    Assessment in a globalized economy: A task-based approach to assess the proficiency of Dutch in specific occupational domains

    For language tests to be in tune with the target context, a constant attention to the shifting characteristics of real-world language use is required. This holds particularly true for language tests in the occupational domain since domestic labor market demands are continuously changing and increasingly met through the recruitment of foreign workers. This paper describes how subject experts were involved in the cyclical validation process of a test of Dutch for the professional domain. A survey of recruitment agents, employers, policy makers, language instructors, examiners, and former test takers indicated that a task-based test targeting the language skills involved in service-oriented work settings such as administration and health care at level B2 of the CEFR was favored. The involvement of subject specialists recruited among the test’s various stakeholders proved to be of vital importance throughout the development and validation process to ensure content validity and avoid biases

    Extended twin study of alcohol use in Virginia and Australia

    Drinking alcohol is a normal behavior in many societies, and prior studies have demonstrated it has both genetic and environmental sources of variation. Using two very large samples of twins and their first-degree relatives (Australia ≈ 20,000 individuals from 8,019 families; Virginia ≈ 23,000 from 6,042 families), we examine whether there are differences: (1) in the genetic and environmental factors that influence four interrelated drinking behaviors (quantity, frequency, age of initiation, and number of drinks in the last week), (2) between the twin-only design and the extended twin design, and (3) the Australian and Virginia samples. We find that while drinking behaviors are interrelated, there are substantial differences in the genetic and environmental architectures across phenotypes. Specifically, drinking quantity, frequency, and number of drinks in the past week have large broad genetic variance components, and smaller but significant environmental variance components, while age of onset is driven exclusively by environmental factors. Further, the twin-only design and the extended twin design come to similar conclusions regarding broad-sense heritability and environmental transmission, but the extended twin models provide a more nuanced perspective. Finally, we find a high level of similarity between the Australian and Virginian samples, especially for the genetic factors. The observed differences, when present, tend to be at the environmental level. Implications for the extended twin model and future directions are discussed

    The Arctic marine ecosystem is changing fast due to climate change, emphasizing the need for solid ecological baselines and monitoring. The polar cod Boreogadus saida functions as key species in the Arctic marine food web. We investigated the stomach content of polar cod from the northern Barents Sea using DNA metabarcoding with the mitochondrial cytochrome c oxidase I (COI) gene in parallel with classical visual analysis. Arctic amphipods and krill dominated the diet in both methods. Yet, metabarcoding allowed for the identification of digested and unidentifiable prey and provided higher taxonomic resolution, revealing new and undiscovered prey items of polar cod in the area. Furthermore, molecular results suggest a higher importance of barnacles and fish (supposedly eggs and pelagic larvae) in the diet than previously recorded. Parasites and, in 6 cases other prey items, were only visually identified, demonstrating the complementary nature of both approaches. The presence of temperate and boreal prey species such as Northern krill and (early life stages of) European flounder and European plaice illustrate the advection of boreal taxa into the polar region or may be indicative of ongoing borealization in the Barents Sea. We show that a combination of visual analysis and metabarcoding provides complementary and semi-quantitative dietary information, and integrative insights to monitor changing marine food webs

    Equine epidermis : a source of epithelial-like stem/progenitor cells with in vitro and in vivo regenerative capacities

    Besides the presence of somatic stem cells in hair follicles and dermis, the epidermis also contains a subpopulation of stem cells, reflecting its high regenerative capacity. However, only limited information concerning epidermis-derived epithelial-like stem/progenitor cells (EpSCs) is available to date. Nonetheless, this stem cell type could prove itself useful in skin reconstitution after injury. After harvesting from equine epidermis, the purified cells were characterized as EpSCs by means of positive expression for CD29, CD44, CD49f, CD90, Casein Kinase 2, p63, and Ki67, low expression for cytokeratin (CK)14 and negative expression for CD105, CK18, Wide CK, and Pan CK. Furthermore, their self-renewal capacity was assessed in adhesion as well as in suspension. Moreover, the isolated cells were differentiated toward keratinocytes and adipocytes. To assess the regenerative capacities of EpSCs, six full-thickness skin wounds were made: three were treated with EpSCs and platelet-rich-plasma (EpSC/PRP-treated), while the remaining three were administered carrier fluid alone (PRP-treated). The dermis of EpSC/PRP-treated wounds was significantly thinner and exhibited more restricted granulation tissue than did the PRP-treated wounds. The EpSC/PRP-treated wounds also exhibited increases in EpSCs, vascularization, elastin content, and follicle-like structures. In addition, combining EpSCs with a PRP treatment enhanced tissue repair after clinical application

    Evaluating genetic traceability methods for captive bred marine fish and their applications in fisheries management and wildlife forensics

    Growing demands for marine fish products is leading to increased pressure on already depleted wild populations and a rise in the aquaculture production. Consequently, more captive bred fish are released into the wild through accidental escape or deliberate restocking, stock enhancement and sea ranching programs. The increased mixing of captive bred fish with wild conspecifics may affect the ecological and/or genetic integrity of wild fish populations. From a fisheries management perspective unambiguous identification tools for captive bred fish will be highly valuable to manage risks. Additionally there is great potential to use these tools in wildlife forensics (i.e. tracing back escapees to their origin and determining mislabelling of seafood products). Using SNP data from captive bred and wild populations of Atlantic cod (Gadus morhua L.) and sole (Solea solea L.), we explored the efficiency of population and parentage assignment techniques for the identification and tracing of captive bred fish. Simulated and empirical data were used to correct for stochastic genetic effects. Overall, parentage assignment performed well when a large effective population size characterizes the broodstock and escapees originate from early generations of captive breeding. Consequently, parentage assignments are particularly useful from a fisheries management perspective to monitor the effects of deliberate releases of captive bred fish on wild populations. Population assignment proved to be more efficient after several generations of captive breeding, which makes it a useful method in forensic applications for well-established aquaculture species. We suggest the implementation of a case by case strategy when choosing the best method

