    Actividad antimicrobiana de germicidas halogenados frente a aislamientos hospitalarios

    Antiseptics and disinfectants are extensively used in hospitals and health care settings for a variety of topical and hard-surface applications. In particular, they are essential part of infection control practices and in the prevention of nosocomial infections. Despite this, few is known about the mode of action of these biocides with respect to antibiotics. In general, the antimicrobial activity can be influenced by many factors such as formulation effects, presence of organic matter, synergy, temperature, dilution and test method. The widespread use of antiseptics and disinfectant products has prompted some speculation on the development of microbial resistance, in particular cross-resistance to antibiotics. The aim of this study was to evaluate microbiological resistance to halogenated compounds by studying the behaviour of the grampositive and gramnegative clinical isolates against halogenated biocides usually applied, with and without organic substance and applying distilled water, potable water and water of 300 ppm hardness as dilution means. The results indicate that the hospital microorganisms show a higher resistance to the biocides than the strain Staphylococcus aureus ATCC 6538, although the effective concentration in clean conditions was lesser than the recommended ones, for all the dilution means. In presence of organic matter the antimicrobial activity was reduced in accordance with the bactericidal concentration of each microorganisrn, due to the oxidant action of these disinfectantsLos antisépticos y desinfectantes se emplean en hospitales para una gran variedad de aplicaciones, tanto tópicas como sobre superficies. Estos compuestos son esenciales en el control y la prevención de las infecciones nosocomiales. A pesar de esto, se conoce bastante menos acerca del modo de acción de estos biocidas que de los antibióticos habitualmente empleados en terapéutica. La actividad antimicrobiana puede ser influenciada por muchos factores tales como la formulación, la presencia de materia orgánica, efectos de sinergia, temperatura, dilución e incluso del método de ensayo. El uso tan difundido de productos desinfectantes y antisépticos ha llevado a algunas especulaciones sobre el desarrollo de resistencia microbiana, y particularmente, resistencia cruzada con antibióticos. Con el propósito de estudiar la resistencia microbiana a germicidas halogenados de uso corriente en ,centros asistenciales, se evaluó el comportamiento de aislamientos hospitalarios tanto gram negativos como gram positivos frente a soluciones de hipoclorito de sodio, iodopovidona y tintura de iodo en presencia y ausencia de sustancias interferentes tales como materia orgánica y cationes. Los multados obtenidos indican que los microorganismos hospitalarios presentan mayor resistencia a los bioddas analizados con respecto al microorganismo de ref- erencia Staphylococcus aureus ATCC 6538. Sin embargo, en condiciones limpias este grupe de biocidas posee una alta eficacia, aún muy por debajo de las concentraciones de uso recomendadas, para todos los medios de dilución estudiados. Con la presencia de materia orgánica la actividad germicida disminuyó en todos los casos en relación directamente proporcional a la concentración, debido a la naturaleza predominantemente oxidante de estos compuestos

    Diagnostic yield of sentinel lymph node biopsy in oral squamous cell carcinoma T1/T2-N0: systematic review and meta-analysis

    The objective of this study was to conduct a systematic review and meta-analysis on the efficacy of sentinel lymph node biopsy (SLNB) in T1/T2-N0 oral squamous cell carcinoma (OSCC). A systematic review of the literature on SLNB until March 2019 was conducted. The review was organized according to the PRISMA protocol, considering the following PICO (population, intervention, comparison, outcome) question: What is the sensitivity of sentinel lymph node biopsy in OSCC? ‘P’ was patients with head and neck squamous cell carcinoma T1/2-N0; ‘I’ was SLNB; ‘C’ was neck treated with elective neck dissection and haematoxylin–eosin histopathology; ‘O’ was sensitivity and specificity. A meta-analysis and meta-regression were performed on the selected studies. The sensitivity of SLNB was up to 88% (95% confidence interval (CI) 72–96%) and specificity was up to 99% (95% CI 96–100%). The area under the summary receiver operating characteristic curve was 0.99 (95% CI 0.98–1.00). In the four studies where immunohistochemistry was performed, both the sensitivity and specificity were higher than in the studies without immunohistochemistry: 93% (95% CI 88–97%) and 98% (95% CI 96–100%), respectively. In conclusion, SLNB is an effective technique for treating patients with some types of stage T1/2-N0 OSCC.     Myxomatosis and rabbit Haemorrhagic disease: A 30-year study of the occurrence on commercial farms in Spain

