    A statistical shape analysis of the neurocranium and long bones

    Morphological variation of skeletal elements, and the potential use of such variation in distinguishing among demographic groups, is often investigated using traditional metric or non-metric assessments. Traditional approaches, however, often fail to sufficiently capture the "true" shape of features, thus also failing to identify potentially important feature characteristics. The development of geometric morphometrics has allowed more comprehensive and accurate three-dimensional data capture which maintains the geometric properties of an object while isolating the effect of size from the data. The aim of this study was to employ the geometric morphometric approach to a 3D digitized sample of 1132 South African individuals from the skeletal collections of the Universities of Cape Town, Stellenbosch, Witwatersrand and Pretoria. Morphological variation among demographic groups was assessed using Generalized Procrustes Analyses applied to the individual bones of the neurocranium and the long bones of the limbs. The ability to distinguish groups based on the detected variation was assessed using Discriminant Function Analysis. The results show that, when size is isolated from the data, only a few elements show sexual dimorphism, while all examined elements show high accuracy in distinguishing among ancestry groups (>74%). When variation is assessed using both parameters, classification accuracies of 70-83% are obtained. Comparison of the different elements shows that the best classification accuracies are based on the frontal bone (84% average) and the humerus (81% average). This study shows that the morphologies of the neurocranium and long bones vary among sex and ancestry groups. This allows assessment of how the combination of variable intrinsic and extrinsic influences can manifest on different parts of the skeleton. In a population as genetically and historically complex as that of South Africa, understanding of the impact of such influences may inform forensic assessments of skeletal material, which is especially valuable considering the high rate of violent crimes and increasing number of unidentified remains being discovered in the country

    Arrested on heating: controlling the motility of active droplets by temperature

    One of the challenges in tailoring the dynamics of active, self-propelling agents lies in arresting and releasing these agents at will. Here, we present an experimental system of active droplets with thermally controllable and reversible states of motion, from unsteady over meandering to persistent to arrested motion. These states depend on the P\'eclet number of the chemical reaction driving the motion, which we can tune by using a temperature sensitive mixture of surfactants as a fuel medium. We quantify the droplet dynamics by analysing flow and chemical fields for the individual states, comparing them to canonical models for autophoretic particles. In the context of these models, we are able to observe in situ the fundamental first transition between the isotropic, immotile base state and self-propelled motility

    Nonadditivity of the Adsorption Energies of Linear Acenes on Au(111): Molecular Anisotropy and Many-Body Effects

    Adsorption energies of chemisorbed molecules on inorganic solids usually scale linearly with molecular size and are well described by additive scaling laws. However, much less is known about scaling laws for physisorbed molecules. Our temperature-programmed desorption experiments demonstrate that the adsorption energy of acenes (benzene to pentacene) on the Au(111) surface in the limit of low coverage is highly nonadditive with respect to the molecular size. For pentacene, the deviation from an additive scaling of the adsorption energy amounts to as much as 0.7 eV. Our first-principles calculations explain the observed nonadditive behavior in terms of anisotropy of molecular polarization stemming from many-body electronic correlations. The observed nonadditivity of the adsorption energy has implications for surface-mediated intermolecular interactions and the ensuing on-surface self-assembly. Thus, future coverage-dependent studies should aim to gain insights into the impact of these complex interactions on the self-assembly of π-conjugated organic molecules on metal surfaces

    Identification of genetic variants associated with Huntington's disease progression: a genome-wide association study

