The genes encoding Arabidopsis ORC subunits are E2F targets and the two ORC1 genes are differently expressed in proliferating and endoreplicating cells

Initiation of eukaryotic DNA replication depends on the function of pre-replication complexes (pre-RC), one of its key component being the six subunits origin recognition complex (ORC). In spite of a significant degree of conservation among ORC proteins from different eukaryotic sources, the regulation of their availability varies considerably in different model systems and cell types. Here, we show that the six ORC genes of Arabidopsis thaliana are regulated at the transcriptional level during cell cycle and development. We found that Arabidopsis ORC genes, except AtORC5, contain binding sites for the E2F family of transcription factors. Expression of AtORC genes containing E2F binding sites peaks at the G1/S-phase. Analysis of AtORC gene expression in plants with reduced E2F activity, obtained by expressing a dominant negative version of DP, the E2F heterodimerization partner, and with increased E2F activity, obtained by inactivation of the retinoblastoma protein, led us to conclude that all AtORC genes, except AtORC5 are E2F targets. Interestingly, Arabidopsis contains two AtORC1 (a and b) genes, highly conserved at the amino acid level but with unrelated promoter sequences. AtORC1b expression is restricted to proliferating cells. However, AtORC1a is preferentially expressed in endoreplicating cells based on our analysis in endoreplicating tissues and in a mutant with altered endocycle pattern. This suggests a differential expression of the two ORC1 genes in Arabidopsis

A web-based Learning Didactical Design for Training Teachers in the Incorporation of Technology to the English Classroom

    Get PDF
The remote teaching and learning for emergency lived during Pandemic Times has demonstrated the urgent need to train teachers in the incorporation of technology into their classrooms. In English Language Teaching (ELT), this need becomes a must since the use of digital tools enables the development of Cultural and Intercultural Competence by approaching authentic language registers in native speakers of English, and variations of the language in speakers of English from other languages.  A conducted literature review shows that models like Mishra and Koehler’s T-Pack, Puentedura’s SAMR, and Salem’s SOSE shed light on how to lead this process of ICT integration to education in general; but there is not a didactical design that describes concrete procedures on how to train English Teachers in this issue. The current article aims at describing and supporting theoretically a conceived Web-based Didactical Design for training English teachers in the use of ICT for ELT, based on the analysis of existing models. The Didactical Design is part of the planned intervention of a study that intends to find out its effects in Teachers’ Digital Competence using the DigCompEdu framework, in three groups of basic, middle, high school, and Tertiary Education English teachers from Colombia

Liver biopsy represents the gold standard for damage evaluation, but noninvasive serum markers that mirror liver fibrosis progression are actual goals both in adults and especially in children. The aim was to determine specific serum markers that correlate with liver fibrosis progression during chronic HCV infection.Liver biopsies and concomitant serum samples from 22 pediatric and 22 adult HCV patients were analyzed. Histological parameters were evaluated. On serum TGF-ß1, tissue inhibitor of matrix metalloprotein inhibitor-1 (TIMP-1), hyaluronic acid (HA) and aminoterminal peptide of procollagen type III (PIIINP) were tested. = 0.022) in adults. The AUROC of TIMP-1 in children to discriminate advanced fibrosis was 0.800 (95%IC 0.598–0.932). In adults, the best AUROCs were that of HA, PIIINP and TGF-ß1 [0.929 (IC95% 0.736–0.994), 0.894 (IC95% 0.689–0.984) and 0.835 (IC95% 0.617–0.957)], respectively. In children, according to the cut off (165.7 ng/mL) value for TIMP-1, biopsies could have been avoided in 72% (18/25). Considering the cut off for HA (109.7 ng/mL), PIIINP (9.1 µg/L), and TGF-ß1 (10,848.3 pg/mL), biopsies could have been avoided in 87% (19/22) of adult patients by using HA and 73% (16/22) using PIIINP or TGF-ß1.In adults given the diagnostic accuracy of HA, PIIINP, TGF-ß1, their combination may provide a potential useful tool to assess liver fibrosis. This first pediatric study suggests that TIMP-1 is clinically useful for predicting liver fibrosis in HCV patients

The anthropometric pattern, demographics data, lifestyle factors and several aspects of morbidity were analyzed for 400 Cuban women ages 40–60 years. More than 85% of women were obese, being these in direct association with morphology typically androgens, more evident in the postmenopausal women. In these women observed less body mass index and overweight, in an equivalent way between pre and postmenopausal women. A moderate consumption of rice, was evidenced, pastas and vegetables, few candies and fatty, what points to or quite acceptable knowledge of the harmful effect of some foods, to weigh that the shadowy use of saturated fats and not saturated it was the most frequent thing. By multifactor approach it is possible to make relation between nutritional patterns, morphology and climacteric symptoms; Cuban women present a different frequencies from an other literature showing the risk of generalist this studies and health’s actions

