    Na+-dependent NO3- uptake in leaf cells of the seagrass Posidonia oceanica (L.) Delile

    Posidonia oceanica (L.) Delile is an endemic Mediterranean seagrass of recognized ecological significance and, as other seagrasses, this species has secondarily adapted to live in the marine environment. In this alkaline medium with a high Na+ concentration (0.5 M), the high inwardly directed electrochemical potential gradient for sodium is used in the seagrass Zostera marina to energize the uptake of nitrate1 and phosphate that usually occur at concentration below 10 ”M. Here we summarize several evidences for the operation of a sodium-dependent high-affinity nitrate transport system at the plasma membrane of the mesophyll leaf cells of P. oceanica. Leaf cells of P. oceanica possess a H+-ATPase as a primary pump, exhibit a plasma membrane potential (Em) of -174 ± 10 mV and show reduced Na+ permeability. The addition of micromolar nitrate concentrations induces membrane depolarizations that show saturation kinetics. Curve fitting of the values renders a semisaturation constant (Km) of 21.3 ± 6.6 ÎŒM and a maximum depolarization (Dmax) of 7 ± 1 mV. In dark conditions, Dmax decreases by fifty percent but no significant effect is observed on the Km value. On the other hand, nitrate induced depolarizations show sodium dependence. The depolarizations induced by 100 ”M NO3- in media containing increasing Na+ concentrations (from 0 to 250 mM) show saturation kinetics, rendering a Km value of 16 ± 5 mM Na+. Moreover, the depolarization induced by 100 ”M NO3- is accompanied by a simultaneous increase of cytosolic sodium, measured by Na+-sensitive microelectrodes, of 0.4 ± 0.2 mM above the resting cytosolic sodium concentration (17 ± 2 mM). Finally, nitrate uptake rates, measured in depletion experiments, decreases by 50% and 80% in dark conditions and in the absence of Na+, respectively, compared with control conditions (0.5 M Na+ and light). All together, these results strongly suggest that NO3- uptake in P. oceanica leaf cells is mediated by a high-affinity nitrate carrier that uses Na+ as the driving ion. 1 Rubio et al. (2005). J. Exp.     A Semiparametric Estimation of Liquidity Effects on Option Pricing

    This paper proposes a semiparametric option pricing model with liquidity, as proxied by the relative bid-ask spread. The nonparametric volatility function with liquidity as an explanatory variable is estimated using the Symmetrized Nearest Neighbors (SNN) estimator rather than the traditional kernel estimator. Moreover, special care is taken in obtaining the smoothing parameter. The in-sample performance of the model turns out to be statistically favorable relative to a competing model without liquidity.     This paper shows the extraordinary capacity of yield spreads to anticipate consumption growth as proxy by the Economic Sentiment Indicator elaborated by the European Commission in order to predict turning points in business cycles. This new evidence complements the well known results regarding the usefulness of the slope of the term structure of interest rates to predict real economic conditions and, in particular, recessions by using a direct measure of expectations. A linear combination of European yield spreads explains a surprising 93.7% of the variability of the Economic Sentiment Indicator. Bajo esta perspectiva se aborda este estudio desde un mĂ©todo mixto - cuantitativo (seguimiento al contenido y actuaciones en redes sociales) y cualitativo (reflexiones y anĂĄlisis de instrumentos con los que se llega a la audiencia por otras vĂ­as)-. Lo anterior permite identificar el diseño de estrategias para captar la atenciĂłn del pĂșblico desde mĂșltiples alternativas de consumo, al tiempo que se incorpora la radio al ecosistema digital, modificando sus rutinas productivas.The Colombian radio industry is living a transition and contents re-thinking process to accommodate to the new mediatic landscape where digital devices and communication platforms compete.     Histone Mutants Separate R Loop Formation from Genome Instability Induction

    R loops have positive physiological roles, but they can also be deleterious by causing genome instability, and the mechanisms for this are unknown. Here we identified yeast histone H3 and H4 mutations that facilitate R loops but do not cause instability. R loops containing single-stranded DNA (ssDNA), versus RNA-DNA hybrids alone, were demonstrated using ssDNA-specific human AID and bisulfite. Notably, they are similar size regardless of whether or not they induce genome instability. Contrary to mutants causing R loop-mediated instability, these histone mutants do not accumulate H3 serine-10 phosphate (H3S10-P). We propose a two-step mechanism in which, first, an altered chromatin facilitates R loops, and second, chromatin is modified, including H3S10-P, as a requisite for compromising genome integrity. Consistently, these histone mutations suppress the high H3S10 phosphorylation and genomic instability of hpr1 and sen1 mutants.     Small and large intestine express a truncated Dab1 isoform that assembles in cell-cell junctions and co-localizes with proteins involved in endocytosis

    Disabled-1 (Dab1) is an essential intracellular adaptor protein in the reelin pathway. Our previous studies in mice intestine showed that Dab1 transmits the reelin signal to cytosolic signalling pathways. Here, we determine the Dab1 isoform expressed in rodent small and large intestine, its subcellular location and co-localization with clathrin, caveolin-1 and N-Wasp. PCR and sequencing analysis reveal that rodent small and large intestine express a Dab1 isoform that misses three (Y198, Y200 and Y220) of the five tyrosine phosphorylation sites present in brain Dab1 isoform (canonical) and contains nuclear localization and export signals. Western blot assays show that both, crypts, which shelter progenitor cells, and enterocytes express the same Dab1 isoform, suggesting that epithelial cell differentiation does not regulate intestinal generation of alternatively spliced Dab1 variants. They also reveal that the canonical and the intestinal Dab1 isoforms differ in their total degree of phosphorylation. Immunostaining assays show that in enterocytes Dab1 localizes at the apical and lateral membranes, apical vesicles, close to adherens junctions and desmosomes, as well as in the nucleus; co-localizes with clathrin and with N-Wasp but not with caveolin-1, and in Caco-2 cells Dab1 localizes at cell-to-cell junctions by a Ca2+-dependent process. In conclusion, the results indicate that in rodent intestine a truncated Dab1 variant transmits the reelin signal and may play a role in clathrin-mediated apical endocytosis and in the control of cell-to-cell junction assembly.     Topological constraints impair RNA polymerase II transcription and causes instability of plasmid-borne convergent genes

    Despite the theoretical bases for the association of topoisomerases and supercoiling changes with transcription and replication, our knowledge of the impact of topological constraints on transcription and replication is incomplete. Although mutation of topoisomerases affects expression and stability of the rDNA region it is not clear whether the same is the case for RNAPII transcription and genome integrity in other regions. We developed new assays in which two convergent RNAPII-driven genes are transcribed simultaneously. Plasmid-based systems were constructed with and without a transcription terminator between the two convergent transcription units, so that the impact of transcription interference could also be evaluated. Using these assays we show that Topos I and II play roles in RNAPII transcription in vivo and reduce the stability of RNAPII-transcribed genes in Saccharomyces cerevisiae. Supercoiling accumulation in convergent transcription units impairs RNAPII transcription in top1Δ strains, but Topo II is also required for efficient transcription independent of Topo I and of detectable supercoiling accumulation. Our work shows that topological constraints negatively affect RNAPII transcription and genetic integrity, and provides an assay to study gene regulation by transcription interference
