonization. The objective of this work was to determine the presence of this fungus as well as the DON content in 50 wheat grain samples. Our results showed that 80% of the samples were contaminated with Fusarium graminearum. Twenty four percent (24%) of the samples contained ≥ 1μg/g DON, 26% ranged from 0,5 and 0,99μg/g, and the remaining 50% had values lower than 0,5μg/g. Correlation was found between the presence of Fusarium graminearum and DON. It is necessary to establish DON limit values in wheat grains for human consumption

    Future journalists’ fight against disinformation: analysis of university training offers and challenges in the Spanish context

    Disinformation has become a global problem affecting mass media, governments and citizens globally. Besides the loss of trust in the media and its weakening influence, exposure to all manner of messages on social media in recent years has paved the way for disinformation, which has become a considerable challenge for journalism. According to the 2022 Edelman Trust Barometer, Spain is one of the countries most concerned about this phenomenon (Edelman, 2022). However, is this concern shared by Spanish journalist associations? What training initiatives are being carried out for future journalists to counter the spread of disinformation? How should fact-checking be taught in university? This study aims to answer these questions by using a dual methodology. First, a review and analysis were undertaken on the different training initiatives for bachelor’s and university-specific master’s degree students. Then, 15 in-depth interviews were conducted with experts, including fact-checkers, experienced journalists and representatives of sectoral associations, to ascertain their views on fact-checking and disinformation. The main results show that Spanish universities offer few training fact-checking-related initiatives, particularly at bachelor’s degree level, although more and more university-specific degrees and master’s degrees on this topic are becoming available. Furthermore, most interviewees view specialised training for the next generation of journalists as a key factor for fighting disinformation, and they provide guidelines to achieve this

    Growth hormone-releasing hormone receptor antagonists modify molecular machinery in the progression of prostate cancer

    Background: Therapeutic strategies should be designed to transform aggressive prostate cancer phenotypes to a chronic situation. To evaluate the effects of the new growth hormone-releasing hormone receptor (GHRH-R) antagonists: MIA-602, MIA-606, and MIA-690 on processes associated with cancer progression as cell proliferation, adhesion, migration, and angiogenesis. Methods: We used three human prostate cell lines (RWPE-1, LNCaP, and PC3). We analyzed several molecules such as E-cadherin, ?-catenin, Bcl2, Bax, p53, MMP2, MMP9, PCNA, and VEGF and signaling mechanisms that are involved on effects exerted by GHRH-R antagonists. Results: GHRH-R antagonists decreased cell viability and provoked a reduction in proliferation in LNCaP and PC3 cells. Moreover, GHRH-R antagonists caused a time-dependent increase of cell adhesion in all three cell lines and retarded the wound closure with the highest value with MIA-690 in PC3 cells. GHRH-R antagonists also provoked a large number of cells in SubG0 phase revealing an increase in apoptotic cells in PC3 cell line. Conclusions: Taken all together, GHRH-R antagonists of the MIAMI series appear to be inhibitors of tumor progression in prostate cancer and should be considered for use in future therapeutic strategies on this malignancy.Junta de Comunidades de Castilla-La ManchaUniversidad de Alcal

    Tumorigenic transformation of human prostatic epithelial cell line RWPE-1 by growth hormone-releasing hormone (GHRH)

    Background: Growth hormone-releasing hormone (GHRH) and its receptors have been implicated in the progression of various tumors. In this study, we analyzed the carcinogenetic potential of exposure to GHRH of a nontumor human prostate epithelial cell line (RWPE-1) as well as its transforming effect in a xenograft model. Methods: We performed cell viability, cell proliferation, adhesion and migration assays. In addition, metalloprotease (MMP)-2 activity by means gelatin zymography, GHRH-R subcellular location using confocal immunofluorescence microscopy and vascular endothelial growth factor (VEGF) levels by enzyme-linked immunoassay were assessed. Besides, we developed an in vivo model in order vivo model to determine the role of GHRH on tumorigenic transformation of RWPE-1 cells. Results: In cell cultures, we observed development of a migratory phenotype consistent with the gelatinolytic activity of MMP-2, expression of VEGF, as well as E-cadherin-mediated cell-cell adhesion and increased cell motility. Treatment with 0.1 µM GHRH for 24 h significantly increased cell viability and cell proliferation. Similar effects of GHRH were seen in RWPE-1 tumors developed by subcutaneous injection of GHRH-treated cells in athymic nude mice, 49 days after inoculation. Conclusions: Thus, GHRH appears to act as a cytokine in the transformation of RWPE-1 cells by mechanisms that likely involve epithelial-mesenchymal transition, thus reinforcing the role of GHRH in tumorigenesis of prostate.Universidad de Alcal

    Effects of irrigation and shoot thinning on the size and phenolics content of developing grape berries (Vitis vinifera L. cv. Tempranillo)