    Human Lyme disease–primarily caused by the bacterium Borrelia burgdorferi sensu stricto (s.s.) in North America–is the most common vector-borne disease in the United States. Research on risk mitigation strategies during the last three decades has emphasized methods to reduce densities of the primary vector in eastern North America, the blacklegged tick (Ixodes scapularis). Controlling white-tailed deer populations has been considered a potential method for reducing tick densities, as white-tailed deer are important hosts for blacklegged tick reproduction. However, the feasibility and efficacy of white-tailed deer management to impact acarological risk of encountering infected ticks (namely, density of host-seeking infected nymphs; DIN) is unclear. We investigated the effect of white-tailed deer density and management on the density of host-seeking nymphs and B. burgdorferi s.s. infection prevalence using surveillance data from eight national parks and park regions in the eastern United States from 2014–2022. We found that deer density was significantly positively correlated with the density of nymphs (nymph density increased by 49% with a 1 standard deviation increase in deer density) but was not strongly correlated with the prevalence of B. burgdorferi s.s. infection in nymphal ticks. Further, while white-tailed deer reduction efforts were followed by a decrease in the density of I. scapularis nymphs in parks, deer removal had variable effects on B. burgdorferi s.s. infection prevalence, with some parks experiencing slight declines and others slight increases in prevalence. Our findings suggest that managing white-tailed deer densities alone may not be effective in reducing DIN in all situations but may be a useful tool when implemented in integrated management regimes

    4E Interacting Protein as a Potential Novel Drug Target for Nucleoside Analogues in Trypanosoma brucei

    Human African trypanosomiasis is a neglected parasitic disease for which the current treatment options are quite limited. Trypanosomes are not able to synthesize purines de novo and thus solely depend on purine salvage from the host environment. This characteristic makes players of the purine salvage pathway putative drug targets. The activity of known nucleoside analogues such as tubercidin and cordycepin led to the development of a series of C7-substituted nucleoside analogues. Here, we use RNA interference (RNAi) libraries to gain insight into the mode-of-action of these novel nucleoside analogues. Whole-genome RNAi screening revealed the involvement of adenosine kinase and 4E interacting protein into the mode-of-action of certain antitrypanosomal nucleoside analogues. Using RNAi lines and gene-deficient parasites, 4E interacting protein was found to be essential for parasite growth and infectivity in the vertebrate host. The essential nature of this gene product and involvement in the activity of certain nucleoside analogues indicates that it represents a potential novel drug target.</jats:p

    The objective of this preliminary study was to investigate the relationship between the housing conditions and the health and welfare of horses. A survey, based on a questionnaire containing 36 multiple choice questions about various aspects of the housing of horses was conducted. A questionnaire was sent via email to approximately 600 horse owners in Flanders. A total of 225 horse owners completed the questionnaire. The study provides a clear picture of the risk factors that horse owners in practice recognize and the link they see between housing related diseases. Although horse owners usually are sufficiently aware of these influences, they are not taken care of in practice. According to fifty percent of the respondents, the major reason is the impracticability of the advice of the veterinarian. According to the horse owners, the main risk factors affecting the health of horses are: draft, the lack of quarantine measures and the presence of (sharp) foreign objects in the stable. As a consequence, more than 50% of the respondents report nasal discharge and coughing as common problems in their horses. Sixty-seven percent of the horse owners are satisfied with the overall management of the stable. However, there is a widespread dissatisfaction with regard to quarantine measures, in case of a disease outbreak (30% of the horse owners) and in case of the introduction of new animals into a group (36%). Fifty percent of the respondents score their own stable infrastructure 8/10 or more while about one out of four is less satisfied (7/10) about the floor and the walls of their stables. The results of this study can help owners and veterinarians to identify housing factors that may increase the risk to health and welfare problems in horses. This should lead to an improved well-being of the modern, often prolonged - housed horse

    Recommendations for dealing with waste contaminated with Ebola virus: a Hazard Analysis of Critical Control Points approach

    Objective To assess, within communities experiencing Ebola virus outbreaks, the risks associated with the disposal of human waste and to generate recommendations for mitigating such risks. Methods A team with expertise in the Hazard Analysis of Critical Control Points framework identified waste products from the care of individuals with Ebola virus disease and constructed, tested and confirmed flow diagrams showing the creation of such products. After listing potential hazards associated with each step in each flow diagram, the team conducted a hazard analysis, determined critical control points and made recommendations to mitigate the transmission risks at each control point. Findings The collection, transportation, cleaning and shared use of blood-soiled fomites and the shared use of latrines contaminated with blood or bloodied faeces appeared to be associated with particularly high levels of risk of Ebola virus transmission. More moderate levels of risk were associated with the collection and transportation of material contaminated with bodily fluids other than blood, shared use of latrines soiled with such fluids, the cleaning and shared use of fomites soiled with such fluids, and the contamination of the environment during the collection and transportation of blood-contaminated waste. Conclusion The risk of the waste-related transmission of Ebola virus could be reduced by the use of full personal protective equipment, appropriate hand hygiene and an appropriate disinfectant after careful cleaning. Use of the Hazard Analysis of Critical Control Points framework could facilitate rapid responses to outbreaks of emerging infectious disease