    Background Huntington's disease is caused by a CAG repeat expansion in the huntingtin gene, HTT. Age at onset has been used as a quantitative phenotype in genetic analysis looking for Huntington's disease modifiers, but is hard to define and not always available. Therefore, we aimed to generate a novel measure of disease progression and to identify genetic markers associated with this progression measure. Methods We generated a progression score on the basis of principal component analysis of prospectively acquired longitudinal changes in motor, cognitive, and imaging measures in the 218 indivduals in the TRACK-HD cohort of Huntington's disease gene mutation carriers (data collected 2008–11). We generated a parallel progression score using data from 1773 previously genotyped participants from the European Huntington's Disease Network REGISTRY study of Huntington's disease mutation carriers (data collected 2003–13). We did a genome-wide association analyses in terms of progression for 216 TRACK-HD participants and 1773 REGISTRY participants, then a meta-analysis of these results was undertaken. Findings Longitudinal motor, cognitive, and imaging scores were correlated with each other in TRACK-HD participants, justifying use of a single, cross-domain measure of disease progression in both studies. The TRACK-HD and REGISTRY progression measures were correlated with each other (r=0·674), and with age at onset (TRACK-HD, r=0·315; REGISTRY, r=0·234). The meta-analysis of progression in TRACK-HD and REGISTRY gave a genome-wide significant signal (p=1·12 × 10−10) on chromosome 5 spanning three genes: MSH3, DHFR, and MTRNR2L2. The genes in this locus were associated with progression in TRACK-HD (MSH3 p=2·94 × 10−8 DHFR p=8·37 × 10−7 MTRNR2L2 p=2·15 × 10−9) and to a lesser extent in REGISTRY (MSH3 p=9·36 × 10−4 DHFR p=8·45 × 10−4 MTRNR2L2 p=1·20 × 10−3). The lead single nucleotide polymorphism (SNP) in TRACK-HD (rs557874766) was genome-wide significant in the meta-analysis (p=1·58 × 10−8), and encodes an aminoacid change (Pro67Ala) in MSH3. In TRACK-HD, each copy of the minor allele at this SNP was associated with a 0·4 units per year (95% CI 0·16–0·66) reduction in the rate of change of the Unified Huntington's Disease Rating Scale (UHDRS) Total Motor Score, and a reduction of 0·12 units per year (95% CI 0·06–0·18) in the rate of change of UHDRS Total Functional Capacity score. These associations remained significant after adjusting for age of onset. Interpretation The multidomain progression measure in TRACK-HD was associated with a functional variant that was genome-wide significant in our meta-analysis. The association in only 216 participants implies that the progression measure is a sensitive reflection of disease burden, that the effect size at this locus is large, or both. Knockout of Msh3 reduces somatic expansion in Huntington's disease mouse models, suggesting this mechanism as an area for future therapeutic investigation

    Entsprechend der Erkenntnis, dass weltweit Regionen von hoher SchutzprioritĂ€t zumeist auch soziale LebensrĂ€ume indigener und lokaler Gemeinschaften sind, wird im internationalen Übereinkommen ĂŒber die Biologische Vielfalt (CBD) explizit gefordert, deren Wissen, das fĂŒr den Schutz und die nachhaltige Nutzung der BiodiversitĂ€t von Bedeutung ist, anzuerkennen, zu bewahren und seine breitere Anwendung zu fördern. Ausgehend von dieser Inwertsetzung fokussiert die umweltethnologische Arbeit die lokale Umsetzung der Konvention durch die Ausweisung von Schutzgebieten und untersucht Möglichkeiten adaptiver Konzepte, die kulturspezifische Wissensformen als Bestandteil indigenen Ressourcenmanagements mit globalen Schutzinteressen zu integrieren versuchen. Basierend auf der Annahme, dass geschĂŒtzte Landschaften auch immer kulturelle Landschaften sind, in die Spuren menschlichen Handelns und Denkens eingeschrieben sind, untersucht die Studie exemplarisch indigenes Wissen von Maya-Q'eqchi' Gemeinschaften im Umfeld von Schutzgebieten in Alta Verapaz, Guatemala. Im Mittelpunkt stehen verschiedene Dimensionen indigenen Wissens, die fĂŒr den Schutz und die Bewahrung natĂŒrlicher Ressourcen von Relevanz sind. Über die materielle Ebene von sichtbaren Wissensinhalten hinaus geht es um symbolische Bedeutungsebenen von Mensch-Umwelt-Beziehungen, die - weil zumeist unsichtbar - im akademischen und umweltpolitischen Diskurs weitgehend unberĂŒcksichtigt bleiben. Beide Wissensdimensionen in einer Analyse zu integrieren und sie in einen Kontext historischer, sozio-ökonomischer und machtpolitischer Strukturen auf lokaler, nationaler und globaler Ebene zu stellen, ist das zentrale Anliegen der Arbeit. Sie zeigt, dass Umweltkenntnisse an spezifische Naturkonzeptionen und Interpretationen von Raum und Zeit, von IdentitĂ€t und KausalitĂ€t gebunden sind, die Menschen Orientierung in ihren wesentlichen Lebensbeziehungen geben. Wie Menschen Natur wahrnehmen und was Natur fĂŒr sie bedeutet, ist an diese kulturspezifischen Wertvorstellungen gebunden. Diese sind eingewoben in ein Weltbild, verstanden als ein meist nicht bewusst reflektiertes philosophisches Konzept mit Deutungsmustern, das nicht nur welterschließend, sondern auch handlungsorientierend ist und als Sinn gebende Instanz das Mensch-Natur-VerhĂ€ltnis definiert. Schutzgebiete sind nicht nur SchauplĂ€tze divergierender Nutzungsinteressen, sondern auch Orte, wo verschiedene Naturkonzeptionen und Weltbilder aufeinander treffen. Davon muss man ausgehen, wenn indigenes Wissen im Sinne der BiodiversitĂ€tskonvention bewahrt werden soll. Die interkulturelle VerstĂ€ndigung ĂŒber verschiedene Naturkonzeptionen ist eine Voraussetzung, globale Schutzinteressen mit NutzungsbedĂŒrfnissen der Bevölkerung auf lokaler Ebene in Einklang zu bringen.
