Mechanisms of dexamethasone-mediated chemokine down-regulation in mild and severe acute pancreatitis

AbstractThis study aimed to investigate the role of therapeutic dexamethasone (Dex) treatment on the mechanisms underlying chemokine expression during mild and severe acute pancreatitis (AP) experimentally induced in rats. Regardless of the AP severity, Dex (1 mg/kg), administered 1 h after AP, reduced the acinar cell activation of extracellular signal-regulated kinase (ERK) and c-Jun-NH2-terminal kinase (JNK) but failed to reduce p38-mitogen-activated protein kinase (MAPK) in severe AP. In both AP models, Dex inhibited the activation of nuclear factor-kappaB (NF-κB) and signal transducers and activators of transcription (STAT) factors. All of this resulted in pancreatic down-regulation of the chemokines monocyte chemoattractant protein-1 (MCP-1) and cytokine-induced neutrophil chemoattractant (CINC). Lower plasma chemokine levels as well as decreased amylasemia, hematocrit and plasma interleukin-1β (Il-1β) levels were found either in mild or severe AP treated with Dex. Pancreatic neutrophil infiltration was attenuated by Dex in mild but not in severe AP. In conclusion, by targeting MAPKs, NF-κB and STAT3 pathways, Dex treatment down-regulated the chemokine expression in different cell sources during mild and severe AP, resulting in decreased severity of the disease

An Accurate and Fully-Automated Ensemble Model for Weekly Time Series Forecasting

Many businesses and industries require accurate forecasts for weekly time series nowadays. However, the forecasting literature does not currently provide easy-to-use, automatic, reproducible and accurate approaches dedicated to this task. We propose a forecasting method in this domain to fill this gap, leveraging state-of-the-art forecasting techniques, such as forecast combination, meta-learning, and global modelling. We consider different meta-learning architectures, algorithms, and base model pools. Based on all considered model variants, we propose to use a stacking approach with lasso regression which optimally combines the forecasts of four base models: a global Recurrent Neural Network model (RNN), Theta, Trigonometric Box-Cox ARMA Trend Seasonal (TBATS) and Dynamic Harmonic Regression ARIMA (DHR-ARIMA), as it shows the overall best performance across seven experimental weekly datasets on four evaluation metrics. Our proposed method also consistently outperforms a set of benchmarks and state-of-the-art weekly forecasting models by a considerable margin with statistical significance. Our method can produce the most accurate forecasts, in terms of mean sMAPE, for the M4 weekly dataset among all benchmarks and all original competition participants.Comment: 1 figure, 9 table

Influence of Recycled Precast Concrete Aggregate on Durability of Concrete's Physical Processes

ABSTRACT: The research presented in this article analysed the influence of incorporating precast concrete waste as an alternative to coarse aggregate in self-compacting concrete to generate new precast elements. The experimental study involved the characterization of recycled aggregate and the design of the mix of the new self-compacting concrete (SCC). The experimental study evaluates the physical processes that affect the durability of concrete with percentages of incorporation such as 20%, 50% and 100% of recycled aggregate. Two types of SCC were manufactured with minimum compressive strength of 30 MPa and 45 MPa. The properties analysed were density of hardened SCC, shrinkage cracking, freeze-thaw resistance, resistance to ageing by thermal shock and abrasion resistance. The results obtained were compared with those of the control concrete, observing great capacity of the SCC under physical aggressions that affect durability. The results of this research show that it is possible to use the recycled aggregate coming from precast pieces in order to the manufacture of self-compacting recycled concrete in the same precast industry. However, high loss of proprieties occurs for a 100% substitution, while for 20% and 50%, the variations with respect to control concrete are smaller. In addition, taking advantage of this waste to incorporate it back into the production chain contributes to more sustainable construction. This research was funded by Junta de Castilla y León (Regional Government) for funding UIC-231 through project BU119P17; MINECO for funding through project BIA2014-55576-C2-1-R; FEDER (European Regional Development Funds) and LADICI

Sustainable bioethanol production using agro-industrial by-products

    Get PDF
This work aimed to evaluate a sustainable bioethanol production by a laboratorial isolate strain of Saccharomyces cerevisiae, along with the use of agro-industrial by-products as carbon source. The effect of several carbon sources and their concentrations was studied using carob pod extract (CPE) and beet molasses (BM) and compared with glucose and sucrose as conventional carbohydrates at different concentrations, 15, 20 and 30 g/l.No significant difference was found between maximum ethanol production obtained with CPE, BM, glucose and sucrose fermentations profiles. It was obtained values of 10.65 g/l and 10.5 g/l ethanol, respectively for sucrose and CPE at 30g/l, which can be improved using higher substrate concentration