     Aim of study: The concentration of phenolics in the grape berries can be influenced by cultural practices such as irrigation or thinning. The main objective of the present study was to evaluate the effect of combinations of these practices on grape size and phenolics content. Area of study: The trial was carried out in an experimental cv. Tempranillo vineyard located in Extremadura, Spain. Material and methods: Two irrigation regimes were considered: rainfed vines (non-irrigated, NIr), and 100% ETc irrigated vines (Ir). For each irrigation treatment, two cropping levels were studied: low shoot-thinning (LT) vs high shoot-thinning (HT) implemented in winter and spring, respectively. Berry weight, and total phenolics, proanthocyanidin, and anthocyanin concentrations were determined at eight stages of berry development in three consecutive years (2014, 2015, and 2016). Main results: Specific weather conditions of each year affected phenolics accumulation differently. In 2014, where maximum temperatures were low and an important rainfall occurred at Stage II, both the NIr-LT and NIr-HT treatments led to the greatest concentrations of total phenolics, proanthocyanidins, and anthocyanins. In 2015, where a little rainfall was registered at Stage II, the berries from the NIr-HT and Ir-HT treatments accumulated the greatest total phenolics and proanthocyanidin contents, but the NIr-LT and NIr-HT treatments led to the greatest accumulation of anthocyanins. Finally, in 2016, where high maximum temperatures and scarce rainfall were registered, the Ir-LT and Ir-HT treatments presented the greatest concentrations of total phenolics, proanthocyanidins, and anthocyanins. Research highlights: A significant effect of irrigation and thinning was observed on berry size and phenolic content, as well as year × thinning interaction

    Pseudopithomyces chartarum associated with wheat seeds in Argentina, pathogenicity and evaluation of toxigenic ability

    Argentina is one of the top 10 world producers and exporters of wheat. In routine surveys of wheat (Triticum aestivum L.) in Buenos Aires Province, Argentina, a new disease was observed in 2012 on seeds of wheat cv. Buck Meteoro. Symptomatic grains (black points) and leaves (chlorosis and spots) were collected during the spring of that year. The objectives of the present study were to identify the causal agent, to investigate its pathogenicity in relation to nine wheat cultivars and to identify the secondary metabolites produced by fungus. Symptomatic grains were plated on potato dextrose agar (PDA). Morphological characterization of colonies and sequencing of the ITS region after DNA extraction identified it as Pseudopithomyces chartarum. For pathogenicity tests, two different isolates, P221 and P224, were inoculated on seedlings of nine wheat cultivars, which showed different disease symptoms, % of grain germination (GG), % of grain discoloration (GD) and % of weak seedlings (WS), suggesting different levels of response against Pseudopithomyces chartarum. Particularly B. Meteoro, Buck Guapo and Klein Proteo cultivars demonstrated major infection tolerance for GD and WS. In contrast, Sy 100 and Klein Pantera were most affected showing weakness, chlorosis, or reduced length of coleoptile, and 50% of necrotic symptoms. Pseudopithomyces chartarum isolates produced secondary metabolites including alternariol, alternariol mono-methyl ether, altertoxin I and altertoxine II. The fungus is a new pathogen of wheat in Argentina that can cause diseases on different cultivars as well as produce mycotoxins.Facultad de Ciencias Agrarias y Forestale

    Analysis of circulating microRNAs and their post-transcriptional modifications in cancer serum by on-line solid-phase extraction-capillary electrophoresis-mass spectrometry

    In this paper, an on-line solid-phase extraction capillary electrophoresis-mass spectrometry (SPE-CE-MS) method is described for the purification, preconcentration, separation, and characterization of endogenous microRNA (miRNA) and their post-transcriptional modifications in serum. First, analysis by CE-MS was optimized using a standard mixture of hsa-miR-21-5p (miR-21-5p) and hsa-let-7g-5p (let-7g-5p). For SPE-CE-MS, a commercial silicon carbide (SiC) resin was used to prepare the microcartridges. Under the optimized conditions with standards, the microcartridge lifetime (>25 analyses) and repeatability (2.8% RSD for the migration times; 4.4 and 6.4% RSD for the miR-21-5p and let-7g-5p peak areas, respectively) were good, the method was linear between 25 and 100 nmol·L-1, and the limit of detection (LOD) was around 10 nmol·L-1 (50 times lower than by CE-MS). In order to analyze human serum samples, an off-line sample pretreatment based on phenol/chloroform/isoamyl alcohol (PCA) extraction was necessary prior to SPE-CE-MS. The potential of the SPE-CE-MS method to screen for B-cell chronic lymphocytic leukemia (CLL) was demonstrated by an analysis of serum samples from healthy controls and patients. MicroRNAs, specifically miR-21-5p and a 23 nucleotide long 5'-phosphorylated miRNA with 3'-uridylation (iso-miR-16-5p), were only detected in the CLL patients