    In this retrospective study, we describe the relative occurrence of clinical myxomatosis, and rabbit haemorrhagic disease (RHD), on 1714 commercial farms visited in Spain, between 1988 and 2018. We determined the annual prevalence based on 817 visits to 394 farms affected by myxomatosis. Myxomatosis was more prevalent from August to March, being lowest in June (3%) and highest in September (8.9%). With regard to RHD, we assessed 253 visits to 156 affected farms. We analyzed mean annual and monthly incidence. Two important RHD epidemics occurred; the first in 1988-1989 due to RHDV GI.1 (also known as RHDV), and the second from 2011 to 2013 due to RHDV GI.2 (RHDV2 or RHDVb). These epidemics occurred at times when effective vaccination had not been carried out. Relative monthly incidence in 2011-2018 was higher from April to August (p < 0.001). The results we obtained from 1404 necropsies on 102 farms did not clearly relate serosanguinous nasal discharge in rabbits with disease caused by GI.2 infection. We also assessed vaccination schedules used on 200 doe farms visited from the end of 2014 to 2018; 95.5% vaccinated against myxomatosis and 97.5% against RHD. Both diseases remain prevalent; however, effective vaccination has produced a steady decline in myxomatosis and RHDV GI.1 and GI.2 on-farm detection. The maintenance of high hygienic standards will be needed to continue and improve this control.     Expressed sequence tags (ESTs) from immune tissues of turbot (Scophthalmus maximus) challenged with pathogens

    Background: The turbot (Scophthalmus maximus; Scophthalmidae; Pleuronectiformes) is a flatfish species of great relevance for marine aquaculture in Europe. In contrast to other cultured flatfish, very few genomic resources are available in this species. Aeromonas salmonicida and Philasterides dicentrarchi are two pathogens that affect turbot culture causing serious economic losses to the turbot industry. Little is known about the molecular mechanisms for disease resistance and hostpathogen interactions in this species. In this work, thousands of ESTs for functional genomic studies and potential markers linked to ESTs for mapping (microsatellites and single nucleotide polymorphisms (SNPs)) are provided. This information enabled us to obtain a preliminary view of regulated genes in response to these pathogens and it constitutes the basis for subsequent and more accurate microarray analysis. Results: A total of 12584 cDNAs partially sequenced from three different cDNA libraries of turbot (Scophthalmus maximus) infected with Aeromonas salmonicida, Philasterides dicentrarchi and from healthy fish were analyzed. Three immune-relevant tissues (liver, spleen and head kidney) were sampled at several time points in the infection process for library construction. The sequences were processed into 9256 high-quality sequences, which constituted the source for the turbot EST database. Clustering and assembly of these sequences, revealed 3482 different putative transcripts, 1073 contigs and 2409 singletons. BLAST searches with public databases detected significant similarity (e-value ≤ 1e-5) in 1766 (50.7%) sequences and 816 of them (23.4%) could be functionally annotated. Two hundred three of these genes (24.9%), encoding for defence/immunerelated proteins, were mostly identified for the first time in turbot. Some ESTs showed significant differences in the number of transcripts when comparing the three libraries, suggesting regulation in response to these pathogens. A total of 191 microsatellites, with 104 having sufficient flanking sequences for primer design, and 1158 putative SNPs were identified from these EST resources in turbot. Conclusion: A collection of 9256 high-quality ESTs was generated representing 3482 unique turbot sequences. A large proportion of defence/immune-related genes were identified, many of them regulated in response to specific pathogens. Putative microsatellites and SNPs were identified.     AIP56: A Novel Bacterial Apoptogenic Toxin