Uraemic toxins impair skeletal muscle regeneration by inhibiting myoblast proliferation, reducing myogenic differentiation, and promoting muscular fibrosis

    Get PDF
17 p.Uraemic toxins increase in serum parallel to a decline in the glomerular filtration rate and the development of sarcopenia in patients with chronic kidney disease (CKD). This study analyses the role of uraemic toxins in sarcopenia at different stages of CKD, evaluating changes in the muscular regeneration process. Cultured C2C12 cells were incubated with a combination of indoxyl sulphate and p-cresol at high doses (100 mu g/mL) or low doses (25 mu g/mL and 10 mu g/mL) resembling late or early CKD stages, respectively. Cell proliferation (analysed by scratch assays and flow cytometry) was inhibited only by high doses of uraemic toxins, which inactivated the cdc2-cyclin B complex, inhibiting mitosis and inducing apoptosis (analysed by annexin V staining). By contrast, low doses of uraemic toxins did not affect proliferation, but reduced myogenic differentiation, primed with 2% horse serum, by inhibiting myogenin expression and promoting fibro-adipogenic differentiation. Finally, to assess the in vivo relevance of these results, studies were performed in gastrocnemii from uraemic rats, which showed higher collagen expression and lower myosin heavy chain expression than those from healthy rats. In conclusion, uraemic toxins impair the skeletal muscular regeneration process, even at low concentrations, suggesting that sarcopenia can progress from the early stages of CKD.Instituto de Salud Carlos II

Gαq activation modulates autophagy by promoting mTORC1 signaling.

    Get PDF
The mTORC1 node plays a major role in autophagy modulation. We report a role of the ubiquitous Gαq subunit, a known transducer of plasma membrane G protein-coupled receptors signaling, as a core modulator of mTORC1 and autophagy. Cells lacking Gαq/11 display higher basal autophagy, enhanced autophagy induction upon different types of nutrient stress along with a decreased mTORC1 activation status. They are also unable to reactivate mTORC1 and thus inactivate ongoing autophagy upon nutrient recovery. Conversely, stimulation of Gαq/11 promotes sustained mTORC1 pathway activation and reversion of autophagy promoted by serum or amino acids removal. Gαq is present in autophagic compartments and lysosomes and is part of the mTORC1 multi-molecular complex, contributing to its assembly and activation via its nutrient status-sensitive interaction with p62, which displays features of a Gαq effector. Gαq emerges as a central regulator of the autophagy machinery required to maintain cellular homeostasis upon nutrient fluctuations. We thank Paula Ramos, Susana Rojo-Berciano, and Laura López for helpful technicalassistance. Strategies to improve swine manure hydrochar: HCl-assisted hydrothermal carbonization versus hydrochar washing

The work focuses on the study of hydrochar upgrading from hydrothermal carbonization (HTC) of swine manure by HClassisted HTC or washing with HCl or acetone, as a post-treatment to conventional HTC. Conventional HTC of swine manure yields a low-quality hydrochar (C content~ 38 wt.%, higher heating value (HHV)~ 15 MJ kg−1, and ash content up to 32 wt.%). HCl-assisted HTC (0.5 M HCl at 230 ℃) substantially reduced the ash content up to ~10 wt.% in the hydrochar and increased the C content to 58 wt.%, reaching a HHV of 23 MJ kg−1. However, the N and S contents remained at values similar to those of the swine manure. Washing post-treatment of conventional hydrochars with HCl or acetone signifcantly improved the C content and the HHV in the range 47–58 wt.% and 19–25 MJ kg−1, respectively, as well as the ash removal with values 7-11 wt.%. Washing the hydrochar with acetone signifcantly reduced the N and S contents, obtaining a carbonaceous material with properties suitable for solid biofuel according to ISO/TS 17225–8, (N17 MJ kg−1; and ash<10 wt.%). Hydrochars obtained by HCl-assisted HTC and HCl/acetone washing post-treatment yielded higher thermal stability, as well as better reactivity and low ash agglomeration indexes than compared to conventional hydrochars. Washing post-treatment with acetone proved to be the best strategy to obtain improved hydrochars from swine manure for industrial use as a solid biofuelAuthors greatly appreciate funding from Spain’s MICINN (PID2019-108445RB-I00), MINECO (PDC2021-120755-I00 and TED2021-130287B-I00), Madrid Regional Government (Project S2018/EMT-4344), and Grupo Kerbest Company.
