    Os Estabelecimentos Assistenciais de Saúde (EAS) são edifícios que passam por constantes revisões em seus projetos arquitetônicos com o objetivo de melhorar os aspectos de qualidade e segurança para os usuários finais destes ambientes. Uma forma de se estabelecer as mudanças estruturais necessárias nos ambientes de saúde é o entendimento das relações entre os usuários e o ambiente construído, com base na atuação de suas funções e necessidades ambientais, uma vez que a participação dos usuários no processo de projeto pode trazer benefícios tanto para a satisfação do usuário como para o próprio desempenho dos edifícios. O uso de mockups de simulações virtuais tem apresentado grandes contribuições na análise e avaliação de projetos de ambientes de saúde por possibilitar a imersão do usuário no processo de concepção do projeto. Dessa forma, este artigo tem como objetivo verificar as potencialidades e limitações da utilização de uma ferramenta físico-digital para auxiliar na representação e comunicação no Co-Design, além de identificar as inconsistências da programação do software e do comportamento dos objetos virtuais. Foi realizado um teste de verificação por meio de uma simulação exploratória de viabilidade e usabilidade com a utilização de mockups virtuais, onde o foco estava nas análises de disposição de equipamentos e mobiliário de um ambiente de saúde. A ferramenta físico-digital demonstrada neste artigo ainda se encontra em fase de desenvolvimento e necessita de ajustes para a sua aplicação final junto aos usuários dos ambientes de saúde.Health Care Establishments (HCE) are buildings that accomplishes constant revisions in their architectural designs with the aim of improving quality and safety aspects for the end users of these environments. One way to establish the necessary structural changes in healthcare environments is to understand the relationships between users and the built environment, based on the performance of their functions and environmental needs, since the participation of users in the design process can bring benefits, both for user satisfaction and for the buildings' own performance. The use of mockups of virtual simulations has presented great contributions in the analysis and evaluation of projects of health environments by enabling the immersion of the user in the process of designing the project. Thus, this article aims to verify the potential and limitations of using a physical-digital tool to assist in representation and communication in Co-Design, in addition to identifying inconsistencies in software programming and in the behavior of virtual objects. A verification test was carried out through an exploratory simulation of feasibility and usability with the use of virtual mockups, where the focus was on the analysis of the arrangement of equipment and furniture in a health environment. The physical-digital tool demonstrated in this article is still under development and needs adjustments for its final application with users in healthcare environments.Los Establecimientos de Salud (ES) son edificios que se someten a constantes revisiones en sus diseños arquitectónicos con el objetivo de mejorar aspectos de calidad y seguridad para los usuarios finales de estos entornos. Una forma de establecer los cambios estructurales necesarios en los ambientes de salud es comprender las relaciones entre los usuarios y el entorno construido, en función del desempeño de sus funciones y las necesidades ambientales, ya que la participación de los usuarios en el proceso de diseño puede traer beneficios tanto para la satisfacción del usuario y para el desempeño de los edificios. El uso de maquetas de simulaciones virtuales ha presentado grandes aportes en el análisis y evaluación de proyectos de ambientes de salud por permitir la inmersión del usuario en el proceso de concepción del proyecto. Por lo tanto, este artículo tiene como objetivo verificar el potencial y las limitaciones del uso de una herramienta físico-digital para ayudar en la representación y comunicación en Co-Design, además de identificar inconsistencias en la programación de software y en el comportamiento de los objetos virtuales. Se realizó una prueba de verificación a través de una simulación exploratoria de factibilidad y usabilidad con el uso de maquetas virtuales, donde el foco estuvo en el análisis de la disposición de equipos y mobiliario en un ambiente de salud.     Evaluating predictive pharmacogenetic signatures of adverse events in colorectal cancer patients treated with fluoropyrimidines

    The potential clinical utility of genetic markers associated with response to fluoropyrimidine treatment in colorectal cancer patients remains controversial despite extensive study. Our aim was to test the clinical validity of both novel and previously identified markers of adverse events in a broad clinical setting. We have conducted an observational pharmacogenetic study of early adverse events in a cohort study of 254 colorectal cancer patients treated with 5-fluorouracil or capecitabine. Sixteen variants of nine key folate (pharmacodynamic) and drug metabolising (pharmacokinetic) enzymes have been analysed as individual markers and/or signatures of markers. We found a significant association between TYMP S471L (rs11479) and early dose modifications and/or severe adverse events (adjusted OR = 2.02 [1.03; 4.00], p = 0.042, adjusted OR = 2.70 [1.23; 5.92], p = 0.01 respectively). There was also a significant association between these phenotypes and a signature of DPYD mutations (Adjusted OR = 3.96 [1.17; 13.33], p = 0.03, adjusted OR = 6.76 [1.99; 22.96], p = 0.002 respectively). We did not identify any significant associations between the individual candidate pharmacodynamic markers and toxicity. If a predictive test for early adverse events analysed the TYMP and DPYD variants as a signature, the sensitivity would be 45.5 %, with a positive predictive value of just 33.9 % and thus poor clinical validity. Most studies to date have been under-powered to consider multiple pharmacokinetic and pharmacodynamic variants simultaneously but this and similar individualised data sets could be pooled in meta-analyses to resolve uncertainties about the potential clinical utility of these markers

    Data on evaluation of some diagnostic parameters of all ELISA kits available in Italy for the serodiagnosis of AIE are presented and discussed. Ten laboratories were involved using a panel of 30 sera ran with 4 commercial kits and 2 in-house kits. Kits were also evaluated using a panel of sera from vaccinated animals at different days post vaccination (p.v.). All sera were also tested in agar gel immunodiffusion (AGID). The parameters evaluated were: diagnostic sensitivity (DSe) and specificity (DSp), Cohen K, weighted Cohen K, coefficient of variation (CV), accordance, concordance. Analysis of these parameters indicates that all kits have a higher sensitivity than AGID, even if a complete evaluation, as indicated by OIE (1) should be carried out

    Detection of Invasive Borrelia burgdorferi Strains in North-Eastern Piedmont, Italy.

    SummaryFollowing reports of human cases of Lyme borreliosis from the Ossola Valley, a mountainous area of Piemonte, north‐western Italy, the abundance and altitudinal distribution of ticks, and infection of these vectors with Borrelia burgdorferi sensu lato were evaluated. A total of 1662 host‐seeking Ixodes ricinus were collected by dragging from April to September 2011 at locations between 400 and 1450 m above sea level. Additional 104 I. ricinus were collected from 35 hunted wild animals (4 chamois, 8 roe deer, 23 red deer). Tick density, expressed as the number of ticks per 100 m2, resulted highly variable among different areas, ranging from 0 to 105 larvae and from 0 to 22 nymphs. A sample of 352 ticks (327 from dragging and 25 from wild animals) was screened by a PCR assay targeting a fragment of the 16S rRNA gene of B. burgdorferi s.l. Positive samples were confirmed with a PCR assay specific for the 5S‐23S rRNA intergenic spacer region and sequenced. Four genospecies were found: B. afzelii (prevalence 4.0%), B. lusitaniae (4.0%), B. garinii (1.5%) and B. valaisiana (0.3%). Phylogenetic analysis based on the ospC gene showed that most of the Borrelia strains from pathogenic genospecies had the potential for human infection and for invasion of secondary body sites