    Photobacterium damselae subsp. piscicida (Phdp) is a Gram-negative pathogen agent of an important fish septicemia. The key virulence factor of Phdp is the plasmid-encoded exotoxin AIP56, which is secreted by exponentially growing pathogenic strains. AIP56 has 520 amino acids including an N-terminal cleavable signal peptide of 23 amino acid residues, two cysteine residues and a zinc-binding region signature HEXXH that is typical of most zinc metallopeptidases. AIP56 induces in vitro and in vivo selective apoptosis of fish macrophages and neutrophils through a caspase-3 dependent mechanism that also involves caspase-8 and -9. In vivo, the AIP56-induced phagocyte apoptosis progresses to secondary necrosis with release of cytotoxic phagocyte molecules including neutrophil elastase. Fish injected with recombinant AIP56 die with a pathology similar to that seen in the natural infection

    Th17-Related Genes and Celiac Disease Susceptibility

    Th17 cells are known to be involved in several autoimmune or inflammatory diseases. In celiac disease (CD), recent studies suggest an implication of those cells in disease pathogenesis. We aimed at studying the role of genes relevant for the Th17 immune response in CD susceptibility. A total of 101 single nucleotide polymorphisms (SNPs), mainly selected to cover most of the variability present in 16 Th17-related genes (IL23R, RORC, IL6R, IL17A, IL17F, CCR6, IL6, JAK2, TNFSF15, IL23A, IL22, STAT3, TBX21, SOCS3, IL12RB1 and IL17RA), were genotyped in 735 CD patients and 549 ethnically matched healthy controls. Case-control comparisons for each SNP and for the haplotypes resulting from the SNPs studied in each gene were performed using chi-square tests. Gene-gene interactions were also evaluated following different methodological approaches. No significant results emerged after performing the appropriate statistical corrections. Our results seem to discard a relevant role of Th17 cells on CD risk

    Hypericum perforatum treatment: effect on behaviour and neurogenesis in a chronic stress model in mice

    <p>Abstract</p> <p>Background</p> <p>Extracts of <it>Hypericum perforatum </it>(St. John's wort) have been traditionally recommended for a wide range of medical conditions, in particular mild-to-moderate depression. The present study was designed to investigate the effect of Hypericum perforatum treatment in a mouse model of anxiety/depressive-like behavior, induced by chronic corticosterone administration.</p> <p>Methods</p> <p>CD1 mice were submitted to 7 weeks corticosterone administration and then behavioral tests as Open Field (OF), Novelty-Suppressed Feeding (NSF), Forced Swim Test (FST) were performed. Cell proliferation in hippocampal dentate gyrus (DG) was investigated by both 5-bromo-2'-deoxyuridine (BrdU) and doublecortin (DCX) immunohistochemistry techniques and stereological procedure was used to quantify labeled cells. Golgi-impregnation method was used to evaluate changes in dendritic spines in DG. Hypericum perforatum (30 mg/Kg) has been administered for 3 weeks and then neural development in the adult hippocampus and behavioral changes have been examined.</p> <p>Results</p> <p>The anxiety/depressive-like state due to chronic corticosterone treatment was reversed by exogenous administration of Hypericum perforatum; the proliferation of progenitor cells in mice hippocampus was significantly reduced under chronic corticosterone treatment, whereas a long term treatment with Hypericum perforatum prevented the corticosterone-induced decrease in hippocampal cell proliferation. Corticosterone-treated mice exhibited a reduced spine density that was ameliorated by Hypericum perforatum administration.</p> <p>Conclusion</p> <p>These results provide evidence of morphological adaptations occurring in mature hippocampal neurons that might underlie resilient responses to chronic stress and contribute to the therapeutic effects of chronic Hypericum perforatum treatment.</p

    While making very substantial changes to the population's working conditions, government strategies to foster economic development in South Korea have historically attempted to keep worker involvement, in terms of influence on the process, to a bare minimum. Applying the Gramscian concept of passive revolution, this article analyses governance mechanisms and production relations over a history of authoritarianism and up to the contemporary period of democratic reform. Trasformismo, which is a strategy of limited concessions, has been provided via vocational training for workers. Despite this attempt at inclusion, it is concluded that workers have not enjoyed full participation in negotiation for their welfare at any time in Korean history
